1.Effect of trichosanthin on mast cell degranulation and histamine release and its relationship with complement activation

The effect of trichosanthin on mast cell degranulation and histamine release were observed with both Alcian blue staining method and fluorescent histamine determination method. It was showed that, ① in vitro trichosanthin showed no direct effect on the mast cell degranulation and histamine release even in the presence of fresh serum; ② in vivo administration on of trichosanthin in mice could cause significant mast cell degranulation and histamine release in inflammatory situ, as shown by the decreased number of Alcian blue stained nondegranulated mast cell in the peritoneal cavities; ③ intra peritoneal injection of trichosanthin in Wistar rats could significantly increase histamine level in the whole blood, while inducing prominent exudation of protein into the peritoneal cavities; ④ the supernatant of peritoneal exudate lavaged 12 hours after trichosanthin injection could cause mast cell degranulation. These results implicate that, although it showed no direct effect on mast cells, trichosanthin could cause mast cell to degranulate and release histamine via in vivo activation of some soluble mast cell degranulating factors. It was discovered in further study that the depletion of complement by cobra venom factor injection revealed no effect on the production of mast cell degranulation factor, suggesting that, in normal animals, the trichosanthininduced complement activation had no significant influence on the mast cell degranulation and histamine release.

3.Protein Expression Profile in Serum of Male Rats Chronically Exposed to Low-level 2,3,7,8-tetrachlorodibenzo-p-dioxin

Objective To explore the chronic effect of low levels of 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD) exposure on the protein expression profile in the serum of rats and to get the further understanding of the molecular mechanism of TCDD toxicity.Methods Thirty-two male SD rats were randomly divided into three TCDD exposed groups treated with TCDD at the doses of 875,350 and 140 ng /kg by gavage,once a week,for 29 consecutive weeks respectively and one control group treated with single corn oil.The proteins in the serum were separated and analyzed by two-dimensional gel electrophoresis and mass spectrometry,the histopathological examination of livers was performed at the same time.Results TCDD induced significant fatty degeneration and necrosis in the livers.Several interesting volume-altered protein spots were identified.Among these proteins,Bal-647 was down-regulated,apolipoprotein E was up-regulated by TCDD and fetuin-like protein was only expressed in the group of 875 ng/kg.Conclusion The differentially expressed proteins may contribute to a better understanding of the toxicity induced by TCDD and may be used as the biomarkers of effect for TCDD exposure.
4.Preparation and Application of Naringin Sensor Based on Molecularly Imprinting Technique

The sensor of naringin(NG), a non-electroactive substance, was prepared based on the molecular imprinting technique.Using cyclic voltammetry technique (scan rate is 100 mV/s), the naringin imprinting sensitive film, poly-o-aminophenol was coated on the surface of a graphite electrode in the presence of naringin which was considered as the template, and characterized by SEM and X-ray reflective spectrophotometry (XRR).Using K_3Fe(CN)_6 as an electroactive marker, the electrochemical properties of the NG sensor were investigated by CV, electrochemical impedance spectroscopy (EIS), differential pulse voltammmetric and chronoamperometric.The results showed that the imprinted electrode was significantly different from the non imprinted electrode in morphologies and electrochemical properties, and a linear relationship between the peak current and the naringin concentration was found in the range of 6.0 × 10 ~(-5)-1.4 × 10 ~(-4) mol/L with a detec tion limit of 1.6 × 10 ~(-5) mol/L.Moreover, the imprinted electrode exhibited a good selectivity and rapid response to the naringin template molecules, as well as an excellent reproducibility(RSD = 1.8 %, n=5).
5.CHARACTERISTIC AND TREATMENT OF ISOLATED TUBERCULOSIS OF THE PANCREAS

Isolated tuberculosis of pancreas is very rare clinical entity. Since 1970, we have treated 9 patients, 3 men and 6 women (average 48.3 years), with isolated tuberculosis of pancreas. 2 patients were diagnosed as pancreatic tuberculosis, 1 patient pancreatic cyst and 6 patients pancreatic carcinoma before operation. They all underwent exploratory laparotomy, and were confirmed to be pancreatic tuberculosis by histopathology. Their common characteristics were that tuberculosis was only localized in pancreas without tuberculosis at extrapancreatic sites and that the treatment of anti-TB drugs had much effect on them. By analysis, it was considered that isolated tuberculosis of pancreas was usually mistaken for pancreatic carcinoma because it localized in pancreas, the contents of masses in pancreas should be noted carefully in order to avoid misdiagnosis and treatment of anti-TB drugs should be taken in time.
6.Research on performance evaluation framework of public health in rural areas

Objective To provide reference for improving the performance of public health by establishing performance evaluation framework of public health in rural areas of China.Methods The performance evaluation framework of public health in rural areas is established by literature research,theoretical derivation and Delphi method.The results of expert consultation are tested by W test of Kendall coordination coefficient and chi-square test.Results The performance evaluation framework of public health in rural areas contains four conceptual modules,ten performance goals and twenty-eight performance evaluation indexes.The positive coefficient of experts is 0.889.The average values of authoritative grade of experts are between 0.681~0.715.The Kendall W uniformity coefficients of the necessity,sensitivity and maneuverability of the indexes are respectively 0.167,0.209 and 0.185.The P values are less than 0.01.Conclusion The performance evaluation framework of public health in rural areas has certain application value.It is necessary to pay attention to scientific nature and applicability in developing performance evaluation of public health in rural areas.
7.A new method for the treatment of refractory fistula after breast augmentation

Objective To explore a new surgical method for the treatment of refractory fistula after breast augmentation and to improve patients healing efficiently.Methods This paper had collected two groups,a total of 178 cases of injected breast augmentation,who had shown the polyacrylamide hydrogel(PAAG)mixture shift,and caused abdominal fistula.Among them,89 cases further adopt new design in the treatment of fistula by using breast muscle membrane tissue flap and improve the effect of fistula treatment after the removal of fistula.And the other 89 cases were token the pure clean the cavity away caused by the abnormal material shift into the chest and abdominal wall.Results It was observed that PAAG widely distributed in the chest and abdominal wall and formatted the fistula connected mammary gland,axillary,chest abdominal wall.The area of lesions lacuna shrink by 80% for the group used tissue flap after 2 weeks,but those not with tissue flap shrink by 45%(P<0.01).At 4th week,all the patients of flap group achieved tissue healed,while the completely heal ratio of control group was only 73%.Compared two groups of healing rate,the group with tissue flap was obviously better than ones only with surgical method.Conclusion It suggests that main reasons for the liquidity bag and huge cavity are PAAG mixture shift into thoracic and abdominal wall and formed chronic fistula and connected the chest and abdominal wall.Local muscle membrane tissue flap can obviously promote the chest and abdominal wall fistula healing.
8.Prognostic values of tumor-stroma ratio and tumor-infiltrating lymphocytes in stagesⅡandⅢcolorectal cancer

Objective:To investigate the prognostic values of tumor–stroma ratio and tumor-infiltrating lymphocytes in colorec-tal cancer. Methods:A total of 218 stageⅡorⅢprimary colorectal cancer patients were analyzed for the tumor–stroma ratio (TSR) and tumor-infiltrating lymphocytes (TIL) by using HE stained histological sections. The relationship between TSR and clinicopatholog-ic variables and the difference in clinical outcomes of different groups were also analyzed. Results:Overall survival rates for the stro-ma-low group were significantly higher than the stroma-high group in stagesⅡandⅢcolorectal cancer (P<0.05). However, the differ-ences in TSR were not correlated with clinicopathologic features such as gender and age (P>0.05). Overall survival rates of patients with high TIL were significantly higher than patients with low TIL (P<0.05). Patients with high TIL had notable better prognosis than patients with low TIL (P<0.05) in the stroma-low group. Conclusion:TSR and TIL are independent prognostic factors for stagesⅡandⅢcolorectal cancer. The combined estimates of TSR and TIL in routine pathology diagnoses may provide more evidence to predict the prognosis of stagesⅡandⅢcolorectal cancer.

All twenty seven patients suffered from Ⅱ grade spondilolisthesis were treated by posterior decompression, fixation with RF system and interbody fusion with TFC. One to three yearsafter operation, pain relief rate was 95%, lumbar fusion ratewas 95%,slip correction was 93.8%, and disc space was increased by 34%. It is concluded that this kind of operation can achieve satisfactory clinical results.
10.Establishment of a rat model of chronic heart failure by transverse aortic constriction and observation of its pathological process

Objective To observe the pathological changes of heart failure caused by transverse aortic constriction in rats.Methods Partial thoracotomy was performed to the second rib and the transverse aortic constriction was performed between the innominate and left carotid arteries to establish a model of heart failure in 24 rats.The same operation was performed on another 8 rats, except for the ligation of the transverse aorta.Echocardiographic assessment, hemodynamic measurement, myocardial histopathological examination and NT-proBNP measurement were performed to the sham group at 12 weeks and model group at 4 weeks, 8 weeks, 12 weeks after the operation.Result At 4 weeks after the operation, NT-proBNP, EF, FS and -dp/dtmax of the model group was significantly increased and LVESV, +dp/dtmax of the model group were significantly decreased (P<0.05).At 8 weeks after the operation, EF and-dp/dtmax of the model group were increased and +dp/dtmax of the model group was significantly decreased (P<0.05).At 12 weeks after the operation, NT-proBNP, EF and +dp/dtmax of the model group were decreased, and LVESV, LVEDV and -dp/dtmax of the model group were increased (P<0.05).The cardiomyocytes became hypertrophic and lined up in disorder at 4 weeks after the operation.Pathologic examination of the myocardial tissue showed connective tissue proliferation and inflammatory cell infiltration at 8 weeks after the operation, and cardiomyocyte apoptosis and collagen fiber deposition at 12 weeks after the operation.Conclusions Transverse aortic constriction induces heart failure in rats.The pathological processes are compensatory hypertrophy at 4 weeks after the operation, initial reaction of decompensation at 8 weeks after the operation, and heart failure at 12 weeks after the operation.
