Objective To investigate the mechanism involved in the brain protection afforded by prenatal hypoxic adaptation by determining the quantitative variation in bcl-2 and bax mRNA expression.Methods Twenty-four Wistar pregnant (22d pregnant) rat were randomly divided into two groups: group I (hypoxia group) and group *** ( control group) . In group Ⅰ the pregnant rats were placed in an airtight cabin specially designed for hypoxic adaptation. When O2 % in the cabin decreased to 15%, the animals were taken out breathing fresh air for 5min and then placed back in the cabin and underwent another episode of hypoxia. In group Ⅱ the animals were placed in the cabin which was not tightly closed and underwent no hypoxia. 7 fetal or newborn rats were taken at 1st, 3rd, 24th, 48th, 72nd, 120th, and 168th h after prenatal hypoxic adaptation from each group and their brains removed for determination of bcl-2 mRNA and bax mRNA. Results In control group the expression of bcl-2 and bax were observed in the brain tissue of normal fetal or newborn rats from the 22nd day in the uterus to the 7th day postpartum during which there were no significant changes in bcl-2 gene expression while bax gene expression gradually decreased with time ( the decrease was of no statistical significance) . In hypoxia group bax gene expression decreased at 8th h after hypoxic adaptation and reached the bottom at 24th h which persisted until 120th h; while bcl-2 gene expression started increasing at 24th h after hypoxic adaptation and persisted until 72nd h. The bcl-2/bax ratio also started increasing at 8th h after hypoxic adaptation and peaked at 24th h and persisted until 72nd h. Conclusions In the brain tissue of fetal and newborn rats which have undergone prenatal hypoxic adaptation, bcl-2 gene expression is elevated, bax gene expression decreases and bcl-2/bax ratio increases. These changes are time -dependent.
6.The effect of immunosuppressive drugs on the activity of pancreatic islet cells

Objective To investigate the effect of immunosuppressive drugs on the activity of pancreatic islet cells, and to explore their role in islet allotransplantation. Method Human pancreatic islet cells were isolated and purified in vitro, and their activity was determined by MTT assay after incubation with immunosuppressive drugs in different concentrations. Results The activity of the islet cells was not reduced after exposure to rapamycin at lower concentrations, although exposure to rapamycin at higher concentrations (≥1ng/ml) was associated with a significant inhibition of activity. No reduction of islet cell activity was observed after exposure to daclizumab and FTY720, at either lower or higher concentrations. Conclusion Rapamycin acts to damage islet cells directly, and the degree of the damage was concentration-dependent. No obvious toxicity to the islet cells was observed after exposure to daclizumab and FTY720.

Objective To study signs of ultrasonography in multiple renal arteritis (MRA) by color Doppler flow imaging(CDFI) and X-ray angiography.Methods 2-D ultrasonography and color Doppler were performed on 23 cases of MRA with 39 diseased blood vessels.The morphology and diameters of stenonsed renal arteries of the kidneys were observed respectively by 2-D ultrasonography and CDFI.CDFI was adopted to observe maximum of velocity(Vmax),minimum of velocity(Vmin),resistance index(RI),acclerate(ACC) and acclerated time(TA) of the renal arteries,and the values were compared with those by X-ray angiography and with those of the normal controls.Results MRA's renal volume and diameter were significantly different from those of controls ( P

The ?-radioactive seed brachytherapy source has been widely employed in the implantation therapy for the prostatic carcinoma and the ophthalmic lesions.In this study the dosimetric parameters for characterization of a low-energy interstitial brachytherapy source 125I were calculated according to dose calculation formalism recommended by AAPM TG-43U1.For data processing,a 0.28 cm active length was used for the geometry function.The dosimetry parameter air-Kerma strength,dose rate constant,radial dose function and anisotropy function were estimated by means of the EGS5 Monte Carlo code.The results obtained from this study are in good agreement with the corresponding values recommended by TG-43U1 and with the data reported by Dolan,et al.
10.Effect of Wuzhi capsules on whole blood concentration of Tacrolimus in renal transplantation recipients

BACKGROUND: It has reported that the combination of Tacrolimus and Wuzhi capsules can increase the peak concentration and area under curve of Tacrolimus recipients, however, the effects of this combination on recipients is poorly understood. OBJECTIVE: To investigate the effects of Wuzhicapsules on the blood concentration of Tacrolimus recipients. METHODS: A total of 38 renal transplantation recipients receiving triple therapy regime (Tacrolimus+mycophenolate +Prednisolone) were involved in the study and were divided into the experimental group (n=21) and control group (n=17). Recipients in the experimental group were taking Wuzhi capsules simultaneously, and those in the control group were not taking Wuzhicapsules. The Tacrolimus dosage to therapeutic concentration window and renal function were compared between the two groups at months 1,3 and 6 after transplantation. The incidence rates of acute rejection and diabetes mellitus, and renal and liver functions were observed at 6 months after transplantation. RESULTS AND CONCLUSION: The Tacrolimus dosage to therapeutic concentration window of the experimental group was significantly smaller than that of the control group (P < 0.05). However, incidence rate of acute rejection, diabetes mellitus, and renal and liver functions had no dramatically difference in 2 groups (P > 0.05). Results suggest that Wuzhi capsules may improve the immunosuppressive efficacy of Tacrolimus in renal recipients, therefore, can decrease the toxic effect of Tacrolimus with reduction dosage.
