1.Protective effects of total extract of astragalus(TEA) onprimary rats hepatocytes injured by CCl4 or H2O2

Aim To study the protective effect of total extract of astragalus(TEA) on primary rat hepatocytes injured by CCl4 or H2O2 and its mechanism.Methods The primary hepatocytes were isolated by Ⅳ collaganase and injured by CCl4 or H2O2.The contents of malondialdehyde(MDA) and glutathion(GSH),glutathion peroxidase(GSHpx) activity of hepatocytes and the AST and /or ALT level in cultural supernatant were determined by general methods. Results (1) The elevation of MDA content of hepatocytes and AST level in supernatant of cultural hepatocytes,and the loss of GSH content and GSHpx activity induced by CCl4 were restored remarkably by TEA(5~80 mg?L-1);(2)The elevation of ALT level in the supernatant of hepatocytes and MDA content of hepatocytes, and the loss of GSH content and GSHpx activity induced by H2O2 were improved significantly by TEA(5~80 mg?L-1) treatment.Conclusion The results suggest that TEA possess direct protective action on primary hepatocyte in vitro injured by CCl4 or H2O2. These might be associated with its anti-oxidative activity.
2.Cytokine storm and liver failure

Cytokine storm plays an important role in the pathogenesis of liver failure.It is closely related to hepatocyte necrosis in the clini-cal course and the prognosis of this disease.With Kupffer cells as a clue,it is elucidated that related cells and cytokines influence each other and jointly function in the development and progression of liver failure,leading to serious liver tissue damage and necrosis.Cytokine -based research can help improve early diagnosis,disease assessment,and individualized treatment for liver failure.
3.Retrospective Analysis of Budd-Chiari Syndrome in 135 Patients

A total of 135 patients diagnosed with Budd-Chiari syndrome(BCS)during August 2004 and February 2007 were reviewed and analyzed retrospectively.The results showed that 105 subiects (77.8%)caused by malignances,while 29 participants(21.5%)had no definite pathogens.The most commonly seen symptom included jaundiee(69.6%),followed by ascites(48.2%),abdominal distention (40.7%)and lower extremity edema(37.0%).Hisher levels of STB,PT and γ-GT were determined.Color Dopple ultrasound,CT and MRI could be helpful in diagnosing BCS;angiography remained the most accurate measurement.Main treatment strategies were stent implantation and/or balloon dilation.BCS is a clinical syndrome with no typical symptoms.which makes its management very difficult.A complete understanding of the condition will help reduce misdiagnosis.Angiography and stent implantation are primary ways to diagnose or treat BCS.
4.Depth of sedation induced by target-controlled infusion of propofol required to inhibit swallowing reflex

ObjectiveTo determine the depth of sedation induced by target-controlled infusion (TCI) of propofol required to inhibit the swallowing reflex.MethodsThirty-five ASA Ⅰ or Ⅱ healthy volunteers,aged 20-60 yr,with body mass index < 30 kg/m2,were enrolled in the study.Sedation was performed with TCI of propofol.The initial target plasma concentration (Cp) of propofol was set at 2.4 μg/ml and the Cp increased or decreased by 0.2 μg/ml every 5 min until the BIS value reached the predetermined level.The initial Cp of propofol was increased or decreased by 0.3 μg/ml in the next subject according to the BIS value in the previous one.The up-and-down sequential experiment was pedormed.The BIS value was set at 65 in the first subject and the ratio of the BIS value between the two consecutive subjects was 1.1.The BIS value was increased or decreased depending on the occurrence of the swallowing reflex in the previous subject.The half-effective BIS value (BIS50) and 95%confidence interval (CI) of propofol inhibiting the swallowing reflex were calculated by up-and-down sequential method.ResultsThe BIS50 of propofol required to inhibit the swallowing reflex was 64.4 (95% CI 59.3-69.8).ConclusionThe BIS50 and 95% CI of propofol administered by TCI and required to inhibit the swallowing reflex is 64,4 (59.3-69.8).
5.Biomechanical Evaluation on Cervical spine stabilizing Effects of Cervical Interbody Spinal Fusion Using Allograft Fusion Cage

Objective\ To evaluate the immediate effects and strength of allograft fusion cage(AFC) used for reconstructing stability of cervical spine. Methods\ Discs of C 5 and C 6 were resected on 8 fresh human cervical spine specimens, and autogenous iliac bone grafts(AIBG), and AFCs were implanted into the intervertebral spaces respectively. Compression test, pull out test and segmental motion measurement were studied.Results\ Comparing with intact and AIBG groups, the range of motions of C5-6 in AFC group were decreased in all directions except for extension; with (502?114) N compressive load,the vertebrae in AFC group were broken while the AFC were intact, but the AIBG were broken at (135?42) N load; with 300 N drawing load,no loosening was found between AFCs and vertebra, but it was found between AIBG and vertebra at 60 N load. Conclusion\ AFC could provide enough support,anti slide ability and could remain or increase the height of intervertebral spaces. It completely meets clinical and biomechanical requirements.
6.Influnce of doxoricin cytotoxicity by lipid peroxidation of docosahexaenic acids

Purpose:Our experiments are dealing with the affect of docosahexaenic acid(DHA) in doxorubicin to cytoxicity effect of MDA-MB-435s, which is of the anti-human breast cancer and whether it is one of the effect mechanisms that it via the lipid peroxidation of DHA.Methods:Add different match such as anti-cancer drugs?PUFAs?oxidan system of sodium ascorbate and ?-methy-1.4-naphoqui-none?antioxidant of Dl-?-tocopherol and so on to the culture cell line of MDA-MB-435s each 24 hours for 6 days, estimate MDA by TBA with cell extracts and estimate the contents of NO via nitrite salt spectrophotography, then draw out a dosage-effect correlation straight line among contents of MDA,NO and cytoxicity. Mitox is used as contraposition to the peroxidation of ADM which is enhanced by DHA for peroxidation of mitox is low.Results:There have been obvious changes on the contents of MDA of the lipid peroxidation has paralled increased while NO has decreased. There is recitilinear correlation relationship among the contents of MDA and NO from cell extracts and the cytoxicity.Conclusions:It is obvious that DHA can add the cytoxicity of cell line of MDA-MB-435s with ADM, particularly after add non-cytoxicity oxidant. It is lipid peroxidation of oncocyte that one of the mechanisms of DHA can enhance anti-breast cancer effect of ADM.
7.Analysis of 32 cases gastrointestinal leakage

Objective To investigate the factors and the preventive methods of gastrointestinal leakage.Method 32 cases of gastrointestinal leakage from 1996 to 2001 were analyzed retrospectively.Results In 32 patients,19 cases were benign diseases and 13 cases were malignant diseases.25 cases were selectively operation and 7 were emergency operation, 11 cases treated by re-operation and 21 cases by other methods.24 cases were cured and 8 cases were died.Conclusions Main cause of leakage was operative method unsuitable.Anastomostic skill was deficiency,local condition was bad and ignored postoperative treatment and so on.Preventive methods should be selected to decreasingintestinal leakage.
8.Expression of fibroblast growth factor receptor-3 during fracture healing

Objective To observe the spatial and temporal expression patterns of FGFR3 gene and analyze the potential roles of FGFR3 during fracture healing and explore the technique of in situ hybridization of paraffin sections of bone tissue. Methods The fracture model of mouse tibia was established by the standard methods. The expression of FGFR3 mRNA was studied by in situ hybridization during the various stages of bone repair. Results There was faint FGFR3 positive-signal in the cytoplasm of prehypertrophic chondrocytes in soft callus on day 5. FGFR3 mRNA was strongly expressed in prehypertrophic chondrocytes and hypertrophic chondrocytes in soft callus on day 7, but disappeared on day 14, 28 because chondrocytes was replaced by osteoblasts. Conclusion FGFR3 mRNA was strongly expressed in prehypertrophic chondrocytes and hypertrophic chondrocytes in the fracture callus, which shows that FGFR3, a gene that regulates the proliferation of chondrocytes during bone development, may participate in the regulation of differentiation of chondrocytes during fracture healing.
9.Imaging diagnosis of monostotic fibrous dysplasia in thoracic and lumbar spine vertebrae.

The X-ray radiograph, CT scan and MRI appearance of 5 patients with pathologically proven fibrous dysplasia in thoracic and lumbar spine vertebrae were retrospectively analyzed. Plain radiographs, CT scans and MR images showed the presentation of eccentric lesion with intact cortex bone and marginal sclerosis in vertebral bodies without involvement of vertebral appendix and extraosseous soft tissue. The lesion masses were round (one being oval-shaped) and radiolucent in plain radiographs and CT scans. Homogeneous long signal was observed on T1 weighted image and strongly enhanced when gadolinium was administered. On T2 weighted MRI, short signal was found in the anterior part of the mass, long signal in the posterior part, and short and slight long signal in the middle part, without partitioning and laminating change. There was a good correlation between radiological features and surgical findings. These findings may be useful to diagnose fibrous dysplasia in spine.

10.The current situation and problems of primary health care personnel since the new round of China's health care reform

Objective: To analyze the current situation and problems of primary health care personnel since the new round of China's health care reform, and to provide a reference for stabilizing the primary health care personnel. Methods: This paper uses descriptive statistical analysis to calculate Gini coefficient and Theil index in order to ana-lyze the equity in headcount, structure and distribution of primary health care personnel among all health care institu-tions nationwide. Results: Between 2008 and 2013, the number of primary health care personnel significantly in-creased and their quality was improved. Moreover, although the Gini coefficient and Theil index of primary health care personnel continuously decreased in China's eastern, central and western provinces the difference inside the east-ern China is the main factor affecting the overall difference. Conclusions:Not only the problems of insufficient num-ber and loss of personnel still coexist in the primary health care personnel in China, but also there are other issues such as the structural imbalance of professional titles is especially prominent, the urban-rural and regional gaps are still large, etc. The main reasons for these issues are related to the irrational incentive mechanism, the inefficiency of the synergistic effects of administrative staffing policy and some reform measures, etc. This paper suggests further re-forming the income distribution system and improving primary governance capability in order to strengthen the policies to synergically attract and stabilize primary healthcare personnel.
