Single nucleotide polymorphisms (SNP) is an important molecular marker in traditional Chinese medicine research, and it is widely used in TCM authentication. The present study created a new genotyping method by combining restriction endonuclease digesting with melting curve analysis, which is a stable, rapid and easy doing SNP genotyping method. The new method analyzed SNP genotyping of two chloroplast SNP which was located in or out of the endonuclease recognition site, the results showed that when attaching a 14 bp GC-clamp (cggcgggagggcgg) to 5' end of the primer and selecting suited endonuclease to digest the amplification products, the melting curve of Lonicera japonica and Atractylodes macrocephala were all of double peaks and the adulterants Shan-yin-hua and A. lancea were of single peaks. The results indicated that the method had good stability and reproducibility for identifying authentic medicines from its adulterants. It is a potential SNP genotyping method and named restriction endonuclease digest - melting curve analysis.

Microbiology 1992;0(01):-
Antimicrobial peptides, a cluster of small peptides secreted by the majority of creatures, have been demonstrated with activity against a wide range of microorganisms including bacteria, protozoa, yeast, fungi, viruses and even tumor cells. These peptides have some features such as broad spectrum , high effi-cacy and stability, little drug resistance. A lack of new antibiotics combined with emerging multi-drug resis-tance issues demands that new antimicrobial strategies be explored for treating these infections. It has been proposed that the antimicrobial peptides might form the foundation for a new class of clinically useful an-timicrobials. We review the advantages of these molecules in construction features and bioactivity, with the focus on the mechanism and clinical applications.

Objective To assess the effects of early correction of anemia with recombinant human erythropoietin (rHuEPO) on the development and progression of left ventricular hypertrophy (LVH) in patients with mild-to-moderate chronic renal insufficiency (CRI) who are not on hemodialysis. Methods A total of 158 patients with serum creatinine from 147μmol/L to 400μmol/L were nrolled in this prospective, multicenter study. Eighty-six patients with hemoglobin (Hb)<110g/L received rHuEPO treatment with a target Hb of ≥110g/L (Group A). Forty patients with comparable Hb concentration (<110g/L) but did not receive rHuEPO (Group B) and 32 patients with Hb≥110g/L and without rHuEPO treatment (Group C) were served as controls. Left ventricular mass index (LVMI) was evaluated by echocardiography at baseline and every 3 months for 2 years. Results There was no difference in age, gender, etiology of renal failure, blood pressure and cardiovascular risk factors among the 3 groups. At baseline, the prevalence of LVH was 72.1% in group A,72.5% in group B and 59.4% in group C. LVMI was inversely correlated with Hb levels (r=0.70, P<0.01). During the 2-year period, the mean LVMI decreased from 142.6±25.7g/m2 to 132.4±18.5 g/m2 in group A, while increased significantly in both group B and group C. The mean Hb concentration increased from 93.8±14.6g/L to 111.2±10.3g/L (P<0.05) in group A, but tended to decrease in group B and group C. There was no significant change of the mean blood pressure, number of anti-hypertensive drugs and serum creatinine concentrations in all 3 groups. However, patients' serum creatinine doubled more often in group B and group C than in group A.Conclusions LVH was common in predialysis CRI patients and was associated with the severity of anemia. Early intervention with rHuEPO may reverse LVH in these patients.

METHODSFrom October 2008 to March 2011, 120 patients with chronic knee synovitis were equally divided into three groups: oral treatment group, external treatment group and combined treatment group. Patients in oral treatment group had a mean age of (56.58 +/- 5.47) years and a course of disease about (7.35 +/- 2.59) months, involving the left knee in 18 cases, the right knee in 17 cases and both knees in 5 cases, who were administered orally with blood-promoting and diuretic Chinese herbal medicines composed of Wu Ling San and Tao Hong Si Wu Decoction. Patients in external treatment group had a mean age of (56.25 +/- 6.35) years and a course of disease about (7.68 +/- 2.76) months, involving the left knee in 16 cases, the right knee in 20 and both knees in 4 cases, who were treated externally with Compound Nanxing Pain Paste. Patients in combined treatment group had a mean age of (55.65 +/- 4.49) years and a course of disease about (7.50 +/- 3.36) months, involving the left knee in 16 cases, the right knee in 18 and both knees in 6 cases, who were given both oral and external treatments. The clinical effects and knee function of the three groups were assessed and compared.
RESULTSFinally, a total of 118 patients were included in the result analysis, including 40 patients in oral treatment group, 39 patients in external treatment group and 39 patients in combined treatment group. In combined treatment group, 27 patients were clinically cured, 9 were improved, and 3 patients were ineffective vs. 15, 16 and 8 patients in external treatment group, and 13, 16 and 11 in oral treatment group. The overall effective rate of combined treatment group was better than that of oral group and external treatment groups (P < 0.05). The function comprehensive score of knee joints in combined treatment group was better than that of oral treatment group, while the function comprehensive score of knee joints in oral treatment group was better than that in external treatment group in terms of demand for assistance, stair-climbing ability, lameness, and swelling pain.
CONCLUSIONCombined use of oral blood-promoting and diuretic Chinese herbal medicines with Nanxing offers a good therapeutic effect on chronic knee synovitis in relieving pain, reducing swelling and improving joint function, while Nanxing could also reduce pain and improve joint function to a certain extent.
