1.Targeted therapy and its application on gastrointestinal stromal tumor

Targeted therapy has gradually become the focus in the treatment of tumor, with the emerge of more and more targeted drugs which has brought the treatment of tumor into a new era. Imatinib, the tyrosine kinase inhibitor, is one of the important types of targeted therapy, has become the standard of care in adjuvant therapy of gastrointestinal stromal tumor(GIST) with extraordinary effects. However, the questions are still on debate incluing the optimal way to evaluate treatment results, the treatment duration, whether it can be intermitted or not, second line therapy for imatinib-intolerance or -resistance and the combination with surgery.

4.Preparation of nano DBM and its properties

[Objective] To observe the structure characteristics of nano decalcified bone matrix(DBM)produced by MICROS super fine mill,and to study the nano-technologies of DBM and the biocompatibility of nano-DBM as bone graft substitutes.[Methods]Improved Urist method was used to produce DBM.The pieces of DBM was pre-porphyrizated by liquid nitrogen frozen ball mill equipment.The DBM powder were further porphyrizated by MICROS super fine mill to be nano-DBM.The structure of nano-DBM was observed by electron microscope.The experiments on acute toxicity,pyrogen and hemolysis were performed according to theTechnical Evaluation Standards of Biomedical Materials and Medical Instruments promulgated by Chinese Ministry of Health.[Results]The DBM was porphyrizated to the size of 50-200 nm in diameter.No toxicity,pyrogen nor hemolysis of nano-DBM was noted.Nano-DBM exhibited excellent biocompatibility.[Conclusion]Nano-DBM produced under the conditions of low or controlled temperature,is a kind of biomaterial which is avirulent,nonirritant,nonpyrogenic,nonimmunological reaction and reveals good biocompatibility.
5.An observation about using Kasai distillate to moderate the dysporia for patients with gynecological operation

Objective To make sure the exact curative effect of using Kasai distillate to moderate the dysporia for patients with gynecological operation. Methods Divided 152 patients with dysporia into observation group and control group randomly, each group have 76 cases. Using Kasai distillate 80 ml in observation group, 40 ml in control group. 6.Clinical study of combination therapy of tamsulosin and solifenacin for benign prostatic hyperplasia with overactive bladder

ObjectiveTo evaluate the efficacy and safety of combination therapy of tamsulosin and solifenacin for benign prostatic hyperplasia ( BPH ) with overactive bladder ( OAB ).Methods82 patients with OAB and coexisting BPH were randomly divided into tamsulosin group ( n = 38 ) and combination group ( n = 44). The ages were from 50 to 75 y ( averaged, 56.8 ± 8.6). Inclusion criteria: the mean frequency of micturition ≥8 times per day, the frequency of nocturia ≥2 times per day, voiding volume <200 ml each time; for the overactive bladder symptom score ( OA BSS), the 3 rd score > 2, total score > 3. The tamsulosin group accepted tamsulosin 0.2 mg once daily, the combination group accepted tansulouxin 0.2 mg and solifenacin 0.2 mg once daily. Both groups were maintained the corresponding therapy for 12 weeks.The international prostate symptom score (IPSS), Qmax, residual urine volume, OAB score and adverse reactions were recorded.ResultsThe values of IPSS ( 19.5 ±2.2 vs 15.6 ±2.4, P =0.027), the voiding symptom score ( 15.6 ± 2.4 vs 3.4 ± 1. 7, P = 0. 022) and Qmax ( 13.7 ± 3.8 vs 16.6 ± 4.1, P = 0. 034 )improved significantly after treatment in tamsulosin group ( P < 0. 05 ). The values of IPSS ( 19.7 ± 2.3 vs 9.7 ± 3.0, P <0. 001), the storage symptom score (13.8 ± 1.9 vs 5.6 ± 1.6, P <0. 001 ), OABSS (10.3 ±1.8 vs 5.3 ±1.3, P <0.001) and Qmax(14.1 ±4.1 vs 17.2 ±3.5, P=0.027) also improved significantly after treatment in combination group ( P < 0. 05 ). The values of IPSS ( 9.7 ± 3.0 vs 15.6 ±2.4, P < 0.001 ), the storage symptom score (5.6 ± 1.6 vs 12.0 ± 1.6, P < 0.001 ) and OABSS ( 5.3 ±1.3 vs 9.7 ± 2.7, P < 0. 001 ) improved significantly in combination group than those in tamsulosin group ( P <0.001 ). There were no difference between two groups in values of the voiding symptom score, Qmax and residual urine volume ( P > 0.05 ). The incidences of adverse reactions in tamsulosin group and combination group were 7.9% (3/38) and 20.5% (9/44) without significant difference. There was no acute urinary retention in both groups.ConclusionsIt is effective and safe for patients with BPH and OAB to accept combination therapy of tamsulosin and solifenacin.
7.Design, synthesis and cholinesterase inhibitory activity of quinoline-polyamine conjugates.

A series of quinoline-polyamine conjugates (8a-8n) were designed, synthesized and evaluated as inhibitors of cholinesterases (ChEs). Some of these compounds had potent ChEs inhibitory activity with IC50 values at micromolar range. Compound 8n exhibited the strongest inhibition on acetylcholinesterase (AChE) with an IC50 value of 8.78 micromol x L(-1), and compound 8i showed the most potent inhibition on butyrylcholinesterase (BChE) with IC50 value of 1.60 micromol x L(-1) which was slightly better than rivastigmine. The structure-activity relationship revealed that the chain length of polyamine and linker played important roles for inhibitory activity. Molecular modeling studies showed that 8i targeted both the catalytic active site (CAS) and the peripheral anionic site (PAS) of cholinesterases.
8.Influence of psychological nursing intervention for quality of life in patients with ovary cancer

Objective To know the influence of psychological nursing intervention for quality of life in patients with ovary cancer. Methods Divided 300 patients with ovary cancer into the intervention group and the control group randomly, there were 150 cases in the each group. Routine nursing cares was ued in the control group, the psychological nursing intervention was used in the intervention group in addi-tion. Compared the quality of life between the two groups by interviewed questionnair. Results After the nursing intervention, the indexes which can indicated the quality of life in the intervention group were bettez than those of in the control group significantly. Conclusions Psychological nursing intervention can ef-fective promote the quality of life of patients with ovary cancer.
9.Unexplained rhabdomyolysis: clinical analysis of twenty three cases

Objective To evaluate clinical features, therapeutic regimen and prognosis of unexplained rhabdomyolysis. Method Clinical manifestations, therapeutic regimen and prognosis were recorded in 23 patients,who were admitted to The First Affiliated Hospital of Nanjing Medical University 13 to 27 August,2010.The 23 patients were diagnozed as unexplained rhabdomyolysis. Results The patients all presented myalgia of upper body,like neck,waist and back,maybe with asthenia, nausea,dyspnea,abdominal pain, red urine or changed color of urine. Laboratory examination: obviously step-up of creatine kinase [CK: (4655 ± 2556) U/L( normal: 25 ~ 190U/L) AST:(141 ±78) U/L(normal:10~45 U/L),LDH:(348± 127) U/L(normal: 110~ 250 U/L)]and myoglobin[( Mb > 1000 μg/L (normal: 0 ~ 50 μg/L)]. Therapeutic regimen included treatment of the underlying diseases, volume repletion, alkalization and dealing with the complications. No patients developed acute renal failure.All the patients recovered. Conclusions Rhabdomyolysis is a syndrome with different clinical manifestations.However, early diagnosis, proper treatment could prevent serious complications,and prognosis is good.
10.Advances in the research of factor Xa inhibitors

Factor Xa is a trypsin-like serine protease playing a pivotal role in the blood coagulation cascade.Factor Xa and its inhibitors are of great importance in the development of orally active antithrombotic agents and have aroused considerable attention from the pharmaceutical industry sector over the years.In this review,the structural characteristics of the factor Xa binding site are discussed and the X-ray information available together with the published structure-activity relationship data is used to identify the molecular interactions that are most important for tight enzyme-inhibitor binding,which would be useful in the structure-based drug design of novel factor Xa inhibitors.
