2.Indoor Air Pollution in Chaffy Dish Restaurant

Objective To investigate the indoor air pollution of chaffy dish restaurant using different kinds of fuels or utensils. Methods The concentration of carbon monoxide (CO),carbon dioxide (CO2) and inhalable particle (IP) were determined 30,60,90,120 and 150 minutes respectively before and after the chaffy dish restaurants were opened in Mar. 2006. Results Before the restaurants were opened,the concentrations of CO and CO2 except IP did not exceed the standard limits. After opened,the concentrations of CO,CO2 and IP in chaffy dish restaurants which use different kinds of fuels or utensils exceed the standard limits in different degrees except the concentration of CO in chaffy dish restaurant using the infrared utensil. The concentration of CO in chaffy dish restaurant that used charcoal as fuel was the highest and more than the standard limits by 7 times. The concentrations of CO,CO2 and IP in chaffy dish restaurants which used the infrared utensil were obviously lower than those in which used the common liquefied gas utensil. The concentration of CO in chaffy dish restaurants which used charcoal as fuel was obviously higher than those used liquefied gas utensils. Conclusion The indoor air pollution of chaffy dish restaurants is extremely severe and it has the potential harm to the health of the customers in restaurants.
3.Arsenic trioxide induced osteosarcoma cell apoptosis in vitro

Objective The clinical results of leukemia improved significantly due to the application of arsenic trioxide(As 2 O 3 ).This compound was used in inducing apoptosis on osteosarcoma cell lines and its mech anism was explored.Methods Human osteosarcoma MG-63cell line was cultured and treated with0.25,0.5,1,2,4and8?mol/L As 2 O 3 .The cell line was measured at24,48,72,96and120h after As 2 O 3 infiltration.MTT assay,phase contrast light microscopy,fluorescence staining,TUNEL,flow cytometry anal-ysis and RT -PCR were used to investigate the inducing apoptosis and inhibitative effect of As 2 O 3 on os-teosarcoma MG-63cell line.Results As 2 O 3 could inhibit the reproduction of osteosarcoma cell line MG-63significantly,the inhibitive rates of MG-63cell line treated with As 2 O 3 over1?mol/L reached70%.Un -der phase contrast light microscopy and fluorescence staining,osteosarcoma cells cultured with As 2 O 3 pre sent -ed typical apoptotic changes.The majority of cells cultured with As 2 O 3 were positive with TUNEL.Flow cy tometry analysis showed a time-dose dependent apoptosis rate and a remarkable G 2 /M phase arrest.The ex pression of c-myc was decreased in MG-63cells treated with As 2 O 3 .Conclusion As 2 O 3 is able to in duce osteosarcoma apoptosis effectively.The expression of decreasing c-myc is an important mechanism in induc-ing osteosarcoma MG-63cells apoptosis.
4.Expression of hTERT in benign and malignant gastric diseases and its clinical significance

Objective: To investigate the expression of human telomerase reverse transcriptase (hTERT)in benign and malignant gastric diseases and its clinical significance. Methods: The expression of hTERT in 126 gastric tissue specimens obtained by surgery and endoscopy were studied by SP immunohistochemical method. The specimens included 10 cases of nomal mucosa,26 of chronic gastritis, 22 of benign ulcer,5 of leiomyoma, 3 of hyperplastic polyps, 50 of gastric cancer, 2 of maligmant lymphoma and 8 of sarcoma. In precancerous lesions,there was 54 cases of atypical hyperplasia,including 18 low grade, 25 of intermediate and 11 high grade. The positive expression rates and relations of clinical characters were analyzed. Results: hTERT was not or seldom expressed in normal and benign gastric diseases, while it was highly expressed in malignant lesions.There was a significant difference between them(P
7.An in vitro skin sensitization test based on THP-1 cell line

Objective To establish an in vitro skin sensitization test,human cell line activation test (h-CLAT),based on THP-1 cell line (a human acute monocytic leukemia cell line),and to assess the sensitizing potency of plant raw materials of chemical and cosmetic products by this in vitro skin sensitization test.Method THP-1 cells were cultured in vitro and exposed to 11 reference skin sensitization chemicals and 9 samples,by monitoring the cell viability,cell surface marker CD54 /CD86 and relative fluorescence intensity of cells surface after the cells was exposures to the substances,and to discover whether there is a positive reaction.At the same time,Buehler test was used to validate the results of samples tested by h-CLAT.Results 11 reference chemicals were distinguished correctly by h-CLAT.Among the 9 samples tested,7 samples were recognized as negative sensitizer and 2 plant extracted substances were identified as suspicious skin sensitizer.The qualitative classification of the 9 samples by h-CLAT test was consistent with the results obtained by animal test.Conclusions The h-CLAT-in vitro test can be used to replace some animal tests for the prediction of soluble skin sensitizing substances.
8.Expression of CD40 in spleen in cirrhotic portal hypertension and its clinical significance

ObjectiveTo investigate the expression and clinical significance of CD40 in the spleen of cirrhotic portal hypertension. MethodsExpression of CD40 was determined with S-P immunohistochemistry in spleen specimens from 50 cases of cirrhotic portal hypertension and 15 healthy individuals. ResultsThe CD40 positive rates in normal spleen and cirrhotic spleen were 86.7% and 36.0%, respectively. There was a significant difference between the 2 groups (P<0.05). There was a negative correlation between the expression of CD40 and Child grades of liver function and cirrhotic splenic types(P<0. 05). Conclusion CD40 might reflect the changes of splenic immune function,which might be one of more exact clinical examination indexes of splenic immune function.

10.Activation of MAPK/ERK and MAPK/P38 is Essential for Proinflammatory Response by Chlamydia trachomatis

Chlamydial infection in human urogenital tract induces inflammation and causes tissue damage and scarring. It is thought that cytokine production by the Chlamydia-infected cells plays a key role in chlamydial disease processes. Although many cytokines have been detected during chlamydial infection, little is known about the molecular mechanisms on how Chlamydia triggers and sustains the inflammatory cytokine cascades. In the current study, chlamydial infection of the human cervical epithelial cell line HeLa cells can induce the production of IL-8, IL-1α, IL-1β and IL-6. Using inhibitors for probing intracellular kinase signaling pathways required for the Chlamydia-induced cytokines, it was found that the Chlamydia-activated MAPK / P38 pathway is required for the chlamydial induction of IL-1α and IL-6 while both the Chlamydia-activated MAPK/ERK and MAPK/P38 pathways contribute to the production of IL-8.
