The Journal of Practical Medicine 2014;(17):2748-2750
Objective To compare the value of liquid-based preparations versus conventional smears in the diagnosis of non-thyroid neck masses from fine needle aspiration. Methods The two different methods were compared by the smear quality and the diagnostic accuracy for the aspiration samples of non-thyroid neck masses of 400 patients. Results The unsatisfactory rate for LBP was lower than that for CS (13.0% vs. 16.0%, P < 0.05). The sensitivity was 94.6% for LBP and 92.9% for CS, and the accuracy was 96.8% for LBP and 95.0% for CS. In the aspect of smear quality, LBP had advantages of quantity of cells, with clear background and easily identifiable cell morphology. Conclusions LBP has more advantages than CS in the identification of non-thyroid neck masses, especially for cervical nodal enlargement. But it has no marked advantages in the diagnosis of salivary gland lesions.

ObjectiveTo explore the effects of short-term high-fat diet on expression of peroxisome proliferator-activated receptor-γ-coactivator 1α (PGC1α) in hypothalamus of rats. Methods18 male SD rats were randomly divided into 2 groups: the normal control group (NC) and the high-fat group (HF). After two-week adaptive feeding, the 2 groups were given control diet and high-fat diet respectively for 3 days. Body weight and energy intake were recorded every day. Random blood glucose of caudal vein was measured before and after intervention. The mRNA and protein levels of PGC1α, UCP2 and CPT1a in hypothalamus were detected by quantitative RT-PCR and Western Blot. ResultsAfter the three-day treatment, the energy intake in HF group was significantly increased compared with that in NC group (75.7 ±5. 02 vs 60.4 ±4.53 kcal/d, P <0.05). However, the body weight, food intake and blood glucose level were not significantly changed. The mRNA level of PGC1α was significantly decreased (P <0.05 ), while no significant difference was observed in protein levels. Conclusion Short-term high-fat diet downregu]ates the expression of PGC1α mRNA in hypothalamus of rats.

BACKGROUND:The mechanism and effect of glycogen synthase kinase 3β(GSK-3β) in the differentiation of cardiac stem cel s into cardiomyocytes are stil unclear, although GSK-3βis closely related to the life activities of cel s.
OBJECTIVE:To investigate the changes of GSK-3βexpression in the treatment of myocardial infarction in rats undergoing cardiac stem cel transplantation.
METHODS:The isolation and culture of cardiac stem cel s were performed in 10 neonatal rats. Lentivirus overexpressing GSK-3βor LacZ (control) was constructed and transferred into cardiac stem cel s. Animal model of myocardial infarction was made in 30 Sprague-Dawley rats. Six weeks after model preparation, rat models were assigned into GSK-3β, LacZ or PBS group. GSK-3βor LacZ overexpressing cardiac stem cel solution or PBS in equal volume was injected into the rat myocardium, respectively. Four weeks after transplantation, the cardiac function and myocardial col agen production in rats were detected and compared.
RESULTS AND CONCLUSION:Compared with the other two groups, the left ventricular ejection fraction was significantly higher, and the left ventricular end diastolic diameter was significantly lower in the GSK-3βgroup (P<0.05). Hydroxyproline content, type I col agen mRNA, and type III col agen mRNA expression were significantly lower in the GSK-3βgroup than the other two groups (P<0.05). Findings from Masson staining showed that the content of blue-stained col agen was significantly lower in the GSK-3βgroup than the LacZ group. Moreover, lowest myocardial infarction size was found in the GSK-3βgroup (P<0.05). Al these experimental findings show that GSK-3 overexpression plays a positive role in promoting the therapeutic effect of cardiac stem cel transplantation.

Objective To investigate the effects of hairpin RNA on the expression of human papillomavirus E6 gene. Methods Plasmids expressing hairpin RNA, sense RNA and anti-sense RNA were constructed. The recombinants were transfected into cervical cancer cell line, CaSKi. Expression of E6 was detected with semi-quantitative RT-PCR and Western blot. Results Hairpin RNA could significantly reduce the expressions of E6 mRNA and protein, which was more efficient than traditional anti-sense RNA did. Conclusion The RNA interference through hairpin RNA can effectively inhibit the expression of target gene, and can be potentially useful in gene therapy of HPV related tumors.

AIM: To evaluate the genotype, muscle histopathology and ultrastructure in dko mice. METHODS: Dystrophin/Utrophin-deficient double knockouts (dko) mice were obtained from university of Oxford, UK. Genotype of filial generation of heterozygote was evaluated by PCR-SSP. HE staining and fluorescent immunohistochemistry by SABC-Cy3 were used to detect striated muscle of dko mouse, and the muscle ultrastructure was observed by transmission electron microscope(TEM). RESULTS: In 112 filial generation mice, there were 28 mdx (25.0%), 26 dko (23.2%) and 58 heterozygote (51.8%), which coincided with the law of Mendelian inheritance. HE staining showed that the myocytes were not very uniform, there were phenomenon of round outline, centrally nucleated fibers, widening interspace, inflammatory cell infiltration and connective tissue proliferation in dko mice. There were no any immunofluorescent expression of dystrophin and utrophin in sarcolemma in dko mice. TEM showed sarcolemma breakage, separation and edema, and loose myofibril texture, inflammatory cell infiltration and connective tissue proliferation in dko mice. CONCLUSION: PCR-SSP is a very quick and accurate way for genotype evaluation of filial generation. The pathophysiology of dko mouse was very similar to Duchenne muscular dystrophy (DMD), and dko mouse is an ideal animal model for study of DMD clinical therapy.
