2.Utility of Fluoxetine in Treating Tinnitus Patients

Objective To evaluate t he efficacy of fluoxetine in treating tinnitus patients with depress. Methods A randomized,open-labeled,controlled clinical tri al was conducted.There were 31 patients in control group and 26 p a tents in fluoxetine group.The efficacy was assessed by tinnitus patients with Ra ting Scale and Hamilton Depression Rating Scale (HAMD). Results The total effective rate of fluoxetine group was 71.11%, and there was significant improvement rate in depression after 1 month f luoxetine treatm ent. Conclusion Fluoxetine is an effective drug for tinnitus p atients with depression.
3.Animal models of brain metastasis:preparation methods and research progress

Brain metastasis ( BM) is a common brain tumor in a-dults, originating from extracranial location in the body. There has been a growing interest in producing suitable animal models for studying BM in vivo. Current BM models take peripheral or brain route for tumor cell inoculation, including intra-cardiac or intra-carotid artery injection or orthotopic injection. Direct im-plantation of patient tumor biopsies into rodent brain bears ad-vantages of clinical relevance. This review presents a compre-hensive introduction to key elements for establishing animal mod-els of BM, with highlights on selections of suitable model ani-mals, brain-seeking tumor cell lines, reasonable inoculation routes, as well as succedent phenotyping methods. In the end, perspectives and future directions in this field are discussed.
4.The inconsistent relation of twin discordancy and the umbilical artery blood flow parameter of systolic/dias-tolic ratio

Objective To study the inconsistent relation of twin discordancy and the umbilical artery blood flow parameter of systolic/diastolic ratio.Methods 358 pregnant women were divided into the two groups of develop-mental nonidentity( n=67 ) and developmental identity ( n=291 ) according to the criterion that the differences of twins weight exceeded 25 percent.The umbilical artery blood flow parameter of systolic/diastolic ratio were observed. Meanwhile,the difference of weight and the umbilical artery blood flow parameter of systolic/diastolic ratio in the group of developmental nonidentity were observed.Results A significant difference of the umbilical artery blood flow parameter of systolic/diastolic ratio were found in the two groups of developmental nonidentity and developmental identity(7.81%vs 32.56%,P<0.05).There were 56 cases that twins weight exceeded from 25 to 35 percent,7 cases that twins weight exceeded from 35 to 45 percent and 4 cases that twins weight exceeded 45 percent in the group of developmental nonidentity which the umbilical artery blood flow parameter of systolic/diastolic ratio were (23.66 ± 5.78)%,(27.16 ±7.25)% and (33.16 ±4.23)% correspondingly with significant difference(F=23.21,P<0.05 ) .Conclusion The umbilical artery blood flow parameter of systolic/diastolic ratio has significance in diagnosis of twin discordancy.

6.Biofeedback therapy for myogenic fecal incontinence

Objective To evaluate the effect of biofeedback therapy for the treatment of myogenic fecal incontinence. MethodsA general assessment about anorectal function was made on 17 cases receiving biofeedback therapy including muscle power training,sensory training and coordination training. Results The clinical scores before and after biofeedback therapy were 1.66?0.23,3.80?0.42 respectively,with an effective rate of 82%. The anus maximum contracting pressure elevated,(73?20) mm Hg vs. (123?30) mm Hg; myoelectric amplitude increased,(122?32) ?V vs. (230?41) ?V;Contracting time prolonged,(4.1?2.0) s vs. (9.4?3.0) s; The sensory threshold was lowered,(50?12) ml vs. ( 20? 10) ml;The feel-contract time increased,(3.1?0.4) s vs. (1.2?0.3) s. Positive rectal contraction reflex was seen in 41% patients before therapy compared with 82% after therapy. Conclusions Biofeedback therapy increases contractility of sphincter,decreases threshold of rectal sensory,and is a therapy of choice for myogenic fecal incontinence.
7.Proportional influence of Th1/Th2 lymphocytes after transferring the recombinant of PcDNA3.0/p27~(kip1) eukaryotic expressing plasmid into asthma model of mice

Objective:To investigate the effects of the proportional imbalance of Th1/Th2 in allergic asthma model by transferring the recombinant of PcDNA3.0/p27kip1 eukaryotic expressing plasmid into allergic asthma model.Methods:KM mice were distributed into three groups:healthy control mice(group A);Ovalbumin(OVA)-sensitized/challenged mice(group B) and OVA-sensitized/challenged mice intratracheally administered with PcDNA3.0/p27kip1 plasmid(plasmid-treated mice,group C).KM mice were sensitized and challenged intratracheally with OVA,then transfected with the recombinant PcDNA3.0/p27kip1 eukaryotic expressing plasmid which was constructed by our laboratory crew through vein injection.The p27 protein expression in lungs 36 h after transfection was analyzed by Western blot;Flow cytometry was used to analyze the proportion of CD4 (+)and CD8(+) T lymphocyte subsets in the spleen.The serum levels of IL-4,IL-5,IL-13 and IFN-? were measured by ELISA.Results:Compared to the group A,the expression of p27 protein was significantly reduced,and the proportion of CD4(+)lymphocytes and serum levels of IL-4,IL-5 and IL-13 were significantly higher but CD8(+)lymphocytes and IFN-? was significantly decreased in group B.However,in group C,the expression of p27 protein was significantly increased;Whereas the proportion of CD4 (+) lymphocytes,serum levels of IL-4,IL-5 and IL-13 were significantly decreased.CD8 (+) lymphocytes and IFN-? were significantly increased than those in group B.Conclusion:p27 could be expressed in lungs of plasmid-treated mice and regulate the proportion of Th1/Th2 subset by means of adjusting cell cycles.This method may serve as a potential therapeutic option for the treatment of asthma.

10.Sorting of side population cells from breast cancer MCF-7 cell line and its biological characteristics

Objective To separate the side population cells(SP) from breast cancer MCF-7 cell line,and observe its biological characteristics.Methods Flow cytometry and Hcechst 33342 dye efflux assay were used to isolate SP cells and non-SP cells from the MCF-7 cell line of human breast cancer.Tumorigenicity of the two subpopulations was observed by a soft agar cloning method.Results The results of FACS analysis indicated that (6.5 ± 0.4 ) %of the MCF-7 cells were SP cells;The vitro colony formation rate of SP cells was(38.5 ±9.4)%,and higher than that of non-SP cells ( 8.4 ± 2.6 ) % ( t =5.34,P < 0,05 ).Concluslon The SP cells sorted from MCF-7 cell line enriched tunor stem cells,which exhibited high tumorigenicity.It indicated that SP cells should play a principal role in breast cancer.
