OBJECTIVETo study the effect of Morinda officinalis (MO) extract on cytoxan (CTX) -impaired spermatogenesis of adult male SD rats.

METHODSWe randomly divided 56 adult male SD rats into seven groups of equal number: blank control, CTX model, CTX + NS, CTX + 10 g/kg MO, CTX + 20 g/kg MO, CTX + 30 g/kg MO, and CTX + 40 g/kg MO. We made the models of impaired spermatogenesis in the SD rats by intraperitoneal injection of CTX and treated the animal models by intragastric administration of MO at the concentrations of 10, 20, 30, and 40 g per kg per d, respectively. After two weeks of medication, we observed the changes in the body weight, testicular and epididymal indexes, and microstructure of the testis tissue, measured the mean seminiferous tubule diameter (MSTD) , and obtained testicular biopsy scores (TBS) in different groups, followed by comparative analyses.

RESULTSAfter treatment, the CTX + NS group showed no remarkable differences in the body weight ([234.83 ± 28.77] g) and epididymal index (2.71 ± 0.34) from those of the four CTX + MO groups, but exhibited a significantly lower testicular index ([12.15 ± 1.04] g) than those in the CTX + 20 g/kg MO ([13.71 ± 0.97] g), CTX + 30 g/kg MO, ([13.30 ± 0.29] g), and CTX + 40 g/kg MO group ([13.48 ± 0.51] g) (P < 0.05). Light microscopy revealed obvious pathological changes of the testis tissue in the CTX + NS group and significantly ameliorated structures of the seminiferous tubules in the CTX + MO 10, 20, 30, and 40 g/kg groups, with the MSTD of (204.78 ± 11.03), (216.55 ± 10.93), (218.03 ± 11.23), and (218.59 ± 14.06) μm, respectively, and the TBS of 9.03 ± 0.39, 9.69 ± 0.26, 9.83 ± 0.18, and 9.89 ± 0.11, respectively, all significantly higher than (189.74 ± 8.55) μm and 5.95 ± 1.21 in the CTX + NS group (P < 0.05). The efficacy of MO extract was increased in a concentration-dependent manner.

CONCLUSIONMorinda officinalis extract can repair cytoxan-induced damage to rat spermatogenesis, with may achieve the best effect at the concentrations of 30 and 40 g per kg per d.

Objective To study the effect of vitamin K3 (VitK3) on the apoptosis of human hepatocellular carcinoma cell line of HEPG2 and its relationship with Caspase-3 and Bcl-2 gene. Method With the treatment of VitK3, time and dosage effect was detected by MTT colorimetry. Appoptosis was observed by morphology, flow cytometry and DNA agarose gelelectrophoresis.Caspase-3 and Bcl-2 mRNA expression was assessed by RT-PCR. ResultVitK3 significantly inhibits HEPG2 cell proliferation and induce apoptosis.Caspase-3 mRNA expression was upregulated. ConclusionVitK3 induces HEPG2 cell apoptosis by way of Caspase-3 gene expression.

Objective To examine the difference in the onset of and recovery from the neuro-muscular (N-M) blockade induced by rocuronium measured by acceleromyography of corrugator supercilli (CS) muscle and adductor pollicis (AP) muscle and the implication for clinical N-M monitoring. Methods Forty ASA Ⅰ - Ⅲ patients aged 32-65yr undergoing elective cholecystectomy or gastrectomy under combined general-epidural anesthesia (CGEA) were randomized to receive rocuronium 0.6 mg?kg-1 (group A, n = 20) or 0.9 mg?kg-1 (group B, n = 20) to facilitate tracheal intubation. Patients with N-M transmission disease, serious heart and lung disease or hepato-renal dysfunction were excluded. Epidural was performed at T8-9 or T9-10 . The level of epidural block was below T4. N-M blockade was measured by acceleromyography (TOF-WATCH SX Organon Teknika) of AP muscle at the thumb and CS muscle at the superciliary arch simultaneously. General anesthesia was induced with midazolam 0.05 mg ? kg-1 , propofol 2 mg ? kg-1 , fentanyl 3 ?g ? kg-1 and rocuronium 0.6 or 0.9 mg ? kg-1 . Tracheal intubation was performed at 80 % depression of T1 . The intubation condition (rated as excellent, good, medium, poor). The onset time (time from injection of rocuronium to maximal depression of T1) and the time of return to 25% and 75% of the control height of T1 were recorded and recovery index (RI) was calculated. Results The onset time of N-M block of CS muscle was (111?36)s at 0.6 mg?kg-1 and (74 ? 26)s at 0.9 mg?kg-1 and that of AP muscle was (106 ? 34) s (0.6 mg?kg-1 ) and (84?28) s (0.9 mg?kg-1 ) . The onset time produced by 0.9 mg?kg-1 was significantly shorter than that produced by 0.6 mg?kg-1 . At 0.6 mg?kg-1 80% depression of T1 of AP muscle was equal to 66% (52%-81 % ) depression of T1 of CS muscle and only 2 out of 10 patients showed good intubation condition. At 0.9 mg?kg-1 80% depression of T1 of AP muscle was epual to 89% (76%-93%) depression of T1 of CS muscle and all patients showed good intubation condition. The recovery of CS muscle from relaxation was faster than that of AP muscle. Conclusion There are differences in the onset of and recovery from the N-M blockade induced by rocuronium between CS and AP muscle. Monitoring N-M blockade of CS muscle is better for intubation.

Objective To evaluate the optimal dosage of misoprostol administered in the rectum for dilation of the cervix. Methods Two hundred and forty women at 40-60 day gestational age without vaginal delivery history were randomly divided into three groups,with 80 cases in each group. Patients received 200,400 or 600μg of misoprostol rectally one hour before electrical vacuum abortions in group A,B and C,respectively. Cervical dilation,blood loss,and drug side effects in the three groups were compared. Venous blood samples were taken before vein anesthesia,and misoprostol acid concentration in the serum was tested by liquid chromatography-mass spectrometry. Results The analgesic rate was 100. 00%in all three dose groups, and cervical dilation rate was 23. 75%,46. 25%and 70. 00%in groups A,B and C,respectively. The severity of drug side effects such as vaginal bleeding and abdominal pain is dose-dependent. Blood concentration of misoprostal acid was(117±65),(206± 98),and(303±149)pg·mL-1 ,in groups A,B and C,respectively. Conclusion The recommended dose of misoprostol is 400 μg administered in rectum. Rectal administration of misoprostol is cheap,safe,and convenient,and therefore could be widely applied.

Objective:To explore influence of low high density lipoprotein cholesterol (HDL‐C) level on kidney func‐tion in patients with coronary heart disease (CHD) after percutaneous coronary intervention (PCI) .Methods :A to‐tal of 300 CHD patients ,who were treated and received PCI in our hospital from Feb 2012 to Jun 2014 ,were select‐ed .According to HDL‐C level ,they were divided into normal HDL‐C level group (n=179 ,HDL‐C≥1. 04 mmol/L) and low HDL‐C level group (n= 121 ,HDL‐C< 1.04 mmol/L) .Baseline data were compared between two groups ,and multi‐factor Logistic regression analysis was used to analyze risk factors for kidney injury after PCI .Re‐sults:Compared with normal HDL‐C level group ,there were significant rise in percentages of overweight (30.73%vs .39.67% ) ,smoking history (29.05% vs .41.32% ) ,myocardial infarction history (35.20% vs .46.28% ) ,left ventricular ejection fraction (LVEF) <50% (18.99% vs .24.79% ) ,anemia (16.20% vs .27.27% ) and kidney in‐jury (12. 85% vs .22. 31% ) in low HDL‐C level group , P<0. 05 or <0. 01. Multi‐factor Logistic regression analysis indicated that age ,myocardial infarction history ,anemia ,LVEF<50% ,diabetes mellitus ,dosage of contrast agent and low HDL‐C level were risk factors for kidney injury in CHD patients after PCI (β=0.499~1.171 ,P<0.05 or<0.01) .Conclusion:Low HDL‐C level is a risk factor for renal function injury in CHD patients after PCI .

Objective Using the frozen human-pooled serum samples,with values assigned by the enzyme reference procedure in laboratory network to explore the way of quantity value transfer from enzyme reference measurement system to routine ones.Methods The study is about quantity value transfer.The frozen human-pooled serum sample was assigned by the enzyme reference laboratory network including six domestic reference procedure laboratories,namely Beijing Chaoyang Hospital affiliate of Capital University of Medical Sciences,Beijing Aerospace general Hospital,Beijing Shijitan Hospital affiliate of Capital University of Medical Sciences,Peking Union Medical College Hospital,Beijing Leadman Biochemistry Co.,Ltd.and Sichuan Maker Biotechnology Co.,Ltd.,which used the reference measurement procedures of creatine kinase (CK) and lactate dehydrogenase (LDH) from the International Federation of Clinical Chemistry and Laboratory Medicine (IFCC).First,the assigned serum was used to calibrate or verify trueness of routine measurement systems from the 49 laboratories of tertiary hospitals in Beijing in Feb,2013 by Beijing Centerfor Clinical Laboratories.Second,the assigned serum and two levels of patient samples were tested in duplicate by laboratory 's measurement systems after calibration with manufacturer's calibrator and with assigned serum.During this procedure,all laboratories data were collected.Harmonization of results was observed after calibration with the assigned serum according to the statistics.Results After calibration,the interlaboratory coefficients of variation (CVs) of two patient samples were decreased from 4.67％ and 5.96％ to 1.86％ and 1.47％ for CK and from 5.72％ and 5.04％ to 3.67％ and 3.62％ for LDH.For peer group of A and B instrument,after calibration,both CVs of the assigned serum and two patient samples were less than 2％ for CK,and less than 4％ for LDH ; For C instrument group,which was not calibrated with the assigned serum,the biases were-4.75％ for CK and-13.1％ for LDH,while the biggest CVs were 3.64％ for CK and 7.57％ for LDH.For peer group of reagents,after calibration,the CVs were decreased from 1.60％-4.44％ to 0.23％-1.62％ for CK and from 0.93％-6.07％ to 1.06％-2.72％ for LDH.Conclusions It is one of the feasible ways for quantity value transfer by using the frozen human-pooled serum samples,which were assigned by the enzyme reference laboratory network for calibration or verification.It can effectively solve the problem of trueness and harmonization of enzyme testing results.In addition,for those measurement systems which are not applicable for this calibration.

Objective In order to investigate the interactions between the activities of daily living of children with post poliomyelitis and depression of their parents.Method 40 cases whose children were with post poliomyelitis were tested by SDSCR.Children's activity of daily living were tested with Barthel's index.Result 65％ of parents whose children with post polimyelitis were depression.Their symptoms of depression were mainly moderate level.There are marked difference comparing with the domestic normal analyzing model.The activities of daily living of the post poliomyelitis's children was worse,the older their parent's age was the higher the incidence of depression occurred was.Conclusion We should pay attention to their parents emotion changes when we carry out the community rehabilitation of post poliomyelitis's children,and strive them for positive coperating our rehabilitation training.
