1. Meta-analysis on effect of Kushen Gel on high-risk cervical HPV infection

Objective: To systematically evaluate the effectiveness of Kushen Gel in the treatment of cervical high-risk human papillomavirus (HR-HPV) infection. Methods: The Cochrane Library, PubMed, Embase, Medline, Wanfang, VIP, CNKI, and CBM databases were electronically searched to collect randomized controlled trials (RCTs) on Kushen Gel in the treatment of the cervical high-risk HPV infection from their inception to December 2019. Two researchers screened the articles, extracted the data, assessed the risk of bias of the eligble studies independently, and performed Meta-analysis with RevMan 5.3. Results: A total of 583 cases were included in five RCTs. The Meta-analysis showed that HR-HPV negative rate [RR = 1.38, 95% CI(1.20, 1.58), P < 0.000 01], reduction of HPV-DNA virus load [MD = -0.70, 95% CI (-0.85, -0.56), P < 0.000 01], and the reduction of postoperative complication rate [RR = 0.45, 95% CI (0.32, 0.65), P < 0.000 1] of applying Kushen Gel alone or applying Kushen Gel combined with microwave/LEEP therapy in the treatment of cervical HR-HPV infection or combined with cervical intraepithelial neoplasia (CIN) were significantly higher than those in the follow-up group; The HR-HPV negative rate of Kushen Gel was higher than interferon group (P < 0.05); There was no significant difference in the one year recurrence rate after LEEP combined with Kushen Gel in the treatment of cervical HR-HPV infection combined with CIN2/CIN3 compared with the follow-up control group (P > 0.05). Conclusion: The therapeutic effect of applying Kushen Gel can increase the rate of clearance of HPV, reduce HPV-DNA load, and reduce postoperative complication. There is no evidence that the use of Kushen Gel after LEEP can reduce the recurrence rate of CIN2/CIN3. Due to the limitation of the quantity and quality of the included studies, the results need to be further verified and updated by more high-quality RCTs.
2.New progress in the clinical application of ultrasound cycloplasty in the treatment of glaucoma

Ultrasound cycloplasty(UCP)procedure using high intensity focused ultrasound(HIFU)is a new non-invasive glaucoma treatment based on miniaturized transducers. In recent years, the safety and efficacy of UCP on various types of glaucoma have been well verified, and it has shown broad application prospects in the treatment of glaucoma. This article collects and reviews the relevant literature on glaucoma treatment by UCP, and summarizes the information on the development of equipment, composition and operation, treatment mechanisms, indications, efficacy and safety, with the purpose to provide relevant basis for clinical applications and future research.
3.Efficacy and safety of ultrasound cycloplasty in the treatment of refractory glaucoma

AIM: To evaluate clinical efficacy and safety of ultrasound cycloplasty(UCP)in the treatment of refractory glaucoma.METHODS:From June 2021 to October 2022, a total of 17 patients(17 eyes)with refractory glaucoma were enrolled in this prospective study, and they all underwent UCP. The patients underwent 6 mo followed-up post-treatment, and the intraocular pressure(IOP), pain grade score, IOP lowering drugs, success rate and occurrence of complications were documented.RESULTS:The IOP was significantly decreased from 51.98±7.80 mmHg before UCP to 32.54±13.21 mmHg at 1 d, 22.38±11.98 mmHg at 1 wk, 22.63±10.78 mmHg at 1 mo, 26.05±9.17 mmHg at 3 mo, and 23.73±9.60 mmHg at 6 mo postoperatively(all P<0.01). The percentage of IOP reduction after treatment was 36.25%, 57.10%, 56.35%, 49.16% and 54.09% at 1 d, 1 wk, 1, 3, and 6 mo, respectively. The pain grade scores were decreased(P<0.01). There was a statistically significant reduction in the use of IOP lowering medications(P=0.008). At 6 mo postoperatively, 2 eyes(12%)were complete success, 11 eyes(65%)were qualified success, and 4 eyes(24%)were failure. The main complication observed was anterior chamber inflammation in 1 eye(6%), foreign body sensation in 2 eyes(12%), subconjunctival hemorrhage in 2 eyes(12%), and conjunctival congestion in 6 eyes(35%). All symptoms spontaneously resolved within 1 wk without requiring any specific treatment. One case of choroidal detachment(6%)occurred on 10 d postoperatively, but recovered after orally treated by prednisone acetate for 1 mo. No other serious complications, such as hyphema, corectopia, synechia or macular edema were reported.CONCLUSION:UCP is safe and efficacious in treating refractory glaucoma, reducing IOP and alleviating ocular pain symptoms, while maintaining a favorable safety profile.
4.Effects of Benzoxazole Derivative PO- 296 on Dendritic Cell Differentiation and Related Indexes

OBJECTIVE: To investigate the effects of benzoxazole derivative 2-(chlorobenzoxazolyl-2-yl)-4,5,6,7-tetrahydro- dihydro-indazole-3-ol (PO-296) on the differentiation of murine bone marrow-derived dendritic cells(DC) and their related indexes as specific surface molecules and inflammatory cytokines. METHODS: Bone marrow nuclear cells of mice were isolated, and immature DC (imDC) was obtained by recombinant mice granulocyte macrophage colony-stimulating factor and recombinant mice IL-4. After pretreated with low-dose, medium-dose and high-dose (1, 5, 25 μmol/L) of PO-296, DC was obtained by lipopolysaccharide induction. Flow cytometry was used to detect the expression of DC specific surface molecules [i.e. the proportion of class Ⅱ major histocompatibility complex (MHC Ⅱ), CD80, CD86 and chemokine receptor 7 (CCR7) positive cells], imDC phagocytosis (i.e. the proportion of dextran positive cells) and DC survival (i.e. the proportion of survival cells). ELISA method was used to detect the levels of inflammatory cytokines (IL-10, IL-12 and TNF-α) in cell culture medium. RESULTS: Compared with imDC group, the proportion of MHC Ⅱ, CD80 and CD86 positive cells were increased significantly in non-loading group (P<0.05). Compared with non-loading group, the levels of IL-10 in cell culture medium were increased significantly in PO-296 groups. The proportions of MHC Ⅱ, CD80 and CD86 positive cells in positive group and PO-296 medium-dose and high-dose groups as well as the levels of IL-12 and TNF-α in cell culture medium in administration groups were decreased significantly (P<0.05). There was no statistical significance in the proportion of CCR7 positive cells, dextran positive cells and survival cells in administration groups, compared with non-loading group (P>0.05). CONCLUSIONS: PO-296 has no obvious cytotoxicity and does not affect the phagocytic function of imDC. At the same time, the compound can inhibit the expression of DC specific surface molecules and regulate the secretion of inflammatory cytokines.
5.Combined local block anesthesia and general anesthesia in cleft lip repairing operation of infants.

OBJECTIVESummarizing the value and key point of combined use of local block anesthesia and general anesthesia in cleft lip repairing for infants.
METHODSLocal block anesthesia and general anesthesia were combined to use in 120 cleft lip repairing operation of infants using Midazolam, Fentangl, Ketamine, Scoline (or Vecuronium) in intubation, and injecting 0.5% - 1% lidocaine of which contained 1/250 000 adrenalinum. While intubation and operation, BP, P, R, SpO2, T and ECG were observed clearly. After operation, extubation was delayed and breathing oxygen was used for infants with breathing airway infection.
RESULTSWhile operation, anesthesia of 120 infants was smooth. After operation, the infants regain conscious quickly and no accident occurred.
CONCLUSIONFor the cleft lip repairing operation to infants, as combined use of local block anesthesia and general anesthesia, anesthesia is smooth, internal environment of body is steady, bleeding is little, regaining consciousness is quick and the infants are safe.

6.Screening of cytotoxic activity against B16 tumor cell of mangrove fungi isolate from Qinglan harbor in Hainan.

Six hundred and eight fungi strains were isolated from seventy-eight samples of mangrove plants and soil that collected from Qinglan harbor. Cyctotoxic activity was detected by observing the growth inhibition or killing of the tumor cells under microscope. The result showed that 81 strains (about 13.32% of the total strains isolated) displayed cytotoxic activity against B16 tumor cell. The most fungi strains were isolated from mangrove plant Sonneratia alba, and most of cytotoxic active fungi strains were isolated from mangrove plant Heritiera littoralis.

7.Study of dry eye after phakic posterior chamber intraocular lens implantation

AIM: To study the clinical changes related to dry eye and evaluate the severity of ocular surface diseases in patients with posterior chamber intraocular lens(ICL)implantation.
METHODS: Prospective study. Totally 50 eyes of 50 cases with myopia who were underwent ICL surgery from September 2019 to October 2019. Ocular surface disease index questionnaire(OSDI), fluoresceine staining of cornea tear break up time(TBUT), Schirmer Ⅰ test(SⅠt), and tear meniscus were measured before and 1wk, 1 and 3mo after surgery.
RESULTS: The scores of OSDI questionnaire and corneal staining were significantly increased at 1wk and 1mo after operation(P<0.05). There was a difference between 3mo and 1wk pastoperatve(χ2=5.267,P=0.022), and recovered to the preoperative level at about 3mo. Corneal staining scores were significantly positively correlated with OSDI questionnaire scores at all time points(rs>0, P<0.05). TBUT decreased significantly at 1wk and 1mo after operation(P<0.05), and returned to the preoperative level 3mo after operation. There was no significant difference in the basal tear volume(SⅠt)and the height of lacrimal river in the middle of lower eyelid between before and after operation(P>0.05).
CONCLUSION: The stability of tear film on the ocular surface will be affected by ICL implantation in the early postoperative period, and the patients will have different degrees of dry eye related symptoms. OSDI questionnaire score can be used to evaluate their subjective discomfort symptoms. However, at 3mo after operation, the stability of tear film on the surface of eyes recovered to the preoperative level. The higher the positive score of corneal staining was, the more obvious the subjective discomfort was.
8.Discussion on opposing needling combined with dragon-tiger fighting needling for chronic pelvic inflammation.

Opposing needling is an acupoint selection method of acupuncture recorded in ('). And the first record of dragon-tiger fighting needling is found in the (), it is a compound reinforcing and reducing manipulation of tonification-purgation method. Both of them are widely used in the treatment of pain syndrome, which are mainly for nerve system and musculoskeletal diseases and seldom for gynecological diseases. By analyzing the pathogenic characteristics of chronic pelvic inflammation, the clinical application of opposing needling combined with dragon-tiger fighting needling for chronic pelvic inflammation is expounded based on the theory of treating different diseases with the same treatment in TCM.

9.Construction of recombinant gene adenovirus encoding enhanced green fluorecence protein-peroxisome proliferator-activated receptor gamma2 fusion protein and its expression in bone marrow mesenchymal stem cells.

OBJECTIVETo construct mouse enhanced green fluorecence protein (EGFP) -peroxisome proliferator-activated receptor (PPAR)gamma2, and to detect EGFP-PPARgamma2 expression in infected mouse bone marrow mesenchymal stem cells (BMSC).
METHODSCut the fragment of PPARgamma2 from the expression plasmid pcDNA flag PPARgamma2, then cloned the gene fragment into pEGFP-C1 and pEGFP-N1 vector. Subsequently, subclone the fragment EGFP-PPARgamma2 from pEGFP-C1-PPARgamma2 into the shuttle plasmid DC315. HEK293 cells were co-transfected with the constructed recombinant shuttle plasmid DC315-EGFP-PPARgamma2 and large adenovirus helper plasmid pBHGlox deltaE1, 3Cre in mediation of liposome. The obtained replication-defective recombinant adenovirus Ad-EGFP-PPARgamma2 was confirmed. Then it was propagated in HEK293 cells. After the BMSC were transfected for 72 h, adipogenic differentiation was demonstrated.
RESULTSHEK293 cells were transfected with the pEGFP-C1-PPARgamma2 or pEGFP-N1-PPARgamma2 in mediation of liposome. The former green fluorescence protein was better than the latter by fluorescence microscope. The recombinant plasmids were digested and identified. Western blot analysis showed the expression of EGFP-PPARgamma2 in vitro. EGFP-PPARgamma2 protein was detectable in the nucleus of BMSC.
CONCLUSIONThe recombinant adenovirus encoding EGFP-PPARgamma2 fusion protein was successfully constructed, which provided a basis for application of EGFP-PPARgamma2 gene to adenovirus-mediated gene therapy.

10.Clinical analysis of maxillofacial patients with diabetes on perioperative period.

OBJECTIVETo investigate the treatment of perioperative period on maxillofacial patients with diabetes.
METHODSThe retrospective analysis was taken on clinical data of 24 cases of maxillofacial patients with diabetes.
RESULTSAll patients recovered without severe complications by controlling blood sugar during whole stage and preventing infection with antibiotics after surgery. The wounds in 21 cases healed in 10-14 days after the operations and that didn't in 3 cases. With the treatment on local wounds, the wounds in 3 cases healed in 14-28 days after the operations.
CONCLUSIONIt's the treatment principle of maxillofacial surgical patients with diabetes to monitor the levels of blood glucose on time, to control the levels of blood glucose rigorously at the whole stage and to prevent complication.
