1.Investigation of Drug Manufacturers’Attitude to the National Assessment Programs of Sampling and Testing Policy

OBJECTIVE:To provide reference for improving the national assessment programs of sampling and testing policy. METHODS:Questionnaire was designed to investigate the drug manufacturers’attitude and suggestions to the national assessment programs of sampling and testing policy;SPSS 18.0 software was adopted to analyze the data;a reliability analysis was conducted for the 17 variables in the table;and a principal component analysis was conducted for the topics with strong correlation. RE-SULTS:A total of 340 questionnaires were sent out,314 were effectively received with the effective rate of 92%. the total internal consistency reliability coefficient Alpha was 0.92;the awareness coefficient of enterprise on the evaluation of sampling and testing was 9.78;4.24 enterprises were relatively agree the evaluation of sampling and testing;the attitude coefficient of enterprise on the improvement of evaluation of sampling and testing was 13.39;and the average score of enterprise on the overall satisfaction scoring was 8.73. CONCLUSIONS:The drug enterprises show high overall recognition for the national assessment programs of sampling and testing policy,and willing to cooperate with the development,and hoping the policy can be improved.
2. Research Progress of Anti-allergy Natural Products

Based on overseas and domestic data bank mainly as Scifinder, ACS publication, CNKI, and CQVIP. The relationships between the types of natural products and their anti-allergic activities have been summarized. The advances of anti-allergic natural products over the past ten years have been reviewed. The pathogenesis of allergic diseases and the diversity of anti-allergy components and natural products were summarized, together with their origins and mechanisms of anti-allergy activities. Many natural products show multiple anti-allergy targets and few side effects, while the research is still far from enough. Several suggestions were proposed for further research on anti-allergy natural medicines on the basis of our review.
3.Using digitized 3Dmodel for assessing characteristics of pelvic and abdominal vessel imaging of CTA and MRA

Objective To construct the digitalized 3 dimensional(3D)model by using the CTA and MRA original images and to assess the differences in the pelvic and abdominal vessel display between CTA and MRA.Methods The original data set in 25 healthy youth female cases of CTA and MRA were collected.The datasets,reconstructed the 3D models of arteries and venous on 3D reconstruction software Mimics10.0 was used to construct the 3D model of pevic and abdominal vessels.The reconstruction situation of abdominal aorta,inferior vena cava,common iliac artery and common iliac venous,internal and external iliac artery and iliac venous,uterine artery,obturator artery and obturator venous,hip artery and its branches,internal pudendal artery,middle sacral artery and venous,ovarian artery and venous and sacral venous plexus were observed.Moreover the chi-square test was used to analyze whether the above vessel display rate having differences between the two kinds of model.Results The abdominal aorta,inferior vena cava,common iliac artery and common iliac venous,internal and external iliac artery and iliac venous and uterine artery almost reached 100 % display.The display rates of CTA model for right and left obturator artery and venous were 92 %,80 %,44 % and 32 % respectively,while which of MRA model were 84 %,72 %,36 % and 4% respectively,the difference was not statistically significant(P =0.382,P=0.508,P =0.564,P =0.382).The sacral venous plexus,including S1,2,3,4 transverse vein,the display rates in 25 cases of CTA 3D models were 4%,8%,0%and 4%respective,while which of MRA models were 76%,92%,96%and 44%respectively,the differences between them were statistically significant(P<0.01,P<0.01,P<0.01,P=0.01).Conclusion Based on the observation of 3D models,the displaying rate of grade 4 pelvic and abdominal vessels has no statistical difference between MRA imaging technology and currently widely used CTA technology,moreover the MRA technology is superior to the CTA technology in presacral vessel net display.

5.Colonization characteristics of endophytic bacteria NJ13 in Panax ginseng and its biocontrol efficiency against Alternaria leaf spot of ginseng.

To reveal the colonization characteristics in host of endophytic biocontrol bacteria NJ13 isolated from Panax ginseng, this study obtained the marked strain NJ13-R which was double antibiotic resistant to rifampicin and streptomycin through enhancing the method of inducing antibiotic. The colonization characteristics in ginseng and its biocontrol efficiency against Alternaria spot of ginseng in the field were studied. The results showed that the strain could colonize in root, stem and leaf of ginseng and the colonization amount was positive correlated with inoculation concentration. Meanwhile, the strain could infect and then transfer in different tissues of ginseng The colonization amount of strain in roots and leaves of ginseng increased first and then decreased. However, the tendency of colonization amount of strain in stems was ascend at first and then descend slowly, and was more than that in roots and leaves along with time, which had a preference to specific tissue of its host. In field experiment, the endophytic bacteria NJ13 was proved to be effective in controlling Alternaria leaf spot of ginseng. The biocontrol efficiency of fermentation broth at the concentration of 0.76 x 10(8) cfu x mL(-1) reached 75.62%, which was close to the controlling level (73.06%) of 0.67 mg x L(-1) 50% cyprodinil WG.

6.Surgical management of acute type A aortic dissection associated with pregnancy

Objective Acute type A aortic dissection associated pregnancy severely threatens the lives of both the mother and her ferus.We retrospectively reviewed our clinical experience with this life-threatening condition in six cases.Methods Between January 2007 and February 2012,6 women with acute type A aotic dissection associated pregnancy were treated by our group,with an average of 3 1 years (range 24 -37 weeks)and a mean gestation weeks of 24.5 (range,12 -38 weeks ).The etiology was Marfan syndrome in 4 cases and gestational hypertension in 2.The pathology was the modified Stanford type A3S in I case,A2C in 2 and A3C in 3.- Five patients were treated surgically and 1 medically.Surgical operations were performed under hypothermic cardiopulmonary bypass or deep hypothermic circulatory arrest,including Bentall procedure in 1case,Bentall + Sun's procedure in 2,ascending aortic replacement + Sun's procedure in 2.Results The woman treated medically and her fetus died from aortic rupture 9 days after admission.The cardiopulmonary bypass and cross clamp time and circulatory arrest time averaged 167 rninites(range,75 -210 minites) and 98 minites(range,83 - 145 minites) and 23.5minites(range,19 -27 minutes),respectively.Five patients treaed surgically survived the operation.Three fetuses survived rand two fetuses died.After a mean follow-up of 2.2 years (range,1 - 3.5 years ),5 patients were doing well.CT angiogram detected nonmal aortic and valvular structures,with no signs of distal dilation.Three babies were normal in development and neurocognitive functios.Conclusion Palients with aortic dissection associated with pregnancy should be operated on ugently and medical treatment carries high risks of aortic rupture and maternal and fetal death.Methods of surgical repair,peffusion techniques and delivery should be chosen based on the underlying aortic pathology and gestational age,so as to maximize the safety of the mother and her baby.
7.Prevention of Retinal Vascular Endothelium and Radiation-induced Rat Optic Nerve Damage with Orifices-dredging and Stasis-removing Therapy

Objective To evaluate the preventive effect of orifices-dredging and stasis-removing therapy on retinal vascular endothelium and optic nerve damage induced by radiation in rats. Methods Thirty-six SD rats were randomly divided into 3 groups, namely blank control group, model group and therapy group. Rats in the model group and therapy group were exposed to X-ray radiation once a week for three weeks. Two weeks before the radiation, therapy group was pretreated with intraperitoneal injection of Astragalus injection ( 4.0 g/kg) , Ligustrazine Hydrochloride injection ( 15 mg/kg) and Xingnaojing injection ( 2.5 mL/kg) once a day, blank control group and model group were given intraperitoneal injection of same volume of saline once a day. Before radiation, one day, and 2, 4 and 6 weeks after radiation, we detected the blood concentrations of endothelin-1 ( ET-1) and Von willebrand factor (vWF) in all of the rats by enzyme-linked immunosorbent assay (ELISA) , and analyzed the dynamic changes and intergroup difference. Demyelination of optic nerve was observed under transmission electron microscope, and demyelination percentage was counted 2, 4, and 6 weeks after irradiation. Results Compared with the blank control group, the blood concentrations of ET-1 and vWF in model group were increased after irradiation (P<0.05) , and the peak value appeared on the fourth week after irradiation. Compared with the model group, the blood concentrations of ET-1 and vWF in the therapy group were decreased weeks 2 after irradiation ( P<0.05) . The demyelination percentage of optic nerve was significantly higher in the model group than that in the blank control group (P<0.05) 2, 4, and 6 weeks after irradiation. Compared with the model group, the demyelination percentage of optic nerve in therapy group showed insignificant changes 2 and 4 weeks after irradiation (P>0.05) , but was decreased obviously 6 weeks after irradiation (P<0.05) . Conclusion Pretreatment with orifices-dredging and stasis-removing therapy can decrease the blood concentrations of ET-1 and vWF in the radiation-induced retinal damage rats, and can reduce the demyelination of optic nerve in irradiated rats.
8.Experimental Research of Effect and Mechanism of Liangxue Tongyu Recipe on Acute Cerebral Hemorrhage in Rats

Objective To observe the effect and mechanism of extracts of Liangxue Tongyu Recipe(LTR) on acute cerebral hemorrhage rats. Methods Twelve SD rats were randomly divided into sham operation group, model group, LTR water-extract group and LTR alcohol-extract group. Rats of sham operation group were injected with 2μL of normal saline into the left caudate nucleus, and rats of the other groups were injected with 2μL (0.25 U/ μL) of type Ⅶ collagenase into the left caudate nucleus to induce the model of acute cerebral hemorrhage. And then the rats of LTR groups were given intra-gastric gavage of LTR water-extract or LTR alcohol-extract. At the end of the experiment, we observed the macroscopic features of rat brain, pathological changes in rat brain under microscope after HE staining, awake time, rectal temperature, neurobehavioral scores and serum-specific enolase (NSE) level. Results Rats except for the sham operation group had various degrees of right hemiplegia symptoms, and the symptoms became severest on the third day of experiment. LTR alcohol-extract group and water-extract group had an effect on improving neurobehavioral scores, shortening awake time, and decreasing rectal temperature and serum NSE level of model rats. Conclusion LTR alcohol-extract and water-extract can relieve acute cerebral hemorrhage induced by type Ⅶcollagenase .
10.Efficacy and oncological safety of bone marrow mesenchymal stem cell transplantation for colitis in inflammatory bowel disease models in mice

BACKGROUND:Transfusion of bone marrow mesenchymal stem cells may become a novel and effective biological therapy for inflammatory bowel disease in clinical practice. Nevertheless, the oncological safety of the treatment is worrisome, and is a key to determine whether mesenchymal stem cells can be widely used in treatment of inflammatory bowel disease, and deserves further investigation. OBJECTIVE:To evaluate the therapeutic effect of bone marrow mesenchymal stem celltransfusion against inflammatory bowel disease in mouse models, and to clarify the effects of mesenchymal stem cells on tumorigenesis of inflammatory bowel disease. METHODS:Mouse model of colitis was established using Balb/c (H-2d) mice exposed to dextran sulfate sodium. Syngeneic bone marrow mesenchymal stem cells were transfused into mouse model through caudal vein. The therapeutic effect of mesenchymal stem cells was compared and observed, and pathological remission of colitis was evaluated. Mouse model of colitis-driven colon carcinogenesis was established using Balb/c (H-2d) mice exposed to dextran sulfate sodium and azoxymethane. Tumor formation within the murine colon was compared and observed after transfusion of mesenchymal stem cells. RESULTS AND CONCLUSION:In models of dextran sulfate sodium-induced colitis, weight loss and fecal occult blood were lessened in the bone marrow mesenchymal stem cellgroup compared with the phosphate buffered saline group. Histological damage score of colitis was less in the bone marrow mesenchymal stem cellgroup:mucosal structure of distal colon was almost intact under microscope, and there was smal area of epithelial defects and cryptal defects. Inflammatory cellinfiltration, proliferation of capil ary and smal vessels could be observed in mucosa and submucosa. Homing and colonization of mesenchymal stem cells in submucosa of inflamed colon could also be observed by in vivo tracing. In the dextran sulfate sodium/azoxymethane model of colitis-driven colon carcinogenesis, the number of intestinal tumors and tumor load were obviously less in the bone marrow mesenchymal stem cellgroup than in the control group. Results indicated that transfusion of bone marrow mesenchymal stem cells can apparently improve colitis lesions of mice with inflammatory bowel disease and inhibit carcinogenesis of colitis, which may provide theoretical support for the biological safety of mesenchymal stem cells transplantation for inflammatory bowel disease.
