1.Construction of Recombinant Mice Factor Ⅶ-pPIC9K Vector and Expression in Pichia

Objective:To construct the yeast expressive vector of rmFⅦ, in which mFⅦ was mutated to inhibit coagulation without affecting the affinity for TF, and express it in Pichia pastoris. Methods:The full length cDNA encoding mFⅦ was amplified from a mouse liver by RT-PCR method, site-direct mutated and restriction enzyme digested as design. Cloning into pPIC9K, electroporation of Gs115, in vivo screen of multiple inserts by G418 resistance, BMGY/BMMY are used for induction and expression of rmFⅦ in pichia pastoris. These proteins were also screened for functional activity. Results: Three different rmFⅦ-pPIC9K yeast expression vectors and it's aim protein were obtained,two kinds of proteins were found to be functional active as design. Conclusion:rmFⅦ protein can be expressed in pichia pastoris and it might facilitate the development of tumor-target molecule, and novel anti-agiogenesis drug study.
2.Effect of water temperature, stocking density and feeding cycle on growth of juvenile Poecilobdella manillensis.

The effect of water temperature, stocking density and feeding cycle on the growth of Poecilobdella manillensis juvenile was conducted P. manillensis was conducted respectively under different conditions: water temperatures(18, 22, 26, 30,34, 38 degrees C and CT), stocking density (75, 125, 200, 275, 350 individual/L) and feeding cycle(2, 4, 6, 8, 12, 16 d). After 30 days, survival rate, weight gain rate, specific growth rate were measured. There was a significant correlation between water temperature and specific growth rate (γ = -0.066x2 + 3.543 1x -38.09, R2 = 0.837 9). Based on the regression equation, the specific growth rate of P. manillensis achieved the maximum (9.461 4) at 26.84 degrees C. And the most optimal water temperature was 26-30 degrees C. Meanwhile, the survival rates of P. manillensis was 0 at 38 degrees C in 3 d. There was significant negative correlation between density and specific growth rate (γ = -0.005 7x + 9.197 3, R2 = 0.998 3) and between feeding cycle and specific growth rate (γ = -0.468 2x + 10.574, R2 = 0.998 8).

3.Effects of HMME-SDT on hypertrophic scar of rabbit ear

Objective To observe the clinical efficacy of HMME-SDT therapy for the treatment of hypertrophic scar (HS) of rabbit ear.Methods 60 white rabbits were randomly divided into five groups.The model group and HMME-SDT treatment group were used to establish the models of hypertrophic scar in ears.Results The effect of HMME-SDT on the fibroblastic density in the hypertrophic scarring models was observed in rabbit ears.The HMME-SDT could lower the fibroblastic density,compared with the model group,with significant difference (P< 0.05).The effect of HMME-SDT on the collagen area density was noted in the hypertrophic scarring models in rabbit ears.The HMME-SDT could lower the collagen area density,compared with the model group,with significant difference from the fourth week of the epithelialization (P<0.01).Conclusions HMME is an effective sonosensitizer.HMME-SDT can significantly inhibit hypertrophic scar of rabbit ear.
4.A new lanostane-type triterpenoid from Cymbopogon citratus.

To study the chemical constituents of Cymbopogon citratus, isolation and purification of constituents were carried out on silica gel, Sephadex LH-20 and prepatative HPLC. The structures of the compounds were identified by physicchemical properties and spectral data analysis. Eight compounds were isolated and identified as 3beta-methoxy lanosta-9(11)-en-27-ol (1), 3beta-hydroxylanosta-9 (11)-en (2), (24S) -3beta-methoxylanosta-9(11), 25-dien-24-ol (3), 8-hydroxyl-neo-menthol (4), (2E)-3,7-dimethyl-2,7-octadiene-1, 6-diol (5), (+)-citronellol (6), 7-hydroxymenthol (7) and ethyl nonadecanoate(8). Compounds 1 is a new one. Compounds 2-3 are obtained from C. citratus for the first time.

5.Comparative study of barium enema, computed tomography and magnetic resonance imaging in preoperative diagnosis of colorectal carcinoma.

OBJECTIVETo compare the diagnostic value of barium enema (BE), computed tomography (CT) and magnetic resonance imaging(MRI) in primary colorectal carcinoma.
METHODSA total of 64 patients with suspected colorectal carcinoma received BE (n=39), spiral CT (n=31) and MRI (n=42). The detective results were compared with the surgical results.
RESULTSAmong 64 patients, 54 cases were pathologically proved as colorectal carcinoma. The diagnostic sensitivity of BE,CT and MRI was 96.9% ,96.2% and 97.1% ,and the overall accuracy was 92.3% 83.9 % and 90.5% respectively. The overall accuracy of CT and MRI for tumor T staging was 73.1% and 82.9% respectively.
CONCLUSIONBE can be considered as a primary approach for diagnosing colorectal carcinoma, CT and MRI be necessary diagnostic approaches. Combined BE with MRI is the best choice for diagnosing of colorectal carcinoma.

6.Three-year efficacy and side effect of adefovir dipivoxil for the treatment of the old patients with chronic hepatitis B virus infection.

OBJECTIVETo investigate 3-year antiviral efficacy and side effect of adefovir dipivoxil (ADV) on the old patients with hepatitis B chronic infection.
METHODS31 HBeAg-negative chronic hepatitis B virus infected old patients (include 8 patients with chronic hepatitis B and 23 patients with liver cirrhosis) with serum HBV DNA levels > 1000 copies/ml, and ALT > 2 times the upper limit of normal, without company with other liver diseases, cancer, renal dysfunction, and autoimmune disease. All the patients were treated with ADV orally (10 mg once daily) for 36 months. HBV DNA and biochemical and blood routine indexes were checked after treated.
RESULTSerum total bilirubin, direct bilirubin, alamine aminotransferase, aspartate aminotransferase and load of HBV DNA decrease significantly after therapy (P < 0. 001). Other biochemical indexs and blood routine are no significant changes (P > 0.05).
CONCLUSIONThe way to treat with ADV is safe and effective for old patients with chronic hepatitis B virus infection.

7.Endorectal ultrasound in evaluation on mesorectal fascia invasion in preoperative rectal cancer

Objective To evaluate the value of endorectal ultrasonography (ERUS) in assessment of mesorectal fascia (MRF) invasion in rectal cancer.Methods Data of 44 patients who accepted preoperative ERUS and total mesorectal excision surgery within a week were retrospective analyzed.There were 18 patients who accepted preoperative neoadjuvant chemotherapy and 26 patients didn't acceped.Taking the pathological diagnosis of circumferential resection margin (CRM) as the gold standard,the diagnostic efficiency of ERUS for the MRF invasion in rectal cancer was evaluated.Results The final pathological T staging was T1 in 2 cases,T2 in 17 cases and T3 in 25 cases.There were 2 cases of CRM positive results,and 42 cases of CRM negative results.With regard to the location of tumor,there were 16 cases located in low,and 28 cases in mid rectum.There were 26 cases located in anterior or antero-lateral wall of rectum,13 cases in posterior or postero-lateral wall,and 5 cases with a circle of rectum.The diagnostic accuracy were 83.33 % (15/18) and 92.31% (24/26) for cases of accepting and not accepting the preoperative neoadjuvant chemotherapy;80.77% (21/26) for cases located in anterior or antero-lateral wall,and 100% (13/13) for cases located in posterior or postero-lateral wall;75.00% (12/16)and 96.43 % (27/28) for low position and mid position tumors.The total diagnostic accuracy was 88.64% (39/44).Conclusion ERUS can be an effective method in preoperative assessment of the MRF invasion in rectal cancer.
8.A case-control study on the influencing factors to mild cognitive impairment among the community-based elderly population

Objective To explore the influencing factors on mild cognitive impairment among the community-based elderly population.Methods A‘n:m'matched case-control study was conducted to analyze the risk factors.COx regression model of survival analysis was selected to deal with non-geometric proportional matched data which was difficult to analyze by logistic regression modeI.Results Four hundred and twenty-three cases together with nine hundred and twenty-five controls were interviewed with an uniformed questionnaire.Through univariate and multivariate cox regression analysis,the odds ratio and 95%CJ of these risk factors appeared to be:physieal labor as 1.396(1.092-1.785);smoking as 1.551 (1.021-2.359);higher level of blood glucose as 1.354(1.102-1.664);HDL-C in the serum as 1.543 (1.232-1.932);LDL-C in the serum as 1.299(1.060-1.592);lower level of estrogen in the serum as 1.263(1.031-1.547);hypertension as 1.967(1.438-2.689);diabete:1.381(1.139-1.675);depressive disorder:1.406(1.110-1.780);cerebral thrombosis as 1.593(1.307-1.943);higher SBP as 1.331(1.129-1.569)and ApoEe4 carrier as 1.462(1.140-1.873)respectively.Odds ratio and 95%CI on protection factors appeared to be:reading newspaper frequently as 0.610(0.503-0.740);frequently doing housework as 0.804(0.665-0.973);frequently engaging in social activities as 0.617(0.502-0.757);reemployment after formal retirement as 0.759(0.636-0.906);having acumen olfaction as 0.900(0.845-0.958);having extrovert personality as 0.829(0.699-0.984);being decisive as 0.811(0.662-0.993).Conclusion The major measures to prevent MCI seemed to be including the following factors as:being intellectuals,engaging in healthy life style and decreasing the risk in developing hypertension,diabetes,depressive disorder and cerebrovascular disease.However.olfactory hypoesthesia,cowardice and haring introvert character,ApoEε4 carrier etc could be treated as early indications to signify MCI.
9.Study on hearing impairment among elderly population in the community of Taiyuan city, Shanxi province

Objective To study the characteristics and influencing factors on hearing impairment among elderly population in the community of Taiyuan city. Methods 384 ageing people above 60 years old were selected from Chaoyang and Guandi community in Taiyuan city by multi-stage sampling. Data on influencing factors of hearing impairment were collected by questionnaire. 5 ml fasting blood samples were drawn to detect the level of glucose, triglyceride and cholesterin in the blood samples. All the objects were tested with binaural hearing. The level of binaural hearing threshold at 0.5 kHz, 1 kHz, 2 kHz, 3 kHz, 4 kHz, 8 kHz were measured by GVSLN-TC-GK2000 hearing-assistant evaluative apparatus. The level of 3 kHz, 4 kHz, average hearing threshold from ear with better audition was chosen as dependent variable. Socio-demographic data, environmental factors and biochemical indicator were chosen as independent variables, t test, ANOVA and accumulative logistic regression were performed to analyze the influencing factors on hearing impairment by software SPSS 13.0. Results The prevalence of hearing impairment among elderly population was 90.9%. The hearing disorder was 78.6% with 1.3% of them using hearing-assistant apparatus. Results from single factor analysis showed that the average levels of 3 kHz, 4 kHz, 8 kHz hearing thresholds were significantly different among elderly with different age, sex, education background and the levels of glucose and cholesterin (P<0.01). Results of accumulative logistics regression showed that except glucosein which was the only one included in regression model of lower median frequency group, all the others were included in regression model of frequency group. Being male, older age and with higher level of glucose and cholesterin in blood were risk factors causing hearing impairment. Higher education level seemed to be a preventive factor. Conclusion Hearing impairment appeared in higher prevalence among the elderly population, suggesting that proper measures should be taken. It is beneficial for abating hearing impairment to decrease the level of glucose and cholesterin in blood.
10.Analysis on the spatial clustering of tuberculosis based on provincial level in China from 2008 to 2010.

OBJECTIVETo study the tuberculosis clustering areas and the changing trend, from 2008 to 2010, so as to provider the reference for tuberculosis control.
METHODSGlobal spatial autocorrelation and SaTScan methods were used to detect and analyse the spatial clustering of total tuberculosis notification rate and the new smear-positive pulmonary tuberculosis notification rate, at the provincial level from 2008 to 2010.
RESULTSThe spatial clustering (SC) phenomenon was significant on total notification rate and new smear-positive pulmonary tuberculosis notification rate from 2008 to 2010 (P < 0.01). The coverages of clustering areas on total notification rate showed a reduction from 19 provinces to 14 provinces, distributed in the south, west and north-east areas of China. The coverages of clustering areas on new smear-positive pulmonary tuberculosis notification rate concentrated in 14 provinces which covered the south and north-east of China.
CONCLUSIONThe disease burden and the risk of transmission in the clustering areas of tuberculosis both located in the south and the north-east of China. The disease burden of tuberculosis was high in the west of China, but not the areas with high risk of transmission.
