2.Experimental autoimmune myasthenia gravis in rabbit induced by peptides T?125~147

Objective To induce experimental autoimmune myasthenia gravis (EAMG) rabbits by using T?125~147 Methods Peptides T?125~147 were synthesized referring to the residue sequence of acetylcholine receptor of Torpedo California and the rabbits were inoculated with the peptides Clinical manifestation was graded in 4 levels Electrophysiological function was assessed by repetitive nerve stimulation (RNS) and single fibre electromyography (SFEMG) tests Anti peptides antibodies were evaluated by enzyme linked immunosorbent assay (ELISA) Student t test was used to analyze the difference between the EAMG and healthy rabbits Results Following the second inoculation,the rabbits appeared weakness Clinical symptoms were improved by neostigmine At 3,5 and 10 Hz,the decrement of compound muscle action potential (CMAP) and the mean jitter (MCD) of the immunized rabbits were higher than in the healthy ones The percentage of decrement of CMAP in order of the control group and the T?125~147 group were:3 Hz: 1 625?1 317,25 375?7 945; 5 Hz: 2 000?1 732,25 625?9 102; 10 Hz: 1 750?1 392,28 875?8 709.Following the above sequence,the MCD were:3 Hz: (9 875?1 126) ?s,(25 875?7 945) ?s; 5 Hz: (11 375?0 916) ?s,(27 500?3 381) ?s; 10 Hz: (12 375?1 061) ?s,(31 000?4 811) ?s Anti peptide antibodies in immunized groups were significantly higher than those in the control group Conclusion T?125~147 might be served as the immunogenic to induce EAMG in rabbits,accompanied by elevation of anti peptide antibodies and the blockage of neuromuscular transmission
3.Update on the molecular targeted therapy for cervical cancer

Cervical cancer is one of the most common and deadliest cancers in females worldwide. Despite the treatment methods of surgery, radiotherapy and chemotherapy are maturing, the prognosis of patients with recurrent, advanced or metastatic cervical cancer remains poor. Molecular targeted therapy provides new hope for these patients. This review focuses on the advances in agents targeting vascular endothelial growth factor pathway, epidermal growth factor receptor, mammalian target of rapamycin, histone deacetylases and cyclooxygenase-2 in cervical cancer.
7.Initial clinical result of the computer-assisted total knee replacement system

[Objective]To evaluate the initial effect of the computer-assisted total knee replacement system.[Method]The 24 computer-navigated knees in our department were involved,and were matched at ratio of 1∶3 by previous cases at sex,age and disease.All cases anteroposterior and lateral projection X-ray films were checked postoperatively.We compared ? and ? angles at anteroposterior films,? and ? angles ant lateral films of all involved cases[Result]Although there was no significant differences existing between two groups,we still observed the obvious reduce risk of malplacement both for femoral and tibial prosthesis,and improvement of mechanical alignment.[Conclusion]By using computer-navigated total knee replacement system during operating,the precise and repeatability could be ensured.
8.Apaf-1 gene transfer and its effects on apoptosis of AGS cell line

Objective Apaf-1 is a key factor involved in activating the Caspase system in apoptosis. The aim of this study was to evaluate the significance of Apaf-1 on the apoptosis process induced by chemotherapeutic agent. Methods We established Apaf-1 hyper- and hypo- expression system by transferred sense and anti-sense Apaf-1 cDNA into cultured AGS cell line. And meanwhile human cyt-c, Apaf-1, caspase-3, bax, and bcl-2 gene expressions were measured by RT-PCR method. Results Cyt-c, caspase-3, and bax gene expressions increased significantly after sense Apaf-1 transfection. On the contrary, anti-sense Apaf-1 transfer has minus effects on cyt-c, caspase-3, and bax gene expressions. No significant changes have been found on bcl-2 gene expression after transfection process.Conclusions Cyt-c pathway may play an important role in the apoptosis induced by chemotherapeutic drugs. The early expression of cyt-c and Apaf-1 may be the key procedure of cyt-c release from mitochondria and of caspases activation.
9.The expression of DNA methyltransferase,demethylase and tumor-associated genes in human gastric cancer

Objective To investigate the relationship of the expression of DNA methyltransferase,demethylase(mbd2) and tumor-associated genes in human gastric cancer. Methods Tissue samples of cancerous,para-cancerous and normal gastric mucosa were obtained surgically from 28 primary gastric cancer patients. The transcription level of DNA methyltransferase 1 (DNMT1),mbd2,methyl-CpG binding protein (MeCP2),p16 INK4A and c-myc were determined by using real-time RT-PCR and RT-PCR. The relationship between the expression of DNA methylation-associated genes and tumor-associated genes was analyzed. Results The mRNA level of DNMT1 was higher and the mRNA level of mbd2 gene was lower in cancerous tissue than that in normal tissue. The expression of c-myc instead of p16 INK4A and MeCP2 was increased in cancer tissues. The mRNA level of c-myc related negatively to mbd2 when gastric cancer developed. However,there was no any close relation between the transcription level of all above genes and tumor biological behavior in human gastric cancer. Conclusion This study indicates that MeCP2 but not DNMT1 may contribute to the regulation of tumor-associated genes expression in human gastric cancer.
10.Isolation and identification of chemical constituents in Hemerocallis fulva (Ⅱ)

Objective To study the chemical constituents in the roots of Hemerocallis fulva. MethodsCompounds were isolated from the CHCl3and n-butanol fraction of the EtOH extract by repeated column chromatography and their structures were identified by spectral analysis and physicochemical properties. Results From the CHCl3 and n-butanol fraction of the EtOH extract in the roots of H. fulva,ten compounds were isolated and identified as terresoxazine (Ⅰ),11-keto-?-boswellic acid (Ⅱ),hopane-6?,22-diol (Ⅲ),hecogenin (Ⅳ),stigmast-4-en-3?-ol (Ⅴ),stigmast-4-en-3-one (Ⅵ),?-feruloyloxy acid (Ⅶ),(E)-3,4-dihydroxyl cinnamic acid (Ⅷ),(E)-p-methyl cinnamic acid (Ⅸ),and vanillic acid (Ⅹ). Conclusion Compounds Ⅰ-Ⅳ,and Ⅶ are isolated and identified from the plants of Hemerocallis L. for the first time.
