1. Study on anti-tumor activity of curcumin analogues with FGFR1 as target

Objective: To search for the small molecule inhibitor with anti-tumor activity by fibroblast growth factor receptor 1 (FGFR1) as target. Methods: The analogue B6 of EF24 was obtained by reforming mono carbonyl curcumin analogues and applied to identifying the target with FGFR1 kinase activity assay. The effect of EF24 and its analogue B6 on the proliferation of HL7702 in normal human and four tumor cells, such as NCI-H460, SGC-7901, A549, and U251; With the concentration of 2.5, 5, and 10 μmol/L, B6 was used to investigate the phosphorylation inhibition of bFGF/FGFR downstream signal protein expression in NCI-H460 cells and Caspase-3 factor expression. Results: With the FGFR1 as target, B6 could inhibit the phosphorylation of FGFR1 in NCI-H460 cells, AKT, and ERK1/2; It also could inhibit the proliferation of cancer cells and promote cell apoptosis. Conclusion: The analogue B6 of EF24 is obtained from the leading compound EF24 with in vitro anti-tumor effect, which provides the basis of looking for the candidate anti-tumor drugs of FGFR1 inhibitor.
2. Research progress on gingerol and zingiberol from ginger

Ginger as a commonly used Chinese materia medica, but also edible, has been widely cultivated in China. Complex chemical components have been found more than 100 kinds, they are mainly sesquiterpenes and monoterpenes. Gingerol and zingiberol are as the active ingredients of ginger. In the literature of ginger, we found that most of them are on gingerol while less on zingiberol; More of them confused or mixed the two. In this paper the related literatures at home and abroad analyze the structure and pharmacological effects as well as the application of gingerol and zingiberol.
3. Alleviation of curcumin analog WZ35 on type 1 diabetes-induced renal injury via inhibiting inflammatory response

Objective: To study the alleviation of curcumin analog WZ35 on type 1 diabetic nephropathy (DN). Methods: Mice were treated with a single ip injection of streptozocin (STZ) to induce type 1 diabetes, while the control animals received the same volume of citrate buffer. The curcumin and its analog WZ35 (20 mg/kg) were ig administration for 9 weeks after the diabete obtained. The body weight and blood glucose were monitored every 7 d. Biochemistry analyzer was used to analyze the creatinine (Cr) and urea nitrogen (BUN) levels in serum. The histopathology of kidney tissue was detected by HE staining. RT-qPCR assay was used to evaluate the gene levels of inflammatory cytokines and chemokines. CD68 staining was used to evaluate the macrophages infiltration in kidney tissue. Results: Compared with negative control, the mice in diabetic group showed the reduced body weight, increased blood glucose, high Cr and BUN levels in serum, renal pathological damage, and increased inflammatory gene, and macrophages infiltration in kidney. While the administration with curcumin and its analog WZ35 could obviously attenuate the DN through inhibiting Cr and BUN in serum while had no effect on the body weight and blood glucose. Compared with curcumin, WZ35 could inhibit the gene expression of inflammatory cytokines and chemokines as well as infiltration of macrophages. Conclusion: The curcumin analog could attenuate DN through inhibiting inflammatory response.

5. Research progress in Polygonati Rhizoma and predictive analysis on Q-marker

Polygonati Rhizoma (PR) is a traditional Chinese herbal medicine, mainly distributed in the North Temperate Zone and North Subtropical Zone, widely spread over the south China. As the progress of PR in pharmacology, medicinal chemistry, and clinical research, the study of Polygonatum's active site and effective components are attached much attention in new drug research and development. In this paper, the resources, chemical composition, pharmacological activity, and clinical application of PR are summarized. Literature research suggests a variety of chemical composition in PR, including polysaccharide, steroidal saponins, terpene, alkaloid, wooden fat element, flavonoids, and phytosterols, in which polysaccharides and steroidal saponins class are the main efficacy components. On the base, the difference of the main chemical composition are compared in P. sibiricum, P. kingianum and P. cyrtonema, to provide experiment basis for Q-maker determining; And the contribution of polygonatum polysaccharide, steroidal saponins and isoflavone compounds to the main therapeutic effects were been studied. It is suggested to carry out the quality standard and Q-marker research in PR-the qualitative and quantitative analysis of polysaccharide, steroidal saponins and isoflavone compounds in the high quality PR and compare the quantitative limit of different batches, to provide scientific data for quality standard setting.
6. Chemical constituents of traditional She medicine Rubi Radix et Rhizoma

Objective: To explore the chemical constituents of traditional She medicine Rubi Radix et Rhizoma (the roots and rhizomes of Rubus chingii in family Rosaceae). Methods: The compounds were isolated and purified by various column chromatography and the structures were identified by physicochemical constant determination and spectral analysis. Results: Nine compounds were isolated and identified as β-sitosterol (1), daucosterol (2), euscaphic acid (3), 11α-euscaphic acid (4), tormentic acid (5), ellagic acid (6), gallic acid (7), ursolic acid (8), and oleanolic acid (9) from 95% ethanol extract of Rubi Radix et Rhizoma. Conclusion: All the nine compounds are isolated from Rubi Radix et Rhizoma for the first time and the compound 4 is first isolated from R. chingii.
7. Curcumin analog exhibited anti-inflammatory activity through inhibiting ERK/JNK and NF-κB signaling pathway

Objective: To study the anti-inflammatory activity and mechanism of mono-carbonyl curcumin analog 22. Methods: The cell cytotoxicity of compound 22 was detected by MTT assay. LPS activated peritoneal macrophages was used as cell model. The effect of compound 22 on inflammatory cytokines protein and gene expression was detected by ELISA and RT-qPCR, respectively. UV-absorbing assay was used to determine the stability of compound 22. The anti-inflammatory mechanism of compound 22 was detected by Western blotting assay. Results: Compound 22 showed no cytotoxicity after incubated cells 24 h. Compared with curcumin, compound 22 dose-dependently inhibited LPS-induced production of inflammatory cytokines TNF-α and IL-6. Compound 22 showed more stability than curcumin in vitro. Meanwhile, compound 22 inhibited LPS-induced inflammatory gene IL-1β, IL-12, IL-6, and TNF-α expression through qRT-PCR assay. Compound 22 inhibited the phosphorylation of ERK/JNK, also reversed the LPS-induced degradation of IκB. Conclusion: Compound 22 exhibits its anti-inflammatory activity through inhibiting ERK/JNK signaling pathway, as well as NF-κB activation.
8. Quantitative analysis on scientific and technical literature of Polygonati Rhizoma in recent five years

To comprehensively understand the developing state of Polygonati Rhizoma (PR) studies in the period of Twelfth-five-year Plan, document retrieval was performed on the literature sources of Chinese Journal Full-text Database and Web of Science during 2011-2015. The literature distribution characteristics of time, journals, authors, author affiliations, author's regions, and research subjects about PR were analyzed statistically by the method of bibliometrics. Ucinet and NetDraw were applied to conducting online co-occurrence graph of keywords. The results showed that China is the leader of research on PR. Distribution of journals is more concentrated, taking the following four journals as representation-Lishizhen Medicine and Materia Medica Research, Chinese Journal of Experimental Traditional Medical Formulae, Seed, and Journal of Anhui Agricultural Sciences. The research subjects have a wide array of sources, including germplasm resources, seed breeding, cultivation techniques, biological property, chemistry, pharmacology, and clinical application. The research institutes and teams have been formed. The study on PR has made remarkable achievements, but the germplasm resources of PR have degenerated continually, seed breeding, cultivation techniques, extraction and isolation technology are less developed. The supply of PR fails to meet the consumption demand. It indicates that the seed propagation, standardized artificial planting, extraction process optimization, food, and health care product development will be hot research areas.
9. Feasibility analysis of coronavirus disease 2019 co-treated by Zedoary Turmeric Oil Injection

Because of no specific drug for the treatment of coronavirus disease 2019 (COVID-19), a number of treatment plans for COVID-19 had been released by national medical treatment authorities. The efficacy of zedoary turmeric oil and its preparation for antiviral and pulmonary fibrosis treatment had been confirmed by many basic studies and clinical applications. It was speculated that Zedoary Turmeric Oil Injection could be used in the clinical diagnose and treatment of COVID-19, especially in the treatment of pulmonary fibrosis caused by pulmonary interstitial changes, antidiarrheal and fever. In addition, the compatibility of Zedoary Turmeric Oil Injection with other antiviral and antibiotic drugs suggested that it could be used to reduce drug-induced liver injury in the treatment of COVID-19 patients, and improve the therapeutic effect, which provided a theoretical basis for the scientific use of zedoary turmeric oil and its injection in the treatment of COVID-19.
10.Application of heterozygous ambiguity resolution primers resolving ambiguous genotyping results of human leukocyte antigen genes

Objective To evaluate the heterozygous ambiguity resolution primers (HARPs) method in resolving ambiguous genotyping results of human leukocyte antigen (HLA) genes in Chinese Hart population, and choose some appropriate HARPs primers. Methods HLA-A, HLA-B and HLA-DRB1 genes of 416 southern Chinese Han individuals were genotyped by sequence-based-typing(SBT) method and then the ambiguous genotyping samples were sequenced again by HARPs primers provided by American Atria company. Results The percentage of ambiguous genotyping samples resolved by HARPs for HLA-A, HLA-B and HLA-DRBI locus was 86.3% (132/153), 73.9% (130/176) and 38.1% (85/223) respectively. Among them, 48.5% (64/132)HLA-A, 80.0% (104/130)HLA-B and all HLA-DRB1(85/85)samples only need one primer, 47.7 % (63/132)HLA-A and 20.0% (26/130)HLA-B samples need two primers. Three to six different HARPs primers can resolve more than 90% ambiguities. Conclusion HARPs is a convenient method and could be a routine method to resolve ambiguities for HLA-A, HLA-B and HLA-DRB1 genes genotyped by SBT in Chinese Han population.
