1.Schwann cells induced neuronal differentiation of adipose-derived stem cells (ADSCs)

Objective To investigate the function of SD rats Schwann cells on the growth and differentiation of adipose-derived stem cells(ADSCs) when they are co-cultured in Transwell culture system. Methods The ADSCs harvested from SD rats were divided into 3 groups. Group A: ADSCs and Schwann cells were cultured in Transwell indirect co-culture system. Group B: ADSCs were induced by β-BME and BHA. Group C: a contrast group. The morphology of ADSCs was observed and immunohistology was pedormed. Results ADSCs of group A and group B were partly differentiated to the cells which had long neurite,and had a positive result in the NF staining, but negative in the GFAP staining. Group A shows no great difference in neurite length compared with group B(P < 0.05), and no great difference in cell quantity compared with group C (P < 0.05). Conclusion Schwann cells of SD rat have a growth supportive and neuronal differentiation inducing effect on ADSCs.
2.Electransfection with telomerase catalytic subunit(hTERT) enhances proliferative capacity of Schwann cells

Objective To transduct with human telomerase reverse transcriptase(hTERT) to Schwann cells via electransfection technique to prolong the life span and enhance the proliferative capacity of Schwann cells.Methods The hTERT RNA was derived from esophagus cancer tissue and pcDNA3.1-hTERT vector was built.With electransfection technique, we tmnsfected vector into ADSCs.The PCR,TRAP-PCR, and PI-annexin V were tested to prove the expression of hTERT in ADSCs.Results After transduction the hTERT RNA in Schwann ceils, TRAP-PCR test, PI-annexin V dying and S100 dying was positive.Conclusion Telomerase catalytic subunit(hTERT) is transduct into Schwann cells, and it can to enhance the prolifertative capacity.
3.Evaluation of clinical characteristics of genitourinary syndrome in postmenopausal women

One hundred and ten postmenopausal women,complaining vaginal discomfort,recurrent urinary tract infections or painful intercourse and visiting the hospital between March 2014 and February 2015 were enrolled in the study.Among them,58 patients received vaginal administration of estriol estriol ointment (group A) and 52 patients received conventional treatment (group B).There were no significant differences in mean age of menopause and mean menopause duration between two groups.The clinical symptoms,vaginal health scores,pelvic organ prolapse were observed after treatment.After 6-month treatment,the female vaginal health score was significantly improved in women group A,the rate of urinary orifice was reduced (78% vs.7%),and the rate of pelvic organ prolapse was decreased (36% vs.21%).The geritourinary syndrome of menopause is a new term for vulvovaginal atrophy with clinical characteristics of multi-system changes in postmenopausal women,including atrophic vaginitis,female urogenital diseases,and pelvic organ prolapse.The new diagnostic criteria are needed for evaluation and management of geritourinary syndrome.
4.Influence of trimetazidine hydrochloride on plasma brain natriuretic peptide and 6min walking distance ;in aged patients with chronic congestive heart failure

Objective:To observe influence of trimetazidine on cardiac function in patients With chronic heart failure (CHF).Methods:According to number table method,a total of 70 CHF patients accorded With inclusion standards Were randomly and equally divided into trimetazidine group and routine treatment group.According to patient's condition,routine treatment group received digitalis,diuretics,angiotensin converting enzyme inhibitors etc CHF routine freatment.Trimetazidine group additionally received trimetazidine based on routine treatment.The treat-ment period Was four Weeks.Plasma brain natriuretic peptide (BNP)level Was measured and patients received 6min Walking test (6MWT)before and after treatment.Results:Compared With before treatment,there Was significant decrease in plasma BNP level in tWo groups after treatment (P<0.01),compared With routine treatment group, there Was significant increase in decreasing amplitude of BNP level [(655.89±135.61)pg/ml vs.(715.60±181.22) pg/ml,P<0.05]in trimetazidine group;compared With before treatment,there Was significant increase in 6min Walking distance (6MWD)in tWo groups after treatment (P<0.01),compared With routine treatment group,there Was significant increase in increasing amplitude of 6MWD [(181.46±51.16)m vs.(226.06±65.18)m,P<0.01] in trimetazidine group.Conclusion:Treatment trimetazidine can significantly reduce plasma brain natriuretic peptide level and improve heart function based on routine treatment in patients With heart failure.
5.Gefitinib in first-fine therapy for 34 cases of advanced non-small cell lung cancer

Background and purpose: It has been proven that gefitinib can be safely and efficiently used to treat advanced non-small cell lung cancer (NSCLC) as a molecule targeted drug. This research was aimed to investigate the efficacy and toxicity of gefitinib as the first-line therapy for advanced NSCLC. Methods: A total of 34 pathologically-confirmed NSCLC patients who were not willing to receive or tolerate traditional cytotoxic drug chemotherapy were enrolled into the study. Gefitinib was orally administered 250 mg daily until disease progression or the occurrence of intolerable toxicity. Results: The objective response rate of gefitinib was 29.4%. The disease control rate was 61.8%. The rate of symptom relief was 47.1%. The median progression-free survival was 3.0 months. The median overall survival was 10.2 months. One-year survival rate was 35.3%. The objective response rate of nun-smoker was higher than smoker (P=0.023). The disease control rate for the patients with rash toxicity after administration of the drug were higher than those without rash (P=0.005). Logistic regression showed that rash was an independent disease control factor (P=0.003). The most common drug-related adverse events were rash and diarrhea. Conclusion: Gefitinib provided another choice to patients who are unwilling or unable to be treated by chemotherapy.
7.Erlotinib versus vinorelbine therapy in chemotherapy-naive elderly patients with advanced non-small-cell lung cancer and EGFR mutation: a randomized, controlled trial

Objective To compare the efficacy and adverse effects of erlotinib versus vinorelbine naive patients with advanced non-small cell lung cancer (NSCLC) and epidermal growth factor receptor (EGFR) mutation.Methods Totally 46 elderly patients with histologically confirmed advanced NSCLC and EGFR mutations (exon 19 dclction or L858R point mutation) were enrolled.Patients were randomly divided into two groups:erlotinib group (43 cases,150mg per day until disease progression or unacceptable toxicities) and control group (21 cases,vinorelbine-based chemotherapy,single vinorelbine chemotherapy or vinorelbine-based double chemotherapy).Results Response rates and disease control rates were significantly improved with erlotinib compared with vinorelbine (78.6% and 88.1% vs.38.1% and 61.9%,respectively,P< 0.05).There was a significant difference in median progression-free survival (11.6 months vs.5.6 months,P<0.05),while no statistical difference in median overall survival with erlotinib compared with vinorelbine (19.0months vs.16.5 months,P=0.193).The most frequent adverse effects were grade Ⅰ or Ⅱ and no patients stopped treatment due to adverse effects and no drug-relatcd death.The primary adverse effects were skin rash (71.4%),diarrhea (31.0%)and liver dysfunction (23.8%) in the erlotinib group and neutropenia (66.7%),nausea or vomit (47.6%),anemia (42.9%),platelet decline (33.3%),constipation (33.3%) and peripheral neuritis (23.8%) in the vinorelbine group.Vinorelbine group versus erlotinib group have more 3-4 level adverse reactions (15/21 vs.7/42)(x2=1.69,P=0.193).Conclusions Erlotinib treatment has advances in PFS,ORR and DCR and tolerability compared with vinorelbine-based chemotherapy in elderly patients with advanced NSCLC and EGFR mutation,while overall survival is in no difference.Erlotinib may be a reasonable first-line treatment option for elderly patients with advanced NSCLC and sensitive EGFR mutation.
8.Calpastatin gene is not associated with late onset sporadic Parkinson' s disease in Chinese Han population

Objective To explore the association between late-onset sporadic Parkinson' s disease (PD) and single nucleotide polymorphisms (SNPs) of Ca2+-dependent protease calpain inhibitor calpastatin (CAST) gene in a Chinese Han population.Methods 370 evaluable patients (221 male,149 female) with PD (mean age 65.2 ± 8.5 years) and 390 neurologically healthy controls (208 male,182 female) matched for gender,ethnicity,and area of residence.PD cases were identified from the PD cohort of the Chinese National Consortium on Neurodegenerative Diseases (www.chinapd.cn).A total of 24 tag-SNPs were genotyped capturing 95% of the genetic variation across the CAST gene.Results There was no association found between any of the polymorphisms and PD in all models tested (co-dominant,dominant-effect and recessive-effect (P > 0.05)).Similarly,none of the common haplotypes was associated with a risk for PD(P > 0.05).Conclusion Results show no significant association between the CAST gene polymorphisms and late onset sporadic PD in the present population.
9.Intraoral approach for the correction of prominent mandibular angle with masseter muscle hypertrophy

Objective To evaluate the surgical corrective results of prominent mandibular angle with masseter muscle hypertrophy by using intraoral approach. Methods One hundred and twenty three cases with various degrees of prominent mandibular angle with masseter muscle hypertrophy were treated through intraoral approach. The basic surgical procedures included masseter muscle reduction (type Ⅰ), mandibular angle osteotomy (type Ⅱ) and angle-splitting osteotomy (type Ⅲ). The type Ⅰ was completed in 4 cases, type Ⅱin 16 cases, type Ⅲ + type Ⅰ in 56 cases, type Ⅲ in 19 cases, type Ⅲ + type Ⅰ in 9 cases, type Ⅲ + type II in 12 cases, type Ⅲ + type Ⅱ+ type Ⅰ in 7 cases. Genioplasty was simultaneously performed in 69 cases.Results The overwidth of the lower face was effectively corrected. The cosmetic results, as determined by both patients and surgeons, were good. No complications, such as facial never injury, or inferior alveolar never injury, occurred in any patients. Conclusions In order to reach good cosmetic results at the patients' lateral view and frontal view, considerations should be determined according to the degrees of the deformities and patient's desires, to choose suitable surgical procedures.
10.Study on 1H-MRS of prefrontal lobe and executive functions in patients with post-concussion syndrome

Objective To identify the metabolic levels in prefrontal lobe in patients with post-concussion syndrome by proton magnetic resonance spectroscopy (1H-MRS), and to explore the relationship between metabolic levels and executive function. Methods The study was conducted in 40 patients with post-concussion syndrome and 20 normal controls. 1H-MRS on prefrontal lobe was performed in patients and controls, the NAA, Cho and Cr were measured and the ratios of NAA/Cr, Cho/Cr and NAA/(Cho + Cr) were determined. They were also evaluated executive functions by verbal fluency test (animal), Wisconsin Card Sorting Test (WSCT) and Tower of Hanoi (TOH). Results Compared with normal controls, the patients with post-concussion syndrome were significantly lower NAA/Cr and NAA/(Cho+Cr) ratios in left prefrontal lobe (P < 0.05). The NAA/Cr ratio in left prefrontal was significantly positive correlated with total scores of verbal fluency (r = 0.66, P < 0.05), categories of WSCT (r = 0.54,P < 0.05) and total score of TOH(r = 0.58, P < 0.05). The NAA/Cr ratio was significantly negative correlated with total errors (r = -0.53, P < 0.05) and persistent errors (r = -0.47, P < 0.05) of WSCT and mean executive time of TOH(r = -0.67, P < 0.05). Conclusions The metabolic levels of NAA in left prefrontal lobe in patients with post-concussion syndrome is significantly decreased , it is one cause of impaired executive functions.
