1.The diagnostic value of B-ultrasonography on congenital ureterostenosis

Objective To study clinical value of ultrasoni c diagnosis in the congenital ureterostenosis. Methods 33 cases of ureterostenosis were measured with B-ultrasonographic examination, the resu lt of B-ultrasonographic examination was compared with the diagnosis of X-ray and surgery. Results The localization rate and accord rate of B -ultrasonographic examination in diagnosis of ureterostenosis(obstruction) were 93.3% and 81.8% respectively. Conclusion B-ultrasonographic examination in diagnosis of congenital ureterostenosis possesses greater advanta ge and better clinical value.
2.Research meteorological environmental factors in children's allergic rhinitis.

OBJECTIVE:
To investigate the possible effects of meteorological and environmental factors on allergic rhinitis of children.
METHOD:
Daily numbers of outpatients consulting general practitioners for AR between Jan 1 2007 and Dec 31 2011 were obtained from the Department of Otolaryngology-Head and Neck Surgery, Shanghai Children Medical Center. Daily maximum, average and minimum temperatures and humidity and wind power for Shanghai of the same 5 years were obtained from the Shanghai Meteorological Office. Air Quality data was provided by the Shanghai Municipal Environmental Protection Monitoring Center on daily SO2, NO2 and PM10, the average of O3 per 8 hours. The data was available as average values derived from the data of 6 state comtrolled monitoring stations distributed across Shanghai. Non-parametric generalized additive model (GAM) was used to analyze the highly non-linear or non-monotonic exposure-response relationship between meteorological and environmental factors and daily outpatient visiting for AR of children.
RESULT:
The daily numbers of AR children in outpatients department in this study were highest in the average daily temperature 11°C and 21°C. It showed two peak. But as the humidity increased, daily numbers of outpatients of AR children went down, indicating that the higy humidity has protective effect. Every 10 microg/m3 increase of environmental pollutants O3, SO2 and PM10 was linked to 1.95%, 1.19% and 0.33% increase in the number of visitors, respectively, suggesting the air pollution may increase the risk of onset of AR in children.
CONCLUSION
Meteorological and environmental factors have important effects on AR in children.

3.Formation and characteristics of crystals on the surface of mineral trioxide aggregate in vitro

Objective:To examine the surface of mineral trioxide aggr eg ate (MTA) when treated in different conditions in vitro. Method: Root blocks were prepared. MTA and other three dental materials (Dycal, Dyr act and amalgam) were respectively filled into the root canals of prepared root blocks. The roots with the materials were maintained in distilled water or simul ated body fluid (SBF) at 37 ℃ for 5 days. Scanning electron microscopy(SEM) and X-ray energy dispersive spectroscopy(EDS) were employed to investigate the mor phological changes and chemical components. Results:There was pr ecipitate of simple crystal units or crystalline structure in the MTA specimens. EDS showed one or two peaks corresponding to calcium or calcium and phosphorus. Conclusions:Apatite may produce and grew by MTA on its surface.
4.Video-assisted subcutaneous mastectomy and immediate reconstruction

Objective To explore the feasibility and cosmetic results of video-assisted subcutaneous mastectomy and immediate reconstruction. Methods Two patients who suffered from breast diseases and need to excise the breasts received a video-assisted total subcutaneous mastectomy and immediate reconstruction in our center. Results Both patients felt satisfied on the cosmetic result. According to the operative experience of these two patents combined with review, the key point of the operation was summarized as follows. The incisions on the anterior axillary line were small and concealing. External retraction method was used to establish the working space. Video-assisted operation was combined with euthyphoria. Blunt dissection was combined with electrical surgery unit and ultrasound knife to isolate the breast skin flap. After breast glandectomy was completed, silica gel prosthesis was put into the interspace behind ectopectoralis. Pressure dressing for 3 weeks was done to prevent the prosthesis upward displacement. Conclusion The video-assisted subcutaneous mastectomy and immediate reconstruction is a safe and feasible operation method with the better cosmetic result.
5.Expression of p73 gene in human non-small cell lung cancer i n vitro

Objective To study the expression of p73 in human non-small cell lung cancer (NSCLC) and the relationship between p73 expression and clinico-pathological parameters. Methods Expression of p7 3 gene was detected by RT-PCR in 32 human NSCLC tissues, tissues adjacent to ca ncer and non-cancer lung tissues. Results p73 gene expression up-regulated substantially and detected in 87.5%(28/32) of human NSCLC tissues while expressed at low level in tissues adjacent to cancer and non-cancer lung tissues. Conclusion Marked up-regulation of p73 gene expres sion is found in human NSCLC.
6.Study on the resistance-related genes of Shigella sonnei with decreased susceptibility to fluoroquinolones

Objective To investigate the resistance-related genes of Shigella sonnei with decreased susceptibility to fluoroquinolones.MethodsA total of 131 strains of Shigella sonnei were analyzed for their antimicrobial susceptibility.Mutations within the quinolone resistance determining regions (QRDR) of gyrA and parC were detected by polymerase chain reaction (PCR) and PCR products were then sequenced. Meanwhile, the plasmid-mediated quinolone resistance (PMQR) genes,qnr and aac(6')-Ib-cr were screened by PCR.ResultsResistance rates of 131 Shigella sonnei isolates to nalidixic acid,tetracycline,ampicillin and trimethoprim-sulfamethoxazole were 100.0%,93.9%,93.2% and 92.8%,respectively.All strains were susceptible to norfloxacin,ciprofloxacin,levofloxacin,while 94% nalidixic acid-resistant Shigella sonnei strains showed reduced susceptibilities to fluoroquinolones.All of nalidixic acid-resistant Shigella sonnei strains presented a single mutation at codon 83 (Ser→Leu) of gyrA genes,but no mutations were detected in parC gene.And PMQR genes qnr and aac (6’)-Ib-cr were not detected.Conclusions The nalidixic acid-resistant Shigella sonnei strains with reduced susceptibility to fluoroquinolones are common in the clinical practice,which may mainly due to a single mutation at codon 83 (Ser→Leu) of gyrA genes.
7.The expression of PPARγmRNA and protein in rats with acute necrotizing pancreatitis

Objective To investigate the changes of PPARr mRNA and protein expressions in rats with ANP. Methods 36 rats were randomly divided into sham group and ANP group. The rats were sacrificed 3 h, 6 h,12 h after ANP induction, the levels of serum amylase were measured, the pancreatic pathological changes were determined and the expressions of pancreatic PPARr mRNA and protein was examined by RT- PCR and immunohistochemistry. Results In ANP group, the level of serum amylase at 6h was (7170.83± 1635.59) U/L, the scores of pathological changes were 6.67±1.03 and 13.00±2.36, which were much higher than those of sham group (P<0.01) ; the PPARr mRNA expression was 0.18±0.05, and there were no obvious differences compared with that of sham group (0.22±0.03 ) ; PPARr protein expression was 4.17 ±0.98, which was significantly higher than that of sham group (1.83±0.71, P<0.05). Conclusions Inflammatory injury resulted in increased deactivation of pancreatic acinar PPARr, meanwhile PPARr gene expression was inhibited by feedback.
8.Content Determination of the Total Naphthoquinone in Oil/Water Emulsion of Shikonin by Ultraviolet Spectrophotometry

OBJECTIVE:To establish a method for content determination of the total naphthoquinone in oil/water emul?sion of shikonin by ultraviolet spectrophotometry.METHODS:The content of the total naphthoquinone was counted in L-shikonin,methanol was used as solvent and the wavelength was set at516nm.RESULTS:The concentration of L-shikonin had good linear correlation with absorbance in the range of8.32~41.60?g/ml(r=1.0000,n=5),the average recovery was98.6%(RSD=0.53%).CONCLUSIONS:The present method is simple,precise and replicable resulting in high recovery and accuracy,it can be used to determine the content of total naphthoquinone in oil/water emulsion of shikonin.

10.A comparative study of four methods for establishing animal models of human breast cancer bone metastasis

Objective To investigate four methods for establishing animal models of human breast cancer bone metastasis. Methods Thirty-two female nude mice aged 4-6 weeks were divided randomly into four groups (n=8 in each group). 5×105 MDA-MB-231 cells were injected into the body via the left second mammary fat pads (group A), the tail veins (group B), the left heart ventricles (group C) and the left tibia marrow cavities (group D), respectively. Tumor formations in situ were recorded in group A. Deaths after the injection were recorded. The surviving nude mice 49 days after the injection were subjected to pathological examination to determine bone metastasis. Results The rate of tumor formation in situ of group A was87.5 %(7/8). One mouse in group C died after the injection of MDA-MB-231 cells. The bone metastasis rate in groups A, B, C and D was zero (0/8), 12.5 % (1/8), 71.4 % (5/7) and 100 % (8/8), respectively. There was statistically significant difference in the bone metastasis rate between group A and group C, group A and group D, group B and group C; and group B and group D. Conclusion Injections of tumor cells via the breast fat pads and tail veins were not suitablemethods to establish animal models of human breast cancer bone metastasis. The bone metastasis model could be established efficiently by injecting tumor cells into the left heart ventricles or the bone marrow cavity of nude mice.
