1.Effect of eupolyphaga seu steleophaga on proliferation of autosomal dominant polycystic kidney disease cyst-lining epithelial cells

Objective:To observe the effect of eupolyphaga seu steleophaga(ESS) on the proliferation of autosom al dom inant polycystic kidney disease(ADPKD) cyst- lining epithelial cells.Methods:ESS serum and control serum were obtained from SD rats gastrogavaged with ESS water solution and physiological solution.ADPKD cyst- lining epithelial cells were treated with RPMI16 4 0 containing:(1) EGF0 ,2 .5 ,5 ,10 ,2 0 ng/ ml;(2 ) ESS serum and control serum(1% ,2 .5 % , 5 % ) ;(3) EGF10 ng/ ml,ESS serum and control serum(1% ,2 .5 % ,5 % ) .The cell proliferation was determined by Brdu incorporation assay. Results:EGF had a stimulating effect on the ADPKD cyst- lining epithelial cells;the ESS serum significantly inhibited the proliferation of ADPKD cyst- lining epithelial cells compared with control serum;ESS serum also significantly inhibited the proliferation of ADPKD cyst- lining epithelial cells activated by EGF(10 ng/ m l) com pared with control serum.All the effects were dose- dependent.Conclusion:Eupolyphaga seu steleophaga may prolong the progression of ADPKD via the inhibitory effect on the proliferation of cyst- lining epithelial cells. 2.Lentivirus-mediated shRNA interference of ghrelin receptor blocks growth of colorectal cancer cells

[ ABSTRACT] AIM:To investigate the therapeutic effects of lentivirus-mediated shRNA targeting growth hormone secretagogue receptor 1a (GHSR1a) on colorectal cancer cell line SW480 both in vitro and in vivo.METHODS: Human GHSR1a sequence was used for the design of shRNA targeting GHSR1a, which was introduced to lentivirus, followed by transfection into SW480 cells.CCK-8 assay was performed to detect cell viability.The mRNA expression of GHSR1a and PTEN in colorectal cancer cells was detected by RT-PCR.The protein levels of GHSR1a, ghrelin, PTEN, p-AKT and p53 were determined by Western blot.Stable GHSR1a silencing in SW480 xenografts in nude mice was established.After the mice were sacrificed and weighted, immunohistochemistry was used to detect the positive expression of Ki-67 and PTEN in the tumors.RESULTS:GHSR1a was over-expressed in the malignant cells in comparison with the normal cells in vitro. The tumor specific lentivirus-mediated shRNA targeting GHSR1a gene and GHSR1a knockdown SW480 cells were success-fully constructed.After transfection with GHSR1a shRNA, the expression of GHSR1a at mRNA and protein levels was markedly inhibited in the SW480 cells.Furthermore, GHSR1a silencing by specific shRNA showed increased PTEN, inhi-bition of AKT phosphorylation and promotion of p53 release in the SW480 cells.In vivo results demonstrated that down-re-gulation of GHSR1a in the SW480 cells significantly decreased the expression of Ki-67 and increased the expression of PTEN in the tumor tissues, leading to a marked reduction in tumor weight in comparison to blank control or negative control
tumors.CONCLUSION:Down-regulation of GHSR1a by shRNA technique inhibits the growth of colorectal cancer cell line and xenograft tumor through activation of the PTEN/PI3K/AKT signaling pathways.
3.Study on the Effect of Aldehydes and Tannic Acid on the Stability of Soft Capsules

Objective To study the influence of aldehydes and tannic acid on soft capsules, approach how to prevent soft capsules from delayed disintegration. Methods Add aldehydes and tannic acid to soft capsules material ingredient, to proceed accelerating stability trial and test the disintegration time. Result Little increase of aldehydes amount had great effect on the disintegration time of soft capsules, different aldehydes and tannic acid had different effect on the disintegration time, adding some organic acid such as fumaric acid to the material would improve disintegrating property. Conclusion The amount of aldehydes and tannic acid contained in the material and ingredient should be controlled strictly when prepare soft capsules so that the disintegration time of capsules could be complying with the Pharmacopeia.
4.Childhood Maltreatment Experiences of Patients with both Obsessive-Compulsive Disorder and Personality Disorders

Objective: To explore childhood traumatic experiences of OCD patients who were also diagnosed personality disorders (PDs) and those without PDs. Methods:SCID-I/P was administrated to diagnose OCD patients. PDQ+4 and PDI-IV were then administrated to 44 OCD subjects to determine whether they also suffered from personality disorders. Childhood maltreatment experiences of the sample were measured with Childhood Trauma Questionnaire (CTQ).50 normal control were also collected and subjected to CTQ-SF tests. Results:32 of 44 (72%) OCD patients also met DSM-IV criteria for personality disorders. OCD patients scored significantly higher than normal control in CTQ-SF (Childhood Truma Questionuaire-28 Short Form) and in all domains with the exception of physical abuse. Comparison between OCD subjects with PDs and normal control, the former scored significantly higher in emotional abuse (55.10/32.54), sex abuse (49.63/36.30), physical neglect (52.03/34.76), and emotional neglect domains (55.84/32.32). Whereas OCD subjects without PDs only scored significantly higher in emotional neglect domains compared with normal control (42.50/28.26).Conclusions:Subjects with PDs experienced more severe traumatic events during their early life. "pure OCD" patients, however, might experienced more emotional neglect.

6.Comparison of four kinds of cholangiographies in the prevention of bile duct injury during laparoscopic cholecystectomy

Objective The efficacies of four cholangiogrphies were compaired with each other in preventing bile duct injury during the procedure of LC. Methods Four cholangiogrphies were used in LC:1. Cold light cholangiography (CLCP); 2. Methylenum coeruleum cholangioguaphy (MCCP); 3. Intraoperative cholangiography (IOCP); 4. Intraoperative endoscopic retrogarde cholangiopancreatography (IERCP). Results The images of CLCP and MCCP were direct and could help operator to identify bile duct structure in LC. The images of IPCP and IERCP were indirect and could not be so helpful. Conclusions CLCP is the only technique that clearly and directly shows the location of the extra hepatic biliary system and may be useful in selected cases with abnormal or uncertain anatomy for the prevention of bile duct injury.

Objective To evaluate the comprehensive benefit about the installation obstructed snails in large irrigation canals for preventing snails spreading in lake-swamp areas of schistosomiasis.Methods The installation obstructed snails was set up in upper reaches of main canal Shilong, Tianmen City Data of snails and schistosomiasis prevalance were collected and analysed. The comprehesive benefit was analysed with the cost-benefit methods of economic hygienics. Results After the installation obstructed snails was set up the snails areas and human infection rate decreased markedly. New patients with acute and advanced schistosomiasis were not found in the irrigation areas. The ratio of comprehesive cost-benefit was 1∶9.84,the ratio of net benefit was 8.84∶1. Conclusion Setting up the installation obstracted snails to cut off the way of snails spreading was the effective measure of control schistosomiasis in lake-swamp areas.
9.Effect of taurine on proliferation and apoptosis of rat mesangjal cells cultured in vitro

Objective To investigate the effect of taurine on proliferation and apoptosis of rat mesangial cells cultured in vitro and explore its possible mechanism. Methods Rat mesangial cells were incubated with different concentration of taurine. Cell proliferation was assessed by MTT colorimetric assay. Cell cycle and apoptosis was analyzed by flow cytometry. Form of apoptosis was examined by vital staining of alcidine orange. The expression of c-jun and c-fos was determined by immunocytochemistry and computed video text analysis system. Results Administration of 5 ~ 50 mmol/L taurine into culture medium had no loxicity to mesangial cells, but could significantly inhibit mesangial cells proliferation in dose-dependent manner, subsequently increased the cells in Go/Gi phase and decreased the cells in S phase. 10.Fluorescence labeling method for evaluating the efficacy of hospital environment cleaning

Objective To evaluate the effectiveness of hospital environmental cleaning practice by fluorescence labeling method.Methods From January to February 2015,312 ward rooms in 7 hospitals were chosen,high touch object surface were labeled with fluorescence,after object surface being cleaned by cleaners,clearance rates of fluorescence labeling were calculated (as baseline survey data),training and on-site guidance for cleaners were performed(intervention measures),fluorescence labeling clearance effect before and after intervention was compared.Results A total of 110 ward rooms were performed baseline survey,the fluorescence labeling clearance rate of 2 856 touched clean surface was only 50.81%,the quantitative evaluation value was 45.70;after intervention,202 rooms were surveyed,fluorescence labeling clearance rate of 3 992 touched clean surface enhanced to 79.23 %,the quantitative evaluation value was 76.30;there was significant difference in fluorescence labeling clearance rate between before and after intervention (x2 =612.14,P<0.05).In the baseline survey,the clearance rates of fluorescence labeling on touched surface of medical instruments,hospital beds,and toilets were 46.07%,37.80%,and 25.20% respectively;after intervention,the clearance rates were 80.59%,75.90%,and 51.70%,respectively.After intervention,fluorescence labeling clearance rates of beepers,toilet seat covers,toilet electrical switches,and chairs were low,the clearance rates of these touched surface in baseline survey were< 30%,after intervention were 47.03 %-68.32%;the clearance rates of other high touch surface were all>75 %.Conclusion Fluorescence labeling method can directly reflect the operation quality of cleaners,and improve the cleanliness of ward environment,it is a simple,inexpensive and objective globally popular method for evaluating hospital environment cleanliness.
