1.Establishment of a sandwich ELISA method for measuring soluble IL-2 receptor

Two kinds of mouse monoclonal antibodies which recognize distinct epitopes of the IL-2 receptor (a chain) were used to establish a sandwich ELISA for measuring soluble IL-2 receptors. The results indicated that this method is sensitive and specific for detecting soluble IL-2 receptors of cell culture supernatant and serum from both healthy donor and patients with leukemia. Therefore this sandwich ELISA method is useful for basic and clinic immunology research.
2.Studies on the requirements for growth of Epstein-Barr Virus thransformed human lymphoblastoid B cells at low cell density

We obtained human lymphoblastoid B cells secreting human antibodies IgM and IgG to HFRS virus by Epstein-Barr virus(EBV)transfomation of PBMC.Conditioned media from the supernatant of a human fibroblast cell CRL1506 culture and irradiated CRL1506 cells as feeder cells could markedly promote the growth and antibody produotion of the human B lymphoblastoid cells at low cell density.It was shown that conditioned media contained a high level of interleukin 6(IL-6) .These results would be very useful for human monoclonal antibody technique including improving human B lymphoblastoid cell growth at low cell density and enhancing cloning efficiency andantibody production.

To study the changes in vestibular function in Parkinson's disease patients, and its significance, eletronystagmography (ENG) was used to examine 54 patients with Parkinson's disease, and also 20 hea thy elderly subjects. The results of vestibular function tests including sensory organization test, nystagmus test, cold caloric test and positional test of these two groups were recorded for statisticed analysis. It was found that the rate of vestibular dysfunction in Parkinson's disease patients were 81 4%, and that of the controls 30%. There was significant difference between the two groups ( P

Objective:To discuss the effect of bone marrow on chemotaxis of aggregation of macrophages in venous thrombosis.Methods:SD adult rats were divided randomly into normal group,sham operation group,thrombus group and rhG-CSF group.Peripheral blood was harvested at 30min before operation and on the 7th and 14th day after operation.Level of MCP-3 in peripheral blood was detected with ELISA.Inferior vena cava specimens of thromus group and rhG-CSF group were collected at postoperative 7th and 14th day.With the application of RT-PCR,expression of MCP-3mRNA in thrombus was detected.Results:The level of MCP-3 in peripheral blood of rhG-CSF group was significantly higher than that before operation and also higher than the levels of MCP-3 in peripheral blood of sham operation group and thrombus group on the 14th day after operation,(P

Most diseases are complex genetic traits caused by multiple genetic and environmental components. It has been proposed that common genetic variations, mainly single nucleotide polymorphisms (SNPs), influence the susceptibility to complex diseases. We have conducted an extensive review on the characters of SNPs, the related website information, and the genotyping methods of SNPs such as direct sequencing, SnaPshot, Taqman, real time quantitative (kinetic) PCR with allele specific amplification, denaturing high performance liquid chromatography, and OLA/PCR.The strategies for studying the relation between SNPs and complex diseases susceptible genes were also reviewed.
7.Study on quality standard for Jianweishu Granule

Objective: To establish the assay methods for paeoniflorin and baicalin in Jianweishu Granules (Radix Scutellariae, Radix Paeoniae Alba, Radix Astragali seu Hedysari, Radix Notoginseng, etc.). Methods: Radix Astragali, Radix Notoginseng; Radix Paeoniae Alba and Herba Dendrobii in Jianweishu Granule were identified by TLC. The paeoniflorin and baicalin in Jianweishu Granule were determined by HPLC on shim pack CLC ODS C 18 Column (6.0mm?150mm,10?m); using methanol water phosphoric acid (47∶53∶0.2) and methanol water (30∶70) as mobile phases, respectively; the flow rate were at 1.0 mL?min -1 ; the detection wavelength were 280nm and 230nm, respectively; the column temperature were at room temperature and 40?C respectively. Results: The average recoveries are 98.94% and 98.67%. RSD are 2.31% ( n =5) and 1.36% ( n =5). Conclusion: The methods are simple, sensitive, accurate and reproducible.

The Wnt/β-catenin signal transduction pathway is essential for regulating cell proliferation, movement, differentiation and embryo development. Researchers have found that Wnt pathway is an important regulation factor in the early stimulation of the cardiogenesis and contacts closely the several key events during cardiac morphogenesis. The article reviews the recent studies about the relation between cardiac development and Wnt pathway in early embryo.
9.Clinical effect of continuous intravenous infusion of sodium nitroprusside combined with dopamine in the treatment of the elderly patients with refractory heart failure

Objective To investigate the clinical effect and safety of continuous intravenous pumping of sodium nitroprusside and dopamine on treating elderly patients with refractory heart failure. Methods Two hundred and seventy elderly patients with refractory heart failure were selected as our subjects and they were randomly divided into sodium nitroprusside group,dopamine group and the combined group( 90 cases for each group). All the patients were given the corresponding symptomatic treatment,including oxygen,diuretics, angiotensin converting enzyme inhibitors,digitalis,aldosterone receptor antagonist,etc. Patients in sodium nitroprusside group,dopamine group were treated sodium nitroprusside or dopamine respectively,and patients in combined group were given both of sodium nitroprusside and dopamine by continuous intravenous pumping. Serum brain natriuretic peptide( BNP ),left ventricular ejection fraction( LVEF ),LV end systolic volume index(LVESVI),left ventricular end diastolic volume index(LVEDVI),left ventricular diastolic early, late filling velocity ratio( E/A ),mean arterial pressure and heart rate( HR ) level were measured or recorded. Results The levels of BNP,LVEF,LVESV,LVEDV,E/A,mean arterial pressure and the level of HR among three group before treatment were no significant differences( F =4. 000,1. 434,0. 503,3. 622,3. 589, 4. 135,2. 752;P﹥0. 05). After treatment,the levels of BNP,LVEF,LVESV,LVEDV and HR in combined group were(201. 6 ±66. 8)pg/L,(45 ± 4)%,(29. 8 ± 4. 8)ml/m2,(45. 1 ± 7. 3)ml/m2,(73. 2 ± 11. 2)times/min,in sodium nitroprusside group were( 298. 6 ± 75. 3 ) pg/L,( 37 ± 4 )%,( 35. 5 ± 4. 6 ) ml/m2 ,( 51. 8 ±7.5)ml/m2,(92.9 ±11.5)times/min,and in dopamine group were(293.7 ±78.2)pg/L,(37 ±3)%, (35. 6 ± 5. 8)ml/m2,(52. 1 ± 7. 3)ml/m2,(93. 6 ± 12. 3)times/min respectively,and the differences were significant(F=41. 683,129. 400,43. 600,28. 800,328. 600;P﹤0. 05). The total effective rate in combination group was 97. 8%( 88/90 ),significantly higher than that of sodium nitroprusside group( 86. 7%,8/90 ) or dopamine group( 88. 9%,80/90 ),and the differences were statistically significant(χ2 =7. 683,P ﹤0. 05). There were 3 cases(3. 3%)with adverse reaction in combination group after treatment,including 1 cases of headache and 2 cases of nausea and vomiting. All above symptoms disappeared after stopping drugs. There was 1 cases(1. 1%)with adverse reaction in sodium nitroprusside group,1 cases(1. 1%)in dopamine group,and there was no significant differences seen among three groups(χ2 =1. 630,P﹥0. 05). Conclusion Continuous intravenous infusion of sodium nitroprusside combined with dopamine on cardiac function in elderly patients with refractory heart failure can improve clinical effect and enhance the medication security.
10.Clinical and genetic analysis in two families with huntington disease

Objective To investigate the relation between CAG expansion length and clinical symptoms in hunting-ton′s disease ( HD) ,and to evaluate the value of CAG age product ( CAP score) in the clinical practice. Methods Two Han Chinese HD family members’ clinical data, including clinical symptoms and signs, the Cambridge cog-nitive examination-Chinese version ( CAMCOG-C) and Hamilton depression rating scale were collected, and these two families pedigree trees were drawn. Volunteers peripheral venous blood was extracted to pursue IT15 genetic test. CAP scores among members whose CAG repeat number was more than 35 were calculated. Results The pedi-gree trees indicated that HD was autosomal dominant inheritance. Among 6 participants who had pursued genetic test,1 was normal and 5 were carring expanded allele; according to fomal clinical diagnosis criteria,2 participants were in manifest period and 3 were in pre-manifest period. Individuals in manifest period had clinical triad,and in-divudals in premanifest period,though hadn’ t been clinically diagnoseded, were characterized by subtle motor,cog-nitive,behavioural or personality changes. Conclusion HD prior to clinical diagnosis may have mild clinical symptoms;the more CAG repeats indicate earlier onset, the heavier the symptoms. CAP score as a parameter, combined with the CAG repeat number and the age, is of great significance for predicting the onset age, clinical tri-al subjects grouping, and give a better understanding of the natural history of HD.
