1.In vitro antifungal activity of closantel against Candida albicans

Objective To determine the in vitro antifungal effects of closantel against Candida albicans.Methods A microdilution method was used to determine the minimum inhibitory concentration (MIC)of fluconazole alone and in combination with closantel against Candida albicans standard strain CAF-2.Ten strains of Candida albicans were cultured in RPMI-1640 liquid culture containing 10% calf serum with or without the presence of closantel at 16 mg/L,followed by the observation of hypha formation.Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) were performed to observe the ultrastructure of Candida albicans CAF-2 strain after exposure to closantel at 16 mg/L for 24 hours.Results Closantel could inhibit the growth of Candida albicans,and enhance the antifungal effect of fluconazole against Candida albicans in vitro.The percentage of Candida albicans forming hypha was 91.2% ± 3.9% in untreated Candida albicans,significantly higher than that in closantel-treated Candida albicans (29.8% ± 5.1%,t =30.24,P < 0.05).As TEM showed,closantel-treated Candida albicans gave a round,oval or pleomorphic appearance,with an irregular budding from the cell surface.Further more,the electron dense layer in the outer layer of cell wall was absent or unevenly distributed,the transparent layer was irregularly thickened,some cell membrane was locally disrupted or collapsed,and intracellular vacuoles increased after closantel treatment.Scanning microscopy revealed a rough surface,sparse and irregular budding of Candida albicans after treatment with closantel.Conclusion Closantel exhibits a promising anti-Candida albicans property in vitro.
2.Therapeutic Observation of Lao Shi Jiu plus Chinese Medication for Gastritis Due to Deficient Cold in Spleen-stomach

Objective To observe the clinical efficacy of Lao Shi Jiu (moxibustion at a group of ten acupoints) plus Wu Zhu Xiang Sha Liu Jun decoction in treating chronic gastritis due to deficient cold in spleen-stomach. Method Ninety patients with chronic gastritis due to deficient cold in spleen-stomach were randomized into a moxibustion-medication group and a Western medication group by using the random number table, 45 cases each. The moxibustion-medication group was intervened by Lao Shi Jiu plus Wu Zhu Xiang Sha Liu Jun decoction, while the Western medication group was prescribed with conventional medications. The clinical efficacy and changes in symptoms were observed after the intervention. Result The total effective rates were respectively 88.9% and 75.6% in the moxibustion-medication group and Western medication group, and the rate in the moxibustion-medication group was significantly higher than that in the Western medication group (P<0.05); the gastroscopy-based total effective rates were respectively 91.1% and 77.8% in the moxibustion-medication group and Western medication group, indicating that the moxibustion-medication group was superior to the Western medication group (P<0.05); the improvement of stomachache in the moxibustion-medication group was more significant than that in the Western medication group (P<0.05). Conclusion The moxibustion-medication group is superior to the Western medication group in comparing the clinical efficacy and improvement of symptoms in treating chronic gastritis due to deficient cold in spleen-stomach.

Objective: To follow up patients received orthognathic surgery Method:108 patients were assessed with SCL-90 five times during the study, that was the time before the operation, after the operation, 3 month, 6 month and 12 months later Result: After operation, three subscores of SCL-90 increased significantly, they were anxiety, depression and hostility From the third month after the operation, the score of SCL-90 began to decrease Both results of the 6th month and 12 th month were significantly lower than that before the operation Patients felt more confident and happy at the end of one year Conclusion:There is a process of psychological adaptation to orthognathic surgery, which lasts for half a year
4.Effects of salvia miltiorrhizae on L-Ca current in ventricular myocyte of guinea pig during normoxia, acute hypoxia and reoxygenation

AIM: To observe the changes of Ca-L current (ICa-L) and to investigate the mechanism of salvia miltiorrhizae (SM) for eliminating Ca~2+ overloaded in cells during acute hypoxia/reoxygenation. METHODS: The whole cell patch clamp technique was applied to study the changes of ICa-L. Different concentrations (32, 320, ~3 200 mg/L) of SM were added to the ventricular myocytes isolated from guinea pigs by enzyme digestion. RESULTS: SM (32, 320, ~3 200 mg/L) decreased the amplitude of ICa-L in a concentration-dependent manner regardless of these cells were under normoxia, hypoxia or reoxygenation. Furthermore, SM at low concentration (32 mg/L) was more effective to hypoxia or reoxygenation-treated cells than that to the cells under normoxia condition. CONCLUSION: These results indicate that SM effectively decreases the abnormal raised amplitude of ICa-L in ventricular myocytes under hypoxia or reoxygenation conditions, preventing Ca~2+ overloaded in the cells. [
6.Determination of Volatile Halohydrocarbons in Drinking Water by Headspace Gas Chromatography

Objective To establish a method to determine 7 kinds of volatile halohydrocarbons in drinking water with headspace gas chromatography. Methods The water samples were heated by water bath,the water head space gas was injected into gas chromatography,separated by DB-624 capillary column and then detected by ECD detector. Results Under the optimal experimental conditions,the methods had a wide linear range,the lowest determination limits was 0.001-0.05 ?g/L,the relative standard deviation was 3.6%-8.9%,the rate of recovery was 84%-104%,the linear range of determination was 0.01-50 ?g/L. Conclusion This method is sensitive,simple,accuracy and is appllicable for determination of 7 kinds of volatile halohydrocarbons in drinking water.

8.Comparative Study on the Process for Extration of Curcumin from Curcuma Longa L. with Different Methods

Objective To compare the five different methods of extration process of Curcuma Long. Methods With the content of curcumin and yield of extract as evaluation index, the best extraction process of Curcuma long L. was optimized. Results The content of curcumin extracted in 80 ℃ ethanol immersion was higher, that was the optimum condition. Conclusion The extractive content of curcumin is higher by this process. It is easy to operate and proved to be stable and feasible.
9.Surgical treatment of postinfarction ventricular septal defect

Objective To review our experience of surgical treatment for postinfarction ventricular septal defect. Methods From 1990 to 2004, 22 patients with postinfarction ventricular septal defect underwent surgical treatment. There were 15 males and 7 females with a mean age of (62.2?7.3) years (from 52 years to 72 years),19 had anterior ventricular septal defect and 3 had posterior VSD, and there were 18 cases with left ventricular aneurysm.10 patients also had introaortic balloon pumping in the early experience. In all the patients cardiac function were in NYHA class III~IV. Results There were 8 operative deaths (35%). There were 7 operative deaths before 1999 and 1 case after 1999. The other patients are clinically asymptomatic after surgery, and the cardiac function is in NYHA class I~II. Conclusion Postinfarction ventricular septal defect is an infrequent but serious complication of myocardial infarction. Surgery is the best way to treat this condition. A coronary angiography should be performed in all patients who can be stabilized hemodynamically and myocardial revascularization should be performed in patients with significant coronary artery stenosis.
10.Early warning value of positive rectal swab culture for bloodstream infection of carbapenem-resistant Klebsiella pneumoniae in liver transplant recipients

Objective:To explore the early warning value of carbapenem-resistant Klebsiella pneumoniae (CRKP) positivity in liver transplantation recipients with rectal swabs, examine the risk factors of CRKP bloodstream infection and provide the relevant treatments.Methods:From June 2018 to December 2019 in Organ Transplantation Research Institute Affiliated Tongji Hospital Tongji Medical College Huazhong University of Science & Technology, 148 cases of liver transplantation recipients with positive CRKP rectal swabbing were recruited. Clinical data were retrospectively analyzed. And the risk factors of CRKP bloodstream infections were examined for intervention and non-intervention groups to observe the effect of interventions of CRKP bloodstream infections.Results:Among them, 23 cases (15.5%) were positive for CRKP and 5 cases (21.7%) were infected with CRKP bloodstream. Rectal swab culture was negative in 125 cases and no bloodstream infection occurred. Long-term use of broad-spectrum antimicrobial agents, severe basic diseases (severe hepatitis), postoperative delayed graft liver function recovery, acute renal failure requiring renal replacement therapy (RRT) and postoperative anti-thymocyte globulin (ATG) induction were risk factors. In intervention group, there were 2 cases (11.1%) of 18 patients with positive CRKP in rectal swab culture in late stage. Among 5 CRKP-positive recipients without intervention, 3 cases (60%) developed later CRKP bloodstream infection. The incidence of bloodstream infection was significantly lower in intervention group than that in non-intervention group ( P<0.05). Conclusions:Rectal swab culture for liver transplantation recipients provides early warning for CRKP bloodstream infection. Interventions for CRKP positive high-risk recipients with rectal swab culture may reduce the occurrence of CRKP bloodstream infection and lower the risk of CRKP bloodstream infection in liver transplantation recipients.
