1.Cloning and bioinformatics analysis of gene F protein binding protein 1 of hepatitis C virus

Objective To clone and bioinformatically analyze the full-length sequence of F protein binding protein 1 (FBP1). Methods Yeast two-hybrid system was employed to obtain an unknown gene and named it as FBP1.The coding sequence of FBP1 was cloned using molecular biological techniques. Results The coding sequence of FBP1 was cloned successfully. Conclusion FBP1, a cellular protein binding to F protein of HCV, plays an important role in the interaction of virus protein and host cell protein.
2.Correlation analysis of Filifactor alocis detection with periodontal status.

OBJECTIVEThe study investigated the epidemiology of Filifactor alocis (F. alocis) in subgingival plaque samples from subjects with different periodontal statuses. The relationship between the prevalence of F. alocis and clinical periodontal parameters was also analyzed.
METHODSSubgingival plaque samples and periodontal data were collected from 68 healthy sites (H groups) in 17 healthy subjects, 64 healthy (G-H group) and 76 diseased sites (G-D group) in 19 patients with chronic gingivitis, and 36 healthy (P-H group) and 56 diseased sites (P-D group) in 14 patients with chronic periodontitis. The plaque samples were analyzed by polymerase chain reaction, and possible correlations between the F. alocis detection rate and the bleeding index, probing depth, or clinical attachment level were determined.
RESULTSThe detection levels of F. alocis increased in both healthy and diseased groups. The lowest level at 30.88% (21/68) was noted in the H group, whereas the highest level at 91.07% (51/56) was obtained from the P-D group. A significant correlation was found between the F. alocis detection levels and periodontal disease condition (P < 0.000 1). Further analyses showed that a significant correlation also existed between the detection level of F. alocis and the abnormal clinical periodontal parameters, namely, bleeding index, probing depth, and clinical attachment loss. The odds ratios were 5.26, 8.85, and 11.65, respectively.
CONCLUSIONF. alocis was found at increased-levels in subjects with periodontal disease. The presence of F. alocis increases the risk of sites with abnormal clinical periodontal parameters.

3.Protective effects of ginseng root saponins on immunity in heat-stressed mice

, 15min),the clearance of reticuloendothelial system (RES) of mice on iv charcoal particles were decreased apparently; serum hemolysin concentration were diminished notably; delayed type hypersensitivity (DTH) reaction induced by sheep red blood cell (SRBC) were inhibited obviously. Ginseng root saponins (GRS) 50, 100 mg ??? kg-1ip at 15min before the heat-stressprotected the immunity of mice from inhibitory effects induced by the heat-stressor. The suppression of the clearance of RES on charcoal particles,serum hemolysin concentration as well as DTH reaction induced by SRBC were all abolished by GRS.
4.Biophysical characteristics of skin in mild to moderate acne

Objective To assess the biophysical characteristics of normal skin and inflammatory skin lesions in mild and moderate acne. Methods Seventy-five mild and moderate acne vulgaris patients were included in the study. One inflammatory lesion measuring 2.0 to 5.0 mm in diameter which occurred within 48 hours prior to the measurement was selected as the target lesion. Trans-epidermis water loss (TEWL), capacitance and a* value were measured in target lesions and lesion-adjacent normal skin. Sebum content immediately,1 and 4 (saturated) hours after face washing was also determined on target lesions and central forehead between the eyebrows. Sebum secretion rate (SSR) was calculated. Results The TEWL and SSR significantly increased in the lesion-adjacent normal skin with the increment of inflammatory lesion number (both P < 0.05), and increased in target lesions with the elevation of a* value (both P < 0.05). The target lesions exhibited a significantly higher TEWL but a lower saturated sebum content and SSR than the adjacent normal skin did (all P <0.05). Conclusions The severity of inflammatory lesions is correlated with the impaired barrier function and increased SSR in facial skin. Compared with the adjacent normal skin, inflammatory skin lesions have a reduced skin barrier function and SSR.
5.Relationship between family intergenerational contact, depressive symptoms and self-reported health status in the elderly

Objective:
To explore the relationship between family intergenerational contact, depressive symptoms and self-rated health status in the elderly, so as to provide insights into promoting health of the elderly and actively coping with population aging.
Methods:
Demographic information, intergenerational contact frequency, self-rated health status and depressive symptoms in the elderly at ages of 65 years and older were collected through the Chinese Longitudinal Healthy Longevity Survey (CLHLS). Structural equation model was established to analyze the effects of intergenerational contact on self-rated health and the mediating role of depressive symptoms between family intergenerational contact and self-rated health in the elderly.
Results:
Totally 15 497 elderly participants were enrolled, with 6 727 males (43.41%) and 8 770 females (56.59%), and there 12 583 individuals lived with family members (81.19%). The median age was 85.00 (interquartile range, 16.00) years. The scores of self-rated health and depressive symptoms were (3.39±0.87) and (2.22±0.56), respectively. The results of structural equation model analysis showed that after adjusting for age, gender and residence, frequent intergenerational contact had a direct positive effect on self-rated health (effect size=0.038, P<0.001), and could improve the depressive symptoms (effect size=0.089, P<0.001) in the elderly. In addition, depressive symptoms had a negative effect on self-rated health in the elderly (effect size=-0.422, P<0.001).
Conclusions
Frequent intergenerational contact can improve the self-rated health status in the elderly, and indirectly improve the self-rated health level by alleviating depressive symptoms.
6.Predictive value of combined preoperative fibrinogen and neutrophil to lymphocyte ratio for the prognosis of pancreatic cancer patients after radical surgery

Objective:To evaluate the prediction value of preoperative Fibrinogen(FIB) in combination with neutrophil-lymphocyte ratio(NLR)for the prognosis of operable pancreatic cancer patients.Methods:The clinical data of 124 patients who underwent radical resection for pancreatic cancer in our hospital from Jan 2010 to Dec 2018 were retrospectively analyzed.The patients were divided into three groups according the high and low NLR, FIB value which defined by the receiver operating characteristic curve (ROC). The clinicopathological data and overall median survival time were compared between the three groups.Results:Univarate analysis showed that age, tumor stage, NLR, FIB and F-NLR score were associated with the prognosis( P<0.05), while multivariate analysis showed that high F-NLR score was the independent prognostic factor. The median survival time of patients with F-NLR scores 0, 1 and 2 group was 30.6, 20.3 and 13.9 months( P<0.05). The prognosis of high F-NLR score was significantly worse than that of low F-NLR score( P<0.05). Conclusions:A high preoperative F-NLR score was a promising predictor for the prognosis of pancreatic cancer patients after radical resection.

8.The mechanisms of protective effects of ginseng root saponins on immunity in heat-stressed mice

The peripheral blood T- lymphocyte percentage, lympocyte percentage in white blood cell (WBC) of mice in heat environment (45C, 15 min) were diminished, and serum corticosterone increased. Ginseng root saponins (GRS) 50, 100 mg/kg were administered ip at 15 min before the heat-stress, the suppression of peripheral blood T-lymphocyte percentage were prevented, but could not inhibit the increase ofserum corticosterone. GRS 50mg?kg-1ip could inhibit the redution of peripheral lymphocyte percentage. GRS 50mg?kg-1 ,reserpine 0.5mg? kg-1or physostigmine salicylate 0.3 mg?kg-1ip abolished the inhibiting effect of heat-stress on DTH reaction in mice.
9.Clinical analysis of systematic 12 + 1-core biopsy of prostate cancer detected by transrectal ultrasound resonance guided prostate system

Objective To compare the dijference of prostate cancer detection rate (PCDR) between 12 + 1-core biopsy and 6-core biopsy of the prostate system guided by transrectal ultrasonography (TRUS).Methods The clinical data of 2 707 patients with prostate biopsy from July 1999 to June 2012 were retrospectively analyzed.These patients were 54 to 92 years old,mean age was 69 years old.The range of PSA was 0.02-158.56 ng/ml,with an average of 16.97 ng/mt.People in the range of PSA 0-4.00,4.01-10.00,10.01-20.00,20.01-30.00,and > 30.00 were 161,826,827,312,581,respectively.The volume of the prostate ranged from 14.1 to 82.6 cm3,mean 47.9 cm3.The 1 603 cases before the July 2009 were performed 6-core biopsy guided by the finger,followed by 1104 TRUS-guided 12 + 1-core biopsy.In addition,after March 2012,60 patients accepeted the MRI examination before prostate biopsy.The strategy of prostate biopsy was medial 6-core,lateral 6-core,the 13th core was positioned at abnormal signal area of TRUS and MRI.Explore the difference of PCDR medial 6-core,lateral 6-core,systematic 12-and 12 + 1-core,and the difference of 13th core and the other 12 cores,with the cut off value of PSA quartile of 30 ng/ml and PV quartile of 46 cm3.Results Comparison of clinical characteristics of prostate biopsy between positive group and negative group was performed,and the result suggested that The positive outcome of prostate cancer biopsy was related with element such as high PSA,old age [(71.7 ±7.1)vs.(68.3 ± 8.1),P =0.008],large fPSA [(8.5 ± 36.4) vs.(2.3 ± 3.4),P < 0.001],small prostate volume [(41.3 ±22.9) vs.(52.3 ±29.3),P <0.001],small value of f/t[(0.12 ± 0.07) vs.(0.17 ±0.10,P <0.001)],high density of PSA [(2.04 ± 9.36) vs.(0.32 ± 0.42),P < 0.001],digital rectal examination [72.0% (522/725) vs.23.1% (457/1 982),P < 0.001],irregular echo level [41.1% (695/1 693) vs.28.0% (284/1 014),P < 0.001],hypoechoic [64.3% (695/1 081) vs.17.5% (284/1 626),P < 0.001],microcalcifcation[56.8% (586/1 032) vs.23.5% (393/ 675),P < 0.001].PCDR of 12 + 1-core biopsy was significantly higher than lateral 6-core biopsy[41.5% (458/1 104)vs.37.0% (408/1 104),P =0.033].However,PCDR of 12-core biopsy had no statistical differences with 6-core biopsy[40.7% (449/1 104) vs.37.0% (408/1 104),P =0.081].PCDR of TRUS-guided biopsy was higher than that of finger-guided biopsy in patients with PSA≤30 ng/ml and PV >46 cm3 [30.0% (254/846)vs.22.2% (284/1 280),P <0.001;31.7% (124/391)vs.18.1% (131/723),P <0.001].PCDR of the 13th core positioned at abnormal signal area of TRUS and MRI was higher than the average PCDR of other 12 cores [70.9% (107/151) vs.56.6% (3 109/5 496),P < 0.001].Conclusion PCDR of TRUS-guided biopsy was higher than that of finger-guided biopsy in patients with PSA≤30 ng/ml and PV >46 cm3.PCDR of 12 + 1-core biopsy was significantly higher than that of lateral 6-core biopsy.However,PCDR of 12-core biopsy had no statistical differences with that of 6-core biopsy.PCDR of the 13th core positioned at abnormal signal area of TRUS was higher than the average PCDR of other 12 cores.
10.Investigation on depression of patients with systemic lupus erythematosus during coronavirus disease 2019 pandemic in Xinjiang

Objective:To explore the depression of patients with systemic lupus erythematosus (SLE) during the coronavirus disease 2019 (COVID-19) pandemic.Methods:Patients with SLE and healthy subjects(control group) from August 16, 2020 to September 2, 2020 in Urumqi, Xinjiang were collected. The depression and sleep quality were assessed by Self rating Depression Scale (SDS) and Pittsburgh sleep quality index (PSQI). The nonparametric data were represented by median (quartile spacing), and the rank sum test was used to compare the continuous variables between the two groups. Pearson correlation test was used for continuous variables, and Spearman correlation test was used for rank variables or classified variables. Statistical product and service solutions.Results:The prevalence rate of depression in the SLE group (41.9%) was significantly higher than that in healthy control group (32.1%). The difference was statistically significant ( χ2= 10.312, P<0.01). The prevalence rate of poor sleep in the SLE group (31.8%) was significantly higher than that in healthy control group (21.6%). The difference was statistically significant( χ2=3.885, P=0.049). The total score of VAS ( OR=1.366, P<0.01) , PSQI( OR=1.198, P<0.01) were the risk factor of depression in patients with SLE. Conclusion:SLE patients have a higher prevalence of depression during pandemic quarantinee-period, pain and poor sleep quality are risk factors. We need to pay attention to the psychological status of SLE patients.
