1.Attempt of seminar in biochemistry teaching for Uygur clinical medicine undergraduate

Objective To discuss the significance of the seminar in biochemistry teaching for Uygur clinical medical undergraduates. Methods 2011 grade totally 86 Uygur clinical medical under-graduates in Xinjiang Medical University were enrolled as the research object and questionnaire was used to make satisfaction survey. Data descript and analysis was performed according to the percentage of each option. Results 86.9%students thought that topic seminar was good;100.0%students thought that semi-nar can help apply and mastery the knowledge;100.0%students considered that seminar conducted dur-ing extracurricular time was necessary to the growth of knowledge;97.8%students considered that search-ing for references in the spare time was helpful;100.0%students were interested in the content of the seminar and thought it was novel and practical;100.0%students thought that seminar can help broaden thinking way;43.5%students supported that contents exceeding the syllabus can be used in examination while 39.1%students opposed; preparation time for examination lasted for 5 to 7 days. Conclusions Seminar applied in biochemistry teaching is very necessary,which can mobilize students' initiatives to learn actively and help consolidate and understand knowledge.
2.The association between polymorphism of parathyroid hormone gene and bone mineral density in Fuzhou postmenopausal women

BACKGROUND:Studies have shown the bone mineral density of postmenopausal women is closely related to parathyroid hormone. But there are differences in different areas.
OBJECTIVE:To investigate the association between BstBⅠ polymorphism of parathyroid hormone gene with bone mineral density in postmenopausal women from Fuzhou area.
METHODS:The bone mineral densities of the lumbar spine, femoral neck, trochanter and Ward’s triangle were measured in 150 postmenopausal women by dual energy X-ray absorptiometry. The genotype of parathyroid hormone gene was determined by polymerase chain reaction-restriction fragment length polymorphism.
RESULTS AND CONCLUSION:(1) The distribution of parathyroid hormone genotypes were BB genotype 68.8%, Bb 24.1%, and bb 7.1%. The B al elic gene frequencies reached 81%, while b was 19%. The distribution fol owed the Hardy-Weinberg equilibrium. (2) Analysis of the relationship between the genotypes and bone mineral density:There was no significant difference in the bone mineral densities of the lumbar spine, femur, neck, trochanter and Ward’s triangle among the three genotypes (P>0.05). BstBⅠ gene polymorphism of parathyroid hormone gene is not correlated to bone mineral density, and there is no enough evidence to support genotype of parathyroid hormone gene as a genetic marker in predicting the risk of developing osteoperosis in Fuzhou postmenopausal women.
3.Secretion of adipokines in miR-26b over-expressed human adipocytes differentiation process

Objective To investigate the inlfuence of overexpression of miR-26b on the secretion of adipokines dur-ing human adipocyte differentiation. Methods Human preadipocytes were infected with the hsa-miR-26b over-expressing lentivirus and were induced to differentiate, and then the levels of adipokines (IL-6, leptin, resistin, TNF-α) at different time points during differentiation were measured by ELISA. Results Compared with control group, decreased secretions of both IL-6 and leptin, and increased secretion of resistin were found during the differentiation of human adipocytes in miR-26b overexpressed group. However, the secretion of TNF-αwas not measured in both groups. Conclusion The miR-26b can improve the inlfammation and insulin resistance of human adipocytes, which will provide potential targets for obesity treat-ment.
4.The application and exploration of PBL mode in the biochemistry teaching of clinical medicine in merging class of minority and Han students

Objective To investigate the effectiveness of problem-based learning in the biochemistry teaching in merging class of minority and Han students.Methods Totally 460 clinical medical students were divided into PBL group which contained 252 students and the traditional teaching group which involved 208 students,respectively.According to each team of seven to eight students,minority and Han students randomly arranged.Control group used classroom teaching mode,experimental group in addition to classroom lectures,had additional 12 hours of PBL teaching,but the theory classes for the two groups of students were taught by the same six teachers with rich teaching experience,and the teaching content and teaching material selection were also the same.At the end of the course,the learning outcomes were evaluated by using descriptive analysis and t test (α=0.05) based on the combination of theoretical examination,experimental practice and the questionnaire survey method.Results Compared with the traditional teaching group,the final scores were higher than those of PBL group (84.72 ± 6.99 and 80.34 ± 7.12,P<0.05).There were also statistically significant between two groups according to different nationality(Minority:85.65 ± 5.27 and 79.70 ± 7.14;Han:83.91 ± 8.26 and 80.95 ± 7.08;P<0.05),and interestingly the increased ratio of scores was higher in minority than that in Han.The questionnaire surveys indicated that the PBL teaching method could enhance professional and comprehensive qualities of students and more than 81.83% students were satisfied with the new teaching mode.Conclusions The combination of tradition and PBL-based teaching methods improved the quality of biochemistry teaching of clinical medicine in merging class of minority and Han students in Xinjiang Medical University.
5.Bioinformatic analysis of the hsa-miR-1908 upstream promoter region

Objective To predict the functions of hsa-miR-1908 promoter using various bioinformatic tools, and to provide clues for further study on transcriptional regulation mechanism of miR-1908 in human adipocytes. Methods The promoter se-quence of miR-1908 was obtained from Ensemble, and then the CpG islands and transcription factor binding sites were pre-dicted by a variety of online bioinformatic tools. Results The length of the miR-1908 promoter sequence was 1 458 bp. The CpG islands, which inhibited the transcription of miR-1908, were located at (438-756) bp, (836-937) bp and (979-1374) bp. Meanwhile, 15 transcription factor binding sites were found in the promoter sequence of miR-1908. Conclusions miRNA up-stream promoter related bioinformatics can not only improve the efficiency of microRNA promoter research, but also provide further important information on transcriptional regulation of miR-1908.
6.The Expression of Beclin-1 and LC3 on Rats with Cerebral Ischemia Reperfusion Injury and the Efficacy of Edaravone

Objective:To study the expressione of autophagy-related protein Beclin-1 and LC3 in cortex and hippocampus of rats after cerebral ischemia reperfusion injury and the efficacy of Edaravone.Methods:Sprague Dawley rats were randomly divided into sham group,model group and Edaravone group.The cerebral ischemia reperfusion model was induced via Zea Longa with blocking the middle cerebral artery of 2 h and reperfusing of 24 h.Animals assigned to sham group were only separated left common carotid artery.Edaravone was injected intraperitoneally at a dose of 1 0 mg/kg at 15 min before reperfusion.The condition of nerve injury of rats was conducted by Neurobehavioral score.The degree of brain injury and success of model were determined based on 2,3,5-triphenyltetrazolium chloride(TTC)staining.The changes of neuron stained by hematoxylin and eosin (HE) in cortex and hippocampus were observed.The expression of Beclin-1 and LC3 was measured by immunohistochemistry.Results:After cerebral ischemia reperfusion the neurobehavioral score of edaravone group was(2.00± 0.67),which was obcviously less than(2.50± 0.53) of model group(P<0.05).The infraction focus and the neuron injury in cortex and hippocampus neurons were also observed in model group,and the edaravone group reduced above expression.The positive rate of Beclin-1 of each group in cortex were (1 1.08± 0.85)%,(33.42± 1.57)% and (25.61± 1.39)%,there was significant difference between model group with sham group and edaravone group (P<0.05).The positive rate of Beclin-1 of each group in hippocampus were (10.34± 0.21)%,(31.82± 1.73)% and (22.74± 1.26)%,there was significant difference between model group with sham group and edaravone group(P < 0.05).The positive rate of LC3 of each group in cortex were (15.33± 0.47)%,(39.72± 1.73)% and (28.53± 1.61)%,there was significant difference between model group with sham group and edaravone group(P<0.05).The positive rate of LC3 of each group in hippocampus were (13.74± 0.37)%,(32.53± 1.43)% and(25.38± 1.23)%,there was significant difference between model group with sham group and edaravone group (P<0.05).Conclusion:The expression of Beclin-1 and LC3 was increased after cerebral ischemia reperfusion.Edaravone may reduce autophagy and brain injury through downregulation the expression of Beclin-1 and LC3.
7.Expression and significance of Gab2 in colorectal cancer tissues

Objective:To investigate the expression and significance of Gab 2 in colorectal cancer tissues .Methods:Immuno-histochemistry was used to detect the expression of Gab 2 in 78 cases of colorectal cancer tissues and 46 cases of the adjacent tissues and to analyze the association of Gab2 expression with the clinicopathologic features of colorectal cancer;The expression of Gab2 in samples from 10 cases of colorectal cancer tissues and matched adjacent nontumorous tissues was detected by Western blot .Results: The results of immunohistochemistry demonstrated that the Gab 2 protein positive expression rate in 78 cases of colorectal cancer was 53.85%;whereas was negative expression or weak in the adjacent tissues , showing a significant difference of comparison within this result (P<0.001) .The expression of Gab2 in colorectal cancer was related with TNM stage and lymph node metastasis (P<0.05) , but no relation with the size and differentiation of tumor (P>0.05) .Western blot showed that the Gab2 protein expression of colorectal cancer cases was significantly higher than that of matched adjacent nontumorous tissues ( P<0.05 ).Conclusion: Gab2 was overexpressed in colorectal cancer .Gab2 maybe play an important role in carcinogenesis and progression of colorectal carcinoma .
8.Genetic polymorphism of 19 STR loci in Xinjiang Barkol Kazakh population.

OBJECTIVE:
To investigate 19 short tandem repeat (STR) loci in Chinese Kazakh population in Barkol County with Goldeneye™20A multiplex amplification system.
METHODS:
DNA samples were screened from 81 unrelated individuals. The 19 loci were D8S1179, D21S11, CSF1PO, D3S1358, D7S820, TH01, D13S317, D2S1338, D18S51, D16S539, TPOX, vWA, D19S433, D5S818, PentaD, PentaE, D6S1043, D12S391 and FGA. The PCR products were analyzed and genotyped by ABI3130XL sequencer.
RESULTS:
These loci were highly polymorphic. The combined power of discrimination was 0.999999999 and the combined paternity of exclusion was 0.999998914.
CONCLUSION
Goldeneye™20A multiplex amplification system is very useful in forensic case investigation for Barkol Kazakh population.

Experimental teaching is an indispensable task in teaching work in colleges and universities.Basic medical experimental teaching is the foundation of clinical practice,techniques,and scientific research.Medical biochemistry experiment is the compulsory experimental teaching content for medical students.The existing basic medical experimental teaching is far from enough for medical students.Therefore,it is very important to tap the potential of students,cultivate their interest in learning,improve the ability to learn on their own,operate manually,be creative,and solve problems,which can start from basic medical experimental teaching and be achieved through the second class activities.
