2.Preliminary study on the effect of tyrosol and farnesol on biofilm formation of Candida albicans

Objective To study the regulation of quorum sensing molecule tyrosol and farnesol on biofilm formation of Candida albicans. Methods Candida albicans biofilms of clinic isolates and standard strain SC5314 were built when quorum sensing molecule existed. And inverted microscope was used to observe the morphology of C. albicans cells. RT-PCR and MTT assay were carried out to investigate the effect of quorum sensing molecule on expression of the two genes (HTA1 and EFG1) and cytoactive. Results Tyrosol could not promote hyphae development and cytoactive of C. albicans biofilms. The expression of HTA1 of C. albicans in biofilms was up-regulated by tyrosol but EFG1 was not. The inhibitory effect of farnesol on hyphae development, cytoactive and gene expression were not changed by addition of tyrosol. Conclusion Tyrosol can make C. albicans biofilms active in early stage. But when tyrosol and farnesol were simultaneously added, the effect of tyrosol were masked by farnesol. And C. albicans cells were more sensitive to farnesol than to tyrosol.
6.Construction and preliminary application of oligonucleotide microarray specialized for pancreatic adenocarcinoma

AIM:To investigate the construction of oligonucleotide microarray specialized for pancreatic adenocarcinoma-associated genes and its preliminary application of detecting differential expressed genes in pancreatic cancer.METHODS:Pancreatic cancer related genes were purposely selected,and oligonucleotide microarray was prepared by spotting oligonucleotide probes on glass slides coated with APS-PDC.Labeled cDNA targets for hybridizations were synthesized by reverse transcription from total RNA in the presence of Cy5-dCTP and Cy3-dCTP,respectively.Hybridized microarray was scanned by Agilent laser scanner,and the aquired image was analyzed by Imagene3.0 software.The intensity ratios of Cy3 and Cy5 were calculated.To confirm the expression profiles of these genes,quantitative reverse transcription-PCR (QRT-PCR) was carried out for CDC25B and TUSC3 genes,and β-actin gene was taken as internal control.The product of PCR was quantitated by comparative Ct method.RESULTS:The signal of microarray hybridization was clear,and the images had a lower background and higher signal-noise ratio.In comparison with normal pancreas,twenty -four differentially expressed genes were identified which included seventeen up-regulated and seven down-regulated genes.The results of QRT-PCR demonstrated that the expressions of CDC25B and TUSC3 in pancreatic cancer were up-regulated and down-regulated respectively,which is consistent with microarray results.CONCLUSION:The oligonucleotide microarray specialized for pancreatic cancer is desirable for its specialty and sensitivity,which can simultaneously and parallelly detect multiple pancreatic cancer-associated genes.

Based on the multi-angle analysis for patent portfolio of G. biloba preparation from the aspects of application time, legal status, technical development route, and patent portfolio layout, this article provides technical reference on research and development of G. biloba preparation, and the author suggest that Chinese applicants learn techniques and layout experiences of other patents fully to enhance the level of research and patent protection level.

10.BIOSYNTHESIS OF POLYHYDROXYALKANOATES BY RHODOCISTA PEKINGENSIS

In this article we report the results on the synthesis and accumulation of PHAs by Rhodocista pekingensis (strain 3-p), a phototroph that was isolated from wastewater treatment plant. Our results showed that the optimal conditions for PHAs accumulation with strain 3-p were as following: 0.01% Yeast Extract, 0.01% NH 4Cl, Acetate 5 g/L, and medium pH of 7.0~7.2.Under optimized conditions, strain 3-p accumulated PHAs up to 60% of its cellular dry weight. Enzymatic activities of ?-ketothiolase, acetoacetyl-CoA reductase, and PHA polymerase were detected and their activities increased as PHA synthesis initialized. Based on these study, we proposed the metabolic pathway of this strain should be:Acetate (or other fatty acids) - Acetyl-CoA --- thiolase Acetoacetyl-CoA --- reductase D (-)-?-Hydroxybutyryl-CoA ----- PHA polymerase PHAs.
