In order to investigate whether Laribacter hongkongensis could be detected in stool samples of Rattus norvegicus in the wild,Rattus norvegicus were trapped alive in an urban community of Guangzhou,China over a period of one year from June 2015 to May 2016,and their stool samples were examined for the presence of L.hongkongensis strains.Isolates were identified based on phenotypic characteristics and 16S rRNA sequence analysis, and were examined for their susceptibility to 19 antimicrobial agents.Further typing of the isolates was performed using multi-loci sequence typing (MLST) analysis.A total of 191 R.norvegicus were trapped alive.L.hongkongensis was identified and successfully isolated from two samples, representing a prevalence of 1.05 %.Although the two isolates possessed similar phenotypic characteristics and have no base difference of 16S rRNA gene, they constituted two new distinct sequence types (STs),ST-163 and ST-164.This is the first report that L.hongkongensis can be detected in the intestinal tract of R.norvegicus.Results suggest that R.norvegicus could serve as carriers of L.hongkongensis and therefore could be another potential source of infection.

A simple and sensitive high performance liquid chromatography with fluorescence detection (HPLC–FD) has been developed for simultaneous quantification of doxorubicin (DOX) and its dipeptide conjugate prodrug (PDOX) in mice plasma. The chromatographic separation was carried out on an Amethyst C18–H column with gradient mobile phase of 0.1%formic acid and 0.1%formic acid in acetonitrile at a flow rate of 1.0 mL/min. The excitation and emission wavelengths were set at 490 and 550 nm, respectively. The method was comprehensively validated. The limits of detection were low up to 5.0 ng/mL for DOX and 25.0 ng/mL for PDOX. And the limits of quantification were low up to 12.5 ng/mL for DOX and 50 ng/mL for PDOX, which were lower than those for most of the current methods. The calibration curves showed good linearity (R2 4 0.999) over the concentration ranges. The extraction recoveries ranged from 84.0%to 88.2% for DOX and from 85.4% to 89.2% for PDOX. Satisfactory intra-day and inter-day precisions were achieved with RSDs less than 9.1%. The results show that the developed HPLC–FD method is accurate, reliable and will be helpful for preclinical pharmacokinetic study of DOX and PDOX.

Chinese Journal of Pathophysiology 2000;0(11):-
Nitric oxide (NO) is a multifunctional biomolecule involved in a variety of physiological and pathological processes, including regulation of blood vessel dilatation and function as a neurotransmitter. However, a large amount of NO is toxic to the host and causes several diseases such as cardiovascular system diseases, septic shock, and diabetes mellitus. Endoplasmic reticulum (ER) stress pathway was first identified as a cellular response pathway induced by the accumulation of unfolded proteins in ER to preserve ER functions. Later it was found that ER stress pathway is also activated by various cellular stresses to protect cells, but when stresses are severe, apoptosis is induced to remove damaged cells. It is reported that NO disturbs ER functions, then ER stress-mediated apoptosis pathway is activated. CHOP/GADD153, which belongs to C/EBP transcription factor family, is induced in this process and mediates apoptosis. ER stress pathway induced by NO is involved in the pathogenesis of various diseases.

AIM: To investigate the clinic effect of the femoral neck fracture (FNF) with percutaneous internal fixation of hollow screw Ti-nails. METHODS: A total of 57 patients with closed FNF were recruited from Department of Orthopaedics in the First Mine Hospital of Pingdingshan Coal Group from February 2001 to December 2005. They were treated with 3 hollow screw Ti-nails (produce by Shanghai Qunli Medical Devices Co., Ltd) in 5-mm diameter under C-arm X-ray monitoring after close reduction by hand, punctured with 4-6 bone round nails in 2.5-mm diameter through skin for prefixation. The patients were encouraged to stretch and flex muscles early and perform articular physiological movement. The criteria of outcomes: Excellent: the patients were completely cured in six months after surgery, no pain was found in diseased coxa, the articular movement was normal, and they could walk freely half one year after surgey. Good: The fracture was cured in 9 months lately or no healing, there was a little ache in diseased coxa, the articular movement was limited, and they could walk normally in 10-12 months after surgery and recover slight work. Bad: The fractures were not cured, there were absorption, shifting, and aching in fracture joint early, and they need walk with walking stick. RESULTS: There were 55 cases followed up for 9 months, 51 cases for 18 months, and 57 cases for 42 months.①The curative effect of FNF by internal fixation was excellent in 27 cases, good in 20 cases and bad in 10 cases. ②Host response: 6 patients suffered from fracture shift due to screw-nail displacement, 4 patients got bone un-union, 6 patients experienced femoral head necrosis, and 2 patients got ache at bone fracture and were limited for articulation movement. No infection and rejection happened among 57 patients. CONCLUSION: The treatment of AO cannula Ti-screws internal fixation under closed reduction is effective for FNF, and no abnormal biocompatibility between materials and host appears in the clinical follow-up.

Objective To explore the cardiac differentiation of human embryonic stem cells(ESCs)in coculturing with human fetal cardiomyocytes. Methods Fetal cardiomyocytes and fibroblasts were obtained from the aborted fetus, and human ESCs were from the inner cell mass(ICM)of the surplus embryos for tube babies, with the treated fibroblast as feeder layer.Human ESCs of passage 2 to 5 were cocultured with fetal cardiomyocytes at the ratio of 1∶2,and the XY chromosome types of these two kinds of human cells were different to each other in order to be marked. At the same time, human ESCs without coculture were used as negative control. The cocultured cells were harvested 5 days later and were double-stained with human XY chromosome probe and cardiac specific antibody(desmin or cardiac troponin I,cTnI).Results On coculture day 5,40%-50% human ESCs expressed cardiac specific antigens, and human ESCs without coculture did not express those antigens. Conclusion Human ESCs could be induced into cardiac differentiation by the coculture with fetal cardiomyocytes, and hopefully they will be the candidate for cell transplantation of heart.
