2.Association of Polymorphisms in Atrial and Brain Natriuretic Peptide Gene with Essential Hypertension

Objective To investigate the associations of atrial natriuretic peptide(ANP)rs5065 and brain natriuretic peptide(BNP)rs198389 gene polymorphisms with human essential hypertension(EH).Methods A total of 976 patients with EH and 976 age-and sex-frequency matched normotensive(NT)control subjects were collected.The ANP rs5065 and BNP rs198389 polymorphisms were genotyped by real-time quantitative polymerase chain reaction(real-time Q-PCR)method--Taqman probe.Results There was significant difference in frequency distribution of AA and AG+GG genotypes,A and G allele of BNP rs198389 between the EH and NT groups(P<0.05).The frequency of AG+GG genotype and G allele was significantly higher in the EH group(30.5% and 17.3%)than in the NT group(26.1% and 14.1%).There was no difference in distribution of ANP rs5065 polymorphism between EH and NT groups(P>0.05).By unconditional Logistic regression analysis of rs198389,before adjusted for the EH risk factors,AG+GG genotype presented a significantly higher risk of EH(P=0.04)than AA genotype,and this risk was still significant after adjusted for the EH risk factors(P=0.02).G allele had more significantly increased risk of EH than A allele(P=0.04).However,there was no significant difference before or after adjusted for the EH risk factors in unconditional Logistic regression analysis of rs198389.Conclusion rs198389 polymorphisms of BNP gene may be associated with EH,but rs5065 polymorphisms of ANP gene may be not associated with EH.

4.Efficacy observation of post-stroke dysphagia treated with acupuncture at Lianquan (CV 23).

OBJECTIVETo verify the clinical efficacy on post-stroke dysphagia treated with acupuncture at Lianquan (CV 23).
METHODSOne hundred and eighty patients were randomized into an acupuncture A group, an acupuncture B group and a rehabilitation group, 60 cases in each one. On the basis of the conventional medication, in the acupuncture A group, acupuncture was applied at Lianquan (CV 23); in the acupuncture B group, acupuncture was applied at Hegu (LI 4) and Neiguan (PC 6) and in the rehabilitation group, the swallowing rehabilitation training was adopted. The treatment was given once a day, 5 times a week, and the 4 weeks of treatment was required in all of the groups. The national institute of health stroke scale (NIHSS) and TV X-ray fluoroscope swallowing scale (VFSS) were used to evaluate neurologic deficit and swallowing function before and after treatment in the patients of each group. The morbidity of pneumonia and clinical efficacy were compared among the groups.
RESULTSThe scores of NIHSS and VFSS were improved apparently after treatment in the patients of the three groups (all P < 0.05) and the results in the acupuncture A group were superior to those in the other two groups (all P < 0.05). The morbidity of pneumonia in the acupuncture A group was lower than that in the acupuncture B group and the rehabilitation group [3.3% (2/60) vs 6.7% (4/60), 8.3% (5/60), both P < 0.05]. The effective rate in the acupuncture A group was better than that in either of the other two groups [95.0% (57/60) vs 81.7% (49/ 60), 75.0% (45/60), both P < 0.05].
CONCLUSIONOn the basis of the conventional medication, acupuncture at Lianquan (CV 23) effectively improves the swallowing function, relieves neurological deficit and reduces the morbidity of pneumonia in the patients of post-stroke dysphagia.

5. Preparation of Lingyi Formula multicomponent microemulsion and its anti-lung cancer activity

Objective: To prepare Lingyi Formula multicomponent microemulsion and evaluate its anti-lung cancer activity. Methods: Lingyi Formula multicomponent microemulsion was prepared by aqueous titration method using polysaccharides solution of Ganoderma lucidum and Coix lachryma-jobi var. ma-yuen as aqueous phase and coix seed oil as oil phase, loading ganoderma triterpenes. The average particle size, Zeta potential, and stability were detected. The results of antitumor efficacy including tumor inhibitory rate, body weight change, immune organ index, and concentration of TNF-α and IL-6 were investigated. Besides, pathological section of tumor tissue and TUNEL labeling were conducted subsequently. Results: The prepared microemulsion displayed spherical surface with mean droplet size of (69.92 ± 8.43) nm, polydispersity (PDI) of 0.060 ± 0.008, and Zeta potential of (-11.30 ± 1.34) mV. Tumor inhibitory rate of microemulsion (57.25%) was significantly higher than that of suspension (45.89%), immune tissue index as well as the concentration of TNF-α and IL-6 were increased significantly. TUNEL labeling and pathological section of tumor tissue showed that the antitumor activity of microemulsion was significantly effective compared with that of suspensions. Conclusion: Lingyi Formula multicomponent microemulsion has a good anti-lung cancer activity.
6.Human Health Risk Assessment of Benzene and Formaldehyde in Different Indoor Environment in Guiyang City

Objective To evaluate the cancer risk of benzene and formaldehyde in different indoor air environments in Guiyang city.Methods From 2004 to 2005,benzene and formaldehyde monitoring was conducted in bedrooms,living rooms, kitchens,offices,classrooms and outdoor environments in winter,spring and autumn,and human health risk assessment was done. Results The mean benzene and formaldehyde concentration in different environments were lower than the China indoor air quality standard except for formaldehyde concentration in offices.The benzene cancer risks of male and female adults were 1.63?10~4 and 1.40?10~(-4)respectively.The formaldehyde cancer risks of male and female adults were 6.05?10~(-4)and 5.23?10~(-4) respectively.The human formaldehyde cancer risk was higher than benzene cancer risk,and the risk for male was higher than that for female.Benzene and formaldehyde cancer risks of different population in different indoor environments were above 1.00?10~(-6), the acceptable level of human cancer risk.Conclusion The benzene and formaldehyde concentrations in different environment in Guiyang city has the high cancer risks to human health.
8.Diagnostic value of CT reconstruction on multilevel lumbar spondylolysis

Objective To explore the prevalence of multilevel lumbar spondylolysis in operation patients,and to evaluate the diagnostic value of CT reconstruction on multilevel lumbar spondylolysis.Methods A total of 312 patients,who had undergone surgical treatment in our hospital from January 2004 to March 2012 for lumbar spondylolysis,were enrolled in this study.There were 147 males (47%) and 165 females (53%),aged from 12 to 76 years (average,44 years).Every patient underwent lumbar spine X-ray examination including anteroposterior,lateral,over-flexion,over-extension and double oblique X-rays,as well as CT examination.The prevalence of multilevel lumbar spondylolysis in all operation patients and the involved segments were recorded and analyzed.Moreover,the clinical manifestation and radiological features were summarized.Results In all enrolled patients,there were 9 patients (2.9%) with multilevel lumbar spondylolysis,including 5 males and 4 females,aged from 21 to 51 years (average,39 years).As for lesion distribution,there were 8 cases of bilateral spondylolysis located at both L4 and L5,and 1 case of bilateral spondylolysis at L5 combined with right spondylolysis of L4 and discontinuity in the base of left pedicle of L4.Among 9 patients,only 3 patients obtained correct diagnosis by anteroposterior and double oblique X-rays of the lumbar spine,while all patients got final diagnosis by sagittal and oblique coronal reconstruction of lumbar spine CT scan.The oblique coronal CT reconstruction could show the details of bone defect in lumbar vertebral isthmus,bony callus,peripheral osteophytes,as well as fibroplasia.For unilateral lumbar spondylolysis,the oblique coronal CT reconstruction could also show the reactive sclerosis in contralateral lamina of vertebra.Conclusion Multilevel lumbar spondylolysis is not rare in operation patients,and L4 and L5 are most common involved levels.Sagittal and oblique coronal reconstruction of lumbar spine CT scan is more accurate in showing the details of lumbar spondylolysis,which is helpful in decreasing the missed diagnosis and improving therapy.
9.Differential diagnosis of ossifying fibroma and fibrous dysplasia

OBJECTIVE To investigate the clinicopathological features of ossifying fibroma (OF) and fibrous dysplasia (FD) , and to evaluate the role of immunohistochemistry in differential diagnosis. METHODS Clinical presentation and morphological study of 40 cases with OF and 26 cases with FD were reviewed. Immunohistochemistry stains for PCNA, Ki-67, Laminin and CK 19 were performed in specimens of 10 cases with FD and 26 cases with OF. RESULTS The average age of OF was 22.2 years and the ratio of male and female was 2.33:1. The average age of FD was 17.2 years and the ratio of male and female is about1:1. The average age of the multibone type of FD was 11.2 years and the ratio of male and female is 1:2. Most of OF occurred in ethmoid sinus (75 %) and 70 % of the patients presented snuffle and hyposmia, while FD inclineed to arise from sphenoid (58 %) and 80.8 %of patients showed eyesight dropping and exophthalmos. The immunohistochemistry stains PCNA was positive in 19 cases (73.1 %) of OF and negative in 10 cases of FD. Ki-67, laminin, CK19 were of little significance. CONCLUSION The gender, age distribution, clinical and pathological features of OF and FD are different from each other. Immunohistochemistry staining for PCNA is of great value in their differential diagnosis.
10.EFFECT OF GRAPE SEED OIL ON SERUM LIPIDS IN EXPER-IMENTAL HYPERCHOLESTEROLEMIC RATS

The effects of grape seed oil on serum total cholesterol (TC) and high density lipoprotein cholesterol (HDL-C) levels were investigated in experimental hypercholesterolemic rats. The grape seed oil containing about 74% linoleic acid, multivitamins and trace elements. Each animal was given 4 ml/kg BW grape seed oil mixed in high fat ration for 40 days. The results in two experiments showed that TC concentration in the serum of rats fed grape seed oil was markedly lowered as compared to control, but serum HDL-C was less lowered. Furthermore, the ratio of HDL-C to TC in the experimental group was also significantly higher than that of control.
