1.Alteration of apoptotic susceptibility and bcl-2 gene in malignant transformation of human bronchial epithelial cells

AIM: To demonstrate the susceptibility of cell apoptosis varies during the progress of cell malig- nant transformation from human being in vitro. METHODS: A SV40T - transfected human bronchial epithelial im- mortalized cell line (called M) was selected in this work, which has acquired some characteristics of malignant trans- formation at the later passage. The alterations of apoptosis and bcl- 2, P53 genes between early and later passage of M cells were investigated by means of TDT labeling in situ, chromosome FISH, RNA and protein testing, etc. RE- SULTS: Incidence of apoptosis induced by cis - platin was significantly lower in later than in early passages of M. Levels of bcl - 2 mRNA and protein in later passages were higher than early passages of M, and overxpression of bcl -2 was accumulated following the development of cellular malignancy. P53 protein level was as high in early as in later passages. CONCLUSION: Overexpression of bcl - 2 decreases the cellular sensitivity to apoptotic inductors plays an important role during progress of carcinogenesis in human bronchial epithelial cancers. The inactivation of P53 protein in the SV40 - T transfected M cell line may be one of reasons of bcl - 2 overexpression, but not associated with the accumulation of bcl - 2 expressed level during cell transformation.
3.Effects of navigation and robot technology on percutaneous ultrasound-guided liver tumor ablation: Accuracy and feasibility of clinical application

OBJECTIVE: To assess the effects of navigation and robot technology on percutaneous ultrasound-guided liver tumor ablation. METHODS:The liver model tumors were placed inside excised calf livers, and the needle was guided into the tumor using either conventional ultrasound-guided method or two new methods-3D navigation and a method based on a robotic assistant. Totally 40 experiments were performed, and the distance (all methods) from the tip of the needle to the planning point in the tumors was measured.RESULTS: Based on the measurements, the robotic assistant performed better than both the 3D navigation and the conventional ultrasound-guided method. While, the 3D navigation performed better than the conventional ultrasound-guided method.CONCLUSTON: This study demonstrates navigation combined with robot technique is feasible to improve the precision of deploying ablation needles, future studies are warranted to optimize the system to meet requirements in clinical practice.
4.Research advances on second primary malignancies of oral cavity following radiotherapy for nasopharyngeal carcinoma

Radiotherapy is the primary treatment modality for nasopharyngeal carcinoma(NPC) which can effectively control the disease.Oral cavity,anatomically near nasopharyngeal region is the main area for the occurrence of complication of radiotherapy.Second primary malignancy (SPM) in oral cavity is an important factor interferencing NPC patients survival rate.The etiology of oral SPM is unclear and,the prognosis is poor.The research of it is still in exploration.
5.Relationship between different degrees of asymptomatic carotid artery stenosis and cognitive impairment

Objective To investigate relationship between different degrees of asymptomatic carotid artery stenosis and cognitive impairment. Methods The cognitive function were evaluated with the montreal cognitive assessment scale (MoCA) in 80 patients with asymptomatic carotid artery stenosis and 56 controls, the total MoCA scores and subprojects scores were compared between the patients with different degrees of ACS and controls. Results The total MoCA score, visual space and executive function, attention and delayed recall had significant difference among the mild, moderate, severe ACS patients and control group (all P < 0.05). There was no statistical significance of the total MoCA score and the scores of all subprojects between the patients of mild ACS patients and the control group (all P > 0.05). The total MoCA score and the scores of visual space and executive function , attention and delayed recall in moderate and severe ACS patients were significantly lower than the control group and mild ACS patients (P < 0.05). Furthermore, the severe ACS patients, compared with the moderate ACS patients , performed worse on the cognitive tasks which involved visual space and executive function, attention and delayed recall (P < 0.05). Conclusions Cognitive function is impaired in the patients with moderate and severe ACS,the neuropsychological characteristics are mainly impaired in visual space and executive function, attention and delayed recall.
6.Effect of implant structural changes on the cortical bone stress distribution and peak of the implant-bone interface:a three-dimensional finite element analysis

BACKGROUND:The biomechanical effect of the implant-bone interface is one of the most important factors for bone resorption. The new structure of the periodontal-ligament-like implants may improve the distribution of the interfacial stress. OBJECTIVE:To discuss the effect of the internal structure changes of traditional implants on the cortical bone stress distribution and peak at the implant-bone interface under different occlusal load conditions, so as to provide a theoretical basis for the optimization design and clinical application of new structure implants. METHODS:Two kinds of digital models, new structure implant (model A) and non-threaded cylindrical implant (model B), were established by Pro/ENGINEER software. Variations of the stress peak and stress distribution of implant-bone interface cortical bone area under the same bone and force environment were analyzed using Ansys software. RESULTS AND CONCLUSION: Under a vertical loading, the stress peak under different forces was reduced by 17.54% in model A compared with model B; under a 45° loading, the stress peak of model A was reduced by 2.59% compared with model B, and it showed an evident tendency of high stress area focusing to the buccal side of model B. Under the chew-simulation loading, the stress peak of model A was lower than that of model B. The biggest difference (0.353 2 MPa) appeared atβ=12°(β is the angle of force direction and the implant axis), and it gradualy reduced atβ > 12°. At the same time, model A had a wider range of application degree compared with model B in two quantitative indicators, including optimal peak stress of promoting bone tissue growth and stress peak of maintaining healthy bone tissue. These results suggest that the optimized structure of implants contributes to improve the cortical bone stress distribution at the implant-bone interface, decrease the peak stress, and reduce the risk of cortical bone absorption in a wider range.

Bacterial biofilm is responsible for a wide variety of nosocomial infections.Many chronic infectious diseases are closely correlated with biofilm formation in the clinic.As the biofilm growth,bacterial clusters detachment and dispersal from mature mixed-species biofilms will result in repeated acute infections.Here reviews three distinct biofilm dispersal strategies: swarming dispersal,clumping dispersal and surface dispersal.Many factors,such as heredity and increased fluid shear in the environment,contribute to this complex and dynamic process.Fundamental investigation upon biofilm will benefit clinical therapy of biofilm-associated infections.
9.The Diagnosis and Differential Diagnosis of Atypical Persistent Hyperplastic Vitreous by Color Doppler Ultrasonics

Objective To explore the color dopplor imaging feature of atypical persistent hyperplastic primary vitreous(PHPV) and its value in differential diagnosis of white pupil. Method Color doppler ultrasonic detection was used to observe various types of white pupil, and the follow up analysis was performed. Results Among 15 eyes of 12 patients,3cases were bilaterally abnormal.Typical PHPV imaging was displayed in 11 eyes and atypical PHPV imaging was showed in 3 eyes.One eyes with retinoblastoma was wrongly diagnosised as PHPV.The arterial flow signal was detected in the central part of hyperplastic vitreous body in 11 eyes. Conclusion The arterial flow signal in hyperplastial vitreous body was a essential evidence in the diagnosis and differential diagnosis of PHPV.
10. Bioanalysis and pharmacokinetics of eight active components from huanglian jiedu decoction in rat plasma by LC-ESI-MS/MS method

Objective: To develop a sensitive and rapid liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) method for the simultaneous determination of wogonin, coptisine, berberine, palmatine, jatrorrhizine, phellodendrine, magnoflorine, and wogonoside in rat plasma and to evaluate the pharmacokinetic characteristics of Huanglian Jiedu Decoction (HJD). Methods: LC separation was performed on an Acquity HSS T3 column (100 mm × 2.1 mm, 1.8 μm) using gradient elution with the mobile phase consisting of acetonitrile and 0.1% formic acid water. The detection was accomplished by using positive electrospray ionization in multiple-reaction monitoring mode. Plasma samples were pretreated by protein precipitation. Results: The method showed a good linearity over a wide concentration range (r2 > 0.99). The lower limits of quantification were 0.20 ng/mL for coptisine and phellodendrine, 0.48 ng/mL for berberine, 0.10 ng/mL for jatrorrhizine, 0.32 ng/mL for magnoflorine, 0.30 ng/mL for palmatine, and 4.80 ng/mL for wogonin and wogonoside, respectively. The intra- and inter-day precision of the analytes was less than 12.11%, while the accuracy was between -14.46% and 4.86%. The mean recovery of all the analytes ranged from 93.10% to 110.91%. Conclusion: This validated method offers the advantages of high sensitivity. It is successfully applied to evaluating the pharmacokinetic properties of HJD. © 2014 Tianjin Press of Chinese Herbal Medicines.
