AIM: To study the effect of crocetin on the formation of advanced glycation end products(AGEs) and receptor for AGEs(RAGE) protein expression in diabetic rats induced by streptozotocin(STZ).METHODS: Rats were injected STZ in tail vessel with a dose of 45(mg?kg~(-1)).3 d later,the rats whose blood glucose were over(11.1)(mmol?L~(-1)) were regarded as diabetic rats,and were divided randomly into two groups: diabetic mellitus(DM) group,crocetin(50(mg?kg~(-1)),po) group.At the same time,8 normal rats were regarded as control group.After 21 d treatment,The levels of fasting blood glucose(FBG),serum fructosamine(FMN) and glycosylated hemoglobin(GHb) were measured.The contents of AGEs in aorta and mesenteric vessel were detected by fluorospectrophotometry.HE staining for mesenteric aorta was performed,and RAGE protein expression was studies with immunohistochemical method.RESULTS: Compared with DM group,crocetin did not decrease the level of FBG.However,it could decrease the levels of FMN and GHb in blood and AGEs in aorta and mesenteric vessel(P
2.Inhibiting effect of montelukast on eosinophilic airway inflammation of asthmatic guinea pigs

Aim To study the effect of montelukast on eosinophilic airway inflammation of asthmatic guinea pigs,and explore possible mechanism of montelukast. Methods Experimental asthma model of guinea pigs was induced by ovalbumin in vivo. The eosinophils in BALF were separated by density gradient centrifugation. The apoptosis of eosinophils was labeled by TdT-mediated dUTP nick end labeling (TUNEL)technique. The content of IL-5 in BALF was detected by enzyme-linked immunosorbent assay (ELISA). The level of ECP in BALF was detected through fluorescence enzyme-labeled assay. Results In BALF of asthmatic guinea pigs model, the number of eosinophils and content of IL-5 and ECP increased obviously, while the apoptosis index of eosinophils was lowered. After treatment with montelukast, the number of eosinophils and content of IL-5 and ECP in BALF of asthmatic guinea pigs decreased significantly, while the apoptosis index of eosinophils was elevated significantly. Conclusion Montelukast can inhibit the eosinophilic airway inflammation of asthmatic guinea pigs. Lowering the levels of IL-5 and ECP in BALF, promoting apoptosis of eosinophils and relieving airway infiltration of eosinophils may be important mechanisms for montelukast to antagonize airway inflammation of asthma.
3.Emergency Interventional Therapy for Ureter Invasion by CACX, Uremia complicated with Hemorrhoea

Objective To discuss the emergency interventional therapy for ureter invasion by CACX, obstructive bilateral hydronephrosis and uremia complicated with hemorrhoea. Methods 5 cases with ureter invasion by CACX, obstructive bilateral hydronephrosis and uremia suffered from hemorrhoea during hemodialysis, who were performed with emergency interventional therapy. Results After therapy, hemostasis were realized in all cases, and all symptoms were alleviated, such as vaginal fluid and fall-swell in pelvis. The short-term total effective rate was 100% . Conclusion Interventional chemoembolization can be used in the treatment of CACX with acute hemorrhoea.
4.The inhibitory effects of skeletal muscle conditioned medium on thoracic malignant tumors

Objective To explore the organic micro environmental effect of skeletal muscles on the proliferation of thoracic malignant cells, its significance in the rarity of metastases in skeletal muscles and the prospect for its clinical applications.Methods Primary culture of new born Wistar rat skeletal muscle cells was established.The murine skeletal muscle conditioned medium(MMCM)was prepared to test its effect on thoracic malignant cell lines of A549、Anip-973,PLA-801C,NCI-H466,Eca109 and benign cell line of BHK-21 by MTT assay.Results Proliferations of thoracic malignant cell lines of A549,Anip-973,PLA-801C,NCI-H466,and Eca109 were significantly restrained when cultured with MMCM,while the proliferation of benign renal cell line(BHK-21)was not affected.Conclusions The conditioned medium of new born Wistar rat skeletal muscle cells could selectively inhibit the proliferation of thoracic malignant cells in vitro.Moreover,it affects tumor cells only and has no apparent effect on normal cells,which differs from most of the chemotherapeutic agents.These findings suggest a sound mechanism in the rarity of metastases in skeletal muscles.A therapeutic agent could be generated from MMCM to complement surgery and/or chemotherapy.
5.Stably upregulating expression of chondromodulin-Ⅰ in bone marrow mesenchymal stem cells

BACKGROUND:Chondromodulin-Ⅰ is expressed mainly in the cartilage, but it is little expressed in mesenchymal stem cells. Combined with the previous study of our group, we concluded that chondromodulin-Ⅰmaybe play an important role in inducing mesenchymal stem cells into chondrocytes accurately.
OBJECTIVE:To construct an expression plasmid stably carrying chondromodulin-Ⅰ to up-regulate the expression of chondromodulin-Ⅰ in bone marrow mesenchymal stem cells.
METHODS:Specific primers were designed in rat cartilage for chondromodulin-Ⅰ gene, then the pcDNA3.1 (+) plasmid expression vector was digested by enzyme and directional connected gene to construct pcDNA3.1(+)/ChM-Ⅰ expression vector. Bone marrow mesenchymal stem cells were obtained from rats using the method of density gradient centrifugation combined with adherent culture. Recombinant plasmid pcDNA3.1(+)/ChM-Ⅰ was transfected into rat bone marrow mesenchymal stem cells with liposome method, and G418 selection was used for stable screen of transfected cells. Reverse transcription-PCR and western blot were used to detect chondromodulin-Ⅰ expression in celllines.
RESULTS AND CONCLUSION:The positive clones were digested by enzyme and were identified and sequenced. The results showed that the reality length and sequence of chondromodulin-Ⅰ gene were consistent with the theoretical values, and reading frame was correct. Reverse transcription-PCR and western blot results showed that the expressions of chondromodulin-ⅠmRNA and protein were markedly up-regulated in bone marrow mesenchymal stem cells. Recombinant plasmid pcDNA3.1(+)/ChM-I was successful y constructed, and transfected into rat bone marrow mesenchymal stem cells. After G418 selection, expression of chondromodulin-Ⅰ was up-regulated stably in rat bone marrow mesenchymal stem cells.
6.Effect of Compound Rhubarb Mixture on contraction of isolated small intestinal smooth muscle of rabbit

OBJECTIVE: To investigate the effect of Compound Rhubarb Mixture on contraction of isolated small intestinal smooth muscle of rabbit. METHODS: Serum containing Compound Rhubarb Mixture was prepared. The contraction curves of isolated small intestinal smooth muscle before and after administration of the serum were recorded by Medlab biological information collecting system. RESULTS: The serum containing Compound Rhubarb Mixture amplified the contraction amplitude of isolated small intestinal smooth muscle of rabbit. The rate of change of contraction amplitude was elevated significantly after administration, while the frequency of contraction did not change obviously. CONCLUSION: The serum containing Compound Rhubarb Mixture can enhance the contractive function of isolated small intestinal smooth muscle of rabbit.
7.Effect and mechanism of emodin on intestinal movement

AIM To examine the effect and mechanism of emodin on intestinal movement of constipation model. METHODS The movement of charcoal powder and the absorption of D-xylose into serum were observed. The electrical p otential difference(PD) related to Na + and glucose transport was measured across the wall of rev erted intestinal sacs. RESULTS Emodin 0 1, 0 2 g?kg -1 incr eased markedly the rate of charcoal powder moving and inhibited the absorption of D-xylose into serum. Emodin 0 2, 0 4, 0 8 and 1 6 g?L -1 decreased PD when ther e was glucose. However, emodin had little effect when glucose was free. CONCLUSION Emodin promotes intestinal movement and this effect is related to Na +,K +-ATPase.

Objective To analyze the reasons of CT misdiagnosis of solid-pseudopapillary tumor of pancreas (SPTP), including wrong orientation, mislocation. Methods The literatures on SPTP were discussed. Results Lesions of SPTP could appear at any position of the pancreas. Conclusion The result is very significant for guiding treatment and judging prognosis.
9.Preparation and antioxidative activities of the sulfated Hunai polysaccharide

Purpose The aim is to sulfonize Hunai polysaccharide fr om p leurotus tuber-rigium(Fr.)Sing. and to evaluate the antioxidative activities of the sulfated po lysaccharide (S-HNP).Methods S-HNP was prepared by the reacti on of Hunai polysaccharide with chlorosulfonic acid-Pyridine. The antioxidative activities o f S-HNP were evaluated as follows: (1) the inhibition effects on Fe2+- Vc inducing the injury of rat liver mitochondria in vitro, (2) the protectiv e ef fect on CuSO4 -Phen-Vc-H2O2 inducing the damage of DNA, (3) the scaven ging effect on O*-2. Results S-HNP could protect mitochondria from lipid peroxidation induced by Fe2+-Vc, i ncluding the inhibitions of the increase of TBARS content, the swelling of mitoc hondria and the decrease of membrane fluidity, and protect DNA from the damage induced by CuSO4-Phen-Vc-H2O2, and scavenge O*-2 generated in the sel f-oxidation of pyrogallic acid. Conclusion S-HNP exhibi ted marked antioxidative activities.
10.The study on the levels of serum uric acid in patients with bipolar disorder

Objective To investigate the serum levels of uric acid (UA) in bipolar disorder patients and their clini?cal significance. Methods Serum levels of UA was measured in 126 bipolar disorder patients including 77 mania pa?tients and 49 depression patients, 69 first-episode schizophrenic patients and 126 healthy controls (control group). Bipo?lar disorder patients was assessed by using the Young Mania Rating Scale (YMRS) and the Hamilton Depression Scale (HAMD). Results The serum UA levels were significantly higher in bipolar group [(349.34 ± 107.21) μmol/L] than in schizophrenic group [(319.71±84.48)μmol/L] and in control group [(280.94±71.90)μmol/L] (P<0.01). There were signifi?cant differences in serum UA levels between manic episode and depressive episode [(366.45 ± 104.01) μmol/L vs. (322.45±107.69)μmol/L]. The serum UA levels were higher in both manic episode and depressive episode groups than in control group (P<0.01). The serum UA levels was not significantly different between patients with and without medication (P>0.05). Correlation analysis indicated that the correlation of serum UA levels with YMRS scores and HAMD scores was not significant in bipolar disorder patients (P>0.05). Conclusions Patients with bipolar disorder has elevated serum uric acid levels which may be a biomarkers of bipolar disorder.
