1.Family function and its relationship with clinical prognosis in patients with major depressive disorder

Objective:To explore characteristics of family function in patients with major depressive disorder (MDD),and to evaluate relationships between family function and prognosis of major depressive episode (MDE).Methods:Forty-six patients with MDD were recruited in the outpatient or inpatient departments of Wuhan Mental Health Center from September 1,2014 to August 31,2015.At the baseline,the patients and their co-resident family members were interviewed for psychiatric screening and diagnosis,and the family function of each patient's family was assessed by Family Assessment Device (FAD).After clinic service or hospitalization,the patients were followed up by telephone until they recovered from the MDE (within 12 months since the follow-up) or for 12 months if they had not achieved remission.Forty-two mentally healthy subjects,with no family members diagnosis for psychiatric diseases,and matched with MDD patients for age,sex,number of children,family roleand socioeconomic status,were recruited from a community.The family function of the MDD families and the controls were compared by independent sample-T test,and the relationship between family function and duration of the MDE was analyzed by Pearson's correlation.Results:MDD families exhibited higher FAD scores in 5 dimensions than control families except for affective involvement and behavior control (P<0.01).Patients with relatively good family function showed significantly shorter duration of MDE and higher proportion of remission within 6 months since the follow-up (P<0.01 and P<0.05).All the dimensions of FAD demonstrated significant positive correlation with the duration of MDE except for the behavior control.Conclusion:Families with MDD patients show impairments in multiple dimensions of family function,and the family functions of MDD patients are correlated with the prognosis of MDE.Improvement of family function may contribute to better prognosis of MDD.
2.Effects of NANC neuro-transmitter on detrusor stability of lower urinary tract with obstraction

Objective To investigate the effects of nonadrenergic noncholinergic (NANC) neuro transmitter on the frequency and strength of the automatic contractility of detrusor strip. Methods Cystometry was performed to observe the occurance of detrusor instability (DI) after 6 weeks of Wistar rats LUT. Then the rats were sacrified and their detrusors were made into strips to observe the effect of NANC vasoactive intestinal peptide (VIP) and adenosine triphoate(ATP) on the frequency and the strength of automatic contractility. Results The occurrence rate of DI was 62% after 6 weeks of obstruction of LUT; VIP and ATP could significantly inhibit the automatic contraction ability of detrusor strip, decreasing the frequency and the strength of the automatic contraction of detrusor ( P 0.05). Conclusions VIP and ATP can significantly inhibit the frequency and strength of the automatic contractibility of normal and obstructive detrusor.
3.Study on the GC-MS Fingerprint Spectrum of Volatile Components in Danqi Piantan Capsule

OBJECTIVE:To establish the GC-MS fingerprint spectrum of volatile components in Danqi piantan capsule. METH-ODS:GC-MS was performed. HPLC conditions:The column was BPX 5,temperature of injection was 250 ℃,respectively;carri-er gas was He,flow rate was 1 ml/min,split injection,split ratio was 50:1 and volume injection was 0.02 μl. MS conditions:ion source was EI,bombarding energy was 70 eV,temperature of ion source and transmission line was 200 ℃ and 250 ℃,respective-ly;scan range was 40-500 amu,solvent delay of 2 min. The peak with largest chromatographic peaks areas was reference peak,com-mon peaks of 10 batches of Danqi piantan capsule were analyzed. Similarity evaluation system for chromatographic figerprint of TCM was adapted for the similarity analysis and NIST standard MS database was used to retrieve and identify the common peak. RESULTS:There were totally 13 common peaks in the 10 batches of Danqi piantan capsule with similarity degree>0.92;accord-ing to the verification,the fingerprint spectrum was a dapted for the similarity analysis and reference fingerprint of 10 batches of Danqi piantan capsule showed good consistency. The No.3 peak was Angelica sinensis unique,the No.2,4,5,7,8,12 peak was Acorus tatarinowii unique. CONCLUSIONS:The established method is specific and stable,and can provide basis for the quality as-sessment for Danqi piantan capsule.
4.Application of the Differentiation of Body Constitution in Bronchiectasis

[Objective] To investigate the relationship between the body constitution and the syndromes of the bronchiectasis patients and provide the basis for the clinical treatment.[Methods] Col ected 80 out-patients and in-patients of bronchiectasis in the first affiliated hospital of Zhejiang Chinese Medical University from May 2011 to March 2012. Al patients met with the inclusion criteria, adopted the standardization of physical scale for physical identifica-tion and dialectical type and analyzed the relationship between constitution and syndrome.[Results] ①The gentleness type, qi-deficiency type and yin-defi-ciency type were more popular in the bronchiectasis, fol owed by the phlegm-wet type, wetness-heat type, yang-deficiency type and qi-depression type. The special inherited type and blood-stasis type were very rare. ②The syndrome of phlegm heat obstructing lung was the top one in the Chinese tradi-tional medical syndromes(48.75%).The frequency of liver-fire attacking lung syndrome, hyperactivity of fire due to yin deficiency syndrome and qi-yin de-ficiency syndrome were 21.25%, 18.75% and 11.25% respectively. ③From different body constitution distribution of TCM syndromes in patients with bronchiectasis, it could be found:gentleness type, qi-deficiency type, yang-deficiency type, phlegm-wet type and wetness-heat type were most presented as phlegm-heat stasis lung syndrome. Patients with qi-deficiency type were most presented as qi-yin deficiency syndrome .Patients with Yin deficiency type were always performed as hyperactivity of fire due to yin-deficiency syndrome. Patients with lung qi-stasis type were liver-fire invading lung syn-drome.[Conclusion] Phlegm-heat stasis syndrome was the main syndrome in patients with bronchiectasis. It conformed to the pathological characteristics of deficiency essence with virtual reality. Different body constitutions decided the different traditional Chinese medical syndromes after the disease onset.
5.Changes of reactive astrocytosis after blockage of heat shock protein 70

Objective To study the changes of reactive astrocytosis after heat shock protein 70 (HSPT0) was blocked by anti-HSP70 antibody. Methods We established cell model of scratch inju-ries by in situ culture and prurification of rat astrocytcs. Anti-HSP70 antibody was added into the nutrient medium at once after injury for intervention (intervention group). Then, immunocytochemical staining of glial fibrillary acidic protein (GFAP) was done at different time points in control group and intervention group to observe astrocytosis and morphologic changes, mRNA expression of GFAP was observed by means of reverse transcriptase-polymerase chain reaction (RT-PCR). Results Compared with the con-trol group, average cell area, average dentritic length and number of dentrities of astrocytes were signifi-cantly reduced in the intervention group(P < 0.05 or P < 0.01), with down-regulated mRNA expression of GFAP (P < 0.05). Conclusion HSP70 plays a facilitative role in reactive astrocytosis after injury of astrocytes. Reactive astrocytosis can be controlled to some extent by blocking HSP70 with anti-HSP70 antibody.

7.Experimental study on preventing obesity by compound rhubarb preparation in rats

AIM: To investigate the effect of compound rhubarb preparation (Kintop) in preventing obesity in rats and its probable mechanism involved. METHODS: Twenty-six newborn SD rats were randomly grouped as rhubarb preparation plus high-energy forage group( n= 8), high-energy forage control group( n= 8) and ordinary forage control group( n= 10). The rats in rhubarb preparation plus high-energy forage group and high-energy forage control group were fed with high-energy forage and those in ordinary forage control group were fed with ordinary forage. The rats in rhubarb preparation plus high-energy forage group were administered by compound rhubarb preparation (40 mg?100 g -1 body weight?d -1 ) from 9th to 17th week. The dynamic changes in body weight, celiac fat weight and adipocytes size were measured. Immunohistochemical analysis of leptin in celiac adipocytes (ABC method) and measurement of serum leptin level (RID method) were performed. RESULTS: The body weight and the wet weights of celiac fat were lower, their adipocytes were smaller and immunohistochemical stainings of leptin were weaker in rhubarb preparation plus high-energy forage group than those in high-energy forage control group. 10.Possible mechanism of referred pain caused by chronic prostatitis

Objective To explore the mechanism of the referred pains in pelvic pain due to chronic prostatitis and the relation between pain due to prostatitis and pelvic floor muscle dysfunction. Methods The retrograde fluorescent double labeling and immunohistochemistry were used. A total of 24 Wistar rats were divided two groups, 12 in each group. In the first group, Propidium Iodide(PI) was injected into the prostate, Bisbenzimide(Bb)into the external anal sphincter. In the second group, PI was injected into the prostate, Bb into genitofemoral, iliohypogastric, ilioinguinal, femoral and lateral femoral cutaneous nerves. Results The fluorescent double labeled cells were found in the L 1 L 2 and L 6~S 3 dorsal root ganglia(DRG) and some of them were confirmed to contain calcitonin gene related peptide(CGRP) and P substance (SP) by immunohistochemistry. Conclusion The findings confirm that the peripheral process of DRG cells dichotomizes into prostate, external anal sphincter and somatic parties, and some of these cells contain CGRP and SP, which indicates the referred pains in pelvic floor may be caused by axon reflex in the peripheral process of DRG cells. Meanwhile, neurogenic inflammation might play an important role in this persistent pain status. Pain due to prostatitis is correlated with pelvic floor muscle dysfunction.
