Tumor metastasis is a primary cause of leading to death of tumor patients,and difficult to control.The relationship between primary tumor(PT)and metastasis tumor(MT)is extremely complicated. Considerable animal experiments and clinical data confirm that treatment of PT encourages the growth of MT, but the mechanism is not still fully clear.Some studies show that the density depression of antiangiogenesis factor and immune escape of tumor cell are main reasons for MT accelerated growth.This article reviews the animal experiment,clinical observation and occurrence mechanism of primary tumor treatment promoting tu- mor metastasis.
2.Protective Effect of Redix Astragail to Intestinal Mucosa Barrier in Infant Rabbits with Intestinal Ischemia-Reperfusion

Objective To study the protective effect of redix astragail on intestinal mucosa barrier in infant rabbits with intestinal ischemiareperfusion.Methods Twenty male infant rabbits were divided into 4 groups randomly:sham control group,intestinal ischemia reperfusion(IIR) group(model group),redix astragail group,control group.Redix astragail group and control group intraperitonerlly injected redix astragail 6 mg/(kg?d)or sodium chloride preoperative.Intestinal mucosa and blood plasma superoxide dismutase(SOD) activity,intestinal mucosa tissue degree of injury were observed in infant rabbits with IIR.Meanwhile,the effect of redix astragail on them was studied.Results After IIR,SOD activity in intestinal mucosa and blood plasma obviously decreased,and intestinal mucosa tissue morphous and intestinal mucosa barrier lesion were progressively aggravated.Redix astragail might increase blood plasma and intestinal tissue SDD activity significantly.Conclusion Redix astragail plays an important role in protecting intestinal mucosa barrier in infant rabbits with IIR.
3.Role of interelenkin-17, -6 and endothelin-1 in statins attenuated no-reflow phenomenon of rat acute myocardial infarction and reperfusion

ObjectiveTo study the relevant effect of proinflammatory cytokines interelenkin-17(IL-17), -6 and endothelin-1 (ET-1) on statins attenuating no-reflow phenomenon after myocardial ischemia-reperfusion in rats.MethodsEighteen healthy male Wistar rats were randomly divided into 3 groups according to body weight: sham operation, injury, preconditioning groups. The preconditioning group was given atorvastatin 2 mg·kg-1 ·d-1 and the other two groups were given the same volume of saline once. After 7 days, the rats were anesthetized with an intraperitoneal injection of chloral hydrate, and then the thoracic cavity was opened. The coronary artery of injury group and preconditioning group were ligated for 60 minutes, and then opened for 15 minutes, to establish the rat acute myocardial ischemia-reperfusion model. The sham operation group was was treated with a seam through the coronary artery without ligation. Eleetrocardiogram was checked before ligation, and ligation was carried out for 15, 30, 45 minutes and then reperfusion for 15 minutes. After reperfusion for 15 minutes, the thioflavine S and Even's were injected from femoral venous, then the heart and blood were obtained(keeping left ventricular only). Hearts were flushed with saline and sliced transversely into five to seven sections. Finally, observed at 365 nm wave length the existence of non-fluorescent areas, which was no-reflow zone. The level of serum IL-17, IL-6 and ET-1 was detected by ELISA. Results The electrocardiogram confirmed that the sham operation group had no ischemic damage and the model of myocardial ischemia- reperfusion was established in preconditioning group and injury group. The noreflow phenomenon could be observed under 365 nm wave length in preconditioning group and injury group. The ligated area[LA%, (57.34 ± 11.49)%, (53.08 ± 8.66)%] of injury group and preconditioning group was higher than that of sham operation group(0, all P < 0.05); the area of no-reflow[ANF%, (48.96 ± 6.94)%, (21.37 ±3.35)%] of injury group and preconditioning group was higher than that of sham operation group(0, all P < 0.05),and the ANF% of preconditioning group was lower than that of injury group(P < 0.05) ; the level of serum IL-17,IL-6 and ET-1[(151.67 ± 11.19) × 10-9, (167.89 ± 5.13) × 10-9, (322.37 ± 19.08) × 10-9 g/L] of injury group was higher than those of sham group and preconditioning operation group[(49.75 ± 14.06) × 10-9, (59.32 ± 5.26) ×10-9, (109.9 ± 12.12) × 10-9, (90.45 ± 11.63) × 10-9, (112.47 ± 10.40) × 10-9 and(198.91 ± 27.88) × 10-9 g/L,P < 0.05], the level of serum IL-17, IL-6 and ET-1 of preconditioning group was higher than those of sham operation group(P< 0.05). Conclusionsno-reflow phenomenon is related with IL-17 and ET-1 which can promote the expression of IL-6, statins decreases the expression of IL-17 and ET-1, and then decreases the on-reflow phenomenon.
4.Prospective study of first night effect on 2-night polysomnographic parameters in adult Chinese snorers with suspected obstructive sleep apnea hypopnea syndrome.

BACKGROUNDA prospective 2-night polysomnographic (PSG) study in Chinese snorers was designed to assess the role of the first night effect (FNE) in PSG parameters and the diagnosis of obstructive sleep apnea hypopnea syndrome (OSAHS).
METHODSSeventy-two snorers from two teaching hospitals underwent overnight PSG on two consecutive nights. The night-to-night variability of PSG parameters were recorded and analyzed.
RESULTSSixty-six patients were analyzed. Among all the PSG parameters, only the total time of stage 2 presented a significant difference between two nights: 219.50 (83.50 - 353.50) vs. 215.25 (59.50 - 342.50) (P = 0.000). Subgroup assessment showed a slight night-to-night difference in about 1 - 2 parameters in the group with the apnea-hypopnea index (AHI) ≥ 20 events per hour as well as the group with AHI < 20 events per hour, but there was no night-to-night difference in the AHI in each group. And slighter FNE was found among patients ≥ 40 years old. There was no significant difference in diagnosis of OSAHS. In the decision of severity, a slight difference was found between the two nights with a Kappa value = 0.531.
CONCLUSIONSOnly mild FNE can be found on two consecutive nights of PSG in adult Chinese snorers, but it has no effect on the diagnosis of OSAHS. A single polysomnographic night may be adequate for the diagnosis of OSAHS.

5.Treatment of anterior dislocation of sternoclavicular joint with Kirschner wire minimally invasive fixation.

OBJECTIVETo investigate the safety and efficacy of close reduction and percutaneous needle fixation for the treatment of anterior dislocation of sternoclavicular joint.
METHODSA retrospective analysis was performed with 6 cases of anterior sternoclavicular dislocation by close reduction and percutaneous needle fixation with Kirschner wire treated from January 2001 to February 2009, including 5 males and 1 female aged from 19 to 45 with an average of 28.8 years old. Among the 6 cases, 4 were on right lateral and 2 were on left lateral. The time from injured to treatment was from 6 hours to 12 days (averaged 4.5 days). The clinical effects were evaluated according to Rockwood scoring and the complications were observed.
RESULTSAll 6 patients were followed-up for 3 to 13 months (averaged 6 months). According to Rockwood scoring,the preoperative score was (7.00 +/- 0.89) points, postoperative score was (13.17 +/- 1.72) points; the results showed excellent in 5 cases and good in 1 case. No local infection, postoperative pain,recurrent dislocation,broken needle, and other complications were observed in this study.
CONCLUSIONThe treatment of anterior sternoclavicular joint dislocation with Kirschner wire minimally invasive fixation is an easy, reliable fixation with less complications.

6.Single-chain urokinase-type plasminogen activator (scu-PA) purification by immuno-affinity chromatography.

The only difference of primary structure between single-chain prourokinase (pro-UK or scu-PA) and two-chain urokinase (UK or tcu-PA) is the cleavage of a single peptide bond (Lys158-Ile159) and transform scu-PA into its active two-chain form. A 13-peptide (Thr-Leu-Arg-Pro-Arg-Phe-Lys-Ile-Ile-Gly-Gly-Glu-Cys), which spans the cleavage peptide bond, was synthesized and linked to KLH (Keyhole limpet hemocyanin). The Balb/c mice were immunized by the conjugated protein with proper adjuvant. According to the Kohler and Milstein's methods, a hybridoma cell line G7 secreting monoclonal antibody specific for scu-PA was obtained. The anti-scu-PA McAb, purified from the supernatant of porous microcarrier hybridoma cell culture, was conjugated to CNBr-activated Sepharose 4B to prepare an immuno-affinity chromatography column. The u-PA was purified only by this affinity column from the supernatant of cultivating the u-PA-producing recombinant CHO cell, the u-PA recovery ratio is 90.4%, the purification factor was about 50, with the specific activity of 1.2 x 10(5) IU/mg, the scu-PA ratio in the u-PA product was 96.3%. Compared to immuno-affinity chromatography, the 3-step process for purifying u-PA (cation-exchange column, gel filtration column and benzamidine affinity column) has a u-PA recovery ratio of about 65%, with a specific activity of 1.0 x 10(5) IU/mg, and an scu-PA ratio of about 90%. These results showed that immuno-affinity chromatography is simple to recover u-PA and effective to separate scu-PA from tcu-PA.

7.Association between the number of peripheral blood stem cells and cardiac function index in patients with ischemic heart failure.

OBJECTIVETo observe the association between the number of peripheral blood stem cells (PBSC) and cardiac function index (NYHA grade, left ventricular ejection fraction, LVEF, N-terminal-proB-type Natriuretic Peptide, NT-pro BNP) in patients with ischemic heart failure (IHF).
METHODSA total of 199 patients hospitalized in our department between August 2007 and April 2008 due to IHF were included in this study. NYHA grade, LVEF (measured by echocardiography), plasma NT-proBNP and number of PBSC were analyzed.
RESULTSPBSC number was significantly decreased in proportion with increasing NYHA grade and PBSC number was significantly lower in patients with NYHA IV than that with class II and III (all P<0.01). LVEF was positively correlated with PBSC number (r = 0.35, P = 0.001), NT-proBNP was negatively correlated with PBSC number (r = - 0.39, P = 0.009). Multiple regression analysis showed that LVEF and NT-proBNP were major determinants of PBSC (Beta = 0.14, - 0.25, 95% CI = -0.01 - 0.02, -0.18 - -0.04, P<0.05, P<0.01).
CONCLUSIONPBSC number was significantly correlated to cardiac function in IHF patients.

8.Study on a novel mutation of B glycosyltransferase gene related with an ABx variant.

OBJECTIVETo explore the molecular basis of an individual featuring an ABx variant of ABO blood group system.
METHODSSerological assays were used to characterize the erythrocyte phenotypes and salivary ABH secretors. All of the seven exons and flanking introns of ABO glycosyltransferase gene were amplified with polymerase chain reaction (PCR). And the products were sequenced bidirectionally following enzyme digestion. Exons 6 and 7 were also subcloned and analyzed for haplotypes of the ABO gene.
RESULTSErythrocytes of the proband have expressed a strong A antigen and a weak B antigen, which was identified as a rare ABx variant in addition with other serological features. Nine heterozygous sites in exon 6 (297A/G) and exon 7 (467C/T, 526C/G, 657C/T, 703G/A, 796C/A, 803G/C, 808T/A, 930G/A) of the coding region of the ABO gene were identified. Based on haplotype analysis, one allele was determined as common A102, whilst another was consistent with B101 except for an 808T>A mutation which has resulted in replacement of phenylalanine with isoleucine at position 270 of glycosyltransferase B.
CONCLUSIONThe 808T>A mutation of the glycosyltransferase B gene may decrease the enzymatic activity and result in the Bx variant.

9.Therapeutic effects of intraarticular injection of ligustrazine on knee osteoarthritis.

OBJECTIVE:
To evaluate the therapeutic effects of intraarticular injection of ligustrazine on knee osteoarthritis (OA).
METHODS:
Seventy-one cases of knee osteoarthritis (82 knees) were randomly divided into ligustrazine (LI) group and sodium hyaluronate (SH) group. The patients were intraarticularly injected ligustrazine or sodium hyaluronate once a week for 5 consecutive weeks, and were followed-up for 3 months. Lequesneos protocol for the evaluation of OA severity and activity was used. The therapeutic effects and changes of Lequesneos index were observed after the treatment.
RESULTS:
There was significant decrease in Lequesneos index in SH group after the treatment (P<0.01), but not in LI group (P>0.05). Three weeks later, there was significant decrease in Lequesneos index in both groups after the treatment (P<0.01), with no significant difference between SH and LI group (P>0.05). After the 5-week treatment, the efficacy rate of the LI group was 82.1%, and that of the SH group was 87.2%. No serious toxic or side effect was observed during the treatment and the follow-up.
CONCLUSION
Intraarticular injection of ligustrazine has a therapeutic effect on knee OA. No adverse effect is observed, but it needs long time to take effect.

10.Effect of compound Rhizoma Smilacinus granules on the expression of Bcl-2 and Bax gene in A549 cell lines of non-small cell lung cancers.

OBJECTIVE:
To determine the effect of compound Rhizoma Smilacinus granules (CRSG) on the expression of Bcl-2 and Bax gene in A549 cell lines, and to explore the mechanism of CRSG on non-small cell lung cancers.
METHODS:
The SD rats were randomly divided into 2 groups: one was administrated with CRSG (n=15), and the other with the same dose of distilled water (n=15). The herbage-containing serum was obtained 2 hours after the 6th treatment. non-small lung cancer A549 cell lines were randomly divided into CRSG group, diamminedichloroplatinum (DDP) group and normal control group, which were cultivated with 10% herbage-containing serum, DDP (3g/mL), and 10% SD rats serum respectively. The cultivated cells were collected after 48 hours, and then RT-PCR technique was used to determine the expression of Bcl-2 and Bax mRNA.
RESULTS:
Compared with the normal control group after 48 h, the level of Bcl-2 mRNA and the rate of Bc-2/Bax decreased in the CRSG group and the DDP group, and the level of Bax mRNA increased with significant difference (P<0.01).
CONCLUSION
non-small cell lung cancer A549 cell lines have a high level of Bcl-2 mRNA and a low level of Bax mRNA. CRSG can significantly down-regulate the expression of Bcl-2 mRNA and the rate of Bc-2/Bax, and obviously up-regulate the expression of Bax mRNA, which probably is one of the molecular mechanisms of CRSG in inhibiting the growth of non-small cell lung cancers.
