1.Osteogenic effects of concentrated growth factors applied in maxillary sinus floor elevation via a lateral window approach with simultaneous implant placement:study protocol for a single-center randomized controlled trial

BACKGROUND:Maxil ary sinus floor elevation via a lateral window approach is the most effective method of overcoming the shortage of bone mass deficiency in atrophic maxil ary posterior region. Bone transplantation is considered to be a prerequisite for the success of maxil ary sinus floor elevation. Platelet-rich plasma and platelet-rich fibrin have been used to accelerate bone formation, regeneration, and repair. However, few in-depth studies are reported on the effects of concentrated growth factors on new bone formation. OBJECTIVE:To investigate the effects of concentrated growth factors applied in maxil ary sinus floor elevation via a lateral window approach with simultaneous implant placement on repair of bone defects and new bone formation. METHODS:This was a randomized double-blind control ed trial, which was performed at the Hospital of Stomatology, Southwest Medical University, China. Forty patients presenting with loss of maxil ary molars and residual bone height of 2-5 mm in the posterior maxil ary region were included in this study. These patients were randomly assigned to either receive concentrated growth factors, hydroxyapatite and autogenous bone (experimental group, n=20) or hydroxyapatite and autogenous bone (control group, n=20) at bone defect sites. X-ray examination was performed 1, 3 and 6 months after surgery to evaluate bone regeneration and repair as per excel ent and good rate. The thickness of the cortical bone at the defect sites was measured using a dental cone beam CT scanner to reflect bone density. The study protocol had been approved by Ethics Committee of Hospital of Stomatology, Southwest Medical University, China, was performed in strict accordance with the Declaration of Helsinki, and had been registered with ClinicalTrial.gov (identifier NCT03046173). Written informed consent had been obtained from each patient prior to enrol ment. RESULTS AND CONCLUSION:This study was completed in 2016. Study outcomes were obtained as fol ows:X-ray images showed that 3 and 6 months after surgery, bone regeneration around the implant was better in the experimental group than in the control group (P<0.01) and better bone regeneration in the experimental group was observed at 6 months than at 3 months after surgery. At 1, 3 and 6 months after surgery, bone density at bone defect site was significantly greater in the experimental group than in the control group (P<0.01). These results suggest that concentrated growth factors applied in maxil ary sinus floor elevation via a lateral window approach with simultaneous implant placement can promote new bone formation and accelerate synosteosis.
3.Osteogenic ability study on co-culture of adipose-derived mesenchymal stem cells and bone marrow mesenchymal stem cells

Objective To assess the osteogenic ability after co-culture BMSC and ADSC in vivo and in vitro.Methods ADSC and BMSC were obtained by adherent screening method and enzymatic digestion method.Flow cytometry was used to confirm the phenotypes of ADSC and BMSC.Oil red O was used to induce MSC to fat.Alkaline phosphatase (ALP) and alizarin red staining were used in osteogenic group.This sample was divided into four groups,no-induced stem cells group;BMSC osteogenic induction group;ADSC osteogenic induction group;co-culture of BMSC and ADSC osteogenic induction group.ALP activities and Calcium absorbance were determined during different periods of osteogenic introduction.OCN and Runx2 expression level were tested via RT-PCR and western blot methods after osteogenic induction for 2 weeks.Furthermore,cells in each group were seeded on HA/CS/PLLA composite scaffolds,and the scaffolds with cells were planted into bone defects in rat models.The rats were sacrificed by overdose anesthesia at 8 weeks after surgery and the scaffolds were removed for further analysis.Results Oil red O staining demonstrated red after adipogenic induction.Alkaline phosphatase and Alizarin red staining showed flaky red under condition of osteogenic induction.There had no statistical change among each group after osteogenic induction for 3 days,and ALP activity significantly increased after osteogenic induction for 5 days.Meanwhile,the ALP activity in co-culture of BMSC and ADSC group was markedly higher than the other three groups.However,there had no significant change in A value of calcium absorbance among each group after osteogenic induction for 7 days,while it increased at 14th day and ALP activity in co-culture of BMSC and ADSC group was significantly higher than the other three groups.After osteogenic induction for 2 weeks,the mRNA expression of OCN and Runx2 in co-culture of BMSC and ADSC group was 78.24±8.11 and 1 180.13±121.16 respectively,and the protein expression of OCN and Runx2 was 6.54±0.59 and 4.43±0.51.These mRNA and protein expression level in co-culture of BMSC and ADSC group enhanced significant compared with the other 3 groups.Histological assay demonstrated that the new bone tissues formed in co-culture of BMSC and ADSC group were 497.75±7.44 μm2,which was larger than that in the other 3 groups at 8 weeks after implantation.Conclusion Co-culture BMSC and ADSC may up-regulated the osteogenic ability in vivo and in vitro.
4.Comparison of decellularizative procedures of bovine pericardium tissue-engineering heart valve scaffold

Objective Compared with different decellularization procedures for their potential of cell removal and the ability to preserve the matrix. Methods Specimens of bovine pericardiums were treated by 3 approaches (detergent and enzyme extraction、trypsin 、Triton-X 100 and sodium-deoxycholate). Tissue samples were then observed by HE staining and scanning electron microscopy to confirm the removal of cells. Von Gieson(VG) staining and Gomori staining were used for showing the integrity of collagen and elastin. DNA content was examined by the method of DNA extraction. Tissue shrinkage temperature and mechanical properties were also studied. Results Completely decellularization were achieved in 3 groups. While trypsin、Triton-X 100 and sodium-deoxycholate caused severe structural destruction and declined of mechanical properties of the matrix. In contrast, detergent and enzyme extraction achieved completely deeellularization and effectively preservation the matrix structure. Conclusion This research demonstrated detergent and enzyme extraction could achieve both complete decellularization and preservation of the matrix structure. This approach may provide an ideal platform for the construction of tissue-engineering heart valves.
5.Cloning and bioinformatic analysis and expression analysis of beta-glucuronidase in Scutellaria baicalensis.

The β-Glucuronidase gene (sbGUS) cDNA firstly from Scutellari abaicalensis leaf was cloned by RT-PCR, with GenBank accession number KR364726. The full length cDNA of sbGUS was 1 584 bp with an open reading frame (ORF), encoding an unstable protein with 527 amino acids. The bioinformatic analysis showed that the sbGUS encoding protein had isoelectric point (pI) of 5.55 and a calculated molecular weight about 58.724 8 kDa, with a transmembrane regions and signal peptide, had conserved domains of glycoside hydrolase super family and unintegrated trans-glycosidase catalytic structure. In the secondary structure, the percentage of alpha helix, extended strand, β-extended and random coil were 25.62%, 28.84%, 13.28% and 32.26%, respectively. The homologous analysis indicated the nucleotide sequence 98.93% similarity and the amino acid sequence 98.29% similarity with S. baicalensis (BAA97804.1), in the nine positions were different. The expression level of sGUS was the highest in root based on a real-time PCR analysis, followed by flower and stem, and the lowest was in stem. The results provide a foundation for exploring the molecular function of sbGUS involved in baicalcin biosynthesis based on synthetic biology approach in S. baicalensis plants.

6.Compilation of questionnaires of research motivation and related psychological factors for medical postgraduates

Objective To develop questionnaires to indentify research motivation and its related psychological factors for medical postgraduates.Methods The questionnaire was developed based on previous literature and case interview by using self-determination theory as the theoretical framework.Delphi method and stratified factor analysis were used to examine the reliability and validity of the questionnaire.Results The questionnaires included 2 sections:research motivation questionnaire and related psychological factors questionnaire.The Cranach's α of each questionnaire was 0.89 and 0.90 and test-retest reliability coefficient was 0.74 and 0.80.The content validity (CVI) of each questionnaire was 0.958 and 0.935,and stratified factor analysis demonstrated that the items of each dimension could account for more than 40% of the accumulated variance.Conclusion The questionnaires have good reliability and validity and could be applied.

Professor Luo Lingjie is a national famous old doctor of traditional Chinese medicine (TCM). He has a set of effective thoughts and methods on health maintenance. The core of the thought is Tiao-Xin, that is to say the emotion regulation is the key to health. The idea and method include three aspects, which include emotion regulation, diet regimen and exercise regimen. The emotion regulation is divided into clean ears, optimistic mood, learn to adapt, maintain psychological balance, and comply with nature. Diet regimen is fell into pay attention to the health food, regular diet, dietotherapy as primary and followed by drug, comply with the four seasons, reasonable to choose the food. There are four principles of exercise regimen, which are the choice of sports according to individual person with the aim to relax the body, move to raise the shape which should be combined with static to repose, proceed in an orderly way and persevere it step by step. The idea and method can also be used for daily personal health and clinical nursing for health maintenance. It is important that the physical and spiritual needs be consistent with long-term practice.
8.Relationship between plasma TRAIL before 20 weeks′gestation and pregnancy induced hypertension

Objective To assess the relationship between maternal plasma tumor necrosis factor-related apoptosis-inducing ligand ( TRAIL ) before 20 weeks′gestation and hypertensive disorder complicating pregnancy (HDCP);and to evaluate the predictive value of plasma TRAIL for HDCP.Methods A 2-phase screening/validation study was designed.In the screening phase , a nested, case-controlled study was performed , the plasma samples collected before 20 weeks′gestation from 20 women who later developed HDCP and 20 age-and gestation week-matched controls were tested in prospective screening test for protein expression profiling during pregnancy and HDCP.Plasma samples were analyzed by a human protein microarray technology designed to detect 507 proteins simultaneously.Differently expressed proteins′functional annotation and clustering were performed by using of Database for Annotation , Visualization and Integrated Discovery ( DAVID) and Gene Ontology ( GO) database.The TRAIL level of plasma samples obtained before 20 weeks′gestation from 53 women who later developed HDCP and 106 similarly matched controls were further validated by ELISA and 62 clinical risk factors were investigated.Logistic regression and ROC analysis were used to evaluate the relationship between TRAIL and HDCP and its predictive value for HDCP.Results In protein microarray analysis , 23 proteins expressed differently before 20 weeks′gestation between the two groups.Further validation results showed that TRAIL levels in HDCP patients were lower significantly (45.7 ±13.1) pg/ml than those in healthy pregnant controls (51.2 ±14.7)pg/ml, P=0.021.Multiple factor logistic regression analysis of 159 pregnancies showed that three features were finally entering the logistic model, they were:anemia (OR=4.87, 95% CI 1.05-24.26), pre-pregnancy BMI (OR=1.72, 95% CI 1.35 -2.19) and TRAIL (OR=0.96, 95% CI 0.92 -0.99).The predictive accuracy of logistic model was 81.8%.The model significantly increases the predictive value (AUC=0.81, 95%CI 0.73-0.87) compared to TRAIL as independent predictor (AUC=0.59, 95%CI 0.51-0.67).Conclusions Totally 23 proteins were expressed differentially before 20 weeks′gestation in plasma of women who later developed HDCP , confirming that HDCP is a heterogeneous disease with different biological changes.The data suggests that plasma TRAIL levels relate with the development of HDCP and its combination with pre-pregnancy BMI and anemia have a high predictive value for HDCP before 20 weeks′gestation.
9.Precolumn derivatization LC-MS/MS method for the determination and pharmacokinetic study of glucosamine in human plasma and urine

A selective precolumn derivatization liquid chromatography-tandem mass spectrometric (LC-MS/MS) method for the determination of glucosamine in human plasma and urine has been developed and validated.Glucosamine was derivatized by o-phthalaldehyde/3-mercaptopropionic acid.Chromatographic separation was performed on a Phenomenex ODS column (150 mm × 4.6mm,5μm) using linear gradient elution by a mobile phase consisting of methanol (A),and an aqueous solution containing 0.2% ammonium acetate and 0.1% formic acid (B) at a flow rate of 1 mL/min.Tolterodine tartrate was used as the internal standard (IS).With protein precipitation by acetonitrile and then the simple one-step derivatization,a sensitive bio-assay was achieved with the lower limit of quantitation (LLOQ) as low as 12 ng/mL for plasma.The standard addition calibration curves suitable for clinical sample analysis showed good linearity over the range of 0.012-8.27 μg/mL in plasma and 1.80-84.1 μg/mL in urine.The fully validated method has been successfully applied to a pharmacokinetic study of compound glucosamine sulfate dispersible tablets in health Chinese volunteers receiving single oral doses at 500,1000 and 1500 mg of glucosamine sulfate,as well as multiple oral doses of 500 mg t.i.d.for 7 consecutive days.
10.Effects of Selenium and Zinc on the Absorption, Excretion and Accumulation of Fluoride in Rats

ve To investigate the effects of selenium and zinc on the absorption, excretion and accumu-lation of fluoride in rats. Methods The contents of fluoride in serum, excrement, urine and bone were determined in Wistar rats drinking distilled water containing 100 mg/L NaF and orally perfused jointly with 0.1 mg/(kg? d) Na2SeO3 and/or 14.8 mg/(kg?d) ZnSO4 one time per two days continuously for 90 days. Results Na2SeO3 and/or ZnSO4 could increase the concentration of fluoride in urine, decrease the concentration of fluoride in serum and the content of fluoride in bone of rats. Exposure to ZnSO4 and joint exposure to Na2SeO3 and ZnSO4 could increase the content of fluoride in excrement. Conclusion ZnSO4 could inhibit the absorption of fluoride in intestine, Na2SeO3 and /or ZnSO4 could promote the excretion of urine fluoride and restrain the accumulation of fluoride in bone of rats.
