1.Nasopharyngeal Carcinoma:Correlation Between Peripheral Deeper Infiltration and Prognosis and Expression of P53 Protein

Objective To investigate the correlation between degree of deeper infiltration of NPC on CT scan,prognosis and expression of P53 protein,in order to further recognize bilogical features of NPC.Methods Collect 70 NPC biopsy specimens,which had performed CT scan before radiotherapy and had detailed clinical records.The expression of P53 protein was examined by means of S-P immunohistochemical technique.The correlation between expression of P53 protein and peripheral deeper infiltration and prognosis was analysed in detailed.Results The results of these cases showed that the positive rate of over expression P53 were 68.57%,which had no relation with the histological types and pathological grades.The positive rate of P53 in Ⅱ、Ⅲ types had a higher positive rate than that of the Ⅰ type of NPC.Expression of P53 protein had no relation with neck lymph node metastasis,but had significant correlation with the size of metastatic lymph node,which is the positive rate of P53 in diameter of metastatic lymph node <4 cm was higher than that of diameter ≥4 cm.Expression of P53 had no relation with local recurrence and distant metastasis after radiotherapy.Conclusion With the tumor infiltrating the deeper layer of nasopharynx,the positive rate of P53 expression is more and more high,and degree of malignant is increasing.The three types on CT scan represents the biological features of NPC.
2.Preliminary investigation on polymorphs of two traditional Chinese medicine reference substances.

Crystal structures of chemical drugs has been being investigated widely. But few attention has been paid to polymorphs-phenomena of active ingredients from Traditional Chinese Medicine(TCM). Taking anhydrous dehydroandrographolide and hydrousprim-O-glucosylcimifugin as example, differences between TCM reference substances (RSs) with different crystal structures were discussed by using microscopy, melting point determination, differential thermal analysis (DTA) and infrared (IR) methods. The results showed that different crystal structures could lead to change of melting points, thermal behaviors and IR spectrum. It's indicated that polymorphs may be considered if different physicochemical properties were obtained when applying TCM RS. Differences of chemical properties of active ingredients from TCM with different crystal structures need further investigation.

3.Survey of natural foci of plague in Xiji county, Ningxia in 2007

Objective To learn the plague's host animals and parasitic flea composition, and to investigate the natural foci of plague in Xiji county in order to provide basic information for plague prevention and control. Methods The Citellus alaschanicus density, nocturnal rodents, the body flea, the burrow track flea, the nest flea were investigated in 8 townships (town) of Xiji county from June 11 2007 to July 25 2007. Specimens of small mammalian, fleas were collected for bacteriological and serological testing. Results The average density of the main host Citellus alaschanicus was 0.85 per hectare. The nocturnal mouse capture rate was 0.80%(24/2987).The survey found 16 species of small mammals that belonging to 3 orders, 9 families and 16 species with Citellus alaschanicus the dominant species. The Citellus alaschanicus had 2.84 fleas per body. Four families and 16 species of fleas were identified in the areas. The Citellus alaschanicus and Citellophilus Tesquorum Mongolicus were the dominant species. Plague bacteriology and serology tests were negative. Conclusions The study shows that the area is suitable for the formation of natural foci of Citellus alaschanicus plague. Surveillance is an important measure for prevention and control of the plague.
4.Analysis of iodized salt monitoring results in Chongqing Municipality from 2001 to 2009

Objective To monitor the quality changes of iodized salt and analyze its impact factor in Chongqing between 2001 and 2009. Methods Salt samples were collected according to the east, west, south,north and center locations in iodized salt production, wholesale and household sectors. Two units in iodized salt production and wholesale segment were sampled from north, south, east and west places and only 1 unit was sampled from the central place. Nine samples were collected every month in each place. If the place had less than 9 units, and then taken all the units. About resident household, 2 townships were sampled from north, south, east and west places, and 1 township was sampled from the central place, then 20 samples were collected from each township. Iodine content was detected by oxidation-reduction assay. The index of mean iodine, qualified rate from factories and wholesale, coverage rate and taking rate of qualified iodized salt in residents were calculated.Significance was analyzed by trend test, analysis of variance and X2 test. Results The qualified rate of iodized salt from the manufacturers was 92.9%(13/14) in 2001 and the rate was 100.0% each year from 2002 to 2009. The qualified rates of iodized salt from the wholesale were 88.7%(282/318) - 99.8%(431/432). The rates of 2001 and 2002 were lower than that of other years(X2 = 4.98 - 45.69, all P< 0.05 or < 0.01). The coverage rate and taking rate of qualified iodized salt in residents were 94.2% (11 154/11 841 ) - 98.9% ( 14 061/14 217), 83.5% (9 887/11 841 ) -95.8% (13 449/14 039), respectively. The rates showed an increasing tendency (F = 9.27, 26.39, all P < 0.05).The districts(counties) with qualified iodized salt consumption rate > 90% kept increasing. The mean iodine from the manufacturers and wholesale were 29.71 - 36.25, and 31.26 - 36.13 mg/kg, respectively. The iodine level showed a descending trend(F = 35.45, 140.59, all P < 0.01 ). The mean iodine level from the inhabitants were 28.84 - 30.98 mg/kg which remained stable (F = 3.05, P > 0.05 ). The iodine level from manufacturers, wholesale to inhabitants showed an descending trend(F = 38.46 - 671.23, all P < 0.01 ). Conclusions The surveillance results of iodized salt shows an increasing tendency in quality of iodized salt, eoverage rate and taking rate of qualified iodized salt. Factors that affect the quality of iodized salt is that the enterprise does not add iodine to salt strictly by the standard.
5.Construction and expression of the recombinant plasmid pET28α-Sj26GST-Sj32 of Schistosoma japonicum in Escherichia coli BL21(DE3)

Objective To construct and express the recombinant plasmid pET28α-Sj26GST-Sj32 of Schistosoma japonicum(Sj) in Escherichia coli BL21 (DE3). Methods Total RNA was extracted from Sj adult worms by ultrasound-breaking, Sj26GST and Sj32 antigen gene was respectively amplified by RT-PCR from the total RNA; Sj26GST-Sj32 fusion gene obtained with gene splicing by overlap extension(SOEing) was cloned into prokaryotic expression plasmid pET28α and transformed into Escherichia coli BL2 (DE3) to construct pET28α-Sj26GST-Sj32;BL21 (pET28α-Sj26GST-Sj32) was induced with isopropyl-β-D-thiogalactopyranosid (IPTG), and the expressed products were analyzed and identified by sodium dodecyl sulfate polyacrylamide gel electropheresis (SDS-PAGE)and Western blotting. Results The 1991 bp Sj26GST-Sj32 fusion gene was successfully amplified by gene SOEing and cloned into pET28α by restriction analysis and PCR identification, the recombinant plasmid pET28α-Sj26GST-Sj32 was successfully constructed; the relative molecular mass of the expressed recombinant protein was approximately 69 × 103 by SDS-PAGE, and the amount of the expressed protein was 25% of the total bacterial proteins; the fusion protein could be recognized by sera from rabbits infected with Sj by Western blotting.Conclusions The recombinant plasmid pET28α-Sj26GST-Sj32 is successfully constructed and highly expressed in Escherichia coli in fused form with His-tag, and the expressed fusion protein shows specific antigenicity.

7.Analysis of sacro-iliac articulation malposition misdiagnosed as single prolapse of lumbar intervertebral disc

ObjectiveTo find out the reason of sacro-iliac articulation malposition misdiagnosed as single prolapse of lumbar intervertebral disc, define the essential point of diagnosing sacro-iliac articulation malposition, and choose the best treatment.MethodsChanges such as X-ray photos of lumbosacral portion and pelvis, CT photos, physiological curvature, pyramid space, joint space, pubic symphysis, and wing of ilium showed in MRI photos, and symptoms, signs of 29 patients with sacro-iliac articulation malposition were analyzed and compared with distribution of nerve in the same part.Results29 patients with sacro-iliac articulation malposition all had bulge of lumbar intervertebral disc showed by CT and MRI photos, 26 had retrograde affection of spinal column, 17 had spinal column sequence changes showed by X-ray photos. After 3-10 times treatment on the basis of sacro-iliac articulation malposition, 19 healed, 8 were effective, and 2 had been better.ConclusionCurative method of sacro-iliac articulation malposition is different with prolapse of lumbar intervertebral disc, although there being many same symptoms and signs, only need conservative treatment.
8.Analysis of Misdiagnosis between Cervical Muscle Strain and Cervical Spondylopathy

ObjectiveTo analyze the reason of cervical muscle strain misdiagnosed as cervical spondylopathy, and confirm the diagnosis and the best treatment.MethodsThe data of cervical X ray (direct, lateral and double oblique), CT and MRI (degeneration degree and lordosis of cervical spine, the changes of vertebral interspace, vertebral joint, intervertebral disc, intervertebral foramen, and transverse foramen) of 32 patients with cervical muscle strain were analyzed corresponding with clinical symptoms and signs. The attribute of the symptoms and signs were distinguished.ResultsIn all patients, besides the common symptoms and signs, there were 26 patients having degeneration and 17 alignment of the cervical spine from X ray, and 29 cases having cervical disc herniation from CT or MRI. After 5~10 times manipulation curing, 22 patients were cured, 8 patients were obviously improved and 2 patients were better than before.ConclusionCervical muscle strain and cervical spondylopathy have some similar symptoms and signs. But their attribute and therapeutics are very different. Manipulation is an effective and stable therapeutic method for cervical muscle strain.
9.Effect of electroacupuncture on calcium-activated chloride channel currents in interstitial cells of Cajal in rats with diabetic gastroparesis

Objective: To investigate the mechanisms of electroacupuncture (EA) at Zusanli (ST 36), Liangmen (ST 21) and Sanyinjiao (SP 6) in intervening diabetic gastroparesis (DGP) based on calcium-activated chloride channel. Methods: Forty Sprague-Dawley rats were randomly divided into four groups, including a normal control group (group A), a model group (group B), an EA group (group C) and a metoclopramide group (group D), with 10 rats in each group. A single intraperitoneal injection of 2% streptozotocin (STZ) combined with 8-week high-glucose high-fat diet was used to establish a DGP rat model. After intervention, gastrointestinal propulsive rate was observed; the expression level of transmembrane protein 16A (TMEM16A) was examined by immunohistochemistry; the Ca2+ concentration in interstitial cells of Cajal (ICCs) was detected by immunofluorescence; and whole-cell patch-clamp technique was applied to detect the current intensity of calcium-activated chloride channel (ICaCC) in ICCs in gastric antrum. Results: After modeling, the blood glucose levels in group B, group C and group D were significantly increased compared with group A (all P<0.01); after intervention, compared with group B, the blood glucose levels in group C and group D were significantly decreased (P<0.05, P<0.01); the intra-group comparison of blood glucose level between after modeling and after intervention found significant difference only in group C (P<0.01). The gastrointestinal propulsive rates in group B, group C and group D were significantly different from that in group A (P<0.01 or P<0.05); the gastrointestinal propulsive rates were markedly higher in group C and group D than in group B (P<0.01, P<0.01). The expressions of TMEM16A in group B and group C were decreased compared with group A (P<0.01, P<0.05); the expressions of TMEM16A in group C and group D were increased compared with group B (P<0.01, P<0.05). The fluorescence intensity of Ca2+ was significantly lower in group B than in group A (P<0.01); the fluorescence intensity of Ca2+ was significantly higher in group C and group D than in group B (P<0.01, P<0.05). ICaCC in ICCs in group B was significantly decreased compared with group A; ICaCC in group C and group D were increased compared with group B. Conclusion: EA at Zusanli (ST 36), Liangmen (ST 21) and Sanyinjiao (SP 6) can significantly improve gastrointestinal motility in DGP rats by up-regulating the ICaCC in ICCs.
10.Scanning electron microscopic study of titanium-hydroxyapatite, the functionally graded-material implanted in rabbits.

OBJECTIVEThe purpose of this study was to observe the tissue tolerance of Ti-HA functionally graded-material (FGM) and the form of the material-bone interface.
METHODSThe sintered Ti -HA FGM, pure HA and pure Ti were respectively implanted into the parietal bone of rabbits. The specimens were observed by SEM at 2, 4, 8 postoperative weeks.
RESULTSIn the early stage, the new bone surrounding the Ti -HA FGM formed earlier with larger amount and better maturity than the pure Ti. The condition was similar to the pure HA. Two months after the operation, direct bonding of material-bone interface was formed between the Ti -HA FGM and the new bone as an integral body. However, there was a little space left between the new bone and the pure Ti.
CONCLUSIONSThe Ti -HA FGM has good tissue tolerance. Its early integration with bone is similar to pure HA and better than pure Ti.
