2.The establishment and identification of human umbilical artery endothelial cells-smooth muscle cells co-culture model
Fudan University Journal of Medical Sciences 2009;36(6):768-775
Objective To establish a human umbilical artery EC-SMC co-culture model, and mimic the morphological and functional characteristics of human arterial wall, for further reseach of the pathological mechanism and therapy of atherosclerosis and imflammatory damage. Methods We secceeded in the primary culture of human umbilical artery endothelial cells (HUAEC) and human umbilical artery smooth muscle cells (HUASMC) by collagenase perfusion digestion and tissue planting, respectively. HUASMCs were incubated in a medium with ascorbic acid at the concentration greater than 50 μg/mL to produce collagen, which was considered as the extracellular matrix for ECs. Then HUAECs were seeded directly upon HUASMCs in a saturate density for sufficient direct physical interaction between ECs and SMCs. The morphological characteristic of EC-SMC co-culture was identified by immunofluorescence staining, and the function of EC-SMC co-culture was identified by Dil-Ac-LDL uptake test. Results The morphological identification showed that the entire surface of HUASMCs was covered by a confluent monolayer confluent monolayer, which indicated that the model had simulated the morphological characteristic of human arterial wall. The results of Dil-Ac-LDL uptake test showed that there was a fluorescent signal in HUAECs. Compared with EC monoculture, the Dil-Ac-LDL uptake of HUAECs was increased significantly in the co-culture system. All the reseach results indicated that there was an interaction between HUAECs and HUASMCs in the co-culture system. Conclusions In the present study, human umbilical artery EC-SMC co-culture model was constructed successfully, which could mimic the morphological characteristic and basic functions of human arterial wall.
3.Study on the correlation between hs-CRP and coronary heart disease as well as acute myocardial infarction
International Journal of Laboratory Medicine 2009;30(1):36-38
Objective To explore the relationship between high-sensitivity C-reactive protein (hs-CRP) and the occurrence of certain diseases such as coronary heart disease (CHD) and acute myocardial infarction (AMI).Methods Sixty-one CHD patients, 45 AMI patients (patients group, excluding high hs-CRP level diseases caused by other etiological factors) and 35 healthy controls (control group) were enrolled in the study. The serum level of hs-CRP and myocardial zymogram was measured in the Hitachi 7060 automatic analyzer by latex-enhance immunoturbidimetry for all subjects, and cTnI of AMI group was detected with immunofluorescence method at the same time. The difference of hs-CRP level was compared and analyzed between control group and patients group; control group before and after treatment. Moreover, the relationship between hs-CRP and myocardial zymogram as well as cTnI was analyzed. Results The level of hs-CRP was significantly higher in patients group than that in control group (P<0.01). There was statistical difference of hs-CRP level before and after treatment in control group (P<0.01). hs-CRP was positively correlative with myocardial zymogram and cTnI.Conclusion The serum level of hs-CRP is significantly elevated in CHD and AMI patients, indicating the measurement of hs-CRP contributes to auxiliary diagnosis, therapeutic effect observation, and prognosis evaluation.
4.Effect of losartan on transforming growth factor β1 in alcoholic nephropathy in rats
Journal of Chinese Physician 2009;11(1):21-24
Objective To investigate the effect of losartan,an angiotensin 1I type 1 receptor antagonist(ATlRA),on transforming growth factor-β1 expression in alcoholic nephropathy in rats. Methods Male adult Wistar rats were random divided into three subgroups and treated daily for 12 weeks as follows, rats were treated with normal saline[20ml/(kg·d)gavage twicely]in vehicle group, rats were treatedwith alcohol[400g/L,8g/(kg·d)gavage twieely]in alcoholic nephropathy, and rats were concomitantly treated with alcohol and Iosartan [200mg/L,ling/(kg·d),gavage]in alcohol+losartan.HE staining,Sirius red staining, electronic microscope, and immunohistochemistry were used to examine histologic changes, collagen fibers deposition, and the expression of TGF-BI.Results Compared with vehicle treated rats, alcoholic nephropathy rats showed tubular vacuolation, granular degeneration, swelling, tubulointerstitial inflammatory cell infil tration, and rarely mesangial proliferation and matrix deposition.Sirius red staining displayed a significant increase in collagen fibers in alias of glomemli, tubulointerstitium and vessels.Concomitant administration of losartan reversed all of above parameters(P<0.05).The upregulated expression of TGF-β1 proteins was significantly suppressed by losartan treatment(P<0.05).Conclusions These findings suggest that alcoholic nephropathy is characterized by the tubular granular degeneration, vacuolation, swelling, and tubulointemtitial inflammato-ry cell infiltration, and this is associated with upregulation of TGF-β1.Losartan treatment offers protection effect by downregulating TGF-β1 expression.

6.The role of reactive oxygen species in the hyperdynamic circulation of cirrhotic portal hypertension
Chinese Journal of Hepatobiliary Surgery 2013;(4):316-320
Oxidative stress is involved in cirrhotic portal hypertension and a close relationship exists between oxidative stress and hyperdynamic circulation. This article elaborates on the function of various reactive oxygen species (e.g.,superoxide anion and hydrogen peroxide) and corresponding anti-oxidants (e.g.,NAD(P) H oxidase inhibitor, the scavengers of superoxide anion, hydrogen peroxide, and vitamin C or E) on the occurrence and development of hyperdynamic circulation in cirrhotic portal hypertension. We observed that the level of reactive oxygen species in cirrhotic portal hypertension was elevated over normal. The potential clinical value is that inhibition or removal of these reactive oxygen species can relieve hyperdynamic circulation and reduce complications for cirrhotic portal hypertension.

8.Research progress of pancreatic protein tumor markers
International Journal of Surgery 2012;39(8):546-549
Pancreatic cancer is a commonly malignant gastrointestinal tumor with an obviously increasing incidence all over the world. Those patients without specific symptoms at early stage had mostly lost the opportunity of surgical therapy when pancreatic cancer was detected at advanced stage, and their prognosis is poor. Therefore,it is rather crucial to improve the early diagnosis of pancreatic cancer.Tumor marker is a considerable tool for early tumor detection and screening. It will help to improve the diagnostic rate of early pancreatic cancer and promote the prognosis of pancreatic cancer if we could find out tumor markers and screen one or a group of pancreatic cancer tumor markers.

Results:Microtensile bonding strength(MPa) between Bis-Core and root canal inner wall and that between root canal inner wall and Chemil Superior glass-ionomer was 20.65?1.72 and 12.89?1.63 respectively(P
10.Micromorphology analysis of bonding interface between material and dentin after root canal rehabilitation
Journal of Practical Stomatology 1995;0(04):-
Objective:To observe the microstructure of interface between material and dentin after root canal rehabilitation. Methods:Three human maxillary incisors extracted within one week were used. After crowns being removed and received root canal treatment, they were used for the study. The root was split into two parts along the long axis, then the bonding interface was made with high speed diamond bur. Bis-Core and Chemil Superior glass-ionomer were stacked on root canal inner wall respectively and the teeth were split along with the long axis with Isomet saw, in order to expose bonding interface. The microstructure of bonding interface of the samples were analyzed by optical microscope and atomic force microscope(AFM). Results: AFM observation of the adhesive interface specimens showed that Chemil Superior glass-ionomer and teeth knitted together completely and no filling invaded into dentin tube. While the bonding interface between Bis-Core and dentin, dentin primer infiltrated into dentin tube and matrix around through the smear layer and colophony protruded. Conclusion:The observations reveal the extent of resin penetration into the dentinal tubules provides a very strong bonding. This is a fundamental cause of resin better bond to tooth.
