1.The Expression of Aquaporin-2,3,4 in Rats' Endolymphatic Sac and Kidney

Objective To confirm the expression of Aquaporin2,3,4(AQP2,3,4,water channel protein) in rats'endolymphatic sacs(ES)and kidneys and to investigate and to compare the effects of anti-diuretic hormone(AVP) and DDAVP([deamino-Cys1,D-Arg8]-Vasopressin,V2-receptor agonist)on the expression of AQP2 in rats' ESs and kidneys.Methods 30 healthy Swards white rats were divided into the negative control,AVP group and dDAVP group respectively.The animals were cardiacally perfused.The temporal bones and kidneys were taken out and sectioned by means of the paraffin-embedded technique.The sections of ESs were labeled with fluorescent antibody by immunohistochemical method,and the kidneys' with avidin-biotin-peroxidase complex method(ABC).The expressions of AQP-2 were confirmed in the ESs of the rats while the different effects of the AVP and DDAVP on the ESs and the kidneys were observed.The slides used were analyzed by the image-analyzer and the subsequent data were statistically studied.Results In the cytomembrane and cytoplasm of ESs' epithelia,the constant and clear fluorescent reaction could be observed in the normal control group with the first antibody of AQP2.Significant feeble fluorescent reaction of the first antibody of AQP-2 was revealed in AVP group and DDAVP group.In comparison with the control group,significant stain was found in AVP group and DDAVP group.Noted were also lower gray(P

Objective To investigated the prevalence of obesity and metabolic syndrome (MS) and the relationship of non-alcoholic fatty liver disease (NAFLD) with MS in Hangzhou of Zhejiang province. Methods A total of 3500 subjects who were negative for hepatitis B surface antigen marker were selected to take physical examinations. Body height, body weight, blood pressure, alanine aminotransferase,fasting blood glucose,high-density lipoprotein cholesterol,and triglyeerides were recorded. B-ultrasonography of the liver and gallbladder was undertaken. Results The prevalence of MS in the NAFLD or normal control group was 25.49% and 8. 61%, respectively ( P < 0. 05 ). In multivariate logistic regression analyses, MS patients were 3. 927 times more likely to be NAFLD-related. Increased body mass index (BMI) and triglyceride were significantly associated with NAFLD among MS patients. Conclusion BMI and triglyceride level may be the main risk factors of NAFLD-related MS.

4.Column-switching high-performance liquid chromatographic method for the determination of morphine and O6-monoacetylmorphine in urine.

OBJECTIVE:
To develop a column-switching high-performance liquid chromatographic method for the determination of morphine and O6-monoacetylmorphine in urine.
METHODS:
Urine samples (1.0 ml) were spiked with 1.0 ml borate buffer, after centrifugation, 1.0 ml of supernate were injected directly into an extraction column (YWG C18 33 mm x 5.0 mm, 10 microns). After a washing step with the extraction mobile phase, the retained morphine and O6-monoacetylmorphine were flushed into the analytical column (Lichrospher 100 CN 125 mm x 4.0 mm, 5 microns) with the mobile phase CH3OH-H2O (60:40). The analytical mobile phase is CH3OH-phosphate buffer (pH6.86) (22:78). The UV detector was set at lambda 286 nm.
RESULTS:
The method shows excellent linearity from 50 to 1,600 ng/ml for morphine and from 100 to 1,600 ng/ml for O6-monoacetylmorphine. The linear correlation coefficients were > 0.999. The relative standard deviations were < 4%. The limits of detection were 40 ng/ml for both morphine and O6-monoacetylmorphine.
CONCLUSION
The method described is sensitive, rapid, reproducible, and simple.

5.Study on the effect of aging on sleep apnea syndrome in rats

Objective To investigate the effect of aging on the sleep apneas syndrome in Sprague-Dawley (SD) rats.Methods 18 male SD rats in a SPF grade were divided into three groups:young (3-month-old rats,n=6),middle-aged(12-14-month-old rats,n=6) and old group (18-20-month-old rats,n=6).The rats were implanted with EEG and ECG electrodes and underwent sleep monitoring.Results During non-rapid eye movement (NREM),the spontaneous apnea index (SPAI) in young,middle-aged and old groups were increased with aging [0.41 (0.00-1.14)times/h,0.76(0.00-6.28)times/h,2.13(1.44-3.87)times/h respectively,(x2=8.801,P=0.012)].There were significant differences in the average time of post sigh apnea in NREM between the three groups [(3.51±0.18)sec vs.(3.84±0.57)sec vs.(4.36±0.57) sec,F=4.729,P=0.026].SPAI in NREM and the average time of post-sigh apnea in rapid eye movement (REM) as well as NREM showed an increase trend with aging,but there was no significant difference between groups (both P >0.05).Conclusions The apnea index,total time of sleep apneas and the average time of postsigh apnea are increased with aging,which is similar to that in human.
7.Clinicopathologic characteristics of eight patients with small cell carcinoma of the cervix

Objective To investigate the clinicopathologic characteristics,therapy and prognostic factors of small cell carcinoma of the cervix.Methods Clinical and pathological data of eight patients were analyzed retrospectively.Results Eight patients all presented with symptoms of abnormal vaginal bleeding or postcoital spotting.One patient had stage Ⅰ b1 disease,2 had stage Ⅰ b2 disease.2 had stage Ⅱ b disease and 3 had stage Ⅲ b disease.Histopathologic findings showed the small tumor cells had scant cytoplasm,round nuclei,absence of nucleoli,and finely dispersed chromatin.Immunohistochemical findings were positive in 7 cases for neuron-specific enolase.Three patients with stage Ⅰ b disease and 1patient with stage Ⅲb disease underwent radical hysterectomy and postoperative chemotherapy.with or without radiotherapy,and the survival period was 64,22,14 and 6 months respectively.Two patients with stage Ⅱ b disease and 2 with stage Ⅲ b disease underwent chemotherapy and radiotherapy,and the survival period was 25,9,10 and 5 months respectively.Conclusions Immunohistochemical analysis using several kinds of neuroendocrine markers is helpful in establishing the correct diagnosis in addition to focusing on characteristic histopathological features.It is necessary to use comprehensive treatment including surgery,chemotherapy and radiotherapy for patients with small cell carcinoma of the cervix.Chemotherapy may play an important role in the treatment.
8.Effect of angiotensin Ⅱ and aldosterone in patients with congenital heart disease by preoperative nursing interventions at high altitude area

Objective To study the effect of angiotensin Ⅱ and aldosterone (ALD) in patients with congenital heart disease by preoperative nursing interventions at high altitude area. Methods 86 patients with congenital heart disease were divided into the experimental group and the control group on average by random. The experimental group was treated with preoperative nursing interventions and routine nursing before operation and the control group was treated with routine nursing only. Angiotensin Ⅱ and aldosterone were detected before anesthesia, 30min after operation and after pulling out of endotracheal tube respectively. Results The concentration of Angiotensin Ⅱ and aldosterone was significantly lower in the experimental group than the control group. Conclusions The risk of open surgical operation for congenital heart disease at high altitude can be reduced by preoperative nursing interventions.

10.Changes in the lymphocyte subgroup after TACE, THPP of primary hepatic carcinoma
