1.Clinical study on ginkgolide nebulised inhalation in treating bronchial asthma.

OBJECTIVETo observe the effect of ginkgolide nebulised inhalation in treating bronchial asthma.
METHODSAsthma patients of mild to moderate degree were randomly divided into three groups: the treated group treated by ginkgolide nebulised inhalation 10 mg/ml, twice daily, for 6 weeks in total, the positive control group, treated by cromlyn sodium nebulised inhalation 20 mg/10 ml, twice daily, for 6 weeks in total and the negative control group treated by normal saline 10 ml, twice daily, for 4 weeks in total. The changes on symptomatic scoring (Chetta' s method), pulmonary function (FEV1, PEF), serum eosinophil count, eosinophil cationic protein (ECP, determined by RIA), as well as airway response to ultrasonically nebulised distilled water (UNDW), and adverse reaction occurred in patients were observed. Results The symptomatic scorings in the treated and the positive control group were reduced from 5.1+/-2.3 and 6.0+/-2.6 to 1.6+/-1.7 and 1.6+/-1.7, respectively (both P<0.01) after 6 weeks of treatment. In the treated group, ECP was reduced from 6.7 microg/L to 4.3 microg/L (P<0.05), FEV1 and PEF were improved (P<0.05, P<0.01), while in positive control group, only improvement of FEV1 was found (P < 0.05). UNDW in the above two groups were reduced (P<0.01). Although decrease of symptomatic scoring was found in the negative control group, yet ECP, pulmonary function and airway response showed no improvement (P >0.05). Adverse reactions revealed were mainly chest stuffiness, stimulating cough, especially in the treated group, but were tolerable to most patients. Conclusion Ginkgolide has the action of fighting against asthmatic airway inflammation, which provides new means for treating bronchial asthma.

2.Comparative study on effect of osthole and genistein on peak bone mass in rats.

OBJECTIVETo compare the ability of osthole (OST) and genistein (GEN) in enhancing bone peak bone mass of rats to prevent osteoporosis.
METHODSThirty-six female one-month-old SD rats of (125 +/- 3) g body weight were randomly divided into three groups, 12 rats in each group, one group was orally administered osthole at 9 mg x kg(-1) d(-1), one group was given genistein at 10 mg x kg(-1) d(-1) and another was given equal quantity of distilled water as the control. The body weight was monitored weekly and the bone mineral density (BMD) of total body was measured every month. All rats were sacrificed after three months, the femoral bone mineral density, the serum levels of osteocalcin (OC) and anti-tartaric acid phosphatase 5b (TRACP 5b) were measured by Elisa. The bone microarchitectures were analyzed with micro-CT and the bone biomechanics properties were tested with universal material machine.
RESULTSNo significant differences were observed between O-treated or GEN group and the control for the food-intake and body weight during three months. However, the rats treated with OST had significant higher BMD for both total body and femur than the control and GEN group. The O-treated rats also had higher level of serum OC and lower level of TRACP 5b. Besides, they owned bigger bone volume/tissue volume, trabecular thickness, trabecular number but smaller trabecular spacing. In the three point bending tests of femurs,they were found to have larger maximum load, the young's modulus and structural model index (SMI).
CONCLUSIONOrally administered osthole could efficiently increase the peak bone mass of rats,which provide new ideas for preventing osteoporosis.

3.Analysis on the characteristics and trend of malignant tumor in Yueqing City, Zhejiang Province from 2012 to 2015

Objective To analyze the characteristics and epidemic trend of malignant tumor in Yueqing City of Zhejiang Province from 2012 to 2015 so as to provide scientific bases for policy making regarding control and prevention of malignant tumor.Methods Descriptive epidemiological analyses were made on the data of malignant tumor in Yueqing City from 2012 to 2015.Results The average annual incidence rate of malignant tumor from 2012 to 2015 was 248.85/105, the incidence rates from 2012 to 2015 were 238.24/105, 242.95/105, 247.15/105, 266.85/105, respectively.The average male incidence rate was 265.82/105, the average female incidence rate was 230.38/105.The former was higher than the latter.The group of 40-year-old and above age accounted for 91.64% of the total number of malignant tumor cases.Leukemia was common among 20-year-old and below age group.Breast cancer, liver cancer and lung cancer were common among 20-to 39-year-old age group.Lung cancer, liver cancer and gastric cancer were common among the age group of 40-year-old and above.The top five incidence rates of malignant tumor were lung cancer, liver cancer, gastric cancer, colorectal cancer and breast cancer, accounting for 56.74% of total cases thereof.Conclusion From 2012 to 2015, the incidence trend of malignant tumor in Yueqing City remained basically stable;the incidence rate remained relatively high and showed a slow rising trend on a yearly basis with the incidence rate of lung cancer increasing most significantly.The male incidence rate of malignant tumor was obviously higher than the female incident rate thereof.For the age group over 40, the incidence rate of malignant tumor increased significantly.It is necessary to pertinently strengthen the relevant health education of malignant tumor and implement the relevant early monitoring, thereby reducing the incidence rate.
4.Magnetic resonance characteristics of endometriosis rat model.

OBJECTIVETo establish a non-invasive, repeatable and dynamic study method in endometriosis rat model using magnetic resonance imaging (MRI), in order to explore the magnetic resonance characteristics of the model.
METHODEndometrium tissues were transplanted into left abdominal walls of unmated adult female SD rats. After surgery, pathological changes were observed and MRI scanning was made for the ectopic lesions.
RESULTThe endometriosis rat model was successfully established and the ectopic lesions imaged strong hyperintense on DWI, hypointense on T1WI, hyperintense on T2WI with a clear border, without enhancement on CE-T1 WI.
CONCLUSIONThe lesions can be clearly observed in the MRI images on the endometriosis rat model established by this method, which facilitates repeat experiments and continuous observation studies.

6.Effect of genistein on rat femoral bone metabolic activity in vitro.

This study is to investigate effects of genistein on rat femoral bone metabolic in vitro. Rat femoral tissues was isolated and randomly divided into two groups including control group and genistein (1 x 10(-5) mol x(-1)) group. Determinations of alkaline phosphatase (ALP) activity, calcium content and osteoprotegerin (OPG), type I-collagen (Collagen-I), RANKL, Runx-2 and bone morphogenetic protein (BMP-2) mRNA expression were done by real-time PCR. The results showed that 1 x 10(-5) mol x L(-1) genistein could increase the activity of ALP and contents of Ca, regulate bone metabolism activity of OPG, RANKL, BMP-2, Collagen-I and Runx-2 mRNA expression level. Genistein can significantly modulate bone metabolism related gene expression level of rat femoral tissue in vitro, and can increase calcium content and the activity of ALP.

7.Investigation on the molecular mechanisms of anti-hepatocarcinoma herbs of traditional Chinese medicine by cell cycle microarray.

OBJECTIVETo design DNA microarray and investigate the molecular anti-tumor mechanism of herbs of traditional Chinese medicine.
METHODcDNA microarrays consisting of 56 probes representing 24 human cell cycle genes were constructed, Four anti-hepatocarcinoma herbs including Radix Linderae, Hebra Artemisiae Annuae, Radix Amebiae, Radix Astragli, were chosen. Effects of herbs on SMMC-7721 cell cycle were observed by flow cytometry assay. Effects of herbs on cell cycle gene expression in SMMC-7721 cells were analyzed by comparing hybridization of Dig-Labeled cDNAs from herb-treated cells and cDNAs from untreated cells.
RESULTExpressions of cell cycle geneswere changed in different degrees after herbs treated. Some genes were down-regulated and some genes were up-regulated. The changes in gene expression agreed with the results of flow cytometry assay.
CONCLUSIONThe results suggest that these herbs may have effects on cell cycle and DNA damage checkpoint genes which may be the mechanism of the herbs, and DNA microarray can be used to investigate the biological function of extracts of traditional Chinese medicine.

8.Experimental study on prevention and treatment of bronchial asthma by compound Chinese herbal monomer recipe.

OBJECTIVETo observe the effect of compound Chinese herbal monomer (CHM) recipe, consisted of ligustrazin (3.75 mg/kg x d), baicalin (7.5 mg/kg x d) and ginkgolide (2 mg/kg x d), on airway atopic inflammation and hyper-responsiveness in asthma.
METHODSModel guinea pigs of asthma were randomly divided into three groups, the model group, the CHM group and the cromlyn sodium (CS) group, they were treated by atomizing inhalation with normal saline, CHM and CS respectively. The eosinophil count and eosinophil cation protein (ECP) in blood and bronchial alveolar lavage fluid (BALF), and total cell count in BALF were measured and compared. And the effect of treatment on the airway hyperresponsiveness and pathology among groups were compared.
RESULTSCHM showed significant inhibition on blood eosinophil count and BALF and total cell count in BALF, showing significant difference (P < 0.05 or P < 0.01) as compared with those in the model group. The level of ECP was not different in the various groups. Airway responsiveness determination showed that CHM has significant inhibitory action on it. And the pathology of airway inflammation in the CHM group was significantly milder than that in the model group.
CONCLUSIONThe compound inhalation liquid consisted of ligustrazin, baicalin and ginkgolide, has the anti-asthmatic airway atopic inflammation and depression on airway hyper-responsiveness, suggesting that components of compound CHM recipe could inhibit the multiple pathogenetic asthmatic inflammation from different angles and on multiple targets, so as to cure asthma effectively.

Currently, exploring the "World-class" disciplinary construction is a vital strategy in China. Since 2015, Shanghai Jiao Tong University School of Medicine started the project of establishing two distinguish teams—the academic clinicians and clinical investigators ("Two-hundred Talent" team), to fuel the development of clinical research. After two years of practice, both advantages and bottlenecks were found. The advantage contains: increasing selection rate among talent projects, increasing influential output, elevating capacity in clinical research. The bottlenecks contain: unbalanced disciplinary development, poor awareness of the importance of clinical research, lack of relevant supporting policies. In order to improve the quality of the clinical research and the two talent clinical research teams, institutional innovation, personalized supporting system, establishment of clinical research center, training of clinical research nurses are essential.
10.Study on the Levels and Clinical Significance of Serum miR-150 in Children with Nephrotic Syndrome

Objective To investigate the levels and clinical significance of serum miR-150 in children with primary nephrotic syndrome(NS).Methods Serum samples were collected from 78 NS children and 79 age-and sex-matched control children in Nanjing General Hospital,Jiangsu Province Hospital of TCM and Nanjing Children Hospital from March 2010 to May 2014.Quantitive Real-time PCR(qRT-PCR)assays were used to determine the concentrations of serum miR-150 in NS and control children.The other lipid and renal function parameters including serum TP,ALB,GLO,TC,TG,Urea,Cr,Uric acid and Uric protein were also assessed.Statistical analyzes were used to evaluate the clinical value of serum miR-150 for NS as well as to assess the clinical association between the levels of serum miR-150 and other clinical parameters.Results The ser-um levels of miR-150 were significantly elevated in NS children[101.4(21.29~336.6)fmol/L(F=3.658,P<0.001)]as compared with controls[34.11(5.53~134.2)fmol/L].ROC curve analysis showed that the area under the receiver operat-ing characteristic curve(AUCROC)was 0.892(95% CI=0.843~0.940).Spearman rank correlations analyzes showed that the levels of serum miR-150 were significantly negatively associated with GLO(r=-0.231,P=0.042)and TG(r=-0.233,P=0.040)in NS children.Multiple linear regression analyses showed that serum miR-150 was independently associ-ated with serum ALB levels(β=0.241,P=0.034;adjusted r2=0.046)after adjustment of other related factors.Further-more,multivariate logistic regression analysis showed that serum miR-150 an independent risk factor for NS after adjusting other factors including age and gender[OR=16.07(95% CI=5.35~48.28),P<0.001].Conclusion The serum levels of miR-150 were markedly elevated in NS children and closely associated with with impaired kidney function as well as lipid pa-rameters,and may have the potential as a novel auxiliary diagnosis marker for assessing the development of NS.
