Objective To study the effects of PIAS3 knocking down on the proliferation, cell cycle and apoptosis of human prostate cancer cell line DU145 in vitro. Methods PIAS3 specific short hairpin RNA(shRNA) expressing plasmid was constructed and named pSilencer4.1/PIAS3.DU145 cells were transfected with pSilencer4.1/PIAS3.The proliferation of DU145 cells was analyzed by MTT assay.Cell cycle and apoptosis of DU145 cells were analyzed by flowcytometry. Results PIAS3 shRNA expressing plasmid was succefully constructed and then confirmed by sequencing.Expression of PIAS3 in DU145 was significantly reduced after pSilencer4.1/PIAS3 transfection.MTT assay showned accelerated proliferation after PIAS3 knocking down, and showned dose-effect curve.Flowcytometry showed cells in S phase increased, cells in G0/G1 decreased and percentage of apoptotic cells decreased after PIAS3 knocking down. Conclusion Knocking down of PIAS3 expression accelerates DU145 cell proliferation and inhibit cell apoptosis in vitro.

Objective To evaluate clinical remission in patients with small bowel Crohn's disease (SBCD) who have received infliximab(IFX) therapy and to evaluate capsule endoscopy combined with ileocolonoscopy for mucosal healing at 14th week of IFX therapy. Methods Clinical data of 23 SBCD patients who received IFX were retrospectively analyzed.Laboratory indices [routine blood tests,C-reactive protein (CRP)and albumin],Crohn's disease activity index (CDAI),Lewis score (LS),Crohn's disease simplified endoscopic score (SES-CD),side effects and complications were compared before IFX treatment and at 14th week of IFX therapy. Results In 23 SBCD patients, both CDAI and CRP levels significantly decreased (P<0.01) while body mass index (BMI) and albumin levels increased at 14th week (P<0.05),compared with those before treatment. The clinical remission rate at 14th week was 91.3% (21/23).There were 8/23 (34.8%)SBCD patients achieving mucosal healing in small bowel,12/21 (57.1%) in terminal ileum and colon, and 7/21 (33.3%) in both small bowel and colon.Twelve patients achieved both clinical remission and biochemical remission at 14th week and all of them achieved mucosal healing in both terminal ileum and colon (SES-CD ≤ 2).However,there were 5 (41.7%) of them still with small bowel inflammation (LS> 135).Conclusion IFX plays a role in promoting clinical remission and mucosal healing in SBCD patients.Mucosal healing of CD patients in terminal ileum and other parts of small intestine are not synchronized. For CD patients with small bowel and colon involved, the evaluation of the whole gastrointestinal tract by capsule endoscopy combined with ileocolonoscopy is recommended on condition that they have no intestinal obstruction or severe stricture.

Objective To investigate the relationship of fascin-1 protein expression with the metastasis of basal cell carcinoma and squamous cell carcinoma. Methods Skin specimens were obtained from 10 normal human controls, 13 patients with basal cell carcinoma (8 nodular variant and 5 superficial variant) and 24 patients with SCC (11 SCC in situ and 13 invasive SCC). Immunohistochemical staining was performed to analyze the expression of fascin-1 protein. The staining results were quantitatively assessed with computer image analysis system (Image-pro Plus 6.0). Results The optical density of fascin-1 averaged 0.1152±0.04574 in SCC in situ, 0.1257±0.03096 in invasive SCC, and 0.0293±0.00981 in normal controls; the expression of fascin-1 was significantly higher in SCC tissue than in normal control skin (both P<0.05). Increased optical density was also observed for fascin-1 in nodular variant of SCC (0.0808 ±0.05642) and superficial variant of SCC (0.0806±0.04346) compared with the normal controls, whereas no statistical difference was observed between nodular and superficial variant of SCC. Conclusion In BCC and SCC, there is an over expression of fascin-1, which may be linked to the local invasion of carcinoma,

Objective To prepare the phmaceutical reference materials of total lactones extract from ginkgo leaf for quantitative analysis. Methods A HPLC determination method was developed to investigate the uniformity and stability of the reference extract of ginkgo leaf total lactones using with bilobalide, ginkgolide A, ginkgolide B and ginkgolide C as the indexes.Three laboratories participated in the collaborative calibration test. Results The four components in reference extract had good uniformity and stability with RSD less than 2.0%;the marked values of the four components had been determined through statistical data analysis which provided by assigned traceability values. The values of bilobalide, ginkgolide A, ginkgolide B, and ginkgolide C were 39.54%, 29.03%, 15.96% and 11.69%, respectively. Conclusion The reference extract of ginkgo leaf total lactones can be prepared for quality control in future quantitative analysis.

Objective To investigate the relationship between findings of proton magnetic resonance spectroscopy(~1H-MRS)and tumor proliferative potential in patents with astrocytoma. Methods Thirty four patients of astrocytomas proved by surgery and pathology were examined by MRI and ~1H-MRS,among them 26 were of grade Ⅰ or Ⅱ(low grade)and 8 of grade Ⅲ or Ⅳ(high grade).Twenty one eases of astrocytomas(17 low-grade and 4 high grade respectively)were homogeneous type.Expression of proliferating cell nuclear antigen(PCNA)was detected by immunohistochemical method of streptavidin- biotin-peroxidase(SP).Results The Cho/NAA and Cho/Cr ratios of high-grade astrocytomas(M=4.895, 4.845 respectively)were significantly higher than those of low-grade astrocytomas(M=2.920, 2.000 respectively)in the 21 homogeneous astrocytomas(Z=-2.597,P=0.009;Z=-2.687,P= 0.007 respectively),and the ratios were found to be significantly correlated to the expression of PCNA(A= 7.880)(r=0.607,P=0.003;r=0.457,P=0.038 respectively). However, the differences of Cho/NAA、 Cho/Cr ratios between the high-grade and low-grade astrocytomas were not significant when 34 cases were included, and no relationship was found between Cho/NAA(M=3.965,2.890 respectively)、Cho/Cr(M= 3.080、1.960 respectively)ratios and the expression of PCNA(A=8.100)(r=0.060,P=0.738;r= 0.125,P=0.480 respectively). Conclusion By measuring the Cho/NAA and Cho/Cr ratios,~1 H-MRS may be a valuable method for presurgical in invo prediction of the malignant degree, tumor proliferative potential and prognosis of homogeneous-type astrocytomas.

The evaluation of permeability in biopharmaceutics classification system of Chinese materia medica (CMMBCS) requires multicomponent as a whole in order to conduct research, even in the study of a specific component, should also be put in the multicomponent environment. Based on this principle, the high content components in Gegen Qinlian decoction were used as multicomponent environmental impact factors in the experiment, and the relevant parameters of intestinal permeability about puerarin were measured with using in situ single-pass intestinal perfusion model, to investigate and evaluate the intestinal permeability of puerarin with other high content components. The experimental results showed that different proportions of baicalin, glycyrrhizic acid and berberine had certain influence on intestinal permeability of puerarin, and glycyrrhizic acid could significantly inhibit the intestinal absorption of puerarin, moreover, high concentration of berberine could promote the absorption of puerarin. The research results indicated that the important research ideas of permeability evaluation in biopharmaceutics classification system of Chinese materia medica with fully considering the effects of other ingredients in multicomponent environment.
