The assurance mode from laser source for medicine is brought forward based on the three-tier mode of the whole assurance system in equipment of scientific research that is in the measurement of quality control personnel, engineering technique personnel and operator. The successful implementation of model will do laboratory equipment for the conditions of security work to a new level in our academy that can play important protection effect in overall research tasks.
2.Sulfuric Acid Modified Poly (GMA-EGDMA) Coating for Solid Phase Microextraction Coupled with High Performance Liquid Chromatography and Its Application to Determination of Four Pharmaceuticals and Personal Care Products in Water

A poly( GMA-EGDMA) coated SPME fiber was prepared using an in-situ polymerization by direct bonding to the surface of a polydopamine-modified stainless steel wire. Then the fiber was modified by sulfuric acid. A novel solid phase microextraction coating coupled to high performance liquid chromatography ( HPLC) method based on the as-prepared fiber was developed for the determination of four pharmaceuticals and personal care products ( PPCPs) in water samples. The influences of extraction parameters, including pH, extraction time, extraction temperature and salt addition were investigated. 3 mL water sample was extracted by the as-prepared fiber for 60 min at 30 ℃, and then desorbed with mobile phase for 30 min, respectively. Desorption solution was analyzed by HPLC-DAD ( diode array detection ) . The results indicated that the extraction yield of the fiber was good for four PPCPs. The linear correlation coefficients were>0. 997 with the linear range of 2-200 μg/L. The limits of detection (S/N=3) were 0. 5-5 μg/L with RSD (n=5) of 4. 1%-11. 9%. The recoveries of four PPCPs at spiked level of 20, 50, 100 μg/L were within the range of 70. 6%-105. 5%. The results showed that this method was easy, green, accurate and precise, and could be used to assay the four PPCPs in real water samples.
3.Effects of kaempferol on learning and memory in rats with chronic cerebral ischemia and possible mechanisms

Aim To investigate the effects and possible mechanisms of kaemperol in the rats with chronic cere-bral ischemia.Methods Chronic cerebral hypoperfu-sion model was produced by permanent occlusion of bi-lateral common carotid arteries (2VO)in rats.After KAE treatment,the rats underwent Morris water maze and prehensile traction test.Neuronal morphology was observed using Nissl and HE staining.The activity of SOD and the content of MDA in brain tissue were de-termined.The DJ-1 protein expression was assayed by Western blot.Results Compared with 2VO model group,KAE significantly improved learning and memo-ry and the grasping ability.In addition,KAE signifi-cantly reduced brain tissue pathological injury induced by 2VO. Furthermore, KAE significantly increased SOD activity and enhanced antioxidant protein DJ-1 ex-pression in brain tissue.Conclusions KAE could sig-nificantly attenuate the cognitive impairment,limb bal-ance dysfunction and pathological injury in rats with chronic cerebral ischemia.The mechanism may be re-lated to improving the antioxidant system in vivo.
4.Study of attribution of multicomponent original medicinal materials in gegen qinlian decoction with intestinal permeability.

The complex level of constructing biopharmaceutics classification system of Chinese materia medica CMMBCS) was the study of traditional Chinese compound, on the premise of insisting that the multicomponent simultaneous determination, when carrying out the study of intestinal permeability, the primary task was to define the source of the components that was absorbed through the intestinal wall, namely, which medicinal material the components belonged to in traditional Chinese compound. The technology of chemical fingerprint and in vitro everted gut sac model were used in this research to make multicomponent an intuitive source attribution which permeated the intestine in the classic formula Gegen Qinlian decoction, and to lay the foundation for the further qualitative and quantitative research of intestinal permeability.

5.MRI Features of Benign Cerebellar Astrocytomas in Children

Objective To study the MR imaging characteristics of benign cerebellar astrocytomas(BCAs) in children. Methods The clinical and MR imaging data of twelve patients with BCAs were reviewed and analyzed retrospectively.Results Of 12 tumors, 7 cases were located in the cerebellar hemisphere, 4 in the vermis, and 1 in the IV ventricle. All children were in company with hydrocephalus rated as extensive (n=9) or moderate (n=3). The tumors ranged in diameter from 23 mm to 68 mm, mean diameter 42 mm. Solid tumors were found in 5 children, and cystic areas occurred in 7 of 12 patients.BCAs were hypointense on T 1-weighted in 10 cases, and hyperintense on T 2WI in 9 cases compared with that of normal brain parenchyma. After contrast injection, BCAs marked (n=2), moderate (n=5), or absent (n=5) enhancement. Hemorrhage and calcification were not common (n=1,0, respectively).Conclusion BCAs has relative characteristic features on MRI in children, and MRI plays an important role in diagnosing and evaluating these tumors preoperatively.
6.Feasibility of proton magnetic resonance spectroscopy of the myocardium in vivo.

OBJECTIVETo investigate the feasibility of proton magnetic resonance spectroscopy ((1)H-MRS) of the myocardium in vivo.
METHODSA total of 100 volunteers were examined with 1H-MRS scan using a 1.5T MR scanner, and the magnetic resonance spectra were analyzed.
RESULTSAll the volunteers completed the examinations, and the spectral peaks were identified with the chemical shifts of water (4.7 ppm), choline (3.2 ppm), creatine (3.0 ppm), and triglyceride (0.9-1.6 ppm), whose presentation ratios were 100%, 86%, 90%, and 98%, respectively. In 86 cases the spectra showed all the chemical shifts. The myocardial metabolite contents were expressed as the ratio of metabolites to water (%), and the contents of choline, creatine, and triglyceride were 1.94%, 1.95%, and 9.41%, respectively.
CONCLUSION(1)H-MRS can be used in detecting the metabolic products of myocardium in vivo.

7.Establishment and application of the molecular-beacon-based asymmetric recombinase amplification for detecting Staphylococcus aureus

Objective To establish a homothermal and fast detecting method on pathogenic bacteria by combining recombinase-aid amplification (RAA) with molecular beacon.Methods The establishment of the methodology.Staphylococcus aureus specific primers were designed from the relative region of the staphylococcal protein A (SPA).Asymmetry amplification was optimized by adjusting the primer concentration ratios.The results of amplification and hybridization were visualized and analyzed by agarose gel electrophoresis and fluorescence detection.The sensitivity was identified by detecting dilute positive plasmids.And the specificity was determined using RAA method by detecting 72 pathogenic bacteria,including Staphylococcus aureus and other Staphylococcus spp.from the Department of Clinical Laboratory of Daping Hospital in December 2016.Besides,the Kappa analysis and the clinical diagnosis efficiency were investigated by analyzing 39 extra strains in the laboratory in December 2016.Results When the concentration ratio of restrictive and non-restrictive primer was 1:20,the yield efficiency of single-stranded DNA (ssDNA) reached the peak.And as for the hybridization efficiency,the asymmetry amplification was higher than symmetry amplification.Twenty copies/μl was proposed as the limits of detection by testing dilute plasmids.And the RAA hybridization method could distinguish Staphylococcus aureus with other Staphylococcus spp.Comparing with traditional detection methods with a Kappa index of 0.860,this method shows a good consistency.By analyzing the 111 bacteria,the sensitivity of the method is 92.5% (37/40),the specificity is 97.2% (69/71),the positive predictive value is 94.9% (37/39),the negative predictive value is 95.8% (69/72),the positive likelihood radio is 33.04,the negative likelihood radio is 0.077,the Youden index is 0.897 and the Kappa index is 0.902.Conclusion Through the combination of asymmetry recombinase-aid amplification optimization and molecular beacon probe,a new method of detecting bacteria DNA with RAA hybridization technique is established,providing the foundation for its clinical application.
8.Rapid detection of beta-thalassemia by LDR-ULP combined with real-time PCR

Objective To establish a new method for rapid detection of β-thalassemia by investigating six clinical common mutation types.Methods Fifty cases of clinical wild-type samples and 42 cases ofβ-thalassemia samples were collected, and β-globin gene was amplified by PCR.Uniform ligation probe ( ULP) specific probes were designed for hybridization reaction to increase the reaction specificity and real-time PCR was performed to increase the sensitivity.After that, PCR products were verified by agarose electrophoresis.After examining the specificity and sensitivity, Kappa test between LDR-ULP method and reverse dot blot( RDB) method was conducted.Results Hybridization efficiency was improved 2.53 times by LDR-ULP hybridization.Each mutant type showed a significant amplification curve, whereas the wild-type had no significant curve within 40 cycles.The limit of determination of this method was 5 pg.The results of 92 cases of peripheral blood samples detected by the method of LDR-ULP and RDB were completely consistent.Conclusion In this study, a simple, inexpensive, rapid new method to detect β-thalassemia were established.
9.Therapeutic effect of brain protein hydrolysate and simvastatin on carotid atherosclerotic plaques in CI patients and its influence on LVEF and CRP level

Objective : To explore clinical value of brain protein hydrolysate (BPH ) and simvastatin in treatment of patients with cerebral infarction (CI) complicated carotid atherosclerotic plaques (CAP).Methods : A total of 90 CI + CAP patients treated in our hospital from Mar 2016 to May 2017 were selected ,randomly and equally divided into routine treatment group and combined treatment group (received simvastatin + BPH based on routine treat-ment) ,both groups were treated for four weeks- Therapeutic effect ,United States National Institutes of Health Stroke Score (NIHSS) ,CAP-related indexes ,hemorheology indexes ,LVEF and CRP before and after treatment and incidence of adverse events were compared between two groups .Results : Compared with routine treatment group after treatment ,there were significant reductions in NIHSS score [ (15-56 ± 6-33) scores vs.(12-14 ± 3-18) scores] ,CAP size [ (1-54 ± 0-32) cm2 vs.(1-34 ± 0-30) cm2 ] ,intima-media thickness [ (1-37 ± 0-08) mm vs. (1-15 ± 0-04) mm] ( P<0-01 all) ,and significant rise in total effective rate (80-00% vs.95-56%, P=0-024) in combined treatment group ; there were significant reductions in hematocrit [ (44-24 ± 3-24 )% vs.(37-72 ± 2-41)%] ,platelet aggregation rate [ (45-62 ± 15-95)% vs.(38-10 ± 2-52)%] ,fibrinogen [ (4-06 ± 1-43) g/L vs. (3-18 ± 1-15) g/L] ,whole blood viscosity [ (2-66 ± 0-77) mPa·s vs.(1-55 ± 0-25) mPa·s] ,plasma viscosity [ (1-45 ± 0-11) mPa·s vs.(1-21 ± 0-16) mPa·s] and serum CRP level [ (4-36 ± 1-03) mg/L vs.(1-70 ± 0-20) mg/L] ,and significant rise in LVEF [ (49-30 ± 3-74)% vs.(56-67 ± 4-37)%] in combined treatment group , P<0-01 all- Incidence rate of cardio-and cerebrovascular events in combined treatment group was significantly lower than that of routine treatment group (4-44% vs.17-18%, P= 0-004 ).Conclusion : Simvastatin combined BPH based on routine treatment can achieve better total therapeutic effect and reduce incidence rate of complications in CI + CAP patients .
10.Impacts of different transurethral prostatic resection procedures on male sexual function: meta-analysis of randomized controlled trials.

OBJECTIVETo compare the impacts of transurethral resection of the prostate (TURP), transurethral electrovaporization of the prostate (TUEVP) and holmium laser enucleation of the prostate (HoLEP) in the treatment of benign prostatic hyperplasia (BPH) on male sexual function.
METHODSWe identified randomized controlled trials on the influence of TURP, TUEVP and HoLEP on the sexual function of BPH patients, and performed meta-analysis on the included data using Revman 5.0.25.
RESULTSNine randomized controlled trials involving 1 050 BPH patients were included in the meta-analysis. The baseline of the study was comparable. TURP affected erectile function less than TUEVP (P = 0.04), but the two had no significant difference in their influence on ejaculatory function. Nor was any significant difference found between HoLEP and TURP in their influence on either erectile or ejaculatory function at 12 and 24 months after surgery.
CONCLUSIONTUEVP induces a higher incidence of erectile dysfunction than TURP, but its influence on ejaculatory function is not significantly different from the latter. HoLEP and TURP have no significant difference in their influence on erectile function and ejaculatory function.
