OBJECTIVE:To establish an HPLC method to determine the content of main component in idrocilamide ointment. METHODS: The samples were separated on HP-ODS column with column temperature maintained at room temperature. The mobile phase consisted of methanol-water (65∶35) with a flow rate of 1.0 mL?min-1. The detection wavelength was set at 276 nm and the injection volume was 20 ?L. RESULTS: The linear range of idrocilamide was 0.05～4 ?g?mL-1,the average recovery rate was 99.97% (RSD=0.80%). CONCLUSION: This method is sensitive,accurate and applicable for the determination of the content of main component in idrocilamide ointment.

AIM:To construct human interleukin-18 receptor ?(IL-18R?) and interleukin-18 receptor ?(IL-18R?) eukaryotic expression vectors named pcDNA3.1-zeocin/IL-18R? and pcDNA3.0/IL-18R? respectively,and then stably transfect pcDNA3.0/IL-18R? into 293 cells in order to establish the cell line stably expressing IL-18R? and cell model which responds IL-18 signal transduction in vitro. METHODS:The experiment was performed at the laboratory of Department of Immunology,Peking University Health Science Center from June 2006 to May 2007. DNA encoding IL-18R? or IL-18R? was amplified and cloned into pcDNA3.1-zeocin or pcDNA3.0 vector,respectively. The recombinant of pcDNA3.0/IL-18R? was transfectd into 293 cells. After screening culture by G418,stable transfected cell line was established and the expression of IL-18R? was identified by Western-blot assay. The stable cell line with the combinant eukaryotic expression vector pcDNA3.1-zeocin /IL-18R? to establish the IL-18 signaling pathway,which was detected by NF-?B-dependent Luciferase. RESULTS:The eukaryotic expression vectors of IL-18R? and IL-18R? were constructed successfully. The gene of IL-18R? stably expressed in 293 cells and cell model of IL-18 signaling transduction in vitro could be established in these stable 293 cell lines. CONCLUSION:The establishment of NF-?B-activated cell model in vitro which responds to IL-18 can provide solid foundation for further experimental studies on IL-18 signaling transduction pathway.

Objective To explore the blood level lead and the related high-risk factors in Uigur and Han children in Urumqi by answer tree. Methods Cluster sampling was used to investigate blood lead level of 988 Uigur and Han children aged 6-10 years,the related high-risk factors were investigated by using a standardized questionnaire,the related high-risk of blood lead level was analyzed by answer tree. Results The geometric mean of blood lead concentration of children was 54.46 ?g/L,in 23.28% of the children,the blood lead concentration was more than 100 ?g/L . No significant blood lead level difference was seen between Uigur and Han children. Such high-risk factors as parents occupation,bad habits and housing condition were associated with the blood lead level. Conclusion The blood lead level and lead poisoning rate of children in Urumqi are higher,many factors may influence the blood lead level of children in Urumqi.

AIM To investigate the effects of apoptosis and the expression of telomerase hTERT mRNA activity of HL-60 cells treated with As 2O 3 in different concentrations. METHODS HL-60 cells was cultured with As 2O 3 in different concentrations for 48 hours, then, cell apoptosis was detected with flow cytometry and the expression of telomerase hTERT mRNA was determined with RT-PCR respectively. Objective To investigate the effect of cosmetic surgery on the facial soft tissue injuries.Methods The clinical data of 52 patients with facial soft tissue injuries from June 2013 to June 2014 were analyzed retrospectively.And plastic surgical techniques were applied to the early treatment of facial soft tissue injuries in patients,according to the size of defect and the degree of deformity of the patients,different plastic surgery treatment was chosen,such as skin flap or skin graft to repair wound surfaces.In this process,one must follow sterile noninvasive principle strictly with emphasis on the technique of plastic surgery such as entire debridement,wound healing application of skin flap and so on.Results Total 49 cases of facial soft tissue injuries were healed by first intention without significant complications,while 4 cases of them had mild scars.Absorbable suture exposition appeared in 3 patients after primary healing.Those sutures were romoved after disinfected and no sequelaes were happened.All patients were recovered with good appearances and functions to the most exent.Conclusions The treatment of facial soft tissue injuries,by applying plastic and aesthetic surgery,could reduce scar and postoperative deformity,and secondary corrective procedures can be avoided in most patients.

Purpose To investigate the expression of c-myc and MDM2 protein in multiple myeloma (MM) and their correlation with clinical stage.Methods Immunohistochemical SP three-step method was used to detect the expression of MDM2 and c-myc protein in 60 cases of MM with different clinical stages and 10 cases of nontumorous bone marrow tissue.Results The positive rate of c-myc protein in MM and nontumorous bone marrow tissue were 71.7％ and 10.0％;the positive rate of MDM2 protein in two tissues were 80.0％ and 20.0％,respectively.The expression difference of two proteins between MM and nontumorous bone marrow tissue was statistically significant (P ＜ 0.05).The expression of c-myc and MDM2 protein was positively correlated with MM clinical ISS stage (P ＜ 0.05),but irrelevant with the age and gender.The expression of c-myc and MDM2 protein in MM was positively correlated (P ＜ 0.05).Conclusion The c-myc and MDM2 protein have a highly expression in MM,and it may be relative to the occurrence of MM and clinical stage.In the MM,c-myc and MDM2 protein may have synergetic functions and promote the development of tumor.
