1.Screening of proteins binding with HBV X promoter by phage display technique from human liver cDNA phage library

Objective To screen the proteins binding with HBV X-promoter and to investigate their potential role in the regulation of replication and expression of HBV DNA. Methods By using HBV X biotinylated promoter DNA as the selective molecule, the T7 select human liver cDNA library was biopanned and positive clones were selected. After screening, positive plaques were performed to amplify for inserted DNA fragment and cloned into pGEM-Teasy vector. Thirty positive plaques were chosen for DNA sequencing. Results Five kinds of known and nine kinds of novel cDNA sequences were obtained. The 5 kinds of known ones are human serum albumin, NADH dehydrogenase subunit 4, prokininase, ubiquitin specific protease 10, and testis enhanced gene transcript. Conclusion The binding proteins of HBV Xp DNA were identified by phage display. The results suggest that this approach provides a new search tool for the study of replication and expression mechanism of HBV DNA.
2.MR perfusion weighted imaging of C6 glioma after antiangiogenic treatment in rats

Objective To observe the therapeutic effect of antiangiogenesis on C6 glioma and to study the practical value of PWI in evaluating the early response of the tumor to the treatment.Methods C6 glioma cells were stereotactically implanted into SD rats intracranially.At the 15 day after tumor implantation,the recombinant human endostatin was administered or stereotactic radiosurgery was performed.Endostatin was injected subcutaneously at the dose of 5 mg?kg -1 ?d -1 ,10 mg?kg -1 ?d -1 ,and 20 mg?kg -1 ?d -1 for 7 days.Stereotactic radiosurgery was given once with the central radiation dose of 25Gy.MR PWI was performed to measure relative cerebral blood volume (rCBV) of the tumor and the normal brain before and 48 hours after treatment.Conventional MRI was performed to calculate tumor volume.Results The tumor volume was (429.0?36.7)mm 3,(305.7?32.8)mm 3,and (277.0?20.6)mm 3 in each endostatin treated group,and(390.0?33.8)mm 3 in SRS treated group,which were lower than that in control group(566.7?135.0)mm 3 (P=0.031).The rCBV in endostatin group was 1.57?0.12,1.30?0.12,and 1.24?0.08,respectively according to three different doses,which were lower than those before endostatin therapy with statistical significance (P
3.Distribution characteristics of hematological malignancies in Harbin

Objective To analyze the prevalence of the various hematological malignancies (HM) in Harbin.Methods Study data was collected from January 2010 to December 2011.All cases were diagnosed and classified on the basis of blood test,bone marrow puncture,histochemical staining and typing and classified by the French American British classification.The age and sex distribution of HM and its subtypes were analyzed.Results Of 2214 Chinese people diagnosed with HM,acute myeloid leukaemia (AML) (33.5 %) and myelodysplastic syndromes (MDS) (29.9 %) were the most prevalent and of 742 AML,the most frequent subtypes were M3.With the growth of age,the rates of chronic lymphocytic leukemia (CLL) and multiple myeloma (MM) were increased,but in acute lymphocytic leukemia (ALL),this trend was reversed.The distribution of HM increased with age between 0-60 years old,in above 60 years old,the frequency of some chronic HM including CLL,chronic myelomonocytic leukemia (CMML),chronic neutrophilic leukemia (CNL) and MM were continued to rise,but other HM subtypes were decrease,lower than in 41-60 years old groups.This study also revealed that M0 and MPAL were more common in 0-20 years as well as ALL.For patients never drinking alcohol and drinking for at least 10 years maybe associated with acute leukemia [OR =1.53 (95 % CI 1.05-2.23)],while smoking wasn' t a substantial risk factor for acute and chronic leukemia (P =0.20,0.48).Conclusions The epidemiology of HM in Harbin indicates that AML is the most prevalent,followed by MDS.Prevalence of CLL and MM increases with the age in patients above 60 years old.Drinking for at least 10 years maybe associated with acute leukemia.

6.Related Studies of Rehabilitation Therapy after the Total Knee Arthroplasty (review)

Rehabilitation is very important to the therapeutic effect of total knee arthroplasty, and it's also the key measure for the knee function to reach the expected effect after the surgery. The most important contents of rehabilitation therapies are the training of muscle strength, range of joint motion, joint position sense and gait. Integrated rehabilitation therapies was used to improve the knee function as possible, such as all kinds of training to improve the knee muscle strength of flex and extend; early training of continued positive motion of knee joint to improve the knee range of motion; the training of knee joint position sense to improve the ability of motion control, posture adjusting and balance keeping; the gait training to adjust the unusual gait.
7.Genome amplification and sequence analysis of human bocavirus 2.

To obtain the genome sequence of human bocavirus 2 (HBoV2), different regions of HBoV2 genome were amplified through PCR in fecal specimens which had been identified as single-positive for HBoV2 in 2010. A genome sequence of HBoV2 (HBoV2-NC, 5444 bp) was obtained after sequence assembly. The phylogenetic analysis showed that HBoV2-NC had the closest evolutionary relationship with HBoV2 Lanzhou strain. The predication of inverted terminal repeats of HBoV2-NC by DINAMelt showed that inverted terminal repeats were contained in HBoV2-NC 5' terminal, which had the typical stem-loop structure in other parvoviruses. Finally, some flanking sequences of HBoV2-NC were amplified by linker-PCR.

8.Analysis of treatment outcomes of elderly pulmonary tuberculosis aged 65 and above in Chongqing from 2015 to 2020

Abstract: Objective To analyze the treatment outcomes of elderly patients with pulmonary tuberculosis in Chongqing, so as to provide reference for the prevention and control strategies of the epidemic of elderly pulmonary tuberculosis. Methods The data of tuberculosis cases aged ≥65 years in Chongqing from 2015 to 2020 were collected from the National Health Insurance Information Project Disease Prevention and Control Information system. Descriptive statistical methods were used to analyze the data. Results The registration rates of elderly active pulmonary tuberculosis patients and etiological positive patients were 110.95/10-5and 32.25/10-5 in 2015 and 84.06/10-5 and 57.29/10-5 in 2020. The annual decline rate of active tuberculosis registration was 5.40%, and the annual increase rate of pathogenic-positive tuberculosis registration was 12.18%. The registration rates of active tuberculosis patients and etiological positive patients in the whole population were 70.75/10-5 and 17.63/10-5 in 2015 and 50.34/10-5 and 29.14/10-5 in 2020. The annual decline rate of active tuberculosis registration was 6.58%, and the annual increase rate of pathogenic-positive tuberculosis registration was 10.57%. From 2015 to 2020, a total of 25 931 cases of elderly pulmonary tuberculosis were registered, of which 21 374 (82.43%) cases were successfully treated and 4 010 (15.80%) cases had unfavorable outcomes. The proportion of cured and death patients showed an increasing trend year by year (χ2trend=313.853, 100.502, P<0.01). From 2015 to 2020, the average annual successful treatment rate of elderly pulmonary tuberculosis in the whole city was 82.43%, with the lowest rate in southeast Chongqing (74.23%), followed by urban areas (81.99%). The success rate of elderly pulmonary tuberculosis treatment in the whole city, west Chongqing, northeast Chongqing and southeast Chongqing showed a downward trend year by year (χ2trend=230.199, 35.278, 108.076, 112.130, all P<0.01), with annual decline rates of 2.77%, 2.26%, 3.0% and 4.12%, respectively. Among the registered elderly patients, female, 65-<75 years old, Han nationality, newly diagnosed, no complications, and negative for etiology (χ2=15.234, 255.910, 146.842, 179.998, 25.575, 131.170, P<0.01) had higher success treatment rates. Conclusions The prevalence of pulmonary tuberculosis in the elderly population in Chongqing City is declining, but the positive registration rate of etiology is increasing annually, and the success rate of treatment is decreasing. Therefore, it is necessary to strengthen the systematic management, publicity and education of elderly patients (especially those in southeast Chongqing, male, positive patients and severe patients) to effectively control the epidemic of tuberculosis in the elderly.
9.Study of surface electromyographic signal features of pelvic floor muscles in patients with complete spinal cord injury

Objective To investigate changes of surface electromyogr8phic(sEM G)signal of pelvic floor muscles in complete spinal cord injury(cSCI)patients with neurogenic bowel dysfunction.Methods Fifteen hospitalized patients with cSCI(observation group)and fifteen normal subjects(control group)were involved in this study.The root mean square(RMS)of sEMG signals were collected at pelvic floor muscles with rectal surface electrode when subjects'pelvic floor muscles were rest(10 s),fleetly contract(2 s×3),continually contract(10 s).Both groups'data of different contracting states of pelvic floor muscles were analyzed and compared.Results The max RMS and average RMS(16.61±2.83,13.52±2.22)at pelvic floor muscles'rest in observation group were higher than that in control group(8.41±5.55,3.45±1.53).There was statistical difference between two groups(P<0.01).In the subjects of observation group max RMS and average RMS(20.24±13.99,13.36±2.39)at continual contraction and average RMS(13.40±2.31)at fleet contraction were nearly the same as RMS value at pelvic floor muscles'rest.There was no statistical difference between these two states(P>0.05).Conclusion The sEMG could be a quantitative index in assessing function of pelvic floor muscles and the neurogenic bowel dysfunction after cSCI.It can supply some clinical value in framing the training of pelvic floor muscles and improving the bowel dysfunction.
10.Effect of gender differences in distal femoral geometry on the clinical outcomes of total knee arthroplasty

Objective To investigate the effect of gender differences in distal femoral geometry on the clinical outcomes of total knee arthroplasty (TKA). Methods From March 2003 to October 2006, 213 cases with 294 osteoarthritic knees who had underwent primary TKA were involved in the study. The study included 87 men (118 knees) and 126 women (176 knees) who had a mean age of 64.4 years. All the cases were assessed with the knee-rating system of Knee Society, the knee range of flexion (ROF), corrected posterior offset (PCO) and anterior condylar offset (ACO) based on sex. Results Each patient had a Minimum of 2 years follow-up. The preoperative Knee Society scores and ROF, the two-year postoperative changes of stability scores and walking ability had no differences between the two groups. While greater improvements of postoperative pain scores and stair-climbing ability were seen in male. Men had better intraoperative ROF than women had ones, but there were no gender differences with regard to the two-year postoperative improvement of ROF. Corrected PCO decreased more markedly in female than in male. Corrected ACO decreased equally in female and male. The difference in the posterior condylar offset after TKA was statistically correlated with the change in pain scores, stair climbing ability and intraoperative ROF, respectively. Conclusion Male had a better outcome than female did after TKA. Femoral components which designed in the light of Caucasian anatomic characteristics couldn't match the native anatomy of distal femurs of Chinese female. Sexual dimorphism in humans and anatomic variations in various ethnic groups should be seriously considered in total knee prosthesis design.
