1.Establishment of a sandwich ELISA method for measuring soluble IL-2 receptor
Chinese Journal of Immunology 1985;0(06):-
Two kinds of mouse monoclonal antibodies which recognize distinct epitopes of the IL-2 receptor (a chain) were used to establish a sandwich ELISA for measuring soluble IL-2 receptors. The results indicated that this method is sensitive and specific for detecting soluble IL-2 receptors of cell culture supernatant and serum from both healthy donor and patients with leukemia. Therefore this sandwich ELISA method is useful for basic and clinic immunology research.
2.Studies on the requirements for growth of Epstein-Barr Virus thransformed human lymphoblastoid B cells at low cell density
Chinese Journal of Immunology 1985;0(02):-
We obtained human lymphoblastoid B cells secreting human antibodies IgM and IgG to HFRS virus by Epstein-Barr virus(EBV)transfomation of PBMC.Conditioned media from the supernatant of a human fibroblast cell CRL1506 culture and irradiated CRL1506 cells as feeder cells could markedly promote the growth and antibody produotion of the human B lymphoblastoid cells at low cell density. It was shown that conditioned media contained a high level of interleukin 6(IL-6) .These results would be very useful for human monoclonal antibody technique including improving human B lymphoblastoid cell growth at low cell density and enhancing cloning efficiency andantibody production.

7.Multifocal visual evoked potential in the detection of glaucomatous field defects
Ophthalmology in China 1994;0(02):-
Objective To evaluate the effect of multifocal visual evoked potential(mfVEP)on the detection of glaucomatous field defects. Design Case-controlled study. Participants 20 healthy individuals and 20 patients with primary glaucoma accepted mfVEP test. Methods 20 healthy individuals and 20 patients with primary glaucoma were tested with RETIscan multifocal VEP System 3.20 and OCTOPUS 101 automated perimetry G2 test program. Amplitudes were measured and interocular response asymmetry coefficient (RAC) was calculated for each site from all participants. The abnormal areas from mfVEP test were compared with field defects from automated perimetry. Main Outcome Measures Amplitude, RAC. Results 90% of suffered eyes showed at least 3 adjacent points had an amplitude decrease or RAC outside 2 standard deviations (SD) from healthy individual database, in which 63% showed at least 3 adjacent points had an amplitude decrease outside 2 SD from normal database while 70% showed at least 3 adjacent points had a RAC outside 2 SD from normal database. 2 patients (2 eyes) had field defects in mfVEP though automated perimetry showed normal field. None of the healthy showd field defects in mfVEP test. Conclusions mfVEP could be used to detect field defects of glaucoma.
8.Anticoagulation Effect of Local Anticoagulation with Citrate in Hemofiltration for Sepsis Patients with Acute Renal Injury
China Pharmacist 2017;20(5):848-850
Objective: To compare the anticoagulation effect of local anticoagulation with citrate in the hemofiltration for sepsis patients with acute renal injury and observe the safety of anticoagulation to provide the basis for clinical anticoagulation of hemofiltration. Methods: Totally 72 patients with sepsis acute kidney injury in the hospital were randomly divided into the control group and the study group with 36 ones in each according to the single and double number. The control group and the study group was respectively treated with heparin sodium and citric acid for anticoagulation with the treatment course of 7 days. The life of filter tube and filter of the two groups were recorded, and the coagulation grade of filter was assessed. The levels of serum creatinine (Scr), blood urea nitrogen (BUN), pH, serum sodium bicarbonate and bicarbonate were measured before and after the treatment. The activated coagulation time (ACT) was measured before the filtration and in 8 h and 24 h after the filtration. The adverse reactions were observed as well. Results: The ratio of blood clotting level at 0-1 grade in the study group was higher than that in the control group, and the ratio of filter coagulation at 2-3 grade in the study group was lower than that in the control group, and the differences were statistically significant (P<0.05).The life of filter pipe and filter in the study group was longer than that in the control group, and the difference was statistically significant (P<0.05).ACTs in 8 h and 24 h after the blood filtration in the study group were longer than those in the control group, and the difference was statistically significant (P<0.05).The levels of BUN and Scr of the two groups after the treatment were lower than those before the treatment, and the differences were statistically significant (P<0.05).There were no significant differences in blood pH, HCO3-, Na + and adverse reactions before and after the treatment between the groups (P>0.05).Conclusion: The application of hemofiltration in the treatment of sepsis patients with acute renal injury can significantly improve renal function. And local anticoagulation has better anticoagulant effect in septic acute renal injury patients with hemofiltration, which can prolong the life of filter tube and filter with higher security.
9.Effects of dexmedetomidine on the perioperative stress in patients undergoing uvulopalatopharyngoplasty
The Journal of Clinical Anesthesiology 2017;33(8):738-741
Objective To observe the effects of dexmedetomidine on the perioperative stress in patients undergoing uvulopalatopharyngoplasty (UPPP).Methods Sixty patients scheduled for uvulopalatopharyngoplasty (UPPP), all males, aged 18-65 years, of ASA physical status Ⅰ or Ⅱ, were selected randomly. Then the patients were divided into dexmedetomidine group (group D) and control group (group C), 30 patients in each group. The patients of group D were administered 1.0 μg/kg bolus dose of dexmedetomidine over 10 min before tracheal intubation, followed by 0.5 μg·kg-1·h-1 infusion for 12 hours in ICU after surgery. And the patients in group C were given normal saline instead as in group D.The levels of MAP, HR, PaO2, PaCO2, norepinephrine (NE), epinephrine (E), cortisol (Cor), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were recorded at time points of entering operating room (T0), tracheal intubation (T1), 30 min after operation (T2), upon entering ICU (T3), and extubation (T4).Results HR of group D was significantly slower than that of group C during T1-T4 (P<0.05).MAP of group D was lower than that of group C during T1-T4 (P<0.05).The levels of NE, E, Cor, IL-6, TNF-α of group D were significantly lower than that of group C during T1-T4(P<0.05).Compared with group C , the incidence of cough reflex during extubation was lower in group D significantly (P<0.05).Conclusion Intravenous infusion of dexmedetomidine can suppress perioperative stress effectively and stabilize hemodynamics for the patients undergoing UPPP without prolonging the recovering time, extubation time and the duration in ICU.
10.Erythropoietin and neuron apoptosis:Signal pathway
Chinese Pharmacological Bulletin 2003;0(12):-
Apoptosis is a main death pattern in many nervous system disease.Erythropoietin(Epo)has been originally characterized as the principal regulator of erythropoiesis by stimulating the proliferation and differentiation of erythroid precursor cells. In recent years, many experimental studies have shown that both Epo and EpoR(erythropoietin receptor)are functionally expressed in the nervous system and that this cytokine exerts a remarkable neuroprotection against nerve cell apoptosis. This article reviews neuroprotective of Epo against apoptosis and outline the involved molecular mechanisms.
