1.Establishment and Application of "Information System for the Introduction and Examination and Approval of New Drugs" in Our Hospital

OBJECTIVE: To strengthen the management on procedures for the introduction,examination and approval of new drugs in hospital.METHODS: The experience on the current procedures for the introduction,examination and approval of new drugs in our hospital was summarized,and the Microsoft.NET technology was used to develop Web-based "information system of introduction,examination and approval of new drugs".RESULTS & CONCLUSIONS: The new system adopted three-level "browser / server" structural pattern and it is composed of external network new drug data collection system and internal network new drug examination and approval system.The system is advanced,stabile,practical,intelligent and safe,and it has simplified the new drug examination and approval procedure,enhanced the working efficiency.Its application is characterized by justice,publicity and transparency.
2.Efficacy and safety of glucocorticoids in community-acquired pneumonia: A systematic review and meta-analysis of randomized controlled trials

Objective To evaluate efficacy and safety of glucocorticoids for community-acquired pneumonia (CAP).Methods We searched relevant randomized controlled trials (RCT) from Pubmed,EMBASE,Cochrane Library,Chinese Journal Full-text Database,Chinese Biomedical Literature Database and traced the related reference to obtain the information that had not been found by using key words of steroids,glucocorticoids,cortisol,corticosteroids,community acquired pneumonia and CAP.The quality of RCT included was evaluated.Meta-analysis was performed with RevMan 5.0 provided by the Cochrance Collaboration.Results Seven RCTs involving 944 patients were included in the meta-analysis.Mean length of hospital stay in glucocorticoids treatment group was significant shorter than that in conventional treatment group (WMD =-1.70,95% CI:-2.01-1.39,Z =10.81,P <0.01).No significant differences were found regarding mortality (RR =0.77,95% CI 0.46-1.27,Z =1.03,P =0.30),mean length of ICU stay (WMD =1.17,95% CI:-1.68-4.02,Z =0.81,P =0.42),the risk of double infection (RR =1.32,95% CI 0.66-2.63,Z=0.79,P=0.43),the incidence of hyperglycemia (RR=1.84,95% CI0.76-4.41,,Z=1.36,P=0.17),the incidence of upper gastrointestinal bleeding (RR =1.98,95% CI O.37-10.59,Z =0.80,P =0.42) between conventional treatment group and glucocorticoids treatment group.Conclusions Glucocorticoids treatment used in patients with community-acquired pneumonia may shorten the course of illness and possess better safety,but could not reduce the mortality.
3.An experimental pathological study of acute intoxication by macleaya cordata

Objective To find out the main target organs, their main pathological changes and to explore the toxic mechanism of the macleaya cordata. Method SD rats were intoxicated by oral administration with extracts of macleaya cordata followed by pathological and electromicroscopic examination of main organs. Results The LD50 was 19.5% when 1.5g/ml macleaya cordata extract was oral administrated. All pocsoned rat's serum GPT, Cr, BUN, CK, CK-MB increased markedly. The EKG were abnormal. The weight of brain, liver, spleen, and kidneys decreased significantly. There were extensive congestion and hemorrhage in heart, brain, liver and kidneys. The mitochondria, endoplasmic reticulum and nuclear membrane of the rat's heart, brain, liver, and kidney were destroyed. Conclusion The results indicated that the toxic target organs of macleaya cordata were the heart, brain, liver and kidneys mainly. The toxic mechanism of macleaya cordata poisoning was the damage of the mitochondrial, endoplasmic reticulum and nuclear membrane directly causd by the macleaya cordata.
4.STUDY ON THE METABOLISM AND SUPPLEMENTATION OF WATER AND ELECTROLYTES TO ATHLETES DURING THE PERIOD OF ACUTE WEIGHT-REDUCTION

A field study on 24 weigbt--lifters and an experiment on rats imitating kuman acute food restrictionwere done in order to find out the metabolism of water and electrolytes during the acute weight-reductionperiod and the effect of appropriate supplementation. The results showed that food restriction caused very significant loss of the water and electrolytes withsigns of muscle cramp, dehydration, ketonuria etc. The experimental results in rats were observed duringfood restriction as following: 1. Serum potham and sodium decreased and the serum chloride and magnesium increased significantly. 2. Potassium and sodium contents of cardiac muscle were significantly lower than those of the control group,but potassium content of skeletal muscles was higber. 3. Blood lactic acid (3 minutes after running on a treadmill for half an hour ) was higber than thatbefote food restriction. Supplementation of appropriate minerals is beneficial to correct the above changes and keep the athletesfit and in good form.
5.Promotion of Function of Endothelial Progenitor Cells with Shexiang Baoxin Pill Treatment under Shear Stress.

The aim of this study was to investigate whether shear stress could promote function of endothelial progenitor cells (EPCs) with Shexiang Baoxin Pill (SBP) treatment in vitro, and to study whether shear stress contributed to vascular injury repair by EPCs. EPCs were isolated and characterized; EPCs' proliferation, migration, adhesion, tube formation and eNOS protein level in vitro were investigated by culturing confluent EPCs in 4 mg/mL SBP under physiological shear stress (15 dyne/cm2) for up to 24 hours. Afterwards, EPCs were transfused into rats after wire-induced carotid artery injury augmented re-endothelialization. The results showed that, compared to the SBP group, the shear stress+SBP group obviously enhanced EPCs proliferation, migration, adhesion, tube formation and eNOS protein expression in vitro (P<0.01). After one week, immunofluorescence staining showed that endothelial regeneration rate obviously enhanced in shear stress+SBP group (P<0.01). The present study demonstrates that shear stress can promote function of endothelial progenitor cells treated with SBP, which improves the vascular injury repair potentials of EPCs.

6.Effect of ethanol precipitation on chlorogenic acid from Flos Lonicerae Japonica

AIM: To test the effect of ethanol precipitation on the purifying technology of chlorogenic acid (CGA). METHODS: Using HPLC, the effect of the extraction of CGA from the crude Flos Lonicerae Japonica powder with three concentrations of ethanol was compared. RESULTS: Ethanol with concentration of 60% was optimal. CGA content was raised from 5.5% to 37.72% by use of ethanol precipitation and extraction in combination. CONCLUSION: The ethanol precipitation can get rid of the impurity in the crude powder of Flos Lonicerae Japonica effectively and the yield of CGA can reach 90.16%.
7.Effect of baicalin on the expression of receptor activator of nuclear factor-?B ligand and osteoprotegerin in human periodontal ligament cells

Objective To study the effect of baicalin on the expression of receptor activator of nuclear factor-?B ligand (RANKL) and osteoprotegerin (OPG) in cultured human periodontal ligament cells (HPDL cells) as well as its action mechanism. Methods We first constructed small interferring RNA (siRNA) eukaryotic expression vector targeted transforming growth factor ?Ⅱ receptor (TGF-?RⅡ),and then transfected it into T cells. Then HPDL cells together with T cells transfected with siRNA or not were placed in medium that had been added with lipopolysaccharide (LPS) and baicalin. They were divided into six groups and cultured for 48 hours. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to observe the effect of baicalin on OPG-RANKL expression in HPDL cells. Results The clone sequence correctly identified by RT-PCR was consistent with the designed target sequence. The recombinant vector was constructed successfully and the expression of TGF-?ⅡR of T cells which had been transfected with siRNA1 was inhibited obviously. Objective:In order to obtain safer HBV DNA vaccine,recombinant kanamycin resistance eukaryotic expression plasmid containing the gene of HBV surface antigen was constructed and used to study humoral immune response in BALB/c mice.Methods:HBV antigen middle protein(preS2+S) encoding gene fragment was isolated from the recombinant eukaryotic expression plasmid pcDNA-S 2S;subcloned into eukaryotic expression vector pVAX1.After confirmed the presence of the gene by restriction endonuclease analysis,it was used to immunize the healthy BALB/c mice at different doses,and the levels of anti-HBs in serum was determined by ELISA.Results:Restriction endonuclease analysis confirmed recombinant plasmid pVAX-S 2S was what was expected.Serum anti-HBs was detected at the 2nd week after DNA vaccination at all three doses(100 ?g,50 ?g,10 ?g per mice)and their titers were increased with time.Quantitative comparison of the serum anti-HBs levels revealed significant(P
