1.Role of IL-6 induced human peripheral blood monocytes tissue factor expression in the pathogenesis of chronic rejection

Objective To study the induction of IL-6 on expression and activity of tissue factor (TF)in peripheral blood monocytes(PBMCs).Methods rhIL-6 100 ng/L and rhIL-6 100 ng/L+ rhIL-6 MoAb 10?g/L were used respectively to stimulate the PBMCs for 24 h,PBMCs without any stimulation as control group,to study the changes of antigen content,mRNA expression and the ac- tivity of the TF.Results The antigen content,mRNA expression and activity of TF were increased significantly in 100 ng/L rhIL-6 group as compared with rhIL-6 100 ng/L+rhIL-6 MoAb 10?g/L and control groups(P<0.01).Conclusions rhIL-6 can induce the increase of antigen expression,activity and mRNA expression in PBMCs,and these effects can be suppressed successfully by rhIL-6 MoAb. This study demonstrated that there was potential relations between cytokines and thrombogenesis, which may play an important role the pathogenesis of chronic rejection.
2.Repair of ear deformity: a study of different methods

Objective To evaluate the surgical effects and influencing factors of ear malformation.Methods 408 cases were reviewed,including surgical methods and its effect in different condition of malformation,in which autogenous rib cartilage was used in reconstruction of ear.Meanwhile,349 cases(401 ears) double-lobe skin flap and 29 cases(29 ears) retroauricular groove flap were designed according to different condition of malformation.All the patients were treated with canalplasty,and 84 cases(84 ears) with mastoid process of pneumatic type were treated with tympanoplasty and canalplasty.Results months and more follow-up obtained in 335 cases (82%),and 6 months to 5 years follow-up in 273 cases(67%).Surgical effect was satisfactory,the shape of reconstructive ear was verisimilitude,and skull auricle angle was suitability;hearing was improved after operation in 335 ears,especially in 84 ears with canalplasty and tympanoplasty,but hearing was not improved after operation in 45 ears(
3.Experience in the teaching of child neurologic training doctors

The training doctors achieved good results in the aspects of medical skills, research ability and comprehensive professional quality.
4.Cloning and bioinformatic analysis and expression analysis of beta-glucuronidase in Scutellaria baicalensis.

The β-Glucuronidase gene (sbGUS) cDNA firstly from Scutellari abaicalensis leaf was cloned by RT-PCR, with GenBank accession number KR364726. The full length cDNA of sbGUS was 1 584 bp with an open reading frame (ORF), encoding an unstable protein with 527 amino acids. The bioinformatic analysis showed that the sbGUS encoding protein had isoelectric point (pI) of 5.55 and a calculated molecular weight about 58.724 8 kDa, with a transmembrane regions and signal peptide, had conserved domains of glycoside hydrolase super family and unintegrated trans-glycosidase catalytic structure. In the secondary structure, the percentage of alpha helix, extended strand, β-extended and random coil were 25.62%, 28.84%, 13.28% and 32.26%, respectively. The homologous analysis indicated the nucleotide sequence 98.93% similarity and the amino acid sequence 98.29% similarity with S. baicalensis (BAA97804.1), in the nine positions were different. The expression level of sGUS was the highest in root based on a real-time PCR analysis, followed by flower and stem, and the lowest was in stem. The results provide a foundation for exploring the molecular function of sbGUS involved in baicalcin biosynthesis based on synthetic biology approach in S. baicalensis plants.

5.Effects of grape seed proanthocyanidin extracts on advanced glycation end product receptor and connective tissue growth factor in the myocardium of diabetic rats

ObjectiveTo investigate the effects of grape seed proanthocyanidin extracts (GSPE) on advanced glycation end product receptor (RAGE),NF-Κb and connective tissue growth factor (CTGF) in the myoeardium of diabetic rats.MethodsTotal 30 streptozotocin (STZ) induced diabetic rats were randomly divided into 2 groups:diabetic group (DM1,n-15) and GSPE (250 mg/ kg,i.g) treated diabetic group (DM2,n= 15).Another two control groups:normal rats(C1,n= 10)and normal rats treated with GSPE (250 mg/kg,i.g) (C2 group,n= 10) were also observed.After 24 weeks,blood was collected to measure fasting plasma glucose (FBG) and RAGE.The protein expression of NF-Κb was determined in myocardial tissue by immunohistochemical staining and Western blot.The protein expressions of RAGE and CTGF were measured by Western blot.Results The levels of FBG and RAGE were significantly higher in diabetic rats than in control rats (P<0.05).After GSPE treatment,RAGE level significantly reduced (P<0.05),but FBG had no change in diabetic rats.The protein expressions of RAGE,NF-Κb and CTGF in the myocardial tissue of diabetic rats had marked increase compared with control rats (P< 0.05),however,their levels significantly reduced after GSPE treatment (P<0.05).ConclusionsGSPE may protect diabetic rats against cardiomyopathy,possibly by decreasing the protein expressions of RAGE,NF-Κb and CTGF.
6.Survey on the knowledge and willingness of genetic counseling and testing in blood relatives of breast cancer patients

Objective To investigate the knowledge and willingness of genetic counseling and testing in blood relatives of breast cancer patients.Methods A total of 922 blood relatives of breast cancer patients finished our questionnaire.Data were devided into different groups according to age,family history of tumor for statistical analysis.Results Most of the respondents were unaware of genetic counseling and genetic testing.However,after a brief introduction,major of them were willing to accept genetic counseling,breast cancer risk evaluation and screening.Specifically,79.8% of them were willing to accept genetic counseling,and 62.3% were willing to accept genetic testing.Most of the respondents would accept inexpensive early genetic screening.For the genetic testing with higher prices,only 37.9% of them would accept it.Supposing a positive genetic testing result,most of them were willing to perform prevention through close follow-upscreening,31.3% of them would choose prophylactic surgery or drugs.Despite being told the confidentiality of the test results,32.9% of them worried about the adverse effects of genetic test.Conclusions Most of the blood relatives of breast cancer patients were unaware of counseling and genetic testing,but had apparent willingness to accept them.Misunderstanding of genetic characteristics,costs and concerning about discrimination are obstacles for the respondents to accept genetic counseling,genetic testing and related screening prevention.
7.An analysis of hospitalization rates of patients with goiter in Lianyungang City Jiangsu Province in 2002-2010

Objective To observe the changes of hospitalization rates of patients with goiter in Lianyungang City in a coastal area with mild iodine deficiency in 2002-2010.Methods Medical reports of patients with goiter from county hospitals were studied at county level with a retrospective method.In accordance with the National Iodized Salt Monitoring Program,nine townships(towns) were taken from each county (district) [total of five counties(districts)]; four villages were selected from each township (town); salt samples were taken from eight households in each village,and salt iodine was determined using direct titration of sodium thiosulfate.Results The hospitalization rate of patient with goiter increased from 1.87/10 million in 2002 to 7.05/10 million in 2009,and then fluctuated to 6.06/10 million in 2010(x2 =281.91,P< 0.01).Among them,female's hospitalization rates were significantly higher than that of male 's(The variation range of x2 values were 35.23-116.04,all P < 0.01),and the hospitalization rates of people over the age of 40 years were significantly higher than that of people less than 40 years of age(The variation range of x2 values were 33.04-263.04,all P < 0.01),and the hospitalization rates of people in urban areas were significantly higher than that of people in rural areas (The variation range of x2 values were 18.35-140.00,all P < 0.01).Average salt iodine was 27.61-30.13 mg/kg in 2002-2010.The coverage rate of iodized salt and the consumption rate of qualified iodized salt both increased year by year (x2 =183.75,211.99,P < 0.01).There was a positive correlation between hospitalization rate of patient with goiter and coverage rate of iodized salt(spearman correlation coefficient was 0.83,P < 0.05).There was a positive correlation between hospitalization rate of patient with goiter and the consumption rate of qualified iodized salt too (spearman correlation coefficient was 0.93,P < 0.05).Conclusions In Lianyungang City,the hospitalization rate of patient with goiter,the coverage rate of iodized salt and the consumption rate of qualified iodized salt all show upward trend in 2002-2010.The relationship among them is worth further study.

9.The changes of cytokines in children with hand-foot-mouth disease

Objective To study the changes of cytokines in children with hand-foot and mouth disease.Methods Sixty-two cases of hand-foot-mouth disease admitted to our hospital from March 2011 to December 2011 were included in this study.Thirty-seven cases were male and 25 cases were female,the youngest one was four month old,and the oldest one was eleven month and 4 years old,the average age was (1.56 ± 0.98) years old.Sixty-two children with hand-foot-mouth disease were devided into ordinary group (n =20),severe group (n =20),critically ill group (n =22).Twenty healthy children served as control group.The plasma levels of pro-inflammatory cytokines [tumor necrosis factor(TNF)-α,interleukin(IL)-1β、IL-6] and anti-inflammatory cytokines (IL-1 0、IL-13、IL-4) were detected.Results The pro-inflammatory and anti-inflammatory cytokines of patients with hand-foot and mouth disease were increased compared with control group.The levels of TNF-α in severe and critically ill groups were (201.45 ± 133.42) ng/L,(220.37 ± 117.54)ng/L respectively,which significantly increased compared with ordinary group [(118.12 ±11205) ng/L] (P <0.05).The levels of IL-1β and IL-6 in critically ill cases were (651.77 ±538.45) ng/L and (88.02 ±50.76) ng/L respectively,which significantly increased compared with severe cases [(551.75 ± 252.75) ng/L,(59.25 ± 28.80) ng/L] and ordinary cases [(270.18 ± 249.79) ng/L,(55.76 ± 28.68) ng/L] (P < 0.05).The level of IL-10 was (137.53 ± 72.07) ng/L in ordinary cases,which increased significantly compared with the severe and critically ill cases[(57.07 ±55.02) ng/L,(73.37 ±56.01) ng/L] (P <0.05).The levels of IL-13 were (507.53 ±210.86) ng/L and (489.67 ± 168.99) ng/L in severe and critically ill cases respectively,which significantly increased compared with ordinary cases [(387.16± 157.43) ng/L] (P <0.05).The levels of IL-4 showed no significant differences among the ordinary,severe and critically ill groups(P > 0.05).Conclusion The pro-and anti-inflammatory cytokines were involved in inflammatory response simultaneously in patients with hand-foot and mouth disease.The increases of TNF-α,IL-1β and IL-6 are consistent with severity of the disease,while factor IL-10,IL-13 are decreased as the severity of the disease.The increase of the pro-inflammatory cytokines and decrease of the anti-inflammatory cytokines are the important sign of the exacerbation.
10.Auto-induction of PcoI-PcoR Quorum-sensing System in Pseudomonas fluorescens 2P24

PcoI-PcoR is a quorum-sensing (QS) system influencing the biofilm formation and rhizosphere colonization in Pseudomonas fluorescens 2P24. The expression of the pcoI, a N-acyl-homoserne lactone (AHL) biosynthase gene, is under the regulation of a number of chromosomal factors, such as the GacS-GacA two-component system. In this paper, we investigated the upstream regulators that influence the transcription of pcoI gene using a chromosomal pcoI-lacZ fusion reporter strain PM203. Cosmids containing genomic DNA of the wild-type strain 2P24 were introduced into the reporter strain PM203 (gacA—, pcoI-lacZ) to screen positive transcriptional regulators of pcoI gene. One of them named pP32-24, which contained a 5-kb Pst I functional fragment was selected. Further analysis identified that the pcoI was the gene responsible for the increase of the pcoI-lacZ expression. The expression of pcoI-lacZ reporter was alsoimproved in both PM101 (pcoI-lacZ) and its gacAmutant PM203 after addition of exogenous AHL, indicating that the expression of pcoI is positively regulated by AHL (autoinduction) in strain 2P24. In addition, deletion mutagenesis and complementation experiments demonstrated that the transcriptional regulator PcoR positively controlled the expression of pcoI and the formation of biofilm. These results suggest that, in strain 2P24, the expression of PcoI-PcoR QS system is auto-inducted, and the transcriptional factor PcoR is involved in the regulation of pcoI transcription and the biofilm formation.
