1.Inhibitory kinetics of paeonol on the activity of mushroom tyrosinase oxidizing L-Dopa

Aim To evaluate the effect of paeonol on the activity of tyrosinase and provide experimental evidence for the treatment of hyperpigmentation disorders. Methods Tyrosinase activity was estimated by measuring the oxidation rate of L-3,4-dihydroxyphenylalanine (L-Dopa). The inhibitory effects of paeonol on the activity of mushroom tyrosinase and Michaelis-Menten kinetics were deduced from the Lineweaver-Burk plots. Results The inhibitory concentration of paeonol leading to 50% enzyme Paeonol is a potential mixed inhibitor of mushroom tyrosinase. The mixed inhibition function may originate from its ability to form a Schiff base with a primary amino group and to chelate copper at the active site of tyrosinase.

3.Presynaptic membrane protein antibody induces epilepsy on rat

Objective To investigate the mechanism of presynaptic protein (Munc18) antibody in inducing epilepsy on rat. Methods Munc18 antibody was continuously micro-injected into CA1 area of hippocampus in Sprague-Dawley (SD) rat (the injection times were nine). The potential epileptic seizure was evaluated according to both electroencephalogram (EEG) and behaviors. HE, Nissle and TUNEL staining were applied on rat brain sections as to detecting the cell damage and apoptosis after tenth week. Results Ten of twelve rats produced epileptic EEG, five cases among these ten rats maintained abnormal EEG after the sixth week since injections. Nine of the above twelve rats had epileptic seizure according to Racine grade one to four, four cases among these nine rats kept seizure after the sixth week since injections. 4.Clinical study of reversed lamellar keratoplasty

Objective To evaluate the therapeutic efficiency of reversed lamellar keratoplasty for bullous keratopathy, Terrien's marginal degeneration and ocular chemical burns.Methods Twenty eyes of patients with bullous keratopathy, twelve eyes with Terrien's marginal degeneration, ten eyes with chemical burns were recruited in this study.Diseased tissues were removed by means of dissection.Reversed lamellae corneal tissues which had been prepared by cryopreservation were thawed and rinsed. Then the prepared posterior corneal lamellae was turned over, endothelium facing upwards. The diameter of the graft should be 0.5~1.0mm larger than that of the recipient bed. The reversed donor posterior lamellae was sutured into the host keratectomy bed with 10-0 nylon sutures continuously or interruptedly.Results Allograft reaction episode occurred only one (chemical burn) of the total 42 eyes enrolled.The grafts of the rest 41 eyes maintained clear or semitransparent for 1~5 years. Graft edema could be observed within 2~4 weeks postoperatively. Postoperative interface separation was not found. For all 20 patients with bullous keratopathy, the symptoms of pain disappeared. The visual acuity improved 2~3 lines of Snellen among eleven of twelve patients with Terrien's marginal degeneration. The visual acuity improved 2~3 lines of Snellen with four patients with chemical burns.Conclusion The technique of reversed lamellar keratoplasty can be applied successfully for tectonic reconstruction of corneal surface such as bullous keratopathy, Terrien' s marginal degeneration and also chemical burns.The effects of therapy was satisfactory.
5.Analysis of preventive measures on controlling of schistosomiasis spreading in Jurong city Jiangsu province

ObjectiveTo investigate the effect of measures on controlling of schistosomiasis in Jurong city Jiangsu province and provide a scientific basis for doing a better job in prevention and control of schistosomiasis.MethodsMonitoring data from 2005 to 2010 was analyzed,including the effect of spraying drugs,changing environment,coupled with health education measures in eradicating oncomelania.Oncomelania areas,positive detection rate of schistosomiasis,and the awareness rates of prevention knowledge of schistosome were observed longitudinally.Correlation of coverage rate of snail controlling and resident positive rate of schistosomiasis was studied.ResultsThere were no infected snails from 2005 to 2010.The densities of living snails and areas with snails decreased year by year.Residents' positive detection rate of schistosomiasis decreased from 0.34%(36/10 551 ) to 0.00%(0/12 372) from 2005 to 2010.The rates of students,and citizens' knowledge of schistosome on prevention of the disease increased from 88.28%(128/145) and 86.44%(204/236) to 96.58%(141/146) and 92.83%(246/265) from 2005 to 2010,respectively.There was a negative correlation between oncomelania areas and drug coverage areas(r =- 0.910,P < 0.05 ); there was no correlation between oncomelania areas and coverage rate of snail control by environmental modification (r =- 0.404,P > 0.05 ); there was no correlation between positive rate of schistosomiasis in resident and coverage rate of snail control with molluscisides,and coverage rate of snail control by environmental modification (r =0.107,- 0.496,all P > 0.05).ConclusionsSnails areas in Jurong city is decreased year by year.Residents' positive detection rate of schistosomiasis infection is maintained at a low level.The effects of oncomelania-controlling,health education,and chemotherapy are remarkable.
6.p38 MAPK signaling pathway is involved in the regulation of receptor-mediated endocytosis

AIM:To investigate the role of p38 MAPK signaling pathway in the receptor-mediated endocytosis.METHODS:The effects of p38 MAPK on the receptor-mediated endocytosis were observed by using Alexa 594-conjugated Transferrin,in the presence of p38 specific inhibitor SB203580 or ERK pathway specific inhibitor PD98059,or using p38 knockout techniques.RESULTS:In the process of receptor-mediated endocytosis,p38 was activated by phosphorylation.Furthermore,the receptor-mediated endocytosis was inhibited by pretreatment with SB203580 or p38 knockout,while pretreatment with PD98059 had no effect on this process.CONCLUSION:p38 MAPK signaling pathway plays a role in the regulation of receptor-mediated endocytosis.
7.The pathogen surveillance of rubella virus in Beijing from 2007 to 2010

Objective To clarify the pathogen for rubella in Beijing from 2007 to 2010. Methods Beijing Center for Disease Preventipn and Control ( CDC ) collected the specimens (including blood, urine and throat swab specimens) frqm clinically diagnosed rubella cases for serological test and virus isolation. The nucleic acid of rubella virus in clinical specimens and isolations was detected by real-time PCR. Results Fifty-five out of 99 blood specimens were positive for anti- rubella IgM. Fifty-one out of 99 clinically diagnosed rubella cases were confirmed as rubella cases by virus isolation. Seventy-two were confirmed as rubella virus infections with real-time PCR method for detecting the nucleic acid of rubella virus in clinical specimens. Compared with the sequences of reference strains of rubella virus, all of detected rubella virus belonged the IE gene type. Conclusion This study indicates that IE gene type virus was the predominant endemic rubella virus in Beijing.
8.Treatment of intraarticular unstable distal radial fractures with locking compression plate in 30 patients

Objective To explore the method and evaluate the effect of locking compression plate (LCP)fixation of intraarticular unstable distal radial fractures.Methods Thirty patients with intraarficular unstable distal radial fractures were treated with LCP.According to AO/ASIF classification,there were 6 cases of type B2 fractures,7 cases of type B3,7 cases of type C1,8 cases of type C2,2 cases of type C3.Results After 6 to 24 months(average 12 months)following up,there were 15 cases graded as excellent,10 eases as good,3 cases as fair,and 2 cases as poor,according to X ray and wrist function assessment.The overall satisfaction rate was 83.3%.Conclusion Volar LCP fixation is a safe and effective treatment for intraarticular unstable distal radial fractures.It could effectively prevent loss of reduction,avoid tendon irritation and other complications if selecting implant angle for locking screw according to the specific intraoperative circumstances.
9.A modified tricuspid annuloplasty for functional severe tricuspid regurgitation

ObjectiveTo retrospectively analyze clinical data of patients who has left-side valvular disease combined with severe tricuspid regurgitation and evaluate the effect of our modified tricuspid annuloplasty with enforcement of artificial felt strip.Methods76 patients who had left-side valvular disease combined with severe tricuspid regurgitation received operations between Jan.2008 and Jun.2010.The average age of the patients was 53.5 years old (32 male and 44 female).Besides the severe tricuspid regurgitation, other combined cardiac impairments included mitral valvar disease (52 cases), aortic valvar disease(5 cases), double valvar disease(19 cases) and left atrial thrombosis(22 cases).6 patients had grade II cardiac function according to the NYHA criteria, while 47 and 23 were in grade III and IV, respectively.Other signs included cyanosis(5cases), jaundice(11 cases), neck vein engorgement(48 cases) , ascites(22 cases), hepatomegaly(41 cases) and pitting edema in the lower limbs(68 cases).The concomitant operative procedures included mitral valve replacement in 52 patients,aortic valve replacement in 5 patients, double valve replacement in 19 patients, removal of left atrial thrombus in 22 patients,left atrium folding in 21 patients and left atrium appendage suture in 68 patients.Left-sided valve disease were corrected first,TAP was performed on the beating heart after the heart had been defibrillated.The anteroseptal commissure was plicated first.A double-armed 3-0 pledgeted suture was taken through the base of the septal leaflet, 5-6 mm from the commissure, extending along the annulus, and out from the point in the anterior annulus 10-12 mm from the anteroseptal commissure.Both ends of the suture was tied until the two Teflon pledgets approximated each other near the commissure.Then a semicircular De Vega type of plicating with a 3-0 prolene was taken, starting just from the anterior annulus near the anteroposterior commissure, and extending clockwise to a point just cephalad to the posteroseptal commissure.The suture was tied with positioning a 27-29 mm valve siser across the tricuspid valve.At last, a 3-5 mm width felt strip was prepared and was sutured to the plicated posterior annulus region with interrupted mattress sutures of 2 to 3 2-0 prolene.A favorable result was considered when TR was not marked by saline injection.Echocardiography was routinely examined one week postoperatively and patients were followed up 6 month after discharge.ResultsThere is no death in all patients.The CVP diminished significantly from 16mmHg preoperatively to 8mmHg postoperatively (P = 0.0021).The systomic pulmonary pressure diminished from 59 mmHg preoperatively to 41 mmHg postoperatively (P = 0.038).Echo one week postoperative showed no tricuspid regurgitation in 56 patients and mild in 18 patients, while 2 had moderate tricuspid regurgitation.The diameter of right atrium diminished significantly postoperatively, too.The ejection fraction was improved even though there was not significant difference as compared with preoperative data.The cardiac function of all patients improved and the signs of right heart failure were alleviated or disappeared.Follow up 1 to 36 months showed no change of the regurgitation except for one become moderate from mild when discharged.No hepatic congestion or edema was observed in all patients.ConclusionThese new modifications make the technique more selective in the remodeling of the tricuspid annulus.It could achieve better coaptation of the anterior leaflet with the others, successful annular reduction, better maintenance of the contractile property of the tricuspid ring, better distribution of pursing force in the more dilated region.It could prevent the tear of the endocardium in the posteroseptal region in the long period of time postoperatively.
10.A new method of distinguishing weak and overlapping signals of proton magnetic resonance spectroscopy.

In this paper, a new method of combining translation invariant (TI) and wavelet-threshold (WT) algorithm to distinguish weak and overlapping signals of proton magnetic resonance spectroscopy (1H-MRS) is presented. First, the 1H-MRS spectrum signal is transformed into wavelet domain and then its wavelet coefficients are obtained. Then, the TI method and WT method are applied to detect the weak signals overlapped by the strong ones. Through the analysis of the simulation data, we can see that both frequency and amplitude information of small-signals can be obtained accurately by the algorithm, and through the combination with the method of signal fitting, quantitative calculation of the area under weak signals peaks can be realized.
