1.The effect of anti-müllerian hormone of serum and follicular fluid in vitro fertilization-embryo transfer

Objective To comprae the difference of Sex hormone,serum and follicular fluid anti Mullerian hormone (AMH) level tube,number of retrieved egg,the number of cleavage rate,fertilization oocytes,the number of available embryos and total embryo on in vitro fertilization and embryo transfer (IVF-ET) of different reaction groups,to explore the relationship between AMH and pregnancy outcome of IVF-EH.Methods Eighty-one cases of IVF-ET assisted pregnancy were divided into low ovarian response group,normal ovarian response group and high ovarian response group according to the response of the super ovulation.Serum sex hormone and AMH of all patients were detected on the third day of menstruation,on the day of HCG injection,oocyte pick-up (OPU) and embryo transfer (ET),AMH in FF were detected too.The difference of oocytes,cleavage rate,embryo quality and the indicators mentioned above among the three groups were compared.All of the above indicators were compared between pregnancy group and unpregnancy group.Results (1) The level of AMH on the third day of menstruation ((1.76 ± 0.47) μg/L,(3.45 ± 1.01) μg/L,(6.34 ± 1.29) μg/L,F =2.435,P<0.001),HCG injection ((1.09±0.13)μg/L,(1.29±0.29)μg/L,(2.64±0.51)μg/L,F=1.542,P=0.001) and embryo planting((1.32±0.33) μg/L,(1.62±0.39) μg/L,(1o 91±0.41) μg/L,F =1.573,P <0.001) all were statistically significant in three groups.On day of OPU,the levels of serum AMH ((0.95±0.21)μg/L,(1.15±0.29) μg/L,(1.74±0.41) μg/L,F =12.573,P<0.001) and AMH in FF((5.82±1.19) μg/L,(6.92±1.05) μg/L,(7.79±1.39) μg/L,F =9.83,P<0.001) were statistically differences in the three groups.(2)The number of oocytes were positively correlated with AFC,based AMH,levels of E2,P and AMH on the day of HCG injection and OPU,E2 and AMH on the day of ET and AMH in FF(P<0.05).(3)AMH in FF in pregnancy group was (6.22±0.82)μ g/L,significantly higher than those without pregnancy group ((5.31 ±0.71)μg/L,P =0.037).Conclusions Levels of AMH in serum and follicular fluid are good predictors of ovarian response in IVF-ET.The level of serum AMH can only predict ovarian response,but can not effectively predict pregnancy outcome.The level of AMH in FF is a good predictor of pregnancy outcome.

3.A New Phenolic Acid from Rhizomes of Abacopteris penangiana

Objective To study the chemical constituents of Abacopteris penangiana.Methods The compounds were separated and purified by various chromatographic techniques and their structures were elucidated on the basis of physiochemical properties and spectroscopic methods.Results Seven compounds were purified and their structures were identified as:(7'Z)-3-O-(3,4-dihydroxy phenylethenyl)-caffeic acid(1),caffeiein B(2),matteucinol(3),protocatechuic acid(4),p-methoxybenzoic acid(5),β-sitosterol(6),and daucosterol(7).Conclusion Compound 1 is a new phenolic acid compound named abacopteric acid,and the other compounds are isolated from the plant for the first time.
4.Effect of curcumin on inflammatory responses in hippocampus during global cerebral ischemia-reperfusion injury in hypertensive rats

Objective To evaluate the effect of curcumin on the inflammatory responses in the hippocampus during global cerebral ischemia-reperfusion (I/R) injury in hypertensive rats.Methods Forty-eight SPF male Wistar-Kyoto rats,aged 8 weeks,weighing 180-200 g,were randomly divided into 2 groups (n =24 each) using a random number table:sham operation group (W-Sham group) and I/R group (W-I/R group).Seventy-two SPF male spontaneously hypertensive rats,aged 8 weeks,weighing 180-200 g,were randomly divided into 3 groups (n =24 each) using a random number table:sham operation group (S-Sham group),I/R group (S-I/R group),and curcumin group (S-Cur group).Global cerebral ischemia was induced by 4-vessel occlusion method (10 min of transient global ischemia followed by reperfusion).Curcumin 100 mg/kg was injected intraperitoneally at 30 min of reperfusion in S-Cur group,and corn oil 5 ml/kg was injected intraperitoneally at 30 min of reperfusion in the other groups.The ability of learning and memory was tested by step-down test at 7 days of reperfusion.Rats were sacrificed at 3 h and 1,3 and 7 days of reperfusion (T1-4) and the hippocampi were removed.The morphological changes of pyramidal cells in hippocampal CA1 area were observed by HE staining.The mean density of pyramidal cells in hippocampal CA1 area was quantified by Nissl staining.The contents of interleukin-lβ (IL-1β),tumor necrosis factor-alpha (TNF-α) and IL-10 in the hippocampus were determined by ELISA.Results Compared with W-Sham group,the ability of learning and memory was significantly decreased,the mean density of pyramidal cells in hippocampal CA1 area was decreased,the contents of IL-1β at T1-3,TNF-α at T1,and IL-10 at T2 were increased,and the contents of IL-10 were decreased at T1,3,4 in W-I/R group,and no significant changes were found in the parameters mentioned above in S-Sham group.Compared with W-I/R group,the ability of learning and memory was significantly decreased,the contents of IL-1β at T1,3 and IL-10 at T2 were decreased,the content of TNF-α was increased at T1,no significant change was found in the mean density of pyramidal cells in hippocampal CA1 area,and the damage to pyramidal cells in hippocampal CA1 area was severe in S-I/R group.Compared with S-I/R group,the ability of learning and memory was significantly increased,the mean density of pyramidal cells in hippocampal CA1 area was increased,the contents of IL-1β at T2,3 and TNF-α at T1-4 were increased,and the content of IL-10 was increased at T2,and the damage to pyramidal cells in hippocampal CA1 area was reduced in S-Cur group.Conclusion Inhibition of inflammatory responses in the hippocampus may be involved in the mechanism by which curcumin reduces global cerebral I/R injury in hypertensive rats.
5.Surveillance of food-borne diseases in Jinshan District from 2014 to 2020

Objective :
To analyze the epidemiological characteristics of food-borne diseases in Jinshan District, Shanghai from 2014 to 2020, so as to provide the evidence for formulating the public health strategy for food-borne diseases control.
Methods :
The medical records of patients with food-borne diseases were collected from 16 monitoring hospitals in Jinshan District from 2014 to 2020, and the basic information, clinical symptoms, history of suspicious dietary exposure and disease diagnosis were extracted. The crowd distribution, temporal distribution, spatial distribution, history of suspected dietary exposure and etiological characteristics of patients with food-borne diseases were descriptively analyzed.
Results :
A total of 1 060 cases with food-borne diseases were reported in Jinshan District from 2014 to 2020, including 1 057 cases with infectious diseases ( 99.72% ) and 3 poisoning cases ( 0.28% ). The male/female ratio of the cases was 0.94∶1, and 47.55% ( 504 cases ) were at ages of 15 to 44 years. Working ( 402 cases, 37.92% ) and farming ( 218 cases, 20.57% ) were predominant occupations, and the detection of food-borne diseases was concentrated between May and October, with two peaks seen in August and May. The suspicious food exposure was predominantly meat and meat products (215 cases, 20.28%), and the suspicious food exposure place was predominantly at home ( 363 cases, 34.25% ). In addition, the positive rate of food-borne infection was 23.03% in 712 samples, including 123 samples with Vibrio parahaemolyticus infections ( 17.83% positive rate ), and 26 samples with Salmonella infections ( 3.65% positive rate ).
Conclusion
Food-borne diseases were highly prevalent in summer in Jinshan District from 2014 to 2020, and infectious cases were predominant. Young people, workers and farmers are at high risk of food-borne diseases, and Vibrio parahaemolyticus and Salmonella were predominant pathogens.
6.Survey on household eye hygiene among primary school students in Jinshan District

Objective:
To investigate the household eye hygiene among students at grades 3 to 5 in primary schools in Jinshan District, Shanghai Municipality, so as to provide insights into the management of myopia among children and adolescents.
Methods:
Students at grades 3 to 5 in primary schools were sampled in Jinshan District using a stratified cluster sampling method from December 2021 to January 2022. The household reading environments were observed, including the height of the learning desk and chair, desk surface color and strength. The duration of reading and writing, duration of watching TV and videos, duration of outdoor activities, duration of sleep and eye use behaviors and habits after school and at weekends were investigated using questionnaires.
Results:
A total of 330 primary school students were surveyed, including 179 boys (54.24%) and 151 girls (45.76%), and there were 36.36% grade 3 primary school students, 36.36% grade 4 students and 27.27% grade 5 students. There were 94.24% of primary school students that used the desk and chair with heights mismatched to students' heights, 25.45% that used desks with dark surface, 26.67% that used desks with light reflection, and 48.48% that used desk lamps with an illumination intensity of <300 lx. There were 56.36% of students with reading and writing duration of 1 h and longer, 15.76% with watching duration of 0.5 h and longer, 86.97% with outdoor activity duration of <1 h and 88.48% with sleep duration of <10 h after school, and 42.42% with reading and writing duration of 2 h and longer, 29.70% with watching duration of 1 hour and longer, 65.45% with outdoor activity duration of <2 h and 55.76% with sleep duration of <10 h at weekends. There were 30.91% of primary school students with 10 cm distance from chest to desk, 26.36% with 33 cm distance from eyes to books and 35.15% with 3 cm distance from fingers to pen points when reading and writing, and 35.45% with >3 m distance from TV, 40.91% with >50 cm distance from computers, and 22.73% with >40 cm distance from cell phones when watching TV or videos.
Conclusions
The household eye use environments remain to be improved, and there are poor eye use behaviors among primary school students at grades 3 to 5 in Jinshan District, Shanghai Municipality; notably, mismatch between the desk and chair height and students' body height and inadequate sleep are common.
7.The clinical evaluation of EOF5 regimen, the combination of epirubicin, oxaliplatin and 5-day continuous infusion of 5-FU, for patients with advanced/metastatic gastric cancer

Background and purpose:Although there is still no standard ifrst line chemotherapy regimen for metastatic gastric cancer (MGC), the doublet and triplet regimens containing platinum and lfuorouracil were most popular worldwidely. The ECF regimen is the classical and one of the most popular treatment choices in this setting, while the marrow suppression, the renal toxicity and poor compliance inhibits its usage. In order to improve its efifcacy and tolerability, this study conducted 2 phaseⅡ trials by modified ECF regimen, the EOF5 regimen (substituting cisplatin with oxaliplatin, shortening the continuous infusion period to 120 h), to treat patients with MGC since 2004. This paper reported the comprehensive results of the 2 studies.Methods:All the patients who enrolled in our previous2 phaseⅡ trials and received EOF5 as ifrst line treatment entered this study. Each patient received the treatment of epirubicin 50 mg/m2 iv d1, oxaliplatin 130 mg/m2 iv gtt d1 and 5-FU 375-425 mg/m2·d-1 civ 120 h, and repeated every 3 weeks. Efifcacy was analyzed every 6 weeks.Results:A total number of 178 patients (all were metastatic patients but 2 advanced patients with unresectable lesions) were included into this study. One hundred and seventy patients were evaluable, and 7 patients (3.9%) achieved complete response (CR), 76 patients (42.7%) achieved partial response (PR), 46.6% patients achieved overall response rate (ORR, CR+PR), and the cases of stable disease (SD) and progressive disease (PD) were 69 (38.8%) and 18 (10.1%), respectively. The median progress free survival (PFS) and overall survival (OS) were 6.0 months (95%CI: 5.2-6.8) and 12.6 months (95%CI: 8.9-16.3), 1-year and 2-year survival rate were 50.9% and 28.0%, respectively. Grade 3, 4 toxicity including: leucopenia (23.0), neutropenia (38.8%), anemia (6.5%), thrombocytopenia (23.5%), nausea/vomiting (14.1%), and peripheral neuropathy toxicity (1.2%). Among 75 patients who received second line treatment, the median survival from second line treatment was 8.0 months (95%CI: 4.8-11.2).Conclusion:EOF5 regimen is a highly effective regimen with moderate and manageable toxicity, and it providesa suitable alternative for the ifrst-line treatment of MGC.
8.Bibliometrics analysis on international scientific and technological cooperation of traditional Chinese medicine based on SCI-E

Objective To analyze the characteristics of SCI papers published in international cooperation of Chinese medicine and to understand the current research situation in this field.Methods Using the scientific citation index extension (SCI-E) database in Web of Science was used as the data source,and this paper retrieves retrieved related papers and highly cited papers on the international cooperation of traditional Chinese medicine during the year 1990-2017,and analyses from the years,cooperative countries and regions,periodicals,fund support,authors' institutions and research content by using the method of Bibliometrics and co co-word clustering.Results The total number of the articles for analyzing was 10 984,and the number of highly cited article was 114,which showed the number of article increased gradually.There are were 145 countries and regions in cooperation.The first three countries are were the United States,Japan and the United Kingdom.A total of 25 core periodicals are were determined.The fund funding rate wais 74.5%.The fund financed institutions with the largest number of cooperative contributions are were both the National Natural Science Foundation of China and the National Institutes of health in the United States.The hot top-subject of cooperative research haswas pharmacology and pharmacology.The breast cancer,ovarian cancer,colorectal cancer and inflammation,such as acupuncture treatment for pain,polycystic ovary syndrome,and depression.Conclusions The international cooperation of traditional Chinese medicine has gone through the embryonic stage,and now it is at the stage of development.
9.RAC2 as a potential prognostic marker in breast cancer:a study based on immunohistochemical analysis and bioinformatics

Objective To investigate the expression,function and prognostic value of Ras-related C3 botulinum toxin substrate 2(RAC2)in breast cancer.Methods The expression profile of RAC2 mRNA was investigated in the TCGA-BRCA cohort,the prognostic value was analyzed using the Kaplan-Meier algorithm,the potential molecular function was investigated by gene set enrichment analysis,the relationship between RAC2 and immune infiltration was analyzed by Estimate and Cibersort algorithms,and the association between Rac2 and mutation spectrum was evaluated by maftools software package.The RNA-seq software package"pRRophetic"was used to predict the effect of chemotherapy drugs.In the validation cohort,Immunohistochemistry(IHC)staining was performed on tumor pathological sections of patients.Kaplan-Meier survival analysis and cox regression analysis were used to analyze the relationship between RAC2 and the prognosis of breast cancer.Results Bioinformatics analysis showed that the expression of RAC2 was up-regulated in estrogen receptor(ER)positive BC.In the Pam50 molecular subtype of BC,its expression decreased in the order of normal-like group,triple negative group,HER2 overexpression group,Luminal A group and Luminal B group.BC patients with high RAC2 expression had better overall survival(OS)and recurrence-free survival(RFS).The results of the validation cohort confirmed that RAC2 can be used as an independent predictor of BC prognosis.RAC2 was involved in the process of immune infiltration of tumor cells,and its high expression was mainly related to the mutations of CCDC168,VPS13C and AHNAK genes.Its up-regulation increased the drug sensitivity of doxorubicin,cisplatin,paclitaxel and gemcitabine.In terms of metabolism,it was negatively correlated with glycolysis and pentose phosphate pathway(PPP),and positively correlated with fatty acid oxidation(FAO)and glutamine metabolism.Conclusion High expression of RAC2 is associated with a good prognosis of BC and plays a role in the metabolism and immune infiltration.Further study on the molecular mechanism of RAC2 may provide new strategies for the prognosis and treatment of BC.
10.A novel histone deacetylase inhibitor induces proliferation inhibition of human colon cancer cells

Objective To examine the anticancer effect of a novel histone deacetylase inhibitor (HDACi), JZ004, on colon cancer cells HCT-8 and HT-29, and to investigate the molecular mechanisms of proliferation inhibition and apoptosis induction of cancer cells treated by JZ 004.Methods Colon cancer cells were treated with a series of concentrations of JZ004 .MTT assay was used to detect the proliferation of cancer cells .The cell cycle distribution and apoptosis were deter-mined by flow cytometry .Rhodamine 123 and DCFH-DA were applied to detect the mitochondrial membrane potential (ΔΨm) and reactive oxygen species ( ROS) production.The protein expressions of acetyl-histone H3, p21, cyclin-dependent kinase(CDK)4, Bcl-2, Mcl-1 and Bax were assayed by Western blotting .Results JZ004 was found to inhibit proliferation and induce apoptosis of colon cancer cells in a time-and dose-dependent manner , accompanied by a dose-dependent hyperacetylation of histone H3.JZ004 induced the cancer cell arrest in G 0/G1 phase by increasing the expres-sion level of p21 while CDK4 was downregulated .JZ004 also increased cellular ROS production and reduced ΔΨm by regu-lating the expressions of Bcl-2 family proteins .Conclusion As a novel HDACi , JZ004 effectively inhibits proliferation and increases ROS production to induce apoptosis of colon cancer cells .The results indicate that JZ004 is a potential compound to be developed as an anti-colon cancer agent for clinic application .
