1.Epigenetic regulation in spermatogenesis.

Spermatogenesis is a process consisting of spermatogonial proliferation, spermatocytic meiosis, and spermiogenesis, and is also considered to be a process in which heterochromatins gradually aggregate and finally reach a highly condensed formation in the sperm head. Recent studies show that epigenetic regulation plays a key role in spermatogenesis. This review discusses the mechanisms of epigenetic regulation in spermatogenesis in three aspects, DNA methylation, histone modification, and noncoding RNAs. These factors are essential for spermatogenesis, fertilization, and embryogenesis by mutual regulation as well as by gene expression regulation, transposon activation, sex chromosome inactivation, and genome imprinting.

2.Comparison between sliding window and step and shoot

Objective To compare and analyze DMLC(Sliding Window,SW)and SMLC(Step and Shoot,SS)for delivering IMRT.Methods 5 patients with nasopharyngeal carcinoma were treated with radical intensity modulated radiation therapy using Varian 23EX and Helios tool on a Varian Eclipse system.Different modalities to deliver IMRT were considered for Sliding Window(SW) and Step and Shoot(SS) techniques using a different number of intensity levels(e.g.5,10 and 20).The total beam-on-time,total delivery time and a number of dose-volume parameters regarding PTV and OARs were considered.Results Comparing with the DVH,it was found that SW was the best of the four modalities in the dose distribution of PTV,but SS was better when considering the protection of OARs.The total beam-on-time(MUs) requirement for SS was 9~23% less than SW,but the total delivery time(in minutes)was about twice as long.Conclusion With the number of intensity level of 10 or more,no differences between SS and SW can be appreciated in the dose distribution of PTV and OARs sparing.Referring to the quality assurance,only leaf position needed to be checked in SS,whereas both leaf position and leaf speed need to be checked in SW,so it is proposed to use SS10 for delivering IMRT.

4.Heart and lung transplantation from the same donor to different recipients at the same time:report of 6 cases

Objective To improve the efficiency of donated organs and to investigate the possi- bility and operation protocol of heart and lung transplantation from the same donor to different reci- pients.Methods The organs of heart and lung were obtained from three donors through the method of orthotopic lavage and whole cutting,and transplanted to end-stage patients with heart diseases or lung diseases.First,the donor was subjected to mechanical ventilation,thoracotomy and lavage of heart and lung.The heart and lung were cut in whole and separated.The recipients were subjected the re- trograde lavage of heart and lung.In the patients receiving unilateral lung transcription,the left and right lungs were isolated.Among 3 recipients of lung transcription,one underwent left unilateral lung transplantation and 2 bilateral lung transplantation.Three cases received orthotopic cardiac transplan- tation by using bicaval orthotopic heart transplantation (BOHT) anastomosis method.Results One recipient of left unilateral lung transplantation recovered well after operation.In one recipient under- going bilateral lung transplantation,right upper lung embolism occurred.After re-operation of cutting the right upper lung on the 9th day of the first one,he recovered from the illness.In another recipient receiving bilateral lung transplantation,anastomotic stenosis of trathea and pulmonary infection occurred after operation.After expectant treatment,he recovered from the illness,but died from serious infection 7 months after operation.One of the heart recipients was found having renal failure of first stage,and recovered from disease after hemodialysis treatment.Quality of life was good in all 3 heart recipients after operation and their heart function achieved the restoration of 0-Ⅰstage.Conclusion The treatment protocol of transplantation from the same donor to different recipients at the same time, which can make full use of the organs from donors,is feasible and has good effect.
5.Allogeneic platelet-rich plasma promotes wound collagen synthesis in diabetic rats

BACKGROUND:Platelet-rich plasma has benefit in diabetic wound healing;however, the effect of al ogeneic platelet-rich plasma is stil unclear. OBJECTIVE:To observe the effect of al ogeneic platelet-rich plasma on col agen synthesis during diabetic wound healing. METHODS:Streptozocin-induced diabetic rats were randomly divided into al ogeneic platelet-rich plasma group and saline control group. Each group had 15 rats. A 1 cm2 ful-thickness skin defect on the rat dorsum was excised. Platelet-rich plasma or saline was applied. Platelet-rich plasma was prepared from the whole blood of al ogeneic healthy rats. Animals of each group were sacrificed on 3, 7, 14 days post operation. The differences of the wound closure rate, morphological character, hydroxyproline content and the relative mRNA expression of the col agen were observed. RESULTS AND CONCLUSION:The wound closure rates were lower in control group on day 3, 7, 14 post operation (P<0.05). Masson staining showed a decreased, disorder, loose col agen fibers distribution in diabetic control wound tissue. The results of the hydroxyproline test showed hydroxyproline content was significantly higher in platelet-rich plasma group at each time point (P<0.01). The relative mRNA expression of type I and III col agen presented a higher expression in platelet-rich plasma treated wound tissue at each time point (P<0.05). And the ratio between type I and III col agen was higher in platelet-rich plasma group at each time point (P<0.05). In summary, al ogeneic platelet-rich plasma can promote diabetic wounds healing, which may attribute to an enhanced col agen synthesis.
6.Effect of Yiqi Yangyin Recipe and Its Different Assembling on Apoptosis Rate and Expression of Associated Protein in Acute Myeloid Leukemia Mice

Objective To observe the effects of Yiqi Yangyin Recipe (supplementing qi and nourishing yin) and its different assembling on apoptosis rate and associated protein expression of c-myc and survivin in mice, and explore its mechanism. Methods NOD-SCID mice were injected with l×106 KG1a cells from the tail vein after irradiation of 60Co. Model mice were randomly divided into control group, whole recipe group, strengthening body resistance group, eliminating pathogens group. The treatment groups were given correspongding administration, and control group was given equal volume of NS. Flow cytometry was applied to measure apoptosis rate. The immunochemistry method was used to detect the expression of c-myc, survivin. Results Apoptosis rate in all treatment groups were increased (P<0.01). The expression of c-myc and survivin in lymph node tissues in all treatment groups were significantly declined compared with control group (P<0.05). The effect of whole recipe group on the all indices was superior to strengthening body resistance group and eliminating pathogens group (P<0.05). Conclusion Yiqi Yangyin Recipe can decrease the expressions of c-myc and survivin, induce apoptosis in leukemia cells, therefore shows the therapeutic effect.

8.Clinical Research of Neck-type Cervical Syndrome Treated with Isometric Neck Strength Training

Objective To evaluate the effectiveness and safety of isometric neck strength training for the treatment of neck-type cervical syndrome.Methods The qualified patients with neck-type cervical syndrome were divided into two groups by stratified randomization: the treatment group(N=55)received isometric neck strength training,and the control group(N=40) received manipulation treatment.Two weeks constituted one treatmeat course.Visual Analog Scale(VAS) and Vernon Cervical Disability Index(VCDI) were used to evaluate the efficacy,and the safety was also monitored.Results All of the 95 enrolled patients were followed up for 3~11.4 months.No aggravation was found in the treatment group,but in the control group one patient developed into cervical spondylotic radiculopathy and one patient developed into sympathetic cervical spondylosis.The frequency of serious neck pain attack within 3 months was 6.65?2.62 times in the control group and 2.27?1.50 times in the treatment group,the difference being significant(P0.05);the result of follow-up on the 3rd month after treatment showed that the difference of VCDI score was significant between the two groups(P
9.A comparative study of three-dimensional high-resolution MR imaging CPR and MRVE displayed in trigeminal neuralgia vascular responsibility

Objective To investigate the value of the three-dimensional high-resolution magnetic resonance imaging curve planar reformation(CPR)combined with virtual endoscopy reconstruction in vascular compression of trigeminal neuralgia.Methods 65 pa-tients with trigeminal neuralgia performed three-dimensional MR doubly excited balanced steady-state free precession sequence (3D-FIESTA-C),and CPR combined with virtual endoscopy reconstruction was used to evaluate the relationship of neurovascular anato-my.And 60 cases with microvascular decompression (MVD)surgery were analyzed.Results 65 cases of 3D-FIESTA-C CPR com-bined with MRVE showed responsibility vascular with a positive rate of 92.3% (60/65).60 cases of all patients with the results of surgery showed responsibility blood vessels and oppression parts with 3D-FIESTA-C CPR combined with MRVE,and it was found that the findings of oppression had a good consistency with MVD with the diagnose accordance rate of 95% (57/60).Conclusion Three-dimensional high-resolution magnetic resonance imaging CPR combined with virtual endoscopy reconstruction can be a new method to clearly and accurately show the anatomical relationship between blood vessels and nerves,which helps to improve the posi-tive rate of detection and to give operative guidance.
10.Prediction of nuclear grade of renal clear cell carcinoma based on MRI texture analysis in combination with imaging features

Objective:To explore the application value of MRI texture analysis in combination with imaging features to predict the WHO/International Society of Urological Pathology (ISUP) nuclear grading in pre-operative patients with clear cell renal carcinoma (ccRCC).Methods:MRI images of 78 patients diagnosed as ccRCC by surgical pathology from July 2016 to July 2020 in First Affiliated Hospital of Fujian Medical University were retrospectively analyzed. According to the WHO/ISUP grading system, the patients were divided into low grade group (49 cases, grade Ⅰ in 2 cases and grade Ⅱ in 47 cases) and high grade group (29 cases, grade Ⅲ in 25 cases and grade Ⅳ in 4 cases), and then were assigned to training set ( n= 63) and validation set ( n=15) in a ratio of 7∶3 using random indicator method. MRI radiological features were evaluated and MRI imaging texture features were extracted. The largest-diameter slice of lesion on cross-sectional images was selected and ROIs were drawn on T 2WI and corticomedullary phase (CMP) images, respectively. Quantitative texture analysis software MaZda was used to extract texture features, including gray-scale histogram, co-occurrence matrix, run-length matrix, gradient, autoregressive model and wavelet transform. The extracted texture features were preliminarily selected by the combination of Fisher, probability of classification errorand average correlation coefficient, and interaction information, and then the reduced texture parameters or imaging features were tested by the independent sample t test, Mann-Whitney U test or χ 2 test. Parameters with statistically significant differences were used to construct a multi-factors binary logistic regression model and the ROC curve was used to analyze its effectiveness in predicting high grade ccRCC. Results:In training set, there were significant differences intumor length, shape and margin, enhancement degree of CMP, vein thrombosis and 47 texture features between the low and high grade ccRCC groups. In the training set, 7 multi-factors binary logistic regression model were constructed, including radiological features model (M1), T 2WI texture features model (M2), CMP image texture features model (M3) and combination radiological features of T 2WI texture features model (M4), combination radiological features of CMP images texture features model (M5), combination T 2WI texture features of CMP images texture features model (M6) and combination of all features model (M7). The area under ROC curve of M7 in predicting nuclear grading of ccRCC was the largest, which were 0.901 (95% CI 0.828-0.974) and 0.820 (95% CI 0.564-0.974) in the training set and validation set, respectively. Conclusion:MRI texture analysis combined with imaging features is hopeful to be an effective preoperative noninvasive method in predicting WHO/ISUP grading of ccRCC.
