1.Inhibition of Src kinase can ameliorate renal interstitial fibrosis in unilateral ureteral obstruction mice

Objective To investigate the effect and mechanism of Src kinase in renal interstitial fibrosis of unilateral ureteral obstruction (UUO) mice.Methods Male C57BL/6J mice were randomly divided into 4 groups,including sham operation group (n=8),sham operation+PP2 group (n=8),UUO operation group (n=8) and UUO operation+PP2 group (n=8).The mice were injected 2 mg/kg PP2 by intraperitoneal everyday after surgery in sham+PP2 group and UUO+PP2 group.PP2 dissolved in 1% DMSO (formulated with normal saline).Sham and UUO group were given equal 1% DMSO.The mice were sacrificed at 7th day.Renal collagen was observed with Sirius red stain.The activities of Src,protein kinase B (PKB,AKT),p38 mitogen-activated protein kinase (p38 MAPK),extracellular signal-regulated kinase (ERK) and the protein expressions of α-smooth muscle actin (α-SMA) and fibronectin (FN) were detected by Western blotting.The expression of collagen I (COL [) was detected by immunohistochemistry and the expressions of matrix metalloprotein 9 (MMP-9),tissue inhibitor of metalloproteinase 1 (TIMP-1),transforming growth factor-β31 (TGF-β31),monocyte chemotactic protein-1(MCP-1),interleukin-6 (IL-6) were measured by ELISA.Resuts Compared with sham mice,UUO mice on 7th day displayed obvious renal fibrosis.Meanwhile,UUO mice had increased expressions of COL Ⅰ and FN,and activities of AKT,ERK and p38 MAPK (all P < 0.05).Their renal expressions of α-SMA,TGF-β1,MMP-9,TIMP-1,MCP-1 and IL-6 were also raised (all P < 0.05).Compared with those in UUO group,in UUO + PP2 group the activities of Src,AKT,p38 MAPK and ERK,and expressions of TGF-β1,MCP-1 and IL-6 decreased (all P < 0.05).Additionally,expressions of COL Ⅰ,FN and α-SMA,collagen deposition and renal fibrosis receded in UUO + PP2 group (all P < 0.05).However,the expressions of MMP-9 and TIMP-1 were not influenced by PP2 treatment.Conclusions Src kinase promotes myofibroblasts accumulation and inflammatory reaction through activating its downstream signaling pathway in the progressing of renal interstitial fibrosis.
4.Basic conditions of skin in female consumers for cosmetics in Chengdu area

Objective To investigate the skin basic conditions of normal female consumers for cosmetics who use a cream of lower than RMB 25 yuan in markets in Chengdu area.Methods Female subjects aged 25 to 45 years were recruited in Chengdu and divided into four age groups.Parameters were noninvasively tested to investigate skin baseline conditions of the two groups,including skin moisturization,trans-epidermal water loss (TEWL),color,texture and pH etc.Meanwhile,subjective scores were acquired by experienced dermatologists and self-questionnaires were collected to investigate subjects' concerns.Results Skin texture,softness,and skin color were found significantly different between age groups.The top 3 skin problems were dryness,skin color and wrinkles; the top 3 ideal skin were moisture,shining and fairness.Conclusions With similar environment and skin care habits,35-year-old is a turning point for females of decreased metabolism in the skin,as seen from skin color,elasticity,and roughness.
5.A method to predict clustered repeats in Salmonella genomes

Objective To develop a simple method identifying and illustrating clustered repeats in bacterial ge-nomes, and to observe the patterns of clustered repeats in Salmonella genomes.Methods Bacterial genomes were cut to be overlapped pieces of identical size with a sliding window strategy .Each piece of genome fragment was aligned against itself with BLAT integrated in PipMaker , which was further used to build collinearity figures . Collinearity figures were analyzed to identify the clustered repeats.Results With the new pipeline CRpred ( Clustered Repeat Predicter) , Salmonella typhimurium LT2 genome was screened, and in 151 clustered repeats were disclosed.Pattern analysis on these repeats indicated that there were five categories, including low-copy simple tandem repeats, high-copy simple tandem repeats, interspaced tandem repeats, reverse-complementary re-peats, and interspaced reverse-complementary repeats.Nine repeat regions in LT2 genome were discovered which could not be simply classified into the 5 categories defined above.Conclusions A new, simple and intuitive strategy is proposed to identify and show clustered repeats in genomes , providing clues for CRISPR , VNTR and oth-er repeat-related studies .
6.Comparative genomic analysis of Salmonella type Ⅲsecreted candidate gene yiiG

Objective To study the distribution and evolution of yiiG, a Salmonella gene encoding a candidate type secretedsubstrate .Methods Salmonella genomes were comprehensively screened for yiiG distribution with se-quence alignment strategies .The evolutionary history of yiiG was traced .Comparative genomic analysis was per-formed to study the evolutionary mechanisms of yiiG gene acquisition , loss and duplication .RNA-seq data were combined to analyzing the correlation between yiiG and other virulence factors .A variety of bioinformatic tools were used for discovering the possible type Ⅲsecretion signals .Results yiiG distributed in S.enterica subsp.enterica but variable in other subspecies of S.enterica.No yiiG was found in S.bongori.Besides Salmonella, only a part of Shigella and E.coli strains were detected with yiiG homologs .The genomic locus of yiiG and its adjacency showed conservation among all Salmonella, E.coli and Shigella strains.In most of the serovars of S.enterica subsp.enteri-ca, there was a head-to-head tandem whole yiiG repeat sequence upstream the yiiG gene, which was renamed as yiiGRrc.RNA-seq analysis showed that yiiG gene expression level was highly correlated with T 3SS-related genes . Bioinformatic prediction also indicated the T 3SS effector signals in YiiG N-terminus.Conclusions yiiG represented an ancient genomic locus , which will be a hot spot where rearrangement events frequently happened .The function of yiiG could potentially be related with Salmonella virulence.Finally, a new protein-encoding gene (yiiGRrc) was newly identified that was closely related with yiiG, providing the target for further understanding the composition , function and function variation of yiiG gene family .
7.Prospective study of risk factors in ventilator-associated pneumonia in mechanically ventilated patients in ICU

Objective To analysis the risk factors in ventilator-associated pneumonia (VAP) in mechanically ventilated patients.Methods Seventy-eight adult inpatients on mechanical ventilation (MV) through oral endotracheal intubation were studied prospectively by observation between June,2007 and May,2010.Clinical associated factors including patients'predisposition related,medical personnel or device related and nutritional state related factors,etc.were recorded and analyzed.Results In 78 ventilated patients,the incidence of VAP was 23.1%,the fatality rate was 22.2%.Preventive antibiotic treatment (OR=6.038; 95% CI:1.319-27.641; P =0.021),applying glucocorticoid (OR =5.385; 95% CI:1.191-24.346; P =0.029) and prealbumin (PA) ≤ 69.7 mg/L (OR =0.975; 95% CI:0.956-0.995 ; P =0.013) were risk factors in VAP.Conclusions The risk factors in VAP were PA ≤ 69.7mg/L,preventive antibiotic treatment and employment of glucocorticoid.
8.Significance and expression of PTEN, MLL gene in T lymphoblastic lymphoma/leukemia

Objective To investigate the significance and expression of PTEN, MLL in T lymphoblastic lymphoma/leukaemia(T-LBL/ALL). Methods Seventy-six cases of T-LBL/ALL were studied by using immunohistochemical EnVision method for PTEN. Fluorescence in-situ hybridization (FISH) for MLL gene (located on chromosome 11q23) was performed to detect its breakage and amplification. Results Among the 76 cases ofT- LBL/ALL, the positive rate of PTEN was 64.47 % (49/76), lower than that in reactivated lymphoid tissue (100 %, 20/20) (λ2= 19.220, P <0.05). PTEN expression was reversely correlated to theclinical stage, Ki-67 index and LDH level (P <0.05). Among the 76 cases, MLL gene with breakage of 11q23 was detected in 13 cases (17.11%), and amplification in 18 cases (23.68 %). Survival rate ot MLL gene breakage group was lower than that of non-breakage group (25.0 %, 43.6 %). Survival rate of MLL gene amplification group was lower than that of non-amplification group too (17.1%, 42.7 %). Both of breakage and amplification were related to prognosis ( λ 2 = 11.357, λ 2 = 4.533; P <0.05). Conclusion Anti-oncogene PTEN down-regulation may play an important role on the development and proceeding of T-LBL/ALL. MLL gene with breakage and amplification of 11q23 are helpful to predict prognosis of T-LBL/ALL. The case with MLL gene breakage and amplification of T-LBL/ALL may have a poor prognosis. It hints this group maybe a subtype of T-LBL/ALL.
9.Research on the relationship between fructose-1, 6-bisphosphatase and chemosensitivity of ovarian carcinoma

Background and purpose: Epithelial ovarian carcinoma is the most malignant tumor in female reproductive system because of its resistance to chemotherapy. Fructose-1, 6-bisphosphatase (FBP1) is a rate-limiting enzyme in gluconeogenesis used to catalyze the hydrolysis of fructose-1, 6-bisphosphate to fructose-6-phosphate and inorganic phosphate, thereby inhibiting the effect of glycolysis in tumor cells. This study aimed to investigate the association between the expression of FBP1 and chemosensitivity. Methods: The expression level of FBP1 in ovarian cancer patients was measured by immunohistochemistry. Results: According to the results of immunohistochemistry in 209 ovarian carcinoma specimens, the percentage of positive FBP1 expression was about 49.3% (103/209). Loss of FBP1 was a negative factor of survival (42.6 months vs 62.1 months, P=0.003). Besides, patients who were sensitive to chemotherapy displayed significantly higher scores of FBP1 expression than patients who were resistant to therapy (P=0.007). Conclusion: The rate-limiting enzyme FBP1 in gluconeogenesis can be used as a biomarker for predicting the chemoresistance and prognosis of ovarian cancer patients.
