AIM:To investigate the construction of oligonucleotide microarray specialized for pancreatic adenocarcinoma-associated genes and its preliminary application of detecting differential expressed genes in pancreatic cancer.METHODS:Pancreatic cancer related genes were purposely selected, and oligonucleotide microarray was prepared by spotting oligonucleotide probes on glass slides coated with APS-PDC.Labeled cDNA targets for hybridizations were synthesized by reverse transcription from total RNA in the presence of Cy5-dCTP and Cy3-dCTP,respectively.Hybridized microarray was scanned by Agilent laser scanner, and the aquired image was analyzed by Imagene3.0 software. The intensity ratios of Cy3 and Cy5 were calculated. To confirm the expression profiles of these genes, quantitative reverse transcription-PCR (QRT-PCR) was carried out for CDC25B and TUSC3 genes, and β-actin gene was taken as internal control. The product of PCR was quantitated by comparative Ct method.RESULTS:The signal of microarray hybridization was clear, and the images had a lower background and higher signal-noise ratio. In comparison with normal pancreas, twenty -four differentially expressed genes were identified which included seventeen up-regulated and seven down-regulated genes. The results of QRT-PCR demonstrated that the expressions of CDC25B and TUSC3 in pancreatic cancer were up-regulated and down-regulated respectively, which is consistent with microarray results.CONCLUSION:The oligonucleotide microarray specialized for pancreatic cancer is desirable for its specialty and sensitivity, which can simultaneously and parallelly detect multiple pancreatic cancer-associated genes.

Objective To investigate the feasibility and clinical application of prospective ECG-gated scan and adaptive cardio se-quence scan of dual-source CT coronary angiography. Methods 255 patients who underwent coronary CT angiography using a dual-source CT scanner were divided into two groups:group A underwent prospective scan, group B underwent retrospective scan. The two groups were further subdivided into low heart rate group and high heart rate group, recorded as A1(n=51),A2(n=89),B1(n=26), B2(n=89),respectively.Group A1 preceded with prospective ECG gated scan;group A2 performed with adaptive cardio sequence scan;group B1 and B2 underwent retrospective scan. All of them applied ATCM (automatic tube current modulation)technique. Effective dose was recorded. The image quality was assessed by two experienced radiologists with a four-point grading scale. Results The excellent or good image quality (score 1 and 2)of the four groups were above 95%,and no statistical differences in images scores was detected in A1 and B1 (P =0.726),A2 and B2 (P =0.079).Effective dose was significantly different in A1 and B1(P <0.001),A2 and B2 (P <0.001),and the former decreased 50% and 40% as compared with the latter. Conclusion Using prospective ECG gated scan and adaptive cardio sequence scan, assessable image quality of coronary artery can be acquired with dual source CT (heart rate<90 beast per minute),and radiation dose can be obviously reduced.

Objective To observe the changes of MEP(motor evoked pontential) and neurologic function after Erigeron beviscapus(EBHM) preconditioning on spinal cord ischemia-reperfusion injury in rabbits, and provide substructural experimental theory. Methods Twenty-four domestic rabbits were randomly divided into four groups with six rabbits each. In sham group (group A) only the abdominal aorta was exposed. In the blank group (group B), the abdominal aorta was occluded 40 minute followed by reperfusion. In the methylprednisolone group (group C), it was occluded 30 minute before the abdominal, methylprednisolone (30 mg/kg) was injected into the rabbit by vein, which was used for two days after the surgeon. In the EBHM group (group D), EBHM(50 mg/kg) was injected into the rabbit by vein, the following procedure was the same as group C. MEP was measured in each observed point and neurologic function was observed everyday for 7days.Results At every observed point after SCI, the MEP changed regularly, and compared with group B, there was significant difference in groups C and D(P<0.05). 7 days later, the neurologic function changed significantly in each group(P<0.05).Conclusion MEP monitoring can reflect the early change on spinal cord ischemia-reperfusion injury. EBHM preconditioning can protect the rabbit from spinal cord ischemia reperfusion injury.

Objective:To explore the regulatory effect of miR-873-5p micro-RNA targeting voltage-dependent anion channel protein 1 (VDAC1) in neurons and its mechanism. Methods:Murine nerve cells were randomly divided in vitro into a control group, a model group, a mimetic negative carrier (miR-con) group and an miR-873-5p group. The epileptiform hippocampal nerve cell model was induced in all of the cells except those in the control group using magnesium-free medium. The control group was normally cultured, while the miR-con and miR-873-5p groups were transfected with miR control and miR-873-5p RNA respectively. Real-time fluorescent quantitative polymerase chain reactions were used to detect the expression of miR-873-5p and VDAC1 mRNA. Western blotting was employed to detect VDAC1, B-cell lymphoma/leukemia-2 protein (Bcl-2), Bcl-2 associated X protein (Bax) and cleared caspase-3 in the neurons. The levels of reactive oxygen species (ROS), malondialdehyde (MDA) and glutathione (GSH) were measured using the DCFH-DA fluorescent probe, the thiobarbituric acid method and enzyme-linked immunosorbent assay respectively. Any apoptosis was detected using flow cytometry, while the targeting of miR-873-5p on VDAC1 was verified using the double fluorescence zymase reporter gene method. Results:Compared with the control group, a significant decrease in the average expression of miR-873-5p, Bcl-2 and in GSH and MDA levels was observed in the model group, but there was a significant increase in the average level of VDAC1, Bax, cleaved caspase-3 and ROS and in the rate of apoptosis. Compared with the miR-con group, a significant decrease in the average expression of Bax, cleaved caspase-3, ROS and in the apoptosis rate was observed in the miR-873-5p group, but there was a significant increase in the average level of Bcl-2, GSH and MDA. Moreover, it was verified that miR-873-5p reduced the expression of VDAC1.Conclusion:miR-873-5p protects damaged neurons by inhibiting their apoptosis through negatively regulating the target gene VDAC1 and the oxidative stress response.
