Chinese Journal of Information on Traditional Chinese Medicine 2013;(10):49-50,51
Objective To explore the extraction and assaying of the alkaloids from Bozhou Peony Paeonia lactiflora. Methods Dry root powder of the medical material was extracted by 90%ethanol. The extraction was first acidified by hydrochloric acid after recovering alcohol, and then extracted by dichloromethane. The acid solution was adjusted pH to alkalescence by ammonia water, then extracted by chloroform to get crude alkaloid. Bozhou Peony alkaloids were confirmed by TLC along with different colouration method, and its content was detected by acid-base titration. Results Four alkaloid-representative salmon patches were observed by colored with improving potassium heptaiodobismuthate method, and two prunosus blotches, four dark prunosus spots were delected by concentrated sulfuric acid and formaldehyde-concentrated sulfuric acid method, respectively. However, no chromogenic spots were found by colored with nitric acid as well with ninhydrin. The content of alkaloids in Bozhou Peony Paeonia lactiflora was about 0.079%. Conclusion Bozhou Peony Paeonia lactiflora contained alkaloid, and may including aconitum alkaloids.

OBJECTIVE:
The purpose of this study is to assess the effects of aging on air-conducted sound elicited cervical vestibular evoked myogenic potential (cVEMP) and ocular vestibular evoked myogenic potential(oVEMP) in normal people.
METHOD:
Sixty normal subjects were recruited for the vestibular evoked myogenic potential examination. Among them, 20-40 year-old group was setted as the first group, 41-60 year-old group was setted as the second group, > 60-year-old group was setted as the third group. Each group included 20 normal subjects. SPSS 16.0 software was used to analyze the data.
RESULT:
As the age growing, P1 latency prolonged, N1-P1 amplitude decreased, and amplitude asymmetric ratio increased in cVEMP; N1 latency prolonged, P1-N1 amplitude decreased, and amplitude asymmetric ratio increased in oVEMP.
CONCLUSION
With the age growing, the time of otolithic organ input pathways prolonged, the function of otolithic organ decreased and the decrease level may be asymeetrical on both sides.

Objective To investigate the effect of DHA on proliferation and killing activity of γδT cells against SW1990 cells in vitro. Methods γδT cells were generated in vitro by stimulating peripheral blood mononuclear cells of healthy donors in RPMI 1640 completed medium containing IPP and IL-2 for 8 d,and then co-cultured with different concentrations of DHA for 48 h.Proliferation rates of γδT cells for each group were detected by MTT method. The perforin, granzyme B and CD107a expression in γδT cells were verified by flow cytometer. The cytotoxic activity of γδT cells against SW1990 cells were analyzed by CCK-8 kit. Results The purity of γδT cells in each group reached 75.46% ±5.32% after 8 d of culture.Compared with the control group, the proliferative capability of γδT cells were enhanced significantly after treated with 50-100 μmol/ml DHA for 48 h,moreover, cytotoxicity against SW1990 cells and perforin, granzyme B and CD107a expression of the γδT cells treated with DHA were higher than the control group. Conclusion DHA could enhance the antitumor activity of γδT cells, which may be associated with the upregulation of perforin and granzyme B expression in γδT cells.

Hypoxia is a general characteristic of most solid malignancies and intimately related to cancer progression. Homeostatic response to hypoxia is primarily mediated by hypoxia inducible factor-1alpha (HIF-1alpha) that elicits transcriptional activity through recruitment P300 coactivator. Targeting the interaction of HIF- alpha and P300 would thus constitute a novel approach for cancer treatment by suppressing tumor angiogenesis and metastasis. Here, a screening assay was developed for inhibitors targeting the interaction between HIF-1alpha and P300. The nucleotide sequence of human HIF-1alpha and P300 were cloned into pBIND and pACT vectors, named pBIND-HIF1alpha and pACT-P300. The interaction of HIF-1alpha and P300 was identified in HEK293 cell using mammalian two-hybrid system. And compound chetomin decreased their interaction in this mammalian two-hybrid system. We further verified HIF-1 inhibition effect of chetomin in U251-HRE cells. Therefore, we established a screening assay combined HIF-1alpha and P300 mammalian two-hybrid system and U251-HRE reporter assay for HIF-1 selective inhibitors.

Objective To explore the efficacy and safety of conversion from calcineurin inhibitor (CNI) to sirolimus (SRL) as major immunosuppressive therapy in lung transplant recipients.Method Retrospective analyses were conducted for the clinical data of all the patients undergoing lung transplantation in Wuxi People's Hospital between January 2011 and December 2014.Sixteen were given conversion treatment with Sirolimus in the postoperative irnmunosuppressive therapy. We analyzed the opportunity and reasons in the conversion treatment, and the safety, effectiveness and complications of the conversion treatment.Result The follow-up period was 8 to 25 months, and the median time of conversion was 6 months after operation (2-18 months). The indications of conversion concluded:malignant tumor (n =8),renal dysfunction (n =5),lymphangioleiomyomatosis (n =1) and intractable diarrhea caused by CNIs (n =2).Four cases suffered from interstitial pneumonitis associated with Sirolimus and one case suffered from spontaneous pneumothorax, and they all conversed back to CNIs.In those patients, cancer recurrence occurred in 4 cases (of them, there were 3 deaths),and 3 patients developed chronic rejection. Those recipients receiving the conversion treatment due to renal dysfunction showed recovery of renal function to some extent. Conclusion It's effective and safe to converse the immunosuppressive therapy based on Sirolimus.Sirolimus should be reduced or withdrawn when interstitial pneumonitis associated with Sirolimus occurred.
