1.Relationship of Human AFP Variant Peptide and Carbohydrate Difference and Clinical Diagnosis of Primary Hepatocellular Carcinoma

AFP from fetal serum and serum of patients with primary hepatic cancer were purified by affinity chromatography. Then, the lentil lectin-reactive and nonreactive variants of these purified glycoproteins were prepared by affinity chromatography with immobilized lectin. Glycopeptides and oligosaccharides were prepared from variants by protease treatment and hydrazinolysis, respectively, and subjected to carbohydrate and amino acid analysis. A small difference in the carbohydrate compositions of each variant was observed. Analysis of chemical compositions of AFP variants is useful for the differential diagnosis between benign liver diseases and primary hepatic cancer.

The goal "Rehabilitation for All" can beachieved only if "rehabilitation in community and service in family" be reached.
3.The value of transurethral cylindrical dilatation of the prostate in the treatment of patients with benign prostatic hyperplasia

Objective:To observe the effects of transurethral split of the prostate (TUSP) on the urodynamics, the sexual function and the quality of life of benign prostatic hyperplasia.Methods:Eighty patients with benign prostatic hyperplasia admitted to the Deqing People′s Hospital of Zhejiang Province from January 2017 to June 2019 were enrolled into the research objects prospectively. According to the random digits table method, they were divided into group A and group B, with 40 cases in each group. The group A was treated with TUSP, while the group B was treated with bipolar transurethral plasma kinetic prostatectomy (TUPKP). The operation time, intraoperative blood loss, bladder irrigation time, urinary catheter indwelling time, hospitalization time of the two groups were compared and surgical effects were evaluated. The changes of hemoglobin (Hb), serum sodium (Na), post-void residual volume (PVR), maximum urinary flow rate (Q max), international prostate symptom score (IPSS) and international index of erectile function score (IIEF-5), the quality of life index (QOL) of 2 groups before and after the operation were measured. Surgical complications were recorded. Results:The operation time, bladder irrigation time, urinary catheter indwelling time and hospital stay in group A were all shorter than that in group B: (15.63 ± 4.17) min vs. (58.79 ± 10.45) min, (6.26 ± 1.17) h vs. (45.51 ± 10.03) h, (3.07 ± 0.68) d vs. (5.67 ± 1.51) d, (3.63 ± 0.43) d vs. (6.08 ± 1.72) d, (18.32 ± 2.79) ml vs. (65.26 ± 20.64) ml, and there were statistical differences ( P<0.05); there were no statistical difference between 2 groups in surgical curative effect grade and Na 1 d after operation ( P>0.05); 1 day after operation, Hb in group A was higher than that in group B: (115.63 ± 9.78) g/L vs. (109.65 ± 8.36) g/L, and there was statistical difference ( P<0.05); PVR, Q max, IPSS, IIEF-5 and QOL 3 and 6 months after surgery were improved in 2 groups ( P<0.05), but the difference between 2 groups was not statistically significant ( P>0.05); the total complication rate in group A was lower than that in group B: 10.00% (4/40) vs. 27.50% (11/40), and there was statistical difference ( P<0.05). Conclusions:TUSP and TUPKP are equally effective in the treatment of benign prostatic hyperplasia, both of which can improve the symptoms of prostatic hyperplasia, improve the quality of life and improve sexual function. But TUSP has less intraoperative bleeding, shorter operation time, faster postoperative recovery, fewer complications, and higher safety.
4.Relationship between mild cognitive impairment and body mass index in older adult patients with type 2 diabetes mellitus

Objective:To investigate the relationship between mild cognitive impairment (MCI) and body mass index (BMI) in older adult patients with type 2 diabetes mellitus (T2DM).Methods:A total of 327 patients with T2DM who received treatment in Wuyunshan Sanatorium from January 2016 to May 2019 were included in the T2DM group. Patients in theT2DM group were subdivided into an MCI group ( n = 73) and a non-MCI group ( n = 254) according to whether they had MCI. An additional 100 older adult volunteers who concurrently received physical examination were included in the control group. Sex, age, years of education, monthly family income, body mass index, living habits (drinking, smoking) and laboratory indexes were compared among the groups. The influential factors of MCI in patients with T2DM were analyzed by logistic regression model. The predictive value of BMI for MCI in older adult patients with T2DM was evaluated by receiver operating characteristic (ROC) curve. Results:Age, monthly family income, the proportion of patients with a history of diabetes mellitus, BMI, fasting blood glucose, glycosylated hemoglobin (HbA1C), low-density lipoprotein cholesterol (LDL-C) in the T2DM group were (73.10 ± 9.56) years old, 8 926 yuan RMB, 189/327, (24.18 ± 2.64) kg/m 2, (6.96 ± 0.88) mmol/L, (7.10 ± 0.84)%, (7.32 ± 0.84) mmol/L, respectively, which were higher than those in the control group [(68.28 ± 8.21) years old, 6 715 yuan RMB, 13/100, (22.30 ± 1.74) kg/m 2, (4.51 ± 0.72) mmol/L, (5.62 ± 0.68)%, (7.04 ± 0.67) mmol/L, t = 4.554, χ2 = 18.601, 61.654, t = 6.668, t = 25.360, 16.077, 3.049, all P < 0.05]. In the MCI group, the proportion of patients having a monthly family income < 5 000 yuan RMB, the proportion of patients having a history of diabetes mellitus, BMI, HbA1c value were 29/73, 60/73, (24.92 ± 2.43) kg/m 2, (7.54 ± 0.88)%, respectively , while they were 70/254, 129/254, (23.77 ± 2.59) kg/m 2, (6.92 ± 0.81)%, respectively in the non-MCI group. There were significant differences in these indexes between MCI and non-MCI groups ( χ2 = 6.144, 22.927, t = 3.389, 5.652, all P < 0.05). Multivariate logistic regression analysis showed that BMI and HbA1c were the influential factors of MCI complicated by T2DM in older adult patients ( OR = 0.274, 0.192, both P < 0.05). Monthly family income and family history of diabetes mellitus were not closely related to the development of MCI in older adult patients with T2DM ( OR = - 0.154, 0.093, both P > 0.05). The ROC curve revealed that when BMI value was 24.49 kg/m 2, Youden index was the largest (0.510), the corresponding sensitivity was 83.86%, and the specificity was 67.12%. The area under the ROC curve was 0.766 [95% CI (0.699 - 0.832)]. Conclusion:BMI is an influential factor of MCI development in older adult patients with T2DM, and may be one of the important indicators for early prediction of MCI.
6.Role of CXCR4 expression in progress and prognosis of breast cancer

CXCR4, a receptor for stromal cell-derived factor-1 (SDF-1, also know as CXCL12) and a member of the chemokine receptor family, has been shown to play an important role in the proliferation, migration, invasion and metastasis of breast cancer cells. CXCR4 expression is an early event in breast cancer development and may serve as a biomarker for early detection and diagnosis of breast cancer. Expression of CXCR4 has been demonstrated to correlate with metastasis and poor prognosis in breast cancer. Inhibition of CXCR4 expression may be a potential target of breast cancer treatment.
7.The expression and clinical significance of MKK4 in osteosarcoma

Objective To explore the clinical significance of expression of MKK4 in osteosarcoma,and to discuss the relationship between MKK4 and osteosarcoma occurrence,development and prognosis.Methods The expression of MKK4 protein was determined with immunohistochemical SP method in 30 cases of osteosarcoma.Results The MKK4 was obviously expressed in both nucleus and cytoplasm in osteosarcoma.The expression of MKK4 in the cytoplasm of osteosarcoma was positively correlated with clinical stage (r =0.72,P =0.000),the expression of MKK4 in the nucleus of osteosarcoma had no correlation with clinical stage (r =0.44,P =0.080).MKK4 expression in the cytoplasm was higher in transfer osteosarcoma than no transfer osteosarcoma(x2 =9.69,P <0.01),while no obvious difference in the nucleus of osteosarcoma(x2 =1.00,P =1.000).Conclusion The MKK4 is highly expressed in the nucleus and cytoplasm of osteosarcoma,suggests that MKK4 may be related with the carcinogenesis development of osteosarcoma.The expression of MKK4 in the cytoplasm of osteosarcoma is related to the malignant,metastasis and prognosis of osteosarcoma,may be used as a potential prognostic indicator of judgment osteosarcoma.
8.Effect of encapsulation in liposomes on toxic effect of bupivacaine on spinal cord in rats

ObjectiveTo study the toxic effect of bupivacaine encapsulated in liposomes on spinal cord in rats.MethodsOne hundred and eight SD rats (200-225 g) in which intrathecal (IT) catheter was successfully implanted without complications were randomly divided into 6 groups ( n =18 each):control group (group C) ; liposome group (group L) ; 0.5% and 1.0% bupivacaine groups (groups B1 and B2 ) and 0.5% and 1.0% bupivacaine encapsulated in liposomes groups (groups LB1 and LB2 ).In groups L,B1,B2,LB1 and LB2,liposome,0.5 % bupivacaine,1.0 % bupivacaine,0.5 % liposomal bypivacaine and 1.0 % liposomal bupivacaine 20 μl were injected IT respectively once a day for 7 consecutive days,while in group C nothing was injected IT.Pain threshold was measured by mechanical stimulation of the plantar surface of hindpaw.Motor function of the hindlimbs was also assessed.The animals were sacrificed at 8 day after IT injection.The lumbar segment of the spinal cord was removed for microscopic examination,detection of neuronal apoptosis (by flow cytometry) and Fos protein expression (by immuno-histochemistry).Results1.0% bupivacaine IT significantly increased the percentage of apoptotic neurons in group B2 as compared with control group.0.5% and 1.0% bupivacaine IT significantly increased the number Fos protein positive cells in group B1 and B2 as compared with group C.1.0% bupivacaine IT induced severe histologic damage including shrinkage of nucleus and vacuole formation in mitochondria.Encapsulation of bupivacaine in liposomes significantly attenuated bupivacaine-induced increase in apoptosis and Fos protein expression and histologic damage in group LB2 as compared with group B2.ConclusionThe encapsulation in liposomes can decrease the neurotoxicity of 1.0 % bupivacaine administered IT in rats.
9.Experimerntal study of etanercept effects the wear debris-induced osteolysis

[Objective]To detect the effect of Etanercept on the Ti particles induced TNF-?IL-1 and IL-6 production by macrophage and to evaluate the validity of the etanercept on treatment of aseptic loosening of prosthesis.[Method]Separate and Cultivate mouse peritoneal macrophages were divide into 5 groups after 24 h.Group A was treated with M? alone,group B with M?+Ti particles,group C with M?+Ti particles + etanercept(10 ng/ml),group D with M? Ti particles + etanercept(100 ng/ml),group E with M? + Ti particles + etanercept(1000 ng/ml).After 18 h,the production of TNF-? IL-1 and IL-6 in culture supernatants was detected by ELISA.[Result]The levels of TNF-? IL-1 and IL-6 production were much higher in group B than those in group A,D,E(P0.05).[Conclusion]Is shows the Ti particles could stimulate M? to excrete profuse TNF-? IL-1 and IL-6 production,etanercept can significantly inhibited the production of TNF-? IL-1 and IL-6 secreted by Ti particles induced macrophages in a dose-dependent manner and hope to be a therapeutic candidate for the prevention of aseptic loosening.
10.Changes of N-acetyl-?-glucosaminidase in Sera of Patients with Hepatoma and Other Diseases

Activities of N-acetyl-?-glucosaminidase (NAG) in sera of normal subjects and patients with hepatocellular carcinoma (HCC) and other diseases were microquantitatively determined by spectrophotometry. The results showed that after the reaction for 4 h, activity of serum NAG in patients with HCC (397.10 ? 174.97 nmol/ml) was obviously higher than that in normal subjects (280.00 ? 63.83 nmol/ml). The elevation of serum NAG in HCC patients had no relationship with their serum a-fetoprotein. However, the increase of serum NAG activity in some patients with benign liver diseases was always accompanied by the abnormality in their liver functions. It is suggested that serum NAG has some referable values in the diagnosis of HCC.
