2.Protective effect of anthocyanidin extracts on light-induced retinal functional and structural damage in rat

Background Light leads to the damage of retinal function and structure by promoting the reproduction of radicals and lipid peroxides when retina is exposed to an intense light environment for a long time.It is necessary to study how to protect the retina against light-induced injury in ophthalmology.Objective The aim of this study was to evaluate the effect of anthocyanidin extracts in preventing retinal photic damage.Methods Eighteen clean male Sprague-Dawley (SD) rats were randomly assigned to normal saline group,anthocyanidin group and mixed (hydrogen-rich saline (HRS) + anthocyanidin,1:1 in volume) group and 6 rats for each.The electroretinogram (ERG) with international standardized 5 items was recorded from all the rats before experiment.Normal saline,anthocyanidin extracts or mixed solution of 5 ml/kg were intraperitoneally injected in the three groups,respectively,for consecutive 5 days.Then the diffused light with the luminance intensity of (5000±300)lx was used to irradiate the right eyes of the rats for consecutive 3 hours during 19:00 through 7:00 in a device made by our laboratory,and the left eyes were covered at the same time.The ERG was repeatedly recorded 5 days after light irradiation.The rats were sacrificed at the end of the experiment and retinal sections were prepared for the histopathological examination.The functional and structural changes of retinas were compared among the different groups.The use of the rats followed the Statement of ARVO.Results The differences of rat body weight were not statistically significant among the three groups (before experiment:F =0.472,P =0.841 ; after experiment:F =0.658,P=0.762).No any significant difference was found in scotopic 0.01 ERG b wave,scotopic 3.0 ERG a and b waves,scotopic 3.0 oscillatory potentials,photopic 3.0 ERG b wave and 3.0 flicker P1wave between the left eyes and the right eyes in the three groups before experiment (P>0.05).The amplitudes of various waves of ERG were significantly declined in the right eyes compared with the left eyes (P<0.05).The mean differential values of scotopic 0.01 ERG b wave,scotopic 3.0 ERG a and b waves,scotopic 3.0 oscillatory potentials were significantly different(F =4.594,P=0.029; F=3.834,P=0.037; F=12.823,P=0.000; F=3.976,P=0.032),but those in photopic 3.0 ERG b wave were not statistically significant (F =1.488,P =0.259).Compared with the normal saline group,the differential values of scotopic 0.01 ERG b wave,scotopic 3.0 ERG a and b waves,scotopic 3.0 oscillatory potentials were all reduced in the anthocyanidin group and mixed group (P < 0.05).The cells of outer nuclear layer were decreased in the right eyes in the three groups compared with the left eyes,especially around the optic nerve head and the upside of the retina,with significant differences between them (P<0.05),and those in the anthocyanidin group and mixed group were significantly less than normal saline group (P<0.05).Conclusions Anthocyanidin has a protective effect on light-induced retinal damage of SD rats.The protective effect of anthocyanidin extracts is similar to the integrated effect of the mixed group.
3.Study on the optimal condition for culturing human stromal cells derived from umbilical cord blood

Objective To investigate the optimal condition for culturing human umbilical cord blood-derived stromal cells(hUCBDSCs),and to observe their biological behaviors.Methods The umbilical cord blood was obtained from the Department of Obstetrics of the authors' hospital.The influence on the growth of hUCBDSCs was determined by the isolation method,the medium and the time of renewal of first medium were analyzed.The status of cell growth was observed under inverted microscope and the morphological characters were studied with the cells stained by Wright's staining.The hUCBDSCs were identified by cytochemistry and immunocytochemistry methods.Results The gelatin precipitation was better than other techniques for isolation.The optimal time for first medium renewal was the fourth day of culturing,and the improved Dexter-type cultural system was better than the classical Dexter-type cultural system in primary culture.The colonies of adherent cells began to form in 9-14 days(with a median of 12.1 days),and the number of colonies reached it maximum in 15-21 days(mean 19.4 days).On day 28,adherent cells spread all over the bottom of dish.On day 28 of culturing,these cells were found under light microscopy to have differentiated into three kinds of cells: fibroblast-like cells,macrophage-like cells and small-round cells.Cytochemistry assay revealed that the positive rate reached 100% with non-specific esterase(NSE) and saccharogen(PAS) staining.26% of the hUCBDSCs were positive with alkaline phosphatase(ALP) staining,but negative with peroxydase(POX) staining.Immunocytochemistry staining revealed that the positive rates of hUCBDSCs for CD31,CD68 and Fn were 96% and 95%,and 94% respectively,and for CD45 was 0%.Conclusion The hUCBDSCs could be successfully cultured in vitro,and it sets a foundation for further study on the clinical application of hUCBDSCs.
5.Association between plasma homocysteine and cystathionine β synthase gene T833C polymorphism in Xinjiang Kazakh population with essential hypertension

Objective To investigate the correlation between the plasm homocysteine (Hcy) levels and cystathionine β synthase (CBS) T833C gene polymorphism in Xinjiang Kazakh with essential hypertension. Methods 239 Kazak patients with hypertension (hypertension group) and 206 with normotensive (control group) were selected for the study. Amplification Refractory Mutation System (ARMS) was used to analyze the polymorphism of CBS gene T833C, TT, TC and CC genotypes and the various sites of T, C allele frequencies in research group and the control group. Results Plasma of Hcy level was higher in the hypertensive group than those of control group, the difference was significant (P<0.05), and the individual plasma of Hcy with TC gene was higher than that with TT gene. The C allele frequencies was significantly higher in EH group than that in controls in Xinjiang Kazakh population, and the difference was significant (P<0.05). The risk of EH group in individuals carrying TC genetype was 2.39 times higher than in individuals carrying TT genetype (OR = 2.39, 95%CI:1.125 ~5.076, P = 0.02). Conclusion Elevated Hcy level may be a risk factor of Kazakhs hypertension in Xinjiang. The Cystathionine β synthase gene of T833C polymorphism may be associated with essential hypertension in Kazak people in Xinjiang.
6.Determination of Three Pyrethroids in Soil by Matrix Solid Phase Dispersion Extraction_Dispersed Liquid Phase Microextraction_Gas Chromatography Mass Spectrometry

A matrix solid phase dispersion extraction_dispersed liquid phase microextraction_gas chromatography_mass spectrometric method has been developed for the determination of three pyrethroids ( tetramethrin, permethrin and deltamethrin) in soil. The optimal conditions for the analysis were as follows. About 0. 5 g soil and 1. 5 g C18 solid phase extraction powder were grinding for 5 min. The mixture was eluted with 10 mL of acetone. The eluent was concentrated to 0. 4 mL, and then mixed with 20 μL of tetrachloromethane and 5. 0 mL of ultrapure water to form a homogeneous cloudy solution. The emulsion was broken by centrifugation. About 1 μL of sediment was injected and analyzed directly by GC_MS. Good linearities for three pyrethroids were ranged from 5 to 200 μg/kg (r2≥0. 9989), and recoveries at three spiked levels were ranged from 86 . 5% to 108 . 0% with RSDs less than 7 . 8%. The LODs of three pyrethroids were 1. 00-1. 48 μg/kg. This method can meet the determination of trace pyrethroids in soil.
7.Investigation of nutrition status and quality of life of perioperative patients with laryngeal cancer

Objective To investigate the nutrition status and quality of life(QOL)in the perioperative patients with laryngeal cancer and to analyze the effect of nutrition status on quality of life.Methods Sixty-six patients with laryngeal cancer were surveyed respectively by self-designed general situation questionnaire,nutritional risk srcreening 2002(NRS2002)and European organization for research and treatment of cancer quality of life core questionnaire(EORTC QLQ-C30).The impact of nutritional status on QOL was analyzed by the multiple linear regression analysis.Results The total score of nutritional risk of perioperative patients with laryngeal cancer was 3.53±1.51 and the incidence of nutritional risk was 28.79%.The nutrition status influenced on QOL, physiological function,cognitive function and pain area(P<0.05).Conclusion Nurses should assess the nutritional status of the patients and correct malnutrition without delay in order to improve the physical function and QOL of the patients.
8.Effect of regional gene therapy with bone morphogenetic protein 7 producing mesenchymal stem cells on spinal fusion in rats

Objective To evaluate effectiveness of ex vivo adenoviral gene transfer creating human bone morphogenetic protein 7-producing mesenchymal stem cells (Ad-hBMP-7 cells) compounded with nano-hydroxyapatite/collagen(NHAC) composite to induce spinal fusion in a rat posterolateral spine fusion model. Methods Mesenchymal stem cells were cultured and transduced by Ad-hBMP-7. Then Ad-hBMP-7 cells were cocultured with NHAC. Totally 56 Wistar rats were divided into four groups. Intertransverse spinal arthrodesis was attempted in four groups of Wistar rats with autogenous bone (group A); NHAC(group B); NHAC and BMSCs (group C); Ad-hBMP-7 cells and NHAC (group D). Each specimen underwent plain radiography, manual palpation and histological analysis. Results All spines in group D had fusion. In contrast, none of the spines in group A and group B had fusion. Three of ten rats in group C achieved fusion at the 12th week post-operatively.Conclusion BMP-7-producing mesenchymal stem cells compounded with NHAC composite may induce intertransverse fusion in the rat spine model.
