1.Correlation analysis of Filifactor alocis detection with periodontal status.

OBJECTIVEThe study investigated the epidemiology of Filifactor alocis (F. alocis) in subgingival plaque samples from subjects with different periodontal statuses. The relationship between the prevalence of F. alocis and clinical periodontal parameters was also analyzed.
METHODSSubgingival plaque samples and periodontal data were collected from 68 healthy sites (H groups) in 17 healthy subjects, 64 healthy (G-H group) and 76 diseased sites (G-D group) in 19 patients with chronic gingivitis, and 36 healthy (P-H group) and 56 diseased sites (P-D group) in 14 patients with chronic periodontitis. The plaque samples were analyzed by polymerase chain reaction, and possible correlations between the F. alocis detection rate and the bleeding index, probing depth, or clinical attachment level were determined.
RESULTSThe detection levels of F. alocis increased in both healthy and diseased groups. The lowest level at 30.88% (21/68) was noted in the H group, whereas the highest level at 91.07% (51/56) was obtained from the P-D group. A significant correlation was found between the F. alocis detection levels and periodontal disease condition (P < 0.000 1). Further analyses showed that a significant correlation also existed between the detection level of F. alocis and the abnormal clinical periodontal parameters, namely, bleeding index, probing depth, and clinical attachment loss. The odds ratios were 5.26, 8.85, and 11.65, respectively.
CONCLUSIONF. alocis was found at increased-levels in subjects with periodontal disease. The presence of F. alocis increases the risk of sites with abnormal clinical periodontal parameters.

3.Effect of ligustrazine on cerebral injury in LPS-induced septic shock rats

AIM: To investigate the effect of ligustrazine (Lig) on cerebral injury in LPS-induced septic shock rats and to explore the underlying mechanism.METHODS: Wistar rats (n =48) were randomly divided into control group, LPS group and LPS +Lig treatment group.The rats in LPS group were randomly divided into 2 subgroups at time points of 6 h and 12 h.After ligustrazine treatment, the venous blood was collected by removal of eyeballs to detect the con-centration of neuron-specific enolase (NSE) using ELISA.The nitric oxide (NO) concentration in the homogenate of brain tissues was examined.The apoptosis in the hippocampus was analyzed by TUNEL staining.The protein expression of Bax and Bcl-2 was determined by Western blot.RESULTS: Ligustrazine inhibited the elevation of NSE and NO concentrations in LPS-induced septic shock rats.Furthermore, ligustrazine administration also attenuated LPS-induced increase in Bax ex-pression and decrease in Bcl-2 expression.CONCLUSION: Ligustrazine decreases the concentration of NSE and NO, and attenuates cerebral injury in LPS-induced septic shock rats.These effects may be related to the regulation of Bax and Bcl-2 expression.
4.Effect of glutaredoxin on oxidative stress of umbilical vein endothelial cell exposed to Porphyromonas gingivalis lipo- polysaccharide.

OBJECTIVEThis study measures the glutaredoxin (Grx) gene and protein expression in umbilical vein endothelial cells upon exposure to Porphyromonas gingivalis (P. gingivalis) lipopolysaccharide (LPS). The involvement of the Akt-signaling pathway is also determined.
METHODSEA-hy926 cells were pretreated with 1,000 ng · mL⁻¹ P. gingivalis LPS for 4, 12, 18, and 24 h, and then real-time reverse transcription polymerase chain reaction was employed to detect Grx1 expression. The effect of Grx on Akt activity was investigated using Western blot for the control, LPS (1,000 ng · mL⁻¹ LPS), and carmus- tine (BCNU) groups (1,000 ng · mL⁻¹ LPS, and the EA-hy926 cells were pretreated with 25 μmol · ml⁻¹ BCNU for 30 min).
RESULTSGene expression of Grx1 significantly increased in LPS group compared with that in the control group. The Grx1 expression reached the peak level in 12 h, and the variation between the expression in 4 and 12 h was significant (P < 0.05). After 12 h, the protein levels of Grx and phosphorylated-Akt (p-Akt) significantly increased in the LPS group (P < 0.05), whereas the BCNU group showed a considerable decrease in both Grx and p-Akt expression levels (P < 0.05). Moreover, a slight difference was observed in the total Akt protein levels in the three groups (P > 0.05).
CONCLUSIONGrx expression increased upon exposure of EA-hy926 cells to the LPS. Akt activity could be inhibited by BCNU (a Grx inhibitor), which indicated that Akt might act as a downstream regulator of Grx.

7.The relationship between DKK1 protein expression and lymph node invasion, and prognosis in patients with intrahepatic cholangiocarcinoma

Objective The objectives of this study were to investigate the relationship between DKK1 protein expression and lymph node invasion,and prognosis in patients with intrahepatic cholangiocarcinoma.Methods From January 2005 to December 2012,78 patients with intrahepatic cholangiocarcinoma were treated with endoscopic resection.Tissue microarray of intrahepatic cholangiocarcinoma and adjacent tissues were done and biochemical indexes were measured before operation.The patients were recruited every 3 months after operation.The Kaplan-Meier method was used to analyze the recurrence-free survival curve and the overall survival curve.The Cox proportional hazards regression model was used to analyze the multivariate analysis and to determine the factors that affect the disease-free survival(RFS)and overall survival(OS).Results There were significant differences in the expression of GGT,hepatic portal lymph node invasion,Child-Pugh grade,MMP9 and DKK1 protein in the negative and positive expression of intrahepatic cholangiocarcinoma(P < 0.05).The positive expression rate of DKK1 protein in intrahepatic cholangiocarcinoma was 35.90% (28/78),and the positive expression rate of DKK1 protein in hilar cholangiocarcinoma was 14.10% (11/78).RFS and OS in 78 cases of intrahepatic cholangiocarcinoma were 51.28% (40/78) and 50.00% (38/78),41.03 % (32/78) and 38.46% (30/ 78),25.64% (20/78) and 23.08% (18/78) after surgery for 1,3 and 5 years,respectively.The results from univariate and multivariate analysis showed that GGT,CA19-9,CEA,tumor size,DKK1 and hilar lymph node invasion were the prognostic factors of OS in patients with intrahepatic cholangiocarcinoma.CEA,tumor size,DKK1 and hilar lymph node involvement were prognostic factors for RFS in patients with intrahepatic cholangiocarcinoma.DKK1 protein expression positive and negative OS curve or RFS curve showed that 5 years after intrahepatic cholangiocarcinoma patients DKK1 protein expression positive and negative OS were 28.20% and 20.51%;RFS were 24.36% and 21.79%.Conclusion The expression of DKK1 protein is closely related to lymph node invasion in patients with intrahepatic cholangiocarcinoma.The expression of DKK1 protein is the prognostic factor of OS and RFS in patients with intrahepatic cholangiocarcinoma.
8.A single-center epidemiological and clinical retrospective study of 8 037 patients with sleep disorders

Objective To assess the epidemiology and clinical characteristics of sleep disorders in a single center in northwest China.Methods Using the International Classification of Sleep Disorders, 3rd Edition, all consecutive patients which were suspected as sleep disorders in Tangdu Hospital between January 2007 and December 2016, were included retrospectively.Results The average age of 8 037 patients was (46.59±15.83) years with male-female ratio 1∶1.24.Chronic insomnia was found in 3 848 (47.9%) patients, obstructive sleep apnea was found in 2 648 (32.9%) patients, narcolepsy was found in 294 (3.7%) patients, Kleine-Levin syndrome was found in 11 (0.1%) patients, circadian rhythm sleep-wake disorders were found in 14 (0.2%) patients, rapid eye movement behavior disorder was found in 193 (2.4%) patients, restless legs syndrome was found in 139 (1.7%) patients, periodic limb movement disorder was found in 109 (1.4%) patients, and other possible sleep disorders were found in 478 (5.9%) patients, respectively.Chronic insomnia and obstructive sleep apnea combided with somatic diseases.Conclusions Patients diagnosed by polysomnography in our single center suggested consultation rate of sleep disorders was increasing in past ten years, of which chronic insomnia and obstructive sleep apnea were dominant and comorbided with somatic diseases.
9.CT and MRI features analysis of pineocytoma: Report of 6 cases

Objective To improve diagnosis accuracy of pineocytoma (PC) by joint analysis of CT,MRI imaging features and differential diagnosis with other lesions in pineal region.Methods Totally 6 pineocytoma patients confirmed surgically and pathologically had their clinical history,CT and MRI data collected and analyzed on lesion morphology,cystic solid changes,existence of necrosis,complications of hemorrhage and or calcification,MRI and enhanced scan of solid component,complications with hydrocephalus and etc.Results Plain scan found 1 case of solid nodule and 5 cases of cystic-solid nodules,2 cases with clearly-bordered lesions and 4 one not as well as 4 cases with significant hydrocephalus and 2 ones with light hydrocephalus.Enhanced scan showed 5 cases of moderate to marked enhancement and one case with no obvious enhancement.CT examination proved there were 1 case of calcification and 1 case of hemorrhage.Conclusion Pineocytoma has the characteristics of benign tumor,and has to be differentiated with other tumors frequently occurring in this region in case of obvious clinical signs due to crushing brain parenchyma or blocking aqueduct cerebri by oversized lesions.
10.Decreased expression of Foxp3 in patients with allergic rhinitis

OBJECTIVE To evaluate the role of forkhead transcription factor p3(Foxp3) in the pathogenesis of allergic rhinitis(AR).METHODS Nasal tissues and peripheral blood mononuclear cells(PBMCs) were obtained from 17 patients with AR and 11 controls.Foxp3 was detected in nasal tissues by immunohistochemisry and real-time reverse transcription polymerase chain reaction(RT-PCR).Foxp3 and CD4+CD25+T cells were evaluated in PBMCs by using ? ow cytometry.RESULTS The numbers of Foxp3+ cells was(44.2?20.5)cells/mm2 and(129.5? 35.6)cells/mm2 in nasal mucosa of two groups(P
