1.Establishment of a two-dimensional electrophoresis technology on brain tissue protein of rat

Objective:To establish a two-dimensional electrophoresis technology on brain tissue protein of rat.Methods:Lysis buffer of different volume was taken to extract brain tissue proteins of rat,and different protein quantities(1mg,2mg,3mg) were taken to establish a two-dimensional electrophoresis.Coomassie brilliant blue was applied to stain protein,and patterns were analyzed.Results:With a molecular mass between 6.5~200ku and isoelectric points(pI) from 3~10,1mg proteins obtained 546 protein spots,2mg 780,and 3mg 805.Pattern of 2mg protein was the best.Conclusion:A two-dimensional electrophoresis technology on brain tissue protein of rat has been established successfully.
2.Emotional Benefits and Its Cardiopulmonary Characteristics during a Single Bout of Exercise

Objective To explore the emotional and cardiorespiratory changes at different stages during a single bout of exercise.Methods Sixty participants were recruited to complete 30 minutes' moderate exercise at a fixed running speed (the speeds were set based on the former self-controled speed test).Self-rated emotional experience was recorded 5 minutes before the exercise intervention,30 minutes after the intervention onset and 10 minutes after the intervention.Meanwhile,participants' physiological indices were monitored throughout the whole process.The data were grouped every five minutes and the data of six groups (t1,t2,t3,t4,t5 and t6)during exercise intervention were analyzed.The reliability of the results was subsequently checked via interview.Results The results showed that during t1 participants' happiness and tidal volume (TV)reached their maximum,but the respiratory frequency (RF) and respiratory quotient (RQ)reached their minimum.During t2 to t5,participants felt the most positive and RF was cooperating with TV appropriately.During t6,the feeling of fatigue hit the highest level;RF and Heart Rate (RT)reached its maximum,but the ratio of VO2/HR reached its minimum.Conclusion During a single bout of exercise,people feel the happiest in the first five minutes with mainly deep breathing.In the subsequent twenty minutes,exercisers experience positive emotion with highest breath efficiency.During the last five minutes,fatigue,as well as low efficient pulmonary respiration and pump function appeared.
3.Preparation of an acellular porcine corneal stroma scaffold and its biocompatibility

BACKGROUND:Studies have found that a variety of biological materials can be used for preparing corneal stroma scaffolds that have good biocompatibility, but research on preparation and biocompatibility of the acel ular porcine corneal stroma scaffold is little. OBJECTIVE:To explore the preparation and biocompatibility of the acel ular porcine corneal stroma scaffold. METHODS:Acel ular porcine corneal stroma scaffold and its extract were prepared. Wel-grown human corneal stromal cel s were selected and cultured in the extract of acel ular porcine corneal stroma scaffold (experimental group) or in the complete medium (control group), respectively. After 1, 2 and 3 days of culture, the proliferation ability of human corneal stromal cel s was detected by MTT assay. In the meanwhile, human corneal cel s were directly seeded onto the acel ular porcine corneal stroma scaffold, and then the cel growth on the scaffold was detected using immunochemical method. RESULTS AND CONCLUSION:The number of human corneal stromal cel s was in a rise with time in the two groups, and absorbance values had no significant difference between two groups at different time points of culture. Human corneal stromal cel s grew wel on the scaffold, and were positive for cel integrinβ1, vimentin, aldehyde dehydrogenase 3A1, as wel as CD34, CDK2 and K-Ras. These results show that the acel ular porcine corneal stroma scaffold has no cytotoxicity, and has good biocompatibility.
4.Selective Enhancement of Open-skill Exercise on Attention Network of College Students

Objectives To investigate the relationship between different exercise types and the attention network.Methods A total of 75 university students were enrolled and randomly divided into an open-skill exercise group,a closed-skill exercise group and an irregular exercise control group,each of 25.They underwent the exercise as their group names implied.Then the attention network test was performed to evaluate their functions of the alerting network,orienting network and executive control network.Results Significant differences were found in the function of the executive control network among the 3 groups,while no significant difference was found among the 3 groups in the functions of the alerting network and orienting network.Conclusions Both open-and closed-skill exercise are associated with better function of executive control network.Furthermore,open-skill exercise showes more benefits than closed-skill exercise in promoting the function of the executive control network.Neither opennor closed-skill exercise bring benefits to the function of the alerting or orienting network.
5.Effect of the total saponins of Rubus parvifolius on the intracellular free calcium in ischemic hippocampal neurons

Aim To study the effect of the total saponins of Rubus parvifolius(TSRP)on the intracellular free calcium levels in ischemic hippocampus neurons.Methods The cytosolic free calcium concentration in ischemic hippocampal neurons was measured by using Ca2+-sensitive fluorescent probe fluo-2/AM with a laser scanning confocal microscope.Results Application of TSRP inhibited free intracellular calcium in ischemic hippocampus neurons in a concentration-dependent manner.Conclusions These results suggest that TSRP might protect hippocampus neurons via attenuating calcium overload induced by ischemia.
6.Study on compared proteomics of focal cerebral ischemia in rats

Aim To establish a model of focal cerebral ischemic tissue of rat brain and explore the injury mechanism of focal cerebral ischemia reperfusion in whole level of proteins.Method The model was established with suture method by reperfusion 24 h after ischemic 2 h according to Koizumi′s method,total brain tissue proteins were extracted with Lysis buffer,proteins were separated by two-dimensional electrophoresis(2-DE),stained by Coomassie brilliant blue,the patterns were gotten,differential proteins were found out,PMS was obtained by MALDI-TOF-MS,and related information of proteins was gained by MS-Fit database.Results A comparative proteomic study of model and normal group was performed.Compared with model group,the normal group gained 23 differential protein spots,13 spots expressed lowly,and 10 spots high,6 protein spots were identified,the relative cerebral ischemic proteins such as Leukotriene A-4 hydrolase,Thyrotropin-releasing hormone-degrading ectoenzyme etc were found out.Conclusions Establishing a 2-DE technology is applied to protein analysis of brain tissue,and the relative proteins of cerebral ischemia are found from proteome aspect.This will contribute to the research on the injury mechanism of cerebral ischemic tissues.
7.Diagnosis of primary neurogenic tumor on chest wall using CT imaging

Objective To discuss manifestations of neurogenic tumors on chest wall on CT imaging and pathology.Methods 7 patients with neurogenic tumors on chest wall confirmed surgically and pathologically were reviewed.To facilitate differential diagnosis,3 cases of malignant sarcoma were reviewed for their features on CT.Results 4 out of 5 cases of neurinoma were benign,with one case showing even density on plain CT scan,3 were poorly even,one case showed obvious evenness after enhancement,and 1 case was poorly evenly enhanced,2 benign cases had compression absorption of surrounding bone of scapula or ribs.One case of moderate malignancy was uneven in plain scan and slightly enhanced after enhancement procedure,and neighboring bone of scapula and rib were compressed and destroyed and absorbed,and neighboring muscular interspace and upper mediastinum were involved.One case showed single lesion of neurofibroma,displaying relatively even density on plain scan and moderate unevenness after enhancement procedure,as well as compression and absorption of neighboring ribs.One case of neurofibroma had multiple lesions,showing uneven density on plain scan.This case did not undergo enhanced scan.The remaining 3 cases were all sarcoma including 2 cases of fibrosarcoma and one case of synovial sarcoma.Conclusion Mass of soft tissues that are situated on chest wall or paravertebral area and with smooth edge are suggestive for benign neurogenic tumor.On the contrary,rough edge and coarse shape and infiltration into neighboring tissues are characteristic for malignant ones.
8.How massage inhibits scar tissue formation in skeletal muscles after blunt injury

Objective To investigate the effect of massage on quadriceps femoris repair after injury by external force and the expression of transforming growth factor β1 (TGF-β1) and collagen-Ⅰ (COL-Ⅰ) mRNA.To explore the molecular mechanisms inhibiting scar tissue formation and promoting muscle repair.Methods Forty New Zealand white rabbits weighing (2.0 ±0.5) kg were randomly divided into a normal control group (A) (n =4),a selfrepair group (B) (n =20,further divided into the 3rd,7th,11th,15th and 19th day time points),and a massage group (C) (n =16,further divided as in group B).In group A the rabbits were not treated,as normal controls.In groups B and C rabbit models of quadriceps femoris injury were prepared using a self-made beater.In group B no massage therapy was given as a natural recovery control; in group C,massage therapy was given after 5 days.Realtime quantitative PCRs were used to detect TGF-β1 and COL-Ⅰ mRNA expression.Resnlts There was no significant difference between groups B and C in the expression of TGF-β1 or COL-Ⅰ mRNA on day 7.At the later time points,expression of both mRNAs in group C was significantly less than in group B.Conclusion Massage can effectively reduce the expression of TGF-β1 and COL-Ⅰ mRNA,inhibit excessive scar formation and promote repair of injured tissue.
9.MRI signs of anterior cruciate ligament tears

Objective To investigate the direct and indirect MRI manifestations of anterior cruciate ligament(ACL)tears and discuss its mechanism and diagnostic value.Methods The study involved 40 patients with ACL tears including 37 males and three females at age range of 16-49 years(mean age of 33 years).Arthroscopy and operation confirmed 28 patients at acute phase of ACL tears,12 at chronic phase of ACL tears,35 with complete ACL tears and five with partial ACL tears.All patients were examined on 1.5T Siemens MRI scanner,with SE,TSE or Medic sequence in multiple directions.The direct and indirect MRI manifestations of ACL tears were retrospectively and statistically analyzed by employing three experienced doctors.Results Among 28 patients with acute tears,the direct MRI signs included signal interruption or non-consecutive(86%),uneven signal(64%)and swelling and thickening ligament(36%).While in 12 patients with chronic teats,the MRI signs manifested swelling ligament(92%),signal interruption or non-consecutive(75%)and uneven signal(58%).The sign of thickening ligament was mainly seen at chronic phase of ACL tears(P <0.01).Of 35 patients with complete ACL tears,80% occurred in the middle part of ACL,with low incidence of upper and lower parts.All five patients with partial ACL tears occurred in the anteriomedialis bundle.The indirect MRI signs of ACL tears included 7-shaped deformity of posterior cruciate ligament(PCL)(34 patients,85%),meniscus exposure(26 patients)including 16 with lateral meniscus exposure(62%),bone injury(15 patients)including segond fracture(8 patients,53%),widened joint space(9 patients)including 78% at chronic phase,and tibia antelocation(23 patients,57%).Among 40 patients,37 patients were diagnosed correctly preoperatively,with accuracy rate of 92%.While three patients with partial ACL tears were missed diagnosis preoperatively,with inaccuracy rate of 8%.Conclusion ACL tear is relatively easy to identify preoperatively according to direct and indirect MRI signs in combination with trauma history.
10.Effects of weightlessness on the apoptosis of hepatocytes in rats

Objective To investigate the effects of weightlessness on the apoptosis of hepatocytes in rats.Methods Eighty-four adult male Wistar rats were randomly divided into weightlessness group and control group according to the random number table.Tail-suspension was used to simulate the weightlessness model.Six rats were sacrificed in each group at the end of day 1-7.Hepatocyte apoptosis and p53 expression were detected by TUNEL assay and immunohistochemistry PV-6001,respectively.All data were analyzed by variance analysis.Results An increase of apoptosis index and up-regulated expression of p53 were detected in weightlessness group.The level of apoptotic index Was significantly higher in weightlessness group than in control group from day 1 to day 5(F=77.608,P<0.05),and the apoptotic indexes of the 2 groups were close at day 6 and day 7.The p53 proteins with positive expression were mainly detected in the nucleus.The level of positive expression of p53 in liver tissue in weightlessness group was higher than that in control group at the early stage(F=113.063,P<0.05).Few hepatocytes with positive expression were detected in control group and weightlessness group at a later stage.Conclusion There is a close relationship between hepatocyte apoptosis,changes of P53 expression and stress response and tolerance to weightlessness.
